In this study, we investigated whether S. baicalensis rhizome ethanol extract (SBRE) has antioxidant capacities against oxidative stress induced cellular damage in the HaCaT keratinocytes. Our results revealed that treatment with SBRE prior to hydrogen peroxide ($H_2O_2$) exposure significantly increased the HaCaT cell viability. SBRE also effectively attenuated $H_2O_2$ induced comet tail formation, and inhibited the $H_2O_2$ induced phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V positive cells. In addition, SBRE exhibited scavenging activity against intracellular ROS generation and restored the mitochondria membrane potential loss induced by $H_2O_2$. Moreover, $H_2O_2$ enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with SBRE. Furthermore, SBRE increased the levels of HO-1 associated with the induction of Nrf2. Therefore, we believed that SBRE may potentially serve as an agent for the treatment and prevention of neurodegenerative diseases caused by oxidative stress.
Objectives The purpose of this study was to investigate the effects of Silbi-san on the antioxidant and fat accumulation inhibition and to analyze the anti-obesity effect by analyzing the changes in serum lipid composition in obese mice. Methods We compared contents of phytochemicals like total polyphenols and total flavonoid and antioxidant activities such as 2,2-dipheny-1-picrylhydrazyl and 2.2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activity. After Silbi-san in 3T3-L1 cells in vitro and mouse adipose tissue ex vivo, we quantified intracellular triglyceride accumulation and lipolysis. Moreover, the anti-obesity activity though inhibiting pancreatic lipase were analyzed. In 3T3-L1 cells, morphological changes showed that control cells had many lipid while cells treated with Silbi-san had less lipid accumulation. 30% EtOH Silbi-san treatment also suppressed the fat absorption by inhibiting the activity of pancreatic lipase and led to high lipolysis through promoting glycerol release. The experimental group was divided into four groups: Normal group fed normal feed, Control group fed 60% high fat diet (HFD) and distilled water, drug group fed 60% high fat diet and 200 mg/kg of Silbi-san water extract, drug group fed 60% HFD and 200 mg/kg of Silbi-san 30% ethanol extract. Results Serum total cholesterol content and serum low density lipoprotein-cholesterol content were significantly decreased in the Silbi-san extract group compared to the control group, serum high density lipoprotein-cholesterol content was significantly increased in Silbi-san extract group. Conclusions In this study, the antioxidant and fat accumulation inhibitory effects of Silbi-san were confirmed.
The present study investigated an ethanol extract of Chaenomeles sinensis fruit (CSF) for possible neuroprotective effects on neurotoxicity induced by amyloid ${\beta}$ protein ($A{\beta}$) (25-35) in cultured rat cortical neurons and also for antidementia activity in mice. Exposure of cultured cortical neurons to $10{\mu}M\;A{\beta}$ (25-35) for 36 h induced neuronal apoptotic death. At $0.1-10{\mu}g/m{\ell}$, CSF inhibited neuronal death, elevation of intracellular calcium concentration ($[Ca^{2+}]_i$), and generation of reactive oxygen species (ROS) induced by $A{\beta}$ (25-35) in primary cultures of rat cortical neurons. Memory loss induced by intracerebroventricular injection of mice with 15 nmol $A{\beta}$ (25-35) was inhibited by chronic treatment with CSF (10, 25 and 50 mg/kg, p.o. for 7 days) as measured by a passive avoidance test. CSF (50 mg/kg) inhibited the increase of cholinesterase activity in $A{\beta}$ (25-35)-injected mice brain. From these results, we suggest that the antidementia effect of CSF is due to its neuroprotective effect against $A{\beta}$ (25-35)-induced neurotoxicity and that CSF may have a therapeutic role for preventing the progression of Alzheimer's disease.
부추뿌리 추출물(Allium tuberosum root extract)에 대한 다양한 생리활성을 조사하여 천연물소재 가능성을 시험하였다. 부추뿌리 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타내었으며 세포독성이 거의 없는 농도에서 부추뿌리 추출물 처리 시, 산화방지 활성(ID50, 6.2 ㎍/mL), 타이로시네이즈 활성저해(ID50, 115.4 ㎍/mL) 및 멜라닌 합성 저해(ID50, 31.5 ㎍/mL)를 나타내었다. 부추뿌리 추출물 농도 의존적으로 타이로시네이즈 발현이 억제가 관찰되었으며, 부추뿌리 추출물이 타이로시네이즈 단백질 발현을 억제시킴으로써 타이로시네이즈 활성저해 및 멜라닌 합성을 저하시키는 효과가 나타난 것으로 판단된다. 또한, 부추뿌리 추출물이 피부탄력 유지에 관련된 엘라스테이즈 활성저해(ID50, 145.1 ㎍/mL)를 보여주었으며 이소케르세틴(isoquercitrin)이 유효성분으로 확인되었다. 이와 같은 결과로 미루어 볼 때 부추뿌리 추출물은 기능성 소재에 활용하기 위한 효과적인 천연물 재료가 될 수 있다고 판단된다.
