로로통(路路通)이 천식유발 chemokine 분비와 호산구 chemotaxis에 미치는 영향 (Fructus Liquidambaris Contributes to the Chemotaxis of Eosinophils and Secretion of Cytokines in A549 Human Epithelial Cells)

  • Objectives: The present study was designated to evaluate the direct effects of Fructus Liquidambaris on suppression of eosinophil activity and on suppression of chemokines such as eotaxin, IL-8, ICAM-1, and VCAM-1, in vitro. Methods: A549 cells were stimulated with TNF-$\alpha$ (100 ng/ml), IL-4 (100 ng/ml) or IL-$1{\beta}$(10 ng/ml) to induce chemokines and adhesion molecules involved in eosinophil chemotaxis. Then after treatment of Fructus Liquidambaris, inhibition effect assay such as ELISA, real-time RT-PCR, and chemotaxis assay was performed. Results: Eotaxin level was suppressed in both protein secretion and mRNA expression. Eosinophil recruitment to lung epithelial cells was also reduced by Fructus Liquidambaris, implying the role of eotaxin in eosinophil recruitment. In addition, expression of IL-8 was also suppressed by Fructus Liquidambaris (p<0.05). However, expression of adhesion molecules (ICAM-1, VCAM-1) related to eosinophil was not affected. The eosinophil migration was inhibited at high concentration of Fructus Liquidambaris (p<0.05). Conclusion: These results suggest that Fructus Liquidambaris may regulate a common signaling pathway of eotaxin and IL-8. FS might be of therapeutic value in diseases such as asthma.

  • The liver is involved in a wide range of activities in vertebrates and some other animals, including metabolism, protein synthesis, detoxification, and the immune system. Until now, various methods have been devised to study liver diseases; however, each method has its own limitations. In situ liver perfusion machinery, originally developed in rats, has been successfully adapted to mice, enabling the study of liver diseases. Here we describe the protocol, which is a simple but widely applicable method for investigating the liver diseases. The liver is perfused in situ by cannulation of the portal vein and suprahepatic inferior vena cava (IVC), with antegrade closed circuit circulation completed by clamping the infrahepatic IVC. In situ liver perfusion can be utilized to evaluate immune cell migration and function, hemodynamics and related cellular reactions in each type of hepatic cells, and the metabolism of toxic or other compounds by changing the composition of the circulating media. In situ liver perfusion method maintains liver function and cell viability for up to 2 h. This study also describes an optional protocol using density-gradient centrifugation for the separation of different types of hepatic cells, allowing the determination of changes in each cell type. In summary, this method of in situ liver perfusion will be useful for studying liver diseases as a complement to other established methods.

Endoscopic Intervention for Anastomotic Leakage After Gastrectomy

  • Anastomotic leaks and fistulas are significant complications of gastric surgery that potentially lead to increased postoperative morbidity and mortality. Surgical intervention is reserved for cases with severe symptoms or hemodynamic instability; however, surgery carries a higher risk of complications. With advancements in endoscopic treatment options, endoscopic approaches have emerged as the primary choice for managing these complications. Endoscopic clipping is a traditional method comprising 2 main categories: through-the-scope clips and over-the-scope clips. Through-the-scope clips are user friendly and adaptable to various clinical scenarios, whereas over-the-scope clips can close larger defects. Another promising approach is endoscopic stent insertion, which has shown a high success rate for leak closure, although vigilant monitoring is required to monitor stent migration. Infection control is essential in post-surgical leakage cases, and endoscopic internal drainage provides a relatively safe and noninvasive means to manage fluids, contributing to infection control and wound healing promotion. Endoscopic suturing offers full-thickness wound closure, but requires additional training and endoscopic versatility. As a promising tool, endoscopic vacuum therapy potentially surpasses stent therapy by draining inflammatory materials and closing defects. Furthermore, the use of tissue sealants, such as fibrin glue and cyanoacrylate, has been reported to be effective in selected situations. The choice of endoscopic device should be tailored to individual cases and specific patient conditions, with careful consideration of the nature of the defect. Further extensive studies involving larger patient populations are required to provide more robust evidence on the efficacy of endoscopic approach in managing post-gastric anastomotic leaks.

