한국인 신생아의 분변 미생물 균총 (Fecal Microflora of Korean Neonates)

  • Probiotic bacteria have been administered to neonates to serve as maturational stimuli for the developing gut and intestinal immune system, establish and develop the intestinal microbiota, and mediate host-microbe interactions; further, these bacteria have shown beneficial effects In the treatment and reduction of the risk of infectious diseases, necrotizing enterocolitis, and atopic disease. An LAB isolation project to identify effective lactic acid bacteria for Korean people is in progress. The average total counts of lactic acid bacteria, lactobacilli, bifidobacteria, and coliforms in the fecal samples from 2 provinces were estimated as 8.31, 5.98, 8.13, and 3.01 CFU/g. Additional samples from other provinces will be analyzed to examine the changes in the lactic bacterial counts according to the area, sex of the neonate, mode of delivery, and type of feeding. A database containing the 16S rDNA sequences and the ribosomal protein profile of all the lactic acid bacteria isolated from fecal samples will be constructed. For the effective use of probiotics, a number of clinical studies are needed to formulate guidelines for strain, subject, purpose, and dose.

The Role of the Immune System in the use of Probiotic Lactic Acid Bacteria in Preventing and Treating Allergic Diseases

  • The immune system is generally divided into the innate and the adopted immune systems, both protecting the body from pathogens. Recently, allergies, a disease associated with an imbalanced immune system, have increased rapidly in developed countries. Prevailing symptoms of allergic diseases are eczema, allergic rhinitis, asthma, inflammatory bowel disease, and food allergy. Probiotic bacteria, mainly consisting of lactic acid bacteria, are used in the prevention and treatment of allergic diseases. The function of them is to stimulate the intestinal immune cells and form a complex signal network to activate other immune cells. Beneficial health effects of probiotics are based on the hygiene hypothesis, which suggests that sanitary environment is important for health, but limited exposure to environmental factors increases allergic diseases. An immunoregulatory effect of probiotic bacteria is demonstrated by controlled trial, animal model, in vitro, in vivo and ex vivo designs. However, the immunoregulatory effect of probiotic bacteria is controversial because it depends on probiotic strains, a dose and a type of diseases. In this review, we discussed clinical evidences on immunoregulatory effects of probiotic bacteria.

Rosemary의 급여가 육계의 생산성, 맹장내 균총 및 깔짚 중 유해가스 발생량에 미치는 영향 (Effects of Dietary Rosemary Supplementation on Performance, Microflora Population in the cecum and Noxious Gas Emission from Litter in Broilers)

  • 본 연구는 육계 생산과정에서의 건조 Rosemary 잎 분말의 급여가 육계의 생산성, 맹장내 균총 및 깔짚 중 유해가스 발생량에 미치는 영향에 대해서 조사하였다. 총 216수의 7일령 육계에 대조구, Rosemary 분말 0.5% (5 g/kg diet, T1) 및 1.0% (10 g/kg diet, T2)로 구분하여 35일령까지 총 4주간의 사양 시험을 실시하였다. 사육기간중 증체중 및 사료효율은 Rosemary 급여에 의해서 유의하게 증가하였지만(P<0.05), 처리구간(T1 Vs. T2)에는 유의한 차이가 인정되지 않았다. 사료섭취량 및 도태율은 대조구와 처리구간에 차이는 인정되지 않았다. 21일령과 35일령 모두 맹장내 Lactobacillus의 수는 Rosemary 급여에 의해서 유의하게 증가하였고(P<0.05), E. Coli 및 Salmonella의 수는 유의하게 감소하였다(P<0.05). 혐기성 총 미생물 수는 Rosemary의 첨가에 의한 영향은 인정되지 않았다. 깔짚 내 Ammonia의 발생량은 Rosemary 첨가에 의해서 유의하게 감소하였다(P<0.05). 그러나 처리구간(T1 Vs. T2)에는 차이가 인정되지 않았다. 이상의 결과를 종합해보면, 육계사양에서 건조 Rosemary 잎의 급여는 사료중 영양소 이용율 증가, 소화관내 유익균의 증가 및 유해균의 감소, 및 배설물 중 $NH_3$의 발생량 감소로 인한 증체량 및 사료효율을 개선시킬 가능성이 생각된다.