본 연구에서는 chaparral 70% 에탄올 추출물을 0.01-100 ㎍/mL의 농도로 HepG2 간세포에 처리하여 세포 사멸, 염증성 사이토카인 분비, 세포 내 지방 축적을 측정하는 in vitro 실험을 통해 chaparral의 간독성 기전을 조사하였다. Chaparral 추출물 처리에 의해 1-100 ㎍/mL의 농도에서 세포 사멸이 관찰되었으며, 염증성 사이토카인인 IL-8과 M-CSF의 분비 및 지방 축적은 10배 더 낮은 농도부터인 0.1-100 ㎍/mL에서 유의적으로 증가하였다(p<0.05). 본 연구결과에서 염증성 사이토카인 분비 및 지방 축적을 통한 간세포 염증은 chaparral 추출물에 의해 유발되는 간독성의 한 형태로 나타났다. 또한 간염 형태의 간독성은 세포 사멸이라는 심각한 독성을 야기시키는 농도보다 낮은 농도에서 발현되어 저농도의 chaparral 섭취에 의해 간염과 같은 간독성이 초래될 수 있음을 시사한다.
Objectives : In this study, we investigated the neuroprotective effects of ethanol extract of Lonicera japonica flower buds (EELJ) on glutamate-induced neurotoxicity in mouse hippocampus-derived neuronal HT22 cells. Methods : After analyzing the cytoprotective effect of EELJ on glutamate in HT22 cells, the inhibitory effect of apoptosis was studied using flow cytometry. In order to analyze the antioxidant efficacy of EELJ, the levels of reactive oxygen species (ROS) and glutathione (GSH) were investigated, and the effects on the activities of superoxide dismutase (SOD) and catalase (CAT) were also analyzed. Furthermore, the effect of EELJ on the expression of apoptosis regulators such as Bax and Bcl-2 in glutamate-treated HT22 cells was investigated. Results : According the current results, pretreatment with EELJ significantly reduced glutamate-induced loss of cell viability and release of lactate dehydrogenase. EELJ also markedly attenuated glutamate-induced generation of intracellular ROS, which was associated with increased levels of GSH, and activity of SOD and CAT in glutamate-stimulated HT22 cells. In addition, EELJ was strikingly inhibited glutamate-induced apoptosis in HT22 cells. Furthermore, the expression of pro-apoptotic Bax was increased and the expression of anti-apoptotic Bcl-2 was decreased in glutamate-treated HT22 cells, while in the presence of EELJ, their expressions were maintained at the control levels. Conclusions : These findings indicate that EELJ protects glutamate-induced cytotoxicity in HT22 hippocampal neurons through antioxidant activity. Therefore, although identification of biologically active substances of EELJ and re-evaluation through animal experiments is necessary, this natural substance is a promising candidate for further research in preventing and treating oxidative stress-mediated neurodegenerative diseases.
This study screened the antimicrobial and anti-inflammatory activities of three extracts [1% acetic acid (HAc), distilled water (D.W.), and ethanol] from the common stalked barnacle Pollicipes mitella. Among the extracts, the 1% HAc extract showed the strongest antibacterial activity against several bacteria, but exhibited no activity against Candida albicans. To improve the degree of separation of the 1% HAc extract, solid-phase extraction was performed using a C18 cartridge with three solvents (D.W., 60A, and 100A). The 1% HAc 60A eluate showed the strongest antibacterial activity and enzyme, salt, and temperature stability, with no hemolytic activity. In addition, strong DNA-binding ability but no bacterial membrane permeability was observed. These results indicate that the P. mitella 1% HAc 60A eluate may contain antibacterial organic compounds that target intracellular components but not bacterial membranes. In addition, the 1% HAc 60A eluate exhibited potent inhibitory activity to reduce the production of inflammatory mediators (nitric oxide and prostaglandin E2) and pro-inflammatory cytokines (tumor necrosis factor-α, interleukin (IL)-6, and IL-1β) with no cytotoxicity. Therefore, the P. mitella 1% HAc 60A eluate has anti-inflammatory activity. Collectively, our results suggest that the P. mitella 1% HAc 60A eluate can be used as a bioactive source with antibacterial and anti-inflammatory activities.
