Peptide Nucleic Acid Probe-Based Analysis as a New Detection Method for Clarithromycin Resistance in Helicobacter pylori

  • Background/Aims: Helicobacter pylori eradication rates are decreasing because of increases in clarithromycin resistance. Thus, finding an easy and accurate method of detecting clarithromycin resistance is important. Methods: We evaluated 70 H. pylori isolates from Korean patients. Dual-labeled peptide nucleic acid (PNA) probes were designed to detect resistance associated with point mutations in 23S ribosomal ribonucleic acid gene domain V (A2142G, A2143G, and T2182C). Data were analyzed by probe-based fluorescence melting curve analysis based on probe-target dissociation temperatures and compared with Sanger sequencing. Results: Among 70 H. pylori isolates, 0, 16, and 58 isolates contained A2142G, A2143G, and T2182C mutations, respectively. PNA probe-based analysis exhibited 100.0% positive predictive values for A2142G and A2143G and a 98.3% positive predictive value for T2182C. PNA probe-based analysis results correlated with 98.6% of Sanger sequencing results (${\kappa}$-value=0.990; standard error, 0.010). Conclusions: H. pylori clarithromycin resistance can be easily and accurately assessed by dual-labeled PNA probe-based melting curve analysis if probes are used based on the appropriate resistance-related mutations. This method is fast, simple, accurate, and adaptable for clinical samples. It may help clinicians choose a precise eradication regimen.

혈중(血中) Thyroxine-결합(結合)-globulin(TBG)의 $T_4$ 결합능(結合能) 측정(測定)에 관한 고찰(考察) (Estimation of the $T_4$ Binding Capacity of Serum Thyroxine Binding Globulin)

  • The most commonly used methods for determining thyroxine binding globulin(TBG) concentration as the total thyroxine-binding capacity utilize electrophoretic seperation of serum. Although technically simple, the electrophoretic method is time consuming and is limited in the number of samples which can be run in a single assay. The author presented a single $T_4$ load ion exchange resin method as an approach to simplify the technique as with clinical practicability and results were analyzed. For construction of the standard curves, serum mixtures were diluted with barbital buffer.which effectively blocked $T_4$-binding to TBPA. For each serum dilution, a constant amount of $T_4-^{125}I$ and increments of unlabelled $T_4$ were added. After incubation in water bath, resin beads were dispensed to the samples which binded all $T_4$ not bound to TBG. The radioactivity in the supernatant was counted in the gamma scintillation counter. Each standard curve was plotted from the percent counts in the supernatant and total $T_4$ in each tube. Unknown samples were diluted to 1:40 and ran at a single $T_4$ loading concentration, and the TBG capacity of the samples was able to be read on the standard isobars. The following results were obtained. 1) Mean and standard deviation for TBG capacity in normal population was $28.6{\pm}5.09{\mu}g\;T_4/100ml$. 2) $24.9{\pm}3.87{\mu}g\;T_4/100ml$ in hyperthyroidism showed low TBG capacity comparing to normal population.(p<0.025) 3) $31.0{\pm}2.40{\mu}g\;T_4/100ml$ in hypothyroidism showed high TBG capacity tendency comparing to normal population. 4) Reversed correlationship existed between TBG capacity and $T_3$ resin uptake(r=-0.624), TBG capacity and serum $T_4$ value (r=-0.859), and TBG capacity and free thyroxine index(r=-0.623). The author assumes that this method of assay is considerably simpler in instrumentation and technique than any other assays traditionally being used, and seems to be more practical for routine clinical laboratory use.

대기 중 다환방향족탄화수소(PAHs)의 측정.분석 신뢰도 향상에 관한 고찰 (Study on Improvement in Reliability of Measurement and Analysis for Polycyclic Aromatic Hydrocarbons in the Atmosphere)