사료내 벤조산 첨가가 이유돼지의 장내 미생물 균총 및 면역체계에 미치는 영향 (Effect of Dietary Benzoic Acid on Beneficial Microflora and Immune Response in the Intestine of Weaning Pigs)

  • 본 연구에서는 이유자돈 사료에 벤조산 첨가가 위장 내 소화물과 방광 내 뇨의 pH, 장내 유익 미생물 균총수 및 장 조직 내 염증성 싸이토카인 유전자 발현양에 미치는 영향을 조사하였다. 자돈기 5주 동안 이유자돈 사료 내 벤조산 0.3%과 0.5% 첨가는 자돈 위 내 pH는 감소시키지는 않았으나, 방광 내 뇨의 pH 수치는 유의적으로 감소하는 것으로 나타났다. Lactobacillus casei 수는 맹장부위에서 벤조산 0.5% 첨가구와 대조구에서 유의적으로 가장 높게 나타났다. Lactobacillus plantarum 수는 맹장부위에서 벤조산 0.5% 첨가구에서 가장 높게 나타났다. 또한, 직장부위에서 L.plantarum 수는 대조구를 제외하고 모든 처리구에서 유의적으로 높게 나타났다. Bacillus subtillis 수는 맹장부위에서 벤조산0.5% 첨가구와 대조구에서 유의적으로 가장 높게 나타났으며, 직장부위에서는 벤조산0.3%와 0.5% 첨가구에서 가장 높게 나타났다. IL-$1{\beta}$ mRNA 유전자 발현 수준은 소장 상단에서는 벤조산 0.5%와 항생제 처리구에서, 소장중부에서는 벤조산 0.3%와 0.5% 처리구에서, 말단부위에서는 벤조산 0.3% 처리구에서 대조구보다 유의적으로 낮게 나타났다. IL-6 mRNA 유전자 발현 수준은 소장중부에서는 벤조산 0.5% 처리구에서 대조구보다 유의적으로 낮게 나타났다. TNF-${\alpha}$ mRNA 유전자 발현 수준은 소장 중단에서 벤조산 0.5%와 항생제 처리구에서 대조구 보다 낮게 나타났다. 본 실험을 통하여, 자돈 사료 내 benzoic acid 0.5% 첨가는 뇨의 pH 감소와 장내 유익 미생물 균총 수 조절 및 소장 내 면역체계 반응에 긍정적인 영향을 미치므로 자돈 건강을 향상 할 수 있으리라 사료된다.

콩 함유 올리고당의 기능적 특성 (Functional Characteristics of Soybean Oligosaccharide)

  • To enjoy a healthy life, it is important to have a well-balanced diet. However, in today's society, there is an increase in the consumption of preprocessed foods and frequency of eating out. Also the western diet, which is becoming move popular worldwide, contains relatively high levels of protein and fat, and a low amount of fiber, Furthermore, the increased availability of favorite foods has created a condition were the individual diet is less variable. With these conditions, it is difficult to maintain a diet that is nutritionally balanced. With these unbalanced diets, which are difficult to change, there has been an increase in adult disease and health problems, such as colon and breast cancer, It is speculated that metabolites for carcinogens are produced from diet components and that intestinal bacteria contribute to the production of these metabolites. Therefore, it is necessary to evaluate the relationships between health, diet, and intestinal microflora. Soybean oligosaccharide is composed of water-soluble saccharides that have been extracted from soybean whey, a by-product from the production of soy protein. This is mainly a mixture of mono-, di-, tri-, and tetrasac-charides, with the principle components being the oligosaccharide raffinose and stachyose. When consumed by humans, the oligosaccharides cannot be digested in the human duodenal and small intestinal mucosa, and these are selectively utilized by beneficial bifidobacteria in intestines. The results of acute and subacute toxicity tests, soy-bean oligosaccharides were nonpoisonous. Soybean oligosaccharides promote the growth of indigenous bifido-bacteria in the colon which by their antagonistic effects, suppress the activity of putrefactive bacteria. Also, they reduce toxic metabolites, detrimental enzymes and plasma lipid, and increase in the frequency of bowel evacuation and fecal quantities. Consequently, soybean oligosaccharides as functional foods components have potential roles in the prevention and medical treatment of chronic adult diseases. The study of processing property and physiological function of soybean oligosacchavides and development of high oligosaccharide variety allow the creation of new and exciting foodstuffs that aye functional healthy.

의약용 프로바이오틱 비스판균의 미생물학적 동정 (Microbiological Identification of Medical Probiotic Bisspan Strain)

  • Beneficial bacteria, which have been used for medical purpose and for medicines for treating intestinal disorders, include strains of Bifidobacterium sp., Lactobacillus sp., Enterococcus sp., Clostridium butyricum, Lactobacillus sporogenes, Bacillus subtilis, Bacillus polyfermenticus and the like. Bacillus polyfermenticuss SCD with is commonly called as Bispan strain has been appropriately used for the treatment of long-term intestinal disorders, since the live strains in the form of active endospores can successfully reach the target intestine. In this study, the identification and characterization of Bispan strain was done using SEM observation, API 50CHB kits, isoprenoid quinone analysis, and fatty acid analysis. These results suggest that Bispan strain is very similar to Bacillus subtilis.

