Distributions of proliferative epithelial cells in gastrointestinal tracts by anti-bromodeoxyuridine monoclonal antibody (Anti-bormodeoxyuridine monoclonal antibody를 이용한 랫드 위(胃)와 장(腸)의 분열 상피세포의 분포에 대하여)

  • The purpose of this stady was to investigate division cells by in vivo bromodeoxyuridine(Brdur) immunohistochemistry for labeling the proliferative epithelial cells in the gastrointestinal tracts of rats. Rats were administrated intraperitonially by twice consecutive injections of 24 hr interval with Brdur(0.05mg/g BW/time) and then were sacrificied at 1 hour after last injection. The specimens were taken from the stomach, small intestine(ileum), and large intestine(colon). The well-oriented crypts and villi in the preparations were examined, The crypt columns and villi were devided into 10 segments from crypt base to surface of the lumen or to villis top. Labeling index(LI) was measured by counting the number of Brdur-positive cells against the total number of crypt column cells in the stomach and large intestine and also against the total numbers of crypt column and it's villi epiterial cells in the small intestine. 1. In the stomach, the LI in each part from segment 1 to segment 10 of the crypt column were 4.2%, 5.0%. 6.6%, 9.0%, 11.3%, 15.3%, 9.3%, 15.6%, 11.3%, 0%, respectively and it's mean LI were 8.7%. The Brdur-positive epithelial cells were predominantly located in the middle regions and middle-upper regions of the crypt columns. 2. In the small intestine, the LI in each part from segment 1 to segment 10 of were 62.4%, 50.9%, 27.8%, 22.5%, 18.6%, 12.1%, 7.5%, 4.3%, 2.5%, 1.4%, respectively and it's mean LI were 21.0%. The Brdur-positive epithelial cells were predominantly located in the lower regions of the crypt columns and tended to be less in the higher regions of the villi than that in the crypt column. 3. In the large intestine, the LI in each part from segment 1 to segment 10 of the crypt column were 19.4%, 29.9%, 34.1%, 41.6%, 41.2%, 32.4%, 25.4%, 15.4%, 10.8%, 1.2%, respectively and it's mean LI were 25.1%, The Brdur-positive epithelial cells were predominantly in the middle and middle-lower regions of the crypt columns. 4. The organs with higher LI were ordered as the large intestine(25.1%), small intestine(21.0%) and stomach(8.7%).

  • The turnover time of the mucosal epithelium in the small intestine(jejunum and ilium) and large interstine(cecum), and the cells in the lamina propria of the small intestine was investigated with the radioautography in mice at various times after single injection of $^3H$-thymidine. Twenty suckling mice were sacrified at each of the following time intervals after injection ; 2 hrs, 1, 3, 5. 7, 14 and 17 days. 1. The labeled index of the epithelial cells in the crypt and the villus of the small intestine averaged 98.7% and 1.3% at 2 hrs, 982% and 1.8% at 1 day, 18.7% and 81.3% at 3 days, 6.3% and 93.7% at 5 days, respectively. The labeled index of the epithelial cells of the crypt-base, the upper-crypt and the mucosal surface in the large intestine averaged 71.8%, 28.2% and 0% at 2 hrs, 45%. 54.2% and 0% at 1 day, 17.2%, 54.5% and 28.2% at 3 days, 10.2%, 32.4% and 57.4% at 5 days, respectively. This result suggested that the turnover time of all the epithelial cells migrating from crypts to villi in the direction of the villus tips was calculated to be less than 5 days, and also the longest turnover time was calculated to be no longer than 7 day. 2. The labeled index of the total cells in the lamina propria of the small intestine averaged 6.2-7% at 2 hrs to 5 days, 4.7% at 7 days 2.6% at 17 days and this index is tend to be decreased moderately at 7 days and severely at 17 days. So this result suggested that the turnover time of the cells with the shorter cycle duration in the lamina propria of the small intestine were less than 5 days and that of the cells with the longer cycle duration more than 17 days.

