  • A recent study showed that comparative sequence analysis of rpoB DNAs could reveal natural relationships in genus Mycobacterium [J Clin Microbial. 37 (6). 1999]. rpoB DNAs showed interspecies variation and intraspecies conservation. Based on these data, we developed polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) protocols which enable species differentiation in genus Mycobacterium. When this assay was applied to 24 clinical isolates identified as M. avium complex (MAC) by biochemical test, these were successfully differentiated into M. avium and M. intracellulare. These results were concordant with those obtained by 16s rDNA analysis. It is the first report that PCR-SSCP analysis of rpoB DNA could be used for species differentiation of MAC strains.

Effects of habitat substrates and companion plants on the growth of Menyanthes trifoliata (서식 기질과 동반종이 조름나물의 생장에 미치는 영향)

  • Bog bean (Menyanthes trifoliata L.) is an endangered species in Korea, inhabiting in montane wetlands. Its' habitats are decreasing due to artificial disturbances and climate change in Korea. Interspecies competitions determine the distribution of a species and it is important to examine the effects of companion species on the growth of a species. To reveal the effects of habitat substrate and companion species on the growth of bog bean, we compared the growth parameters of bog bean at three substrate conditions where plant communities were formed: water column, Sphagnum mat, and paddy soil. Bog bean had the highest relative coverage at Sphagnum mat condition and there are few species whose height were higher than it. In contrast, there were many species larger than bog bean at paddy soil condition. The maximum coverage of bog bean was the largest at water column condition, and the lowest at paddy soil condition. Our results indicate that the growth of bog bean was more restricted by interspecific competition at paddy soil condition than at Sphagnum mat condition. Those results suggest that bog bean avoid competitions by expansion of rhizome rather than elongation of its height. For conservation or creation of bog bean habitats, it is necessary to offer the environment where other species cannot survive well.

Understory Vegetation Structure by Altitude and Azimuth Slope and Indicator Species Analysis in Mt. Gyebang (계방산의 고도와 사면방위별 하층식생구조 및 지표종 분석)

  • This study was conducted to investigate composition of understory vegetation and indicator species by altitude and slope azimuth in Mt. Gyebang designated as Protected Area for Forest Genetic Resource Conservation and National Park. Tracheophytes were 350 taxa; 80 families, 203 genera, 303 species, 38 varieties, 5 forma and 4 sub-species in research area. The species of greatest importance value were Tripterygium regelii (9.143%), Acer pseudosieboldianum (7.594%), Symplocos chinensis for. pilosa (6.347%) in the shrub layer and were Sasa borealis (8.653%), Isodon excisus (2.936%) and Carex siderosticta (2.897%). In the herb layer as a result of NMS analysis, the distribution range of the major species were found to be affected by the altitude (shrub layer: $R^2$ > 0.3, herb layer: $R^2$ > 0.6). The result of plexus diagram analysis showed that Acer pseudosieboldianum was associated with Magnolia sieboldii, Acer barbinerve, Euonymus oxyphyllus etc. in the shrub layer; Meehania urticifolia was associated with Aconitum jaluense, Veratrum oxysepalum, Prunus padus etc. in the herb layer. The significant indicator species were analyzed for 60 species by the altitude and investigated for 30 species in accordance with the slope azimuth. As a consequence of MRPP, interspecies composition along the altitude group was heterogeneous and the species composition according to the azimuth slope was extremely different between the NE and SW.

  • Technetium labeled isonitrile analogues are widely used as myocardial perfusion imaging agents. We synthesized and characterized a new isonitrile compound, ethyl 3-isocyanobutyrate(EIB). Proton and $^{13}C$ NMR spectroscopy and thin layer chromatography with a $C_{18}$ coat was performed. EIB was easily labeled with $^{99m}TcO_4^-$- with sodium dithionite. The labeling efficiency measured by RP-HPLC was over 95%. The labeled product was stable with dilution in normal saline and with prolonged incubation at room temperature. There was no formation of secondary products or free $^{99m}TcO_4^-$. In vivo kinetics study of $^{99m}Tc$ (I) labeled EIB in rabbits showed adequate myocardial uptake, good contrast against lung background, and relatively rapid liver clearance. The heart to lung ratio was over 2.5 and the heart to liver ratio was approximately from 0.4 to 5 at 60 minutes post injection. Hepatic clearance of $^{99m}Tc-MIBI$ was faster ($t_{1/2}$=6 minutes) than that of $^{99m}Tc-MIBI$. In vivo kinetics observed in dog was similar to that in rabbit but there was faster gallbladder filling, and thus lower liver background. SPECT imaging of the canine myocardium showed favorable imaging characteristics. However, biodistribution in mice demonstrated a myocardial % injected dose/organ of less than 0.1%. This was thought to be due to interspecies difference in plasma esterase activity. In human plasma, $^{99m}Tc$ ( I ) labeled EIB was stable for at least 2 hours, without production of secondary products by HPLC. We conclude that ethyl 3-isocyanobutyrate may be a potential new myocardial perfusion imaging agent and deserves further investigation as to its usefulness for clinical use.