A Case Study on the Brand Development of Odor-reducing Feed Additives

  • In the past, antibiotics and antimicrobial substances have been used for the purpose of promoting the growth of livestock or treating livestock, but various problems such as the presence of livestock products or resistant bacteria have emerged. Recently, regulations on the use of antibiotics have been strengthened worldwide, and probiotics are attracting attention as an alternative. Probiotic microorganisms have already been used for human use, such as intestinal abnormal fermentation, diarrhea, and indigestion. In livestock, beneficial microorganisms are increasing in use for the purpose of improving productivity, such as promoting livestock development and preventing diarrhea. Therefore, it is advisable to understand livestock probiotics in deeper and think about effective uses. The role of probiotics in the livestock sector is made with microorganisms themselves, so it is a substance that promotes livestock growth and improves feed efficiency by settling in the intestines of livestock, suppressing the growth of other harmful microorganisms, helping digestion and absorption of ingested feed, and helping to synthesize other nutrients. There is a need for a probiotic that suppresses intestinal bacteria by supplying probiotics used as a means to minimize the effects of stress in livestock management, thereby suppressing disease outbreaks by maintaining beneficial microorganisms and suppressing pathogenic microorganisms. The purpose of this paper is to develop a brand of feed additive probiotics to improve health conditions due to increased feed intake, improve the efficiency of use of feed nutrients, inhibit the decomposition and production of toxic substances, increase immunity, reduce odor in livestock, and improve the environment. We investigated and analyzed feed additive probiotics already on the market, and developed the naming and logo of suitable feed additive probiotic brands in livestock. We hoped that the newly developed product will be used in the field and help solve problems in the livestock field.

근채류의 기호도와 장내세균의 유해효소 억제효과 (The Preference and Inhibitory Effect of Root Vegetables on β-Glucuronidase and Tryptophanase of Human Intestinal Bacteria)

  • 식이섬유를 함유한 근채류의 기호도를 조사하고 장내유산균 증식효과와 장내유해균이 생산하는 ${\beta}$-glucuronidase와 tryptophanase 저해효과를 측정한 결과는 다음과 같다. 1. 조사대상자가 가장 선호하는 근채류는 고구마(4.05)였으며 남자의 선호도는 감자(3.88), 고구마(3.68), 더덕(3.77), 무(3.59), 양파(3.56)의 순으로 나타났으며 여자는 고구마(4.17), 감자(4.03), 양파(3.76), 무(3.61), 더덕(3.55)의 순으로 나타났다. 2. 여자가 남자보다 '근채류가 건강에 좋다'고 인식하고 있었으며 건강에 좋다고 생각하는 근채류는 남자의 경우 마늘 26.4%, 당근 16.7%, 감자 15.3%, 더덕 12.5%의 순으로 나타났고 여자의 경우 마늘 26.7%, 더덕 17.3%, 감자 13.9%, 당근 10.4%의 순으로 나타났다. 3. 근채류 추출물 0.5%를 첨가한 배지에서 장내유익균인 B. breve K-110의 증식효과는 감자(139%), 더덕(145%), 마(164%), 토란(144%), 연근(147%)에서 높았다. 장내유익균(B. breve K-110)과 유해균(E. coli HGU-3와 Bacteroides JY-6)을 혼합배양시 근채류 추출물 0.5% 첨가한 배지에서 장내유해균이 생산하는 유해효소인 ${\beta}$-glucuronidase와 tryptophanase 활성저해 효과는 감자, 더덕, 마, 토란에서 높게 나타났다. 감자, 더덕, 마, 토란추출물을 0.5% 첨가한 배지에 사람의 장내세균총을 배양하였을 때 이들 유해효소의 활성은 50%이상의 억제효과가 나타났다. 4. Sephadex G-100 column chromatography에 의하여 분리한 감자, 마, 더덕, 토란추출물의 고분자 분획에서 장내유익균인 B. breve K-110의 증식효과와 ${\beta}$-glucuronidase와 tryptophanase 활성저해 효과가 높게 나타났다. 5. 분리한 고분자 분획의 ninhydrin 발색반응은 감자, 마, 더덕, 토란에서 음성을 나타내어 아미노산이 아닌 것을 확인하였고 요오드반응은 마, 더덕, 토란에서 음성을 나타내었고 감자에서는 양성을 나타내었다. 이상과 같은 결과에 의하면 근채류중 감자, 마, 더덕, 토란이 유산균을 효과적으로 증식시켜 장내 pH를 저하시킴에 의해 ${\beta}$-glucuronidase, tryptophanase와 같은 장내의 유해효소를 억제할 것이라고 기대되어진다. 그러므로 이들 근채류는 장내의 유해효소에 의해 발생하는 대장암등의 질병예방과 장내균총의 불균형에 의해 야기될 수 있는 설사와 변비같은 질병의 예방에도 효과를 나타낼 것으로 생각된다.