스테인레스강 Overlay 용접부의 Disbonding에 관한 연구 1

  • Many pressure vessels for the hot H$\sub$2//H$\sub$2/S service are made of 2+1/4Cr-1Mo steel with austenitic stainless steel overlay to combat agressive corrosion due to hydrogen sulfide. Hydrogen dissolves in to materials during operation, and sometimes gives rise to unfore-seeable damages. Appropriate precautions must, therefore, be taken to avoid the hydrogen induced damages in the design, fabrication and operation stage of such reactor vessels. Recently, hydrogeninduced cracking (or Disbonding) was found at the interface between base metal and stainless weld overlay of a desulfurizing reactor. Since the stainless steel overlay weld metal is subjected to thermal and internal-pressure loads in reactor operation, it is desirable for the overlay weld metal to have high strength and ductility from the stand point of structural safety. In section III of ASME Boiler and Pressure Vessel Code, Post-Weld Heat Treatment(PWHT) of more than one hour per inch at over 1100.deg. F(593.deg. C) is required for the weld joints of low alloy pressure vessel steels. This heat treatment to relieve stresses in the welded joint during construction of the pressure vessel is considered to cause sensitization of the overlay weld metal. The present study was carried out to make clear the diffusion of carbon migration by PWHT in dissimilar metal welded joint. The main conclusion reached from this study are as follows: 1) The theoretical analysis for diffusion of carbon in stainless steel overlay weld metal does not agree with Fick's 2nd law but the general law of molecular diffusion phenomenon by thermodynamic chemical potential. 2) In the stainless steel overlay welded joint, the PWHT at 720.deg. C for 10 hours causes a diffusion of carbon atoms from ferritic steel into austenitic steel according to the theoretical analysis for carbon migration and its experiment. 3) In case of PWHT at 720.deg. C for 10 hours, the micro-hardness of stainless steel weld metal in bonded zone increase very highly in the carburized layer with remarkable hardening than that of weld metal.

MicroRNA-152-5p inhibits proliferation and migration and promotes apoptosis by regulating expression of Smad3 in human keloid fibroblasts

  • Keloids are the most common pathological form of trauma healing, with features that seriously affect appearance and body function, are difficult to treat and have a high recurrence rate. Emerging evidence suggests that miRNAs are involved in a variety of pathological processes and play an important role in the process of fibrosis. In this study, we investigated the function and regulatory network of miR-152-5p in keloids. The miRNA miR-152-5p is frequently downregulated in keloid tissue and primary cells compared to normal skin tissue and fibroblasts. In addition, the downregulation of miR-152-5p is significantly associated with the proliferation, migration and apoptosis of keloid cells. Overexpression of miR-152-5p significantly inhibits the progression of fibrosis in keloids. Smad3 is a direct target of miR-152-5p, and knockdown of Smad3 also inhibits fibrosis progression, consistent with the overexpression of miR-152-5p. The interaction between miR-152-5p and Smad3 occurs through the Erk1/2 and Akt pathways and regulates collagen3 production. In summary, our study demonstrates that miR-152-5p/Smad3 regulatory pathways involved in fibrotic progression may be a potential therapeutic target of keloids.

필리핀 국제결혼이주여성의 초국가적 행태에 관한 연구 (Intermarriage Migration and Transnationalism focused on Filipina Wives in South Korea)

  • This study is to explain the nature of transnational activities being involved in by Filipina intermarriage migrants in Korea by examining the institutional backgrounds of market, society and the state. The increasing number of Filipina intermarriage with Korean coincides with the advance of liberal market economy, which governs internal and bilateral interactions between and among the three institutions in both countries. While existing various reasons for engaging in intermarriage, a significant number of Filipina wives in Korea ventured into it with uncertain expectations that they might earn better lives and could support their families. Such hopes usually turn out in vain when they meet the real lives in Korea. It is mainly because their spouses in Korea would rather be those who left behind in the marriage market due to their lack of competitiveness. Filipina wives are also suffering from social isolation caused by language and other barriers such as family relations or rural life they might settle in. Their transnational activities usually tend to be their effort to breakthrough their unexpected condition of difficult lives in Korea. They usually make use of transnational sort of community activities to cultivate chances to engage in bread earning activity. Migrant's transnational activity has a great impact on sociocultural changes in the country of origin and of arrival. Transnational activity provides migrants with economic opportunities, and uplifts self-esteem as well. Intermarriage couples, especially with Southeast Asian wives, and their offsprings show a tendency of downward assimilation to Korean society. Korean state policy toward them should not simply apply undiscriminated assimilation theory, but take into account their possible strength of transnational identity with which they could find a means to integrate themselves successfully into the mainstream Korean society.

대규모 멀티미디어 서버에서 효율적인 데이터 이동/중복 기법 (An Efficient Data Nigration/Replication Scheme in a Large Scale Multimedia Server)

  • 최근 멀티미디어 데이터의 품질이 높아짐에 따라 서버는 더 큰 용량의 저장 공간과 더 높은 I/O 대역폭을 요구하고 있다. 이러한 대규모 멀티미디어 서버에서는 각 객체의 인기도에 따른 접근 빈도의 차이로 인한 디스크 간의 부하 균형 문제가 성능에 중요한 영향을 미친다. 따라서 이 문제를 해결하기 위해 여러 데이터 중복 기법들이 제안되었다. 본 논문에서는 멀티미디어 서버에서 사용되는 대표적인 데이터 중복 기법인 동적 중복 기법보다 저장 효율이 뛰어나고 성능이 우수한 이동/중복 기법을 제안한다. 이 기법은 기존 중복 기법의 단점인 각 객체의 복사본 수를 줄임으로써 중복으로 인해 추가로 요구되는 저장 공간을 크게 감소시킨다. 또한 각 객체에 대한 요청들 간의 간격을 줄임으로써 캐슁 효과를 높여서 기존 중복 기법에 비해 동시에 지원할 수 있는 최대 사용자 수를 증가시킨다.