DPPH를 이용한 인동덩굴 추출물의 자유라디칼 소거활성($FSC_{50}$) 측정 결과, 인동덩굴 에틸아세테이트 분획물 ($77.25{\mu}g/ml$)이 50% 에탄올 추출물($152.00{\mu}g/ml$)보다 약 2배 높은 활성을 나타내었다. Luminol 발광법을 이용한 인동덩굴 추출물의 활성산소 소거활성(총 항산화능, $OSC_{50}$) 측정 결과, 인동덩굴 에틸아세테이트 분획물($0.33{\mu}g/ml$) > 인동덩굴 50% 에탄올 추출물($1.12{\mu}g/ml$) > L-ascorbic acid ($4.95{\mu}g/ml$) 순서로 높게 나타났다. $^1O_2$으로 유도된 적혈구 파괴에 대한 세포 보호 효과(${\tau}_{50}$) 측정 결과, 인동덩굴 추출물의 농도에 의존적으로 세포 보호 효과가 나타났으나, 고농도에서는 세포 보호 효과가 감소하는 경향을 나타내었다. 인동덩굴 50% 에탄올 추출물은 $10{\mu}g/ml$의 농도(46.0 min)에서, 인동덩굴 에틸아세테이트 분획물은 $1{\mu}g/ml$의 농도(52.3 min)에서 가장 높은 세포 보호 효과를 나타내었다. 인동덩굴 추출물 및 분획물의 세포 독성 평가 결과, 본 실험에서는 $0.4-12.5{\mu}g/ml$의 농도 범위를 설정하여 세포 보호 효과를 평가하였다. 과산화수소로 유도된 각질 형성 세포인 HaCaT 세포 보호 효과는 인동덩굴 50% 에탄올 추출물 및 에틸아세테이트 분획물 $12.5{\mu}g/ml$의 농도에서 각각 70.0, 78.6%의 세포 생존율을 나타냈으며, 대조군(65%)과 비교하여 유의한 세포 보호 효과를 가지고 있음을 확인하였다. UVB를 조사한 HaCaT 세포의 세포 내 ROS 소거 활성에서 인동덩굴 50% 에탄올 추출물 및 에틸아세테이트 분획물 모두 농도 의존적으로 ROS를 소거하였으며, $12.5{\mu}g/ml$ 농도에서 각각 45.2, 63.3%의 ROS 소거율을 나타내었다. 이를 통해 인동덩굴 에틸아세테이트 분획물이 50% 에탄올 추출물보다 높은 세포 내 ROS 억제능이 있음을 확인하였다. TLC와 HPLC 크로마토그램을 이용한 인동덩굴의 에틸아세테이트 분획물의 성분 분석을 통해 caffeic acid, luteolin, apigenin, coumaric acid를 확인하였다. Caffeic acid와 luteolin의 뛰어난 환원력과 킬레이팅 작용이 인동덩굴 추출물의 활성산소 소거활성과 세포 보호 효과에 크게 기인했을 것으로 사료된다. 인동덩굴 50% 에탄올 추출물과 에틸아세테이트 분획물의 총 페놀성 화합물 및 총 플라보노이드 함량 측정 결과를 통해, 인동덩굴 에틸아세테이트 분획물이 50% 에탄올 추출물보다 많은 페놀성 화합물, 플라보노이드를 함유하고 있기 때문에 더욱 우수한 항산화 활성을 나타내었음을 확인하였다. 본 연구에서는 자외선에 의해 생성되는 다양한 ROS에 대한 인동덩굴 추출물 및 분획물의 항산화 활성을 평가하였고 이상의 결과를 통해 인동덩굴 추출물이 식물유래 천연 항산화제로서 가치가 있음을 시사하였다. 뿐만 아니라, 인동덩굴의 항산화 기능성 화장품 소재로서의 개발 가능성을 확인하였다.
