Relationship between the nucleolar cycle and chromatoid body formation in the spermatogenesis of $Phrynops$ $geoffroanus$ (Reptilia Testudines)

  • The nucleolus is a distinct nuclear territory involved in the compartmentalization of nuclear functions. There is some evidence of a relationship between nuclear fragmentation during spermatogenesis and chromatoid body (CB) formation. The CB is a typical cytoplasmic organelle of haploid germ cells, and is involved in RNA and protein accumulation for later germ-cell differentiation. The goal of this study was to qualitatively and quantitatively describe the nucleolar cycle during the spermatogenesis of $Phrynops$ $geoffroanus$ (Reptilia Testudines), and compare this nucleolar fragmentation with CB formation in this species through the use of cytochemical and ultrastructural analysis. Qualitative analysis showed a fragmentation of the nuclear material after pachytene of the first meiotic division in the primary spermatocytes. Quantitative analysis of the nucleolar cycle revealed a significant difference in the number of nucleoli and in the size of the nucleolus between spermatogonia and early spermatids. Using ultrastructural analysis, we recorded the beginning of the CB formation process in the cytoplasm of primary spermatocytes at the same time as when nuclear fragmentation occurs. In the cytoplasm of primary spermatocytes, the CB was observed in association with mitochondrial aggregates and the Golgi complex. In the cytoplasm of early spermatids, the CB was observed in association with lipid droplets. In conclusion, our data show that the nucleolus plays a role in the CB formation process. During spermatogenesis of $P.$ $geoffroanus$, the CB is involved in some important biological processes, including acrosome formation and mitochondrial migration to the spermatozoon tail and middle piece region.

  • Planarians show negative phototaxis and have extensive regenerative ability, including the ability to regenerate the brain. Recently the process of regeneration of the planarian brain has been divided into three steps based on the expression of neural markers. In this study, we have analyzed the process of recovery of the light response during head regeneration. Although morphological observations indicated that regeneration of the eyes and optic nerves appeared to be completed by the fourth day, the recovery of the evasion behavior against light was not recovered within 4 days after amputation. Functional recovery of the evasion behavior could be detected starting 5 days after amputation and then gradually recovered. We previously identified genes which are specifically expressed in the brain after the recovery of morphological structures. This characteristic suggested that these genes may be involved in functional recovery of the brain. To investigate the function of these genes, we performed gene knockout analysis using the RNA interference method. The results clearly indicated that these genes are involved in the functional recovery of the visual system.

Risk Ranking for Tunnelling Construction Projects in Malaysia

  • Tunnelling has become a preferred method of construction for road and highway projects in countries with a lot of hilly slope geological conditions such as found in Malaysia. However the construction works of a tunnelling project are usually complicated and costly, which consequently impose great risks to the parties involved. This paper identifies the key significant risks and sub-risks for tunnelling construction projects in Malaysia through a case study. Interview has been used as the solitary means to determine the significant risks from contractor's eleven key project personnel who were directly involved in the tunnelling construction such as consultant, construction manager and tunnel engineers. The importance of the risks identified is then prioritised and ranked via the Analytic Hierarchy Process (AHP)'s pairwise comparison approach to determine their criticality towards a successful delivery of project. As a result, three key risks have been identified as significant for the tunnelling case study project, namely health and safety, cost overrun in construction and time overrun in construction. Two sub-risks each of the latter categories, which are cost underestimation and unforeseen events (cost overrun in construction) as well as plant and machinery failure and delay in material delivery (time overrun in construction), have occupied the top five overall risk ranking.

  • Biosynthesis of nanoparticles has acquired particular attention due to its economic feasibility, low toxicity and simplicity of the process. Extracellular synthesis of nanoparticles by bacteria and fungi has been stated to be brought about by enzymes and other reducing agents that may be secreted in the culture medium. The present study was carried out to determine the underlying mechanisms of extracellular silver nanoparticle synthesis by Pseudomonas hibiscicola isolated from the effluent of an electroplating industry in Mumbai. Synthesized nanoparticles were characterized by spectroscopy and electron microscopic techniques. Protein profiling studies were done using Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (1D-SDS PAGE) and subjected to identification by Mass Spectrometry. Characterization studies revealed synthesis of 50 nm nanoparticles of well-defined morphology. Total protein content and SDS PAGE analysis revealed a reduction of total protein content in test (nanoparticles solution) samples when compared to controls (broth supernatant). 45.45% of the proteins involved in the process of nanoparticle synthesis were identified to be oxidoreductases and are thought to be involved in either reduction of metal ions or capping of synthesized nanoparticles.

  • The development of routine techniques for the isolation and in vitro maintenance of conducting airway epithelial cells in a differentiated state provides an ideal model to study the factors involved in the regulation of the expression of mucocilicary differentiation. Several key factors and conditions have been identified. These factors and conditions include the use of biphasic culture technique to achieve mucociliary differentiation and the use of such stimulators, the thickness of collagen gel substratum, the calcium level, and vitamin A, and such inhibitors, the growth factors EGF and insulin, and steroid hormones, for mucous cell differentiation. Using the defined culture medium, the life cycle of the mucous cell population in vitro was investigated. It was demonstrated that the majority of the mucous cell population in primary cultures is not involved in DNA replication. However, the mucous cell type is capable of self-renewal in culture and this reproduction is vitamin A dependent. furthermore, differentiation from non-mucous cell type to mucous cell type can be demonstrated by adding back a positive regulator such as vitamin A to the “starved” culture. Cell kinetics data suggest that vitamin A-dependent mucous cell differentiation in culture is a DNA replication-independent process and the process is inhibited by TGF-${\beta}$1.