  • In this study, various techniques for measurement and analysis of PAHs in the ambient air were verified in order to select a more reliable method. Sampling and analysis of PAHs were done by the EPA TO-13a method. QA/QC of the measurement was conducted to minimize errors in sampling and analyzing processes. The linearity of calibration curve of the PAH standards was good ($R^2{\geq}0.99$). Audit accuracy was evaluated using 5 internal standards of PAHs ($Naphthalene-d_8,\;Acenaphthene-d_{10},\;Phenanthrene-d_{10},\;Chrysene-d_{12},\;Perylene-d_{12}$). Relative standard deviations of the internal standard of the PAHs were ranged from 6.22% for $acenaphthene-d_{10}$ to 8.11% for $chrysene-d_{12}$. To evaluate the surrogate recoveries, two field surrogate standards of PAHs ($fluoranthene-d_{10},\;benzo(a)pyrene-d_{12}$) and two extract surrogate standards of the PAHs ($fluorene-d_{10},\;pyrene-d_{12}$) were spiked into all samples before field sampling and sample extraction, respectively. Recoveries of field the surrogate standards ranged from $80.4{\pm}12.2%$ for $fluoranthene-d_{10}$ to $66.2{\pm}12.8%$ for $benzo(a)pyrene-d_{12}$. Extraction recoveries of the surrogate standards ranged from $70.4{\pm}10.2%$ for $fluorene-d_{10}$ to $77.6{\pm}10.8%$ for $pyrene-d_{10}$. The detection limit of benzo(a)pyrene among 16 PAHs standards for quantitation was 20 pg.

  • Giant pulmonary bulla (GPB) is a rare manifestation of emphysema and usually enlarges gradually over time, occasionally resulting in complications. Hence, more often than not, the surgical intervention of a Bullectomy is the standard method of treatment for GPB. However, there are case reports that show the complete resolution of GPB after its inflammation process even without surgical intervention. A 51-year-old man was admitted to our clinic due to pleuritic pain. After a chest X-ray and CT scan, a new air-fluid level within the GPB was revealed in the right upper lobe of his lung. His clinical status had improved promptly with intravenous antibiotics. A one-year follow-up study showed the GPB was completely resolved.

  • 페놀은 보통 대기로 배출되며 반감기가 비교적 짧은 물질로서 환경에 농축되는 경우는 드물다. 하지만, 누출사고가 발생되어 수계나 토양에 오염될 경우 페놀에 오염된 농수산물의 안전성을 확보하기 위하여 페놀을 정확하게 검출할 수 있는 시험법을 확립하고자 하였다. 식품 중 페놀을 초음파추출기로 추출하고 GPC로 정제하여 GC/MS로 분석하는 시험법을 확립하였으며, 시험법의 유효성 확인을 위한 실험 결과들이 AOAC 가이드라인에서 제시하는 Criteria를 만족함으로써 시험법의 신뢰성을 확보할 수 있었다. 확립된 시험법은 식품 중 페놀의 오염조사 및 이행률 조사에 활용될 수 있다.

  • A local-step optimization method is proposed to supplement the global-step optimization methods which adopt online update mode of internal weights and error energy as stop criterion in learning of multilayer perceptrons (MLPs). This optimization method is applied to the standard online error backpropagation(EBP) and the performance is evaluated for a speaker verification system.

  • A new quantitative analysis of chloride by the HERETIC NMR method which does not need internal or external references was described. The results showed that the use of HERETIC peak corresponding $500\;{\mu}g/mL$ of chloride calibration showed less than 4 % standard deviations from 50 to $5000\;{\mu}g/mL$ range of chloride concentrations.

  • In this paper, we suggest more precise performance analysis method of turbo interleavers based on two criteria; performance bounds like Union Bound and weight frequency of codewords. In order to present our new method, we employ block, pseudo random, and so-called prime interleavers in compliance of 3GPP standard, respectively. 3GPP complied turbo encoder, decoder, and AWGN channel are implemented by using MATLAB for our performance analysis. According to our analysis, both criteria should be taken into account coincidently to predict the performance of newly designed interleavers.

Gas Chromatography법에 의한 Quercetin의 미량분석에 관한 연구 (Microquantitative Analysis of Quercetin by Gas Chromatography)

  • This investigation was intended to develope the quantitative gas liquid chromatographic method for quercetin. The method involves derivatization of the sample with trimethylsilylimidazole containing cholesterol as an internal standard and injection into a gas chromatograph with 3% OV-17 column. The calibration curve for the quercetin was linear over a sample in the range of 4-16.mu.g. The minimum detectable amount using FID in $1.6{\times}10^{-10}$ a.f.s. was determined to be $6.64{\times}10^{-10}mol$.

On-Line Determination of dn/dc for Size Exclusion Chromatography Coupled with a Light Scattering Detector

  • The internal standard method, which is widely used in chromatographic techniques, is applied to the size exclusion chromatography for the on-line determination of dn/dc value of the sample. This method is found to provide the dn/dc value with suitable accuracy in determining the absolute molecular weight and the molecular weight distribution of the polymers when a light scattering detector is used.