  • The purpose of this study was to investigate the effects of Angelica gigas on jejunal survival, endogenous spleen colony formation and jejunal crypt cells of mice irradiated with Gamma-ray irradiation. The subject of this study includes 42 mice which were divided into each 7 groups. Angelica gigas experiment groups were Angelica gigas + Gamma-ray(10Gy), Angelica gigas + Gamma-ray(3Gy), Angelica gigas. Gamma-ray(1 Gy), Gamma-ray control (10Gy), Gamma-ray control(3Gy), Gamma-ray control(1Gy), Normal groups. In the present study to evaluate the effect of Angelica gigas on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells of mice Gamma-ray with each dose of Gamma-ray irradiation. The results of this study were as follows: In low-dose(1Gy) Gamma-ray radiation were treatment of Angelica gigas showed significantly increased(p<0.05) on the cell death apoptosis in crypt, intestine crypts survival of intestine after gamma-ray irradiation. High-dose(10Gy) Gamma-ray, treatment of Angelica gigas showed significantly increased(p<0.05) on the leukocyte. The above results suggest that Angelica gigas were immunostimulating effectively reduced Gamma-ray irradiation.

  • This experimental study was carried out to investigate the immunostimulating effect of Acanthopanax, as Oriental rhizomata herbs, on jejunal survival, endogenous spleen colony formation, apoptosis in jejunal crypt cells and lipid peroxidation in the liver of mice following Gamma-ray irradiation. The subject of this study includes 72 mice which were divided into each 7 groups. Acanthopanax experiment groups were Acanthopanax. Gamma-ray(lOGy), Acanthopanax. Gamma-ray(3Gy), Acanthopnax. Gamma-ray(1Gy), Gamma-ray control(1OGy), Gamma-ray control(3Gy), Gamma-ray control(1Gy), Normal groups. The results of this study were as follows : Treatment with Acanthopanax showed significantly increased(p<0.05) on the cell death apoptosis in crypt, intestine crypts survival of intestine in mice following low-dose(1Gy) Gamma-ray radiation. And that significantly increased(p<0.05) on jejunal crypt survival and reduced(p<0.05) on lipid peroxidation in mice following high-dose(1OGy) Gamma-ray radiation. The above results suggest that Acathopanax were immunostimulating effectively reduced Gamma-ray irradiation.

Effects of Supplementing Different Levels of a Commercial Enzyme Complex on Performance, Nutrient Availability, Enzyme Activity and Gut Morphology of Broilers

  • A trial was conducted to study the influence of different levels of a commercial enzyme complex on performance, nutrient availability, blood parameters, digestive tract measurements, amylase and trypsin activity of the digestive tract and gut morphology in broilers fed the typical diets in north China. There were four treatments: the control diet and the other three enzyme complex supplemented diets which were 180 mg/kg, 360 mg/kg and 720 mg/kg enzyme complex supplemented to the control diet, respectively. The birds fed the diets supplemented with 180 mg/kg and 360 mg/kg enzyme complex had better performance and nutrient availability, the activities of amylase and trypsin in the digestive tract in the two treatments were improved, the villus height and surface area of villus in the small intestine increased and the crypt depth and epithelial thickness of small intestine decreased. Relative weights of pancreas and relative weights and lengths of small intestine decreased. However, the addition of 720 mg/kg enzyme complex had no effects on these parameters and increased crypt depth and epithelial thickness of the small intestine. The data suggested that suitable supplementation of enzyme complex was beneficial for the birds, while excess enzyme complex inhibited secretion of endogenous enzyme and destroyed the structure of the small intestine.