Present Status of Fermented Milk Products in Japan

  • Fermented milks have been recognized as healthy foods since ancient times, but those using intestinal bacteria such as Bifidobacterium and lactic acid bacteria (LAB) are even more valuable from the standpoint of view of maintaining health. They have also now come to be recognized as important in the field of preventive medicine. Although advances in the medical sciences in the last 50 years have significantly increased the human lift span, an unfortunate fact is that many of us are now living long enough to experience chronic disorders such as coronary heart disease, hypertension, osteoporosis, diabetes and cancer. In recent years there has been renewed interest in health promotion and disease prevention by incorporating probiotic bacteria into foods to counteract harmful bacteria in the intestinal tract. Therefore, there are now a wide variety of commercial products containing prospective probiotics that claim health-promoting effects, such reductions in large botvel carcinogens and mutagens, antitumor properties, cholesterol-lowering effects, increased lactose digestion, relief from con-stipation, stimulation of immunocomponent cells and enhancement of phagocytosis. Two well-known representative probiotic is LAB and Bifidobaclerium. Traditional probiotic dairy strains of LAB which have been designated as GRAS (Generally Recognized As Safe) bacteria have a long history of safe use and most strains are considered comestible microorganisms with no pathogenic potential. Accordingly, there is considerable interest in extending the range of foods containing probiotic organisms from dairy foods to infant formulas, baby foods, and pharmaceuticals. In addition, the ingestion of probiotics, prebiotics, and symbiotic as wll as combinations of pro- and prebiotics has recently aroused renewed interest as enhancing the beneficial relationship between the host and intestinal microflora in both healthy and diseased indivisuals. Non-communicable chronic diseases such as cancer, cerebral hemorrhage, is chemic heart disease, and diabetes mellitus has recently been recognized as adult diseases in Japan as well as other countries. and are considered to be inevitably associated with aging. These diseases occur as a result of individual life styles. The Japanes Government. Ministry of Health, Labor and Welfare has proposed substituting the term 'adult diseases' with 'lifestyle-related diseases'. It has emphasized the importance of prevention rather than treatment. since the well-known increase in the elderly population in Japan is predicted to result in a variety of socioeconomic problems. n this lecture on the Present status of fermented milk products in Japan, I will report a strategy for the development of fermented milk products in Japan from the standpoint of view of research in Japan on LAB and Bifidobacteria. They could play an important role in preserving human health by controlling intestinal microflora capable of producing toxic effects on the host.

Glycine max Merr enhances the viability and adhesion ability of Lactobacillus buchneri in gastrointestinal condition in vitro.

  • Probiotics are microorganisms that have beneficial effects on the health of the host. The health promoting effect by probiotics influences suppressing harmful bacteria, prevention of constipation, blood cholesterol reduction and regulation of blood pressure. Prebiotics are used to promote the growth or activity of microorganisms. Synbiotics, which are a mixture of probiotics and prebiotics, synergize in the intestines by complementing each other. Synbiotics not only improves the viability of the probiotics while passing through the gastrointestinal tract, maintain intestinal homeostasis, but also regulate balance of harmful and useful bacterial growth. Glycine max Merr (GMM) has been widely used in Asian countries to treat cancer, obesity, oxidative stress and imbalanced immune diseases. In addition, it has been reported that dietary fiber-rich grains promote bowel movements and prevent constipation. In this study, we investigated the viability of LactobacillIus buchneri (L.buchneri) strains, known as lactic acid bacteria under conditions of gastric fluid and intestinal fluid to determine the suitability of L.buchneri as probiotics. The adhesion ability of L.buchneri to caco-2 cells was also confirmed. The present studies showed that GMM extract promoted the growth and activity of L.buchneri strains as prebiotics. Also, this results suggested that the mixture of L.buchneri and GMM extract can helps maintain intestinal health and healthy body as synbiotics and health functional food material.