  • 초고층건물의 구조설계 정보에는 설계작업 프로세스 모델, 설계정보 전산모델, 기본설계 프로그램 및 설계 도서생성 프로그램 개발 등 여러 가지 특성화된 부분으로 구성되어 있다. 기존의 구조 설계정보는 통합적으로 관리되는 기능이 없기 때문에 건설 정보 교환에 대한 공유가 제대로 이루어지지 않고 있다. 이러한 설계정보의 데이터 수정 및 공유를 합리적으로 처리하기 위해서 통합설계에 대한 필요성이 증가하고 있다. 또한, 초고층 구조설계에서는 건설정보의 표준화가 이루어진 후 합리적인 정보교환을 위해서도 통합설계의 필요성이 대두되고 있다. 본 연구에서는 초고층건물의 구조설계를 위하여 STEP을 이용한 초고층 구조설계의 개념을 소개하고 초고층건물의 구조설계방법에 대한 질량 엔티티 및 기둥축소량 엔티티, 사용성평가 엔티티를 제안하고자 한다.

공리설계와 반응표면모델에 의한 형상제어 압연기의 추력모델 개발 (Approximate Model of Thrust of Pair-Cross Mill using Axiomatic Design and Response Surface Model)

  • Rolling process to fabricate a strip with even thickness is significant to enhance the quality of the strip. The thickness of a strip can be effectively controlled by pair-cross mills. However, pair-cross mill generates thrust in the axial direction of roller and causes skewness, deflection, twist and even accidental roll chock failure. Therefore, accurate estimation of the thrust of the pair-cross mill during rolling process is necessary to monitor the failure of roll and the quality of products. An empirical equation given by Mitsubishi Heavy Industry (MHI) is hitherto employed, where the thrust is expressed in terms of rolling force, reduction ratio and crossed angle. However it turns out that the MHI empirical equation provides somehow inaccurate and unsuitable thrust in practical rolling processes. Moreover, we learn that three parameters involved in MHI equation are coupled each other. In this paper, axiomatic design principle is employed to select appropriate parameters involved in approximate equation in order to make parameters uncoupled. A quadratic equation using response surface method with new parameters is suggested. The accuracy of the approximate model is examined by comparing with real experimental data.

  • In nursing, a practice discipline, it has been argued whether the mastery of clinical competence depends on types of learning styles, learning experience, and the use of specialized skills. All these problems are not limited to nursing education. Later educators identified the importance of reflective learning which is a vital element in any form of learning and that teachers and students need to consider how they can incorporate some forms of reflection in the courses. The purpose of this study is to review educational articles for understanding better what reflection in learning is, to identify the theme which is of important relevance to professional practice, from the book, Educating the reflective practitioner, and to discuss the theme within nursing education. Reflection in learning was defined by Dewey(1933) as the process which is involved the perception of relationships and connections between the parts of an experience. This experience is passed on when two people becoming involved with each other in a conversation. schon(1987) emphasized that learning conversation, which is a part of the interaction of student and teacher, is an important factor of the process of reflection-om-actopm. In clinical nursing education, good relationships between teacher and student, faculty's role, interpersonal skills are critical in learning conversation. Then Practing nurses who accept the need to choose nursing actionss on the basis of reflection, who accepet the necessity for understanding and being able to communicate the reasons for action are a powerfful force for the development of nursing into an increasingly more effective profession for the benefit of patients.

Global Approaches to Identify Genes Involved during Infection Structure Formation in Rice Blast Fungus, Magnaporthe grisea

  • The ascomycete Magnaporthe grisea is a pathogen of rice blast and is known to form specialized infection structures called appressoria for successful infection into host cells. To understand the molecular mechanism underlying infection process, appressorium-related genes were identified through global approaches including EST sequencing, differential hybridization, and sup-pression subtractive hybridization. EST database was generated on >2,000 cDNA clones randomly selected from appressorium stage cDNA library. Large number of ESTs showed homology to known proteins possibly involved in infection-related cellular development (attachment, germination, appressorium formation, and colonization) of rice blast fungus. The 1051 ESTs showing significant homology to known genes were assigned to 11 functional categories. Differential hybridization and suppression subtractive hybridization were applied to identify genes showing an appressorium stage specific expression pattern. A number of genes were selected as up-regulated during appressorium formation compared with the vegetative growing stage. Clones from various cDNA libraries constructed in different developmental stages were arrayed on slide glass for further expression profiling study. functional characterization of genes identified from these global approaches may lead to a better understand-ing of the infection process of this devastating plant disease, and the development of novel ways to protect host plant.

Analysis of Gene Expression in Mouse Spinal Cord-derived Neural Precursor Cells During Neuronal Differentiation

  • The differentiation of neural precursor cells (NPCs) into neurons and astrocytes is a process that is tightly controlled by complicated and ill-defined gene networks. To extend our knowledge to gene networks, we performed a temporal analysis of gene expression during the differentiation (2, 4, and 8 days) of spinal cord-derived NPCs using oligonucleotide microarray technology. Out of 32,996 genes analyzed, 1878 exhibited significant changes in expression level (fold change>2, p<0.05) at least once throughout the differentiation process. These 1878 genes were classified into 12 groups by k-means clustering, based on their expression patterns. K-means clustering analysis revealed that the genes involved in astrogenesis were categorized into the clusters containing constantly upregulated genes, whereas the genes involved in neurogenesis were grouped to the cluster showing a sudden decrease in gene expression on Day 8. Functional analysis of the differentially expressed genes indicated the enrichment of genes for Pax6- NeuroD signaling.TGFb-SMAD and BMP-SMAD.which suggest the implication of these genes in the differentiation of NPCs and, in particular, key roles for Nova1 and TGFBR1 in the neurogenesis/astrogenesis of mouse spinal cord.