Immunohistochemical Studv on the Gastrin, Somatostatin and Serotonin Cells in the Gastric and Small Intestinal Mucosa of Rat during Development (발생기 흰쥐 위와 소장점막의 gastrin, somatostatin 및 serotonin세포에 대한 면역조직화학적연구)

  • The developmental changes of three enteroendocrine cells, i.e. gastrln, somatostatin and serotonin, of gastric and small intestinal mucosa in pre- and postnatal rat were examined by peroxidase-antiperoxidase (PAP) method. In the course of development, gastrin cells were obsenred in the pyloric gland region and the whole part of small intestine, while somstostatin and serotonin cells in the whole gastric gland region and small intestine. More entroendocrine cells were detected in the pyloric gland region and duodenum than in the other portion. In the stomach, gastrin, somatostatin and serotonin ceils were first obsenred in the pyloric Bland region on 17, 19 and 19 days of gestation respectively. The small intestinal gastrin and serotonin cells were first appeared in the duodenum and iriunum on 17 and 15 days of gestation respectively, and somatostBtin cells in duodenum on 17 days of gestation. The number of cells examined from the stomach were increased from fetal to weanling period and showed a decrease during adult period: the notable increase was shown at the end of suckling period or at early weanling period. The cells of the small intestine increased from fetal to suckling period, especially, these cells markedly increased at the end of fetal period or at early suckling period, and decreased from weanling period. The shape of these cells was oval or fusiform during fetal period. In the stomach, most of gastrin cells turned out to be oval and open-type from suckling period, while the remaining two tripes of cells were oval and open- or closed-type. In the small intestine, 311%Ves of cells examined were changed to fusiform and open-type from the end of fetal period. Three types of cell were distributed over the stratified epithelium on 15 and 17 days of gestation. In the stomach, these cells were distributed lower gastric pit and gland from the following fetal period, and were detected mainly on the upper part of gland from suckling period, and then obsenred on the whole part of gland. In the small intestine, most of cells distributed over only between epithelium of villi on 19 days of gestation, increased in number on the crypt from following fetal period, and also observed abundantly in the crypt at adult period.

Histopathological and immunohistochemical studies on the intestinal lesions related to the infected age in spontaneous canine parvovirus enteritis (개 파보바이러스장염의 감염일령에 따른 병변의 병리조직학적 및 면역조직화학적 관찰)

  • Ninety seven cases of histopathologically diagnosed spontaneous canine parvovirus enteritis(CPE) were studied gross pathologically, histopathologically, immunohistochemically, to investigate histopathological types of small intestinal lesions, and antigen distributions in each pattern related to the infected age. And also, reliability of histopathological method in diagnosis of CPE was inspected with immunohistochemistry. The results were as follows : 1. Age-related occurring ratio in histopathologically diagnosed CPE was 53.6% in 4-8 weeks, 26.8% in 9-15 weeks, 8.25 in 16-19 weeks and 11.3% in 20-45 weeks of the clog age. 2. In histopathologic classification based on patterns of villi/crypts lesions of small intestine(jejunum), the ratio of A type (initial phase of necrosis of crypt epithelia, desquamated epithelial cells in the dilated lumen of the crypt) was 20.6%; the ratio of B type(middle phase of atrophy and fission of the villi, collapse of the mucosa, loss of normal crypt structure) was 62.9%, and C type(regenerative phase of the crypt architecture) was 16.5%. 3. The ratio of A, B, C type in 4-8 weeks old, respectively, was 23.5%, 61.5%, 15.4%; in 9-15 weeks old was 19.2%, 65.4%, 15.3% in 16-19 weeks old was 25.0%, 75.0%, 0.0%; and in 20-45 weeks old was 9.0%, 54.5%, 36.4%. 4. The antigen distribution in the nuclei of the crypt epithelial cells was higher than of the cytoplasm and numerous desquamated epithelial cells in dialated crypts in A type; The antigen cytoplasm and numerous desquamated epithelial cells in dialated crypts in A type; The antigen distribution in the nuclei of the collapsed crypt epithelial cells was not higher than that of the cytoplasm, crypts were lined by and filled with released viral antigens from the destructed epithelial cells in B type; and its distribution was also higher than in the epithelial cells adjacent to the tips of the villi, but it was not reacted in the regenerative crypt epithelial cells in C type. 5. Immunohistochemically detected antigen ratio in the small intestine of histopathologically diagnosed CPE was 94.6%, and this result indicates that histopathological diagnosis is very reliable method in diagnosis of CPE.