본 연구에서는 범부채 뿌리 50% 에탄올 추출물 및 에틸아세테이트 분획물을 제조하고 이들의 항산화 및 항균 활성, 세포 보호 효능을 평가하였다. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) 자유라디칼 소거 활성($FSC_{50}$) 측정 결과, 50% 에탄올 추출물은 $621.5{\mu}g/mL$, 에틸아세테이트 분획물은 $253.0{\mu}g/mL$이었다. Luminol 발광법을 이용한 총 항산화능($OSC_{50}$)은 추출물과 분획물에서 각각 13.6 및 $3.0{\mu}g/mL$이었다. 항균 활성 측정에서 Staphylococcus aureus 및 Candida albicans에 대한 에틸아세테이트 분획물의 최소저해농도(minimum inhibitory concentration, MIC)는 각각 156 및 $1,250{\mu}g/mL$으로 나타났으며, 화장품에 사용하는 기존 방부제인 methyl paraben보다 유사하거나 더 높은 활성을 보여주었다. $^1O_2$로 유도된 세포 손상에 대한 보호 효과(${\tau}_{50}$)에서 50% 에탄올 추출물은 $4{\sim}64{\mu}g/mL$ 농도 범위에서 농도 의존적으로 세포 보호 활성을 나타냈다. $16{\mu}g/mL$ 농도에서 50% 에탄올 추출물, 에틸아세테이트 분획물 및 (+)-${\alpha}$-tocopherol의 ${\tau}_{50}$은 각각 36.4, 45.0 및 45.8 min이었으며, 에틸아세테이트 분획물은 $^1O_2$로 유도된 세포 손상에서 (+)-${\alpha}$-tocopherol과 유사한 세포 보호 활성을 나타냈다. UVB로 유도된 HaCaT 세포 손상에서 에틸아세테이트 분획물은 $8{\mu}g/mL$에서 세포 내 활성산소종(reactive oxygen species, ROS)을 최대 45.9%까지 감소시켰다. 과산화수소로 유도된 HaCaT 세포 손상에서도 에틸아세테이트 분획물은 $0.5{\sim}8.0{\mu}g/mL$에서 세포 생존율을 유의적으로 증가시켰다. 범부채 뿌리 에틸아세테이트 분획물의 성분 분석 결과, irisflorentin, irigenin, tectorigenin, resveratrol, iridin 및 tectoridin 등의 플라보노이드 및 폴리페놀 성분이 확인되었다. 결론적으로 범부채 뿌리 추출물 및 분획물은 화장품의 천연 항산화 및 항균 소재로서 적용 가능성이 있음을 시사한다.
본 연구에서는 초임계 공정에 의해 복령피를 추출하였고, 에탄올 복령피 추출물과 비교하여 항염, 미백, 항산화효과를 측정하였다. 또한 선정된 추출물의 제형과 약물전달 펩티드를 활용하여 효율적인 경피투과 방법을 제시하였다. 복령피의 항암, 항염성분으로 알려진 Pachymic acid를 지표성분으로 HPLC 분석 결과 에탄올 복령피 추출물 보다 초임계 복령피 추출물이 2배 이상 높게 추출되는 것으로 나타났다. 복령피의 항산화효과를 확인하기 위하여 DPPH 소거능과 ABTS 소거능은 복령피 에탄올 추출물이 초임계 복령피 추출물보다 더 낮은 농도에서 $IC_{50}$을 나타내었다. 하지만 RAW 264.7 세포 내 NO(Nitric oxide) 생성량을 측정한 결과에서는, 에탄올 복령피 추출물보다 초임계 복령피 추출물이 동일 농도에서 더 낮은 NO 생성량을 나타냈다. 또한 B16 melanoma 세포에 복령피 추출물을 $20{\mu}g/mL$ 농도를 72시간 처리 후 세포 내외 멜라닌 합성을 확인한 결과에서는, 에탄올 복령피 추출물과 초임계 복령피 추출물 모두 효과가 있었으며 초임계 추출물이 더 낮은 멜라닌 함량을 보였다. NO 생성량 실험에서 활용된 RAW 264.7 세포에서는 $800{\mu}g/mL$농도에서도 독성이 나타나지 않았다. 하지만 B16 melanoma 세포에서는 $50{\mu}g/mL$에서도 에탄올 추출물과 초임계 추출물 모두 60 % 미만의 생존율을 나타냈다. 초임계 복령피 추출물을 대상으로 리포좀 제형과 약물전달 펩티드를 활용하여 Franz diffusion cell 실험 결과에서는, 리포좀 제형과 약물 전달 펩티드가 초임계 복령피 추출물을 경피 투과를 증진시켜 주는 것으로 나타났다. 초임계 복령피 추출물이 다양한 효능의 화장품 소재로서 개발될 가능성이 있다고 판단된다.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에

제 5 장 계약 해지 및 이용 제한

일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