rhGM-CSF(Leucogen)의 비감염성 상처 치유 효과에 관한 연구 (The healing effect of rhGM-CSF on uninfected wounds)

  • Background: rhGM-CSF has been shown to enhance the migration and proliferation of endothelial cells and to promote keratinocyte growth. This study was tried to evaluate the effect of rhGM-CSF dressing on the uninfected wounds. Methods: Thirty Sprague-dawley white mice(250-300g) were selected in this study. The number of wound with the diameter of 5 mm, was 3 in left and 3 in right at the symmetric sites, respectively. The site of rhGM-CSF dressing was decided by a randomization. rhGM-CSF($Leucogen^{(R)}$) was diluted in the distilled water($5{\mu}g/mL$). The experimental wound group was dressed by l mL of distilled water mixed with rhGM-CSF and control wound group was dressed by l mL of distilled water. The dressing was done, every 24 hours. The criteria of comparison were the duration of wound healing duration, histologic findings and the bacterial culture of wound sites. Results: The duration of wound healing was $10.3{\pm}1.7days$ in experimental group and $10.2{\pm}2.8days$ in control group, without significant difference. There was no specific difference of histologic findings between both groups. The pathogen was not found, at all. Conclusion: It seems to be that rhGM-CSF has no prominent effect on the uninfected wound healing in the mice without immune suppression.

저잡음 · 고신뢰성 Differential Paired eFuse OTP 메모리 설계 (Design of Low-Noise and High-Reliability Differential Paired eFuse OTP Memory)

  • 본 논문에서는 power IC에서 파워가 ON되어있는 동안 입력 신호인 RD(Read) 신호 포트에 glitch와 같은 신호 잡음이 발생하더라도 파워-업(power-up)시 readout된 DOUT 데이터를 유지하면서 다시 읽기 모드로 재진입하지 못하도록 막아주는 IRD(Internal Read Data) 회로를 제안하였다. 그리고 pulsed WL(Word-Line) 구동방식을 사용하여 differential paird eFuse OTP 셀의 read 트랜지스터에 수 십 ${\mu}A$의 DC 전류가 흐르는 것을 방지하여 blowing 안된 eFuse 링크가 EM(Electro-Migration)에 의해 blowing되는 것을 막아주어 신뢰성을 확보하였다. 또한 program-verify-read 모드에서 프로그램된 eFuse 저항의 변동을 고려하여 가변 풀-업 부하(variable pull-up load)를 갖는 센싱 마진 테스트 기능을 수행하는 동시에 프로그램 데이터와 read 데이터를 비교하여 PFb(pass fail bar) 핀으로 비교 결과를 출력하는 회로를 설계하였다. $0.18{\mu}m$ 공정을 이용하여 설계된 8-비트 eFuse OTP IP의 레이아웃 면적은 $189.625{\mu}m{\times}138.850{\mu}m(=0.0263mm^2)$이다.

악성 위출구 폐쇄 치료의 선택: 스텐트 삽입술 혹은 수술적 우회술? (Pyloric Obstruction with Advanced Gastric Cancer: Stent vs. Bypass)

  • 악성 위장관 협착에 의한 폐쇄 증상의 개선을 위하여 과거에는 고식적인 수술적 우회술을 주로 시행하였으나, 최근 내시경적 스텐트 삽입술이 개발되고 기술적 진보와 임상 경험이 축적됨에 따라 새로운 치료 방침의 하나로 확립되었다. 스텐트 삽입술은 고식적 우회술에 비해 시술이 간편하여 성공률이 높고 비침습적이며 단기간에 증상을 개선시키고 시술과 관련된 이환률이 적다는 장점이 있으며 시술에 필요한 입원기간이 짧고 비용면에서 더욱 효과적일 수 있다. 반면 종양의 스텐트 내발육 혹은 위산이나 담즙에 의한 스텐트 손상, 스텐트 일탈 등의 합병증이 발생하여 폐쇄 증상이 조기에 재발하고 재시술이 필요할 수 있다는 문제점이 있어 최근에는 스텐트 일탈을 예방하고 종양의 내발육을 극복할 수 있는 다양한 종류의 스텐트가 개발되어 임상에 응용되고 있다. 일반적으로 악성 위장관 폐쇄에 의한 폐쇄 증상의 완화를 목적으로 하는 스텐트 삽입술은 기대 여명이 6개월 이하인 환자에서 효과적이고 그 이상의 생존이 기대되는 환자에서는 고식적 우회술을 시행하는 것이 적합한 치료 방침으로 생각된다. 본 종설에서는 악성 위출구 폐쇄 환자의 치료에서 쟁점이 되고 있는 스텐트 삽입술과 수술적 우회술 중 어떤 치료가 보다 적절한가를 판단하기 위해서 기술적 측면, 임상 증상의 개선, 합병증, 비용 대비 효과 등을 문헌 고찰을 통해 비교함으로써 적절한 치료 방법의 선택에 대한 해답을 찾고자 한다.