Coexpression of PCNA and p21 for DNA repair in small intestinal crypt cells of mouse with 60Co γ-rays irradiation (방사선을 조사한 마우스의 소장 음와세포에서 DNA 수복을 위한 PCNA와 p21의 발현 양상)

  • The irradiation of radioactive ${\gamma}-ray$ induces apoptosis of radiosensitive organs for homeostasis. In this study, we investigated the repair mechanisms for homeostasis in the small intestine after cell damage by $^{60}Co\;{\gamma}-ray$ irradiation. The apoptosis was most frequently observed in the crypt cells of the small intestine after four and six hours by radioactive ${\gamma}-ray$ irradiation, and the frequency of apoptosis was proportional to the amount of irradiation. Also, the number of apoptotic cells was coincident with expression pattern of p53. Interestingly, PCNA (proliferating cell nuclear antigen) which is engaged in DNA replication and repair was expressed in apoptotic cells of small intestinal crypts. Also, it was observed that cell-cycle regulator p21 which is known to induce cell-cycle arrest is co-expressed in the same apoptotic cells of irradiated small intestinal crypt cells. These findings suggest that the co-expression of PCNA and p21 proteins, which may lead to resistance to DNA damage through cell-cycle arrest is closely associated with repair of damaged gastrointestinal cells after ${\gamma}-ray$ irradiation.

  • Intestinal histopathological changes due to infection with Echinostcma hortense (Trematoda) were studied in rats after experimental infection with the metacercariae. The metacercariae were obtained from the tadpoles of Rana nigrcmaculata, a second intermediate host infected in the laboratory. Total 18 albino rats(Sprague-Dawley) were given 200 matacercariae each and sacrificed on the day 1, 3, 7, 11, 22 or 44 post-infection(PI) Segments of- the small intestine at 1, 3, 5, 8 and 30 cm posterior to the pylorus(PTP) were rejected and studied histopathologically. 1. The flukes were seen to have intruded into the intervillous space in the upper small intestine at early stages(1∼3 days PI), however, they were located mainly in the intestinal lumen at later stages(7∼44 days PI) . The flukes were sucking and destroying the epithelial layers of villi with their oral and ventral suckers. 2. Histopathological changes of the intestine were recognizable in as early as 1∼3 days after infection, and the changes became severer as the infection progressed. 3. The intestinal mucosa was histopathologically characterized by villous atrophy and crypt hyperplasia throughout the infection period. Major villous changes were blunting, fusion, severe destruction and loss of epithelial layers of villi. Villous/crypt(V/C) height ratio was remarkably reduced from 3 : 1 in controls to 1 : 1 in severely infected animals. In the stroma of villi, inaamma- tory cell infiltrations, vascular congestion, edema, and/or fibrosis were recognized. The goblet cells were increased in number after 11 days PI. It was revealed in the present study that the pathological changes in the intestine of rats infected with E. hortense were chieay confined to the mucosal layer of the upper small intestine, however, the changes were very severe accompanying remarkable destruction of villi and loss of mucosal integrity, and persistent until 44 days PI.

Feline panleukopenia virus infection in imported cats

  • The cases of feline panleukopenia virus (FPLV) infection were diagnosed in three imported cats. All cats died within one week after mild emaciation, depression and anorexia. One cat showed yellowish watery diarrhea. At necropsy, all cats had segmental hemorrhage on the serosa and mucosa of the small intestine. Histopathologically, severe diffuse necro-hemorrhagic enteritis was observed in small intestine especially in jejunum and ileum. The crypts of Lieberkuhn were dilated and contained necrotic epithelia. Severely damaged epithelia of crypts were transformed into bizarre shapes. Multifocal lympholysis and lymphoid depletion were found in Peyer's patches and other lymphoid tissues. Direct fluorescent antibody (FA) test revealed the characteristic FPLV antigen in the cytoplasms of crypt epithelial cells. Based on the clinical signs, characteristic pathologic findings and FA test, these cases were diagnosed as FPLV infection. In our best knowledge, this study is the first case report for FPLV infection in imported cats in Korea.