Natural hybridization of Iris species in Mt. Palgong-san, Korea (팔공산 금붓꽃 계열의 자연 잡종 현상)

  • Series Chinensis, Genus Iris, endemic to the far regions of East Asia, consists of four species and related varieties. This series is divided into two major groups (I. rossii and I. minutiaurea complex). In this study, the ITS region and matK gene sequences within nuclear ribosomal DNA and plastid DNA were analyzed in order to investigate the phylogenetic relationships among the I. minutiaurea complex (I. minutiaurea, I. odaesanensis, and I. koreana) and the taxonomic identities of a putative hybrid in Mt. Palgong. In the internal transcribed spacer (ITS1, 5.8S, and ITS2) region, a total of 106 cloned genomic sequences from three taxa were obtained to study the intragenomic polymorphisms of the ITS regions. Three taxa revealed high levels of intragenomic polymorphisms, indicative of incomplete nrDNA concerted evolution. This incomplete ITS concerted evolution in the series Chinensis may be linked to the recent species divergence and frequent interspecies hybridization of the series Chinensis. In the matK gene, three taxa were fairly separated by eleven variable sites. In eight individuals collected on Mt. Palgong, putative hybrids between I. odaesanensis and I. minutiaurea were clustered in the I. minutiaurea clade in the NJ (neighbor-joining) tree based on the matK gene. However, in the ITS tree, some of them were clustered in the I. odaesanensis clade and others were clustered in the I. minutiaurea clade. Therefore, the individuals on Mt. Palgong were formed by the hybridization between two taxa (I. odaesanensis and I. minutiaurea) and not through the lineage of I. koreana.

In vitro Callus and Somatic Embryo Induction of Six Hosta Species Native to Korea

  • Hosta is a genus of the family Asparagaceae and distributed in East Asia. There are six Hosta species (Hosta capitata (Koidz.) Nakai, H. clausa Nakai, H. jonesii M.G.Chung, H. minor (Baker) Nakai, H. venusta F.Maek., and H. yingeri S.B.Jones) native to Korea and among them, four species (H. minor, H. jonesii, H. venusta and H. yingeri) are endemic to the Korea peninsula. Hosta is generally propagated by seed, crown division or tissue culture. However, tissue culture is a more efficient method to mass proliferation, a new cultivar development and disease-free plantlet production in a limit time. Hence, we conducted this study to evaluate the influence of various plant growth regulators (PGRs) treatments on the induction of callus and somatic embryo of the six Hosta species. Leaf, petiole and root were used to select optimum tissue culture explants. Petiole explants thus only were used for callus induction and somatic embryogenesis with TDZ (0.1, 0.5 or 1.0mg/L) and NAA (0.1 or 0.5 mg/L) combinations. After 12 weeks of culture, the highest rate of somatic embryogenesis was achieved on modificated MS medium containing 1.0 mg/L TDZ and 0.1 mg/L NAA in H. capitata and H. minor (15.5%, respectively), 0.1 or 0.5 mg/L TDZ and 0.1 mg/L NAA in H. jonesii (22.2%), 1.0 mg/L TDZ and 0.5 mg/L NAA in H. yingeri (26.7%), and 0.1 mg/L TDZ and 0.5 mg/L NAA in H. venusta (53.3%). H. clausa showed very low effect on somatic embryogenesis by PGRs; 2.2%. There was interspecies difference to PGRs respond for callus and somatic embryo induction. Regenerated multiple shoots and plantlet of H. minor, H. jonesii, H. venusta and H. yingeri were obtained via somatic embryogenesis.

Age and Growth of Three Species of Genus Gymnogobius from Korea (한국산 날망둑속(屬) 3종(種) (망둑어과(科))의 연령과 성장)

  • Three species, Gymnogobius urotaenia, G. sp. 1 and G. sp. 2, which were collected at Hosanriver, Hosan-ri, Samcheock-si, Gangwon-do, Korea, were examined for study of the age and growth. There was no differences between sexes in growth (p>0.05) except in age 0 of G. urotaenia, However, there were differences among the three species (p<0.0001). namely, G. sp. 1 was longer by about 5~10 mm (SL) than the other two species in each age group, while G. sp. 2 was shortest in each age group: G. rotaenia, at age 0 was less than 45 mm, at age 1 to be 45~60 mm, at age 2 to be 60~80 mm, at age 3 to be over 80 mm, and its maximum size was 105.0 mm. G. sp. 1, at age 0 was less than 55 mm, at age 1 55~65 mm, at age 2 65~85 mm, at age 3 over 85 mm and its maximum size was 105.3 mm. G. sp. 2, 0 age was less than 40 mm, at age 1 40~55 mm, at age 2 55~75 mm, at age 3 over 75 mm and its maximum size was 85 mm. Changes of body parts with increasing of body length were examined and analyzed. Covariance analysis showed interspecies differences in the following characters: body depth, head length, caudal peduncle depth, upper jaw length and pelvic fin ray length. G. urotaenia had the highest K-value (the relative growth coefficient) in head length and pelvic fin ray length, and G. sp. 1 had the highest in body depth, caudal peduncle depth and upper jaw length. However, G. sp. 2 had the lowest K-value in these 5 characters.

Establishment of the Korean Tolerable Daily Intake of Bisphenol A Based on Risk Assessments by an Expert Committee

  • Recently, reproductive and neurobehavioral effects of bisphenol A (BPA) have been documented, and thus a review was requested for BPA management direction by the government. Therefore, this study was performed to establish a Korean tolerable daily intake (TDI) for BPA. An expert committee, consisting of specialists in fields such as toxicology, medicine, pharmacology, and statistics, was asked to evaluate BPA health based guidance values (HbGVs). Although many toxicological studies were reviewed to select a point of departure (POD) for TDI, rat and mouse reproductive studies by Tyl et al. (2002, 2006), which were performed according to GLP standards and OECD guidelines, were selected. This POD was the lowest value determined from the most sensitive toxicological test. The POD, a NOAEL of 5 mg/kg bw/day, was selected based on its systemic toxicity as critical effects. An uncertainty factor of 100 including interspecies and intraspecies differences was applied to calculate the TDI. According to the evaluation results, a TDI of BPA for Korean was suggested at 0.05 mg/kg bw/day. In addition, the BPA exposure level based on food consumption by the Korean population was estimated as 1.509 ${\mu}g/kg$ bw/day, and the HI was evaluated at 0.03 when the TDI of 0.05 mg/kg bw/day was applied. This HI value of 0.03 indicated that hazardous effects would not be expected from BPA oral exposures. Although highly uncertain, further studies on low dose neurobehavioral effects of BPA should be performed. In addition, it is recommended that the 'as low as reasonably achievable' (ALARA) principle be applied for BPA exposure from food packaging materials in newborn infants and children.

Antifungal susceptibility of Candida spp isolated from bovine mammary glands and teat cups of milking machines (Candida속 균의 항진균성약제에 대한 감수성)

  • In vitro antifungal susceptibility test was carried out on 53 strains of Candida spp isolated from milk of dairy cows with subclinical mastitis and teat cups of milking machines, Nystatin, clotrimazole, miconazole, econazole, 5-fluorocytosine, cycloheximide, haloprogin and griseofulvin were tested by the agar dilution method. The 84.8% to 98.2% of Candida strains were inhibited by clotrimazole, econazole and miconazole at $${\leq_-}25{\mu}g/ml$$, and clotrimazole was most active. Interspecies differences of antifungal susceptibility were recognized and these were as follows. C albicans was most sensitive to clotrimazole (GM-MIC, $5.49{\mu}g/ml$) followed by 5-fluorocytosine, econazole and miconazole. C pseudotropicalis and C guilliermondii were notably sensitive to haloprogin, clotrimazole, miconazole, cconazole, 5-fluorocytosine, and haloprogin (GM-MIC, $0.17{\sim}0.19{\mu}g/ml$) was most active. C krusei was most sensitive to cycloheximide (GM-MIC, $0.54{\mu}g/ml$) followed by clotrimazole, haloprogin, miconazole and econazole. C parapsilosis was somewhat sensitive to econazole, cycloheximide, clotrimazole, and econazole (GM-MIC, $7.26{\mu}g/ml$) was most active. C tropicalis showed very low sensitivity to all tested drugs (GM-MIC, $${\geq_-}20.32{\mu}g/ml$$).

Evaluation of DNA Double Strand Breaks in Human and Mouse Lymphocyte Following ${\gamma}-Irradiation$ (${\gamma}-Ray$ 조사에 따른 사람의 정상임파구와 마우스 정상임파구의 DNA Double Strand Break 발생율에 대한 비교분석)

  • The evaluation of radiation-induced DNA double strand breaks (DSB) was made following irradiation of human lymphocytes, murine lymphocytes and EL-4 leukemia cells over a wide dose range of $^{60}Co\;{gamma}-rays.$ In lipopolysaccharide (LPS) or phytohemagglutinin (PHA)-stimulated murine lymphocytes, the slopes of the stand scission factor (SSF) revealed that lymphocytes with LPS increased DNA DSB formation by a factor of 1.432 (p<0.005). Furthermore, strand break production was relatively inefficient in the T lymphocytes compared to the B lymuhocytes. And EL-4 leukemia cells were found to form significantly more DNA DSB to a greater extent than normal lymphocytes (p<0.005). The in vitro studies of the intrinsic radiosensitivity between human lymphocytes and murine lymphocytes showed similar phasic kinetics. However, murine lymphocytes were lower in DNA DSB formation and higher in the relative radiation dose of 10 percent DNA strand breaks at 3.5 hours following ${gamma}-irradiation$ than human lymphocytes. Though it is difficult to interpret these results, these differences may be result from environmental and genetic factors. From our data, if complementary explanations for this difference will be proposed, the differences in the dose-effect relationship for the induction of DSB between humans and mice must be related to interspecies variations in the physiological condition of the peripheral blood in vitro and not to differences in the intrinsic radiation sensitivity of the lymphocytes. These results can be estimated on the basis of dose-effect correlation enabling the interpretation of clinical response and the radiobiological parameters of cytometrical assessment.
