The Effects of Scrophulariae Radix on the Injured Tracheal Tissue Induced $SO_2$ in Rats (현삼(玄蔘)이 $SO_2$로 손상된 흰쥐의 호흡기 조직에 미치는 영향)

  • Objective : This experiment was performed in order to study the effects of Scrophulariae Radix on the injured tracheal tissue induced So, in rats. Methods : Healthy adult male rats weighting about 250g were divided into 4 groups - the Normal group, the Control group, the group of Scrophulariae Radix administration for 5 days after $SO_2$, gas exposure(Sample I), and the group of Scrophulariae Radix administration for 10 days before and for 5 days after $SO_2$, gas exposure(Sample II). Results : In the trachea Control group, the lesion of the ciliated epithelium was severe and the mucus secretion of the respiratory tract was increased significantly. In the trachea of Sample I group, the lesion of the ciliated epithelium and the mucus secretion of the respiratory tract were decreased compared with Control group. In the trachea of Sample II group, the lesion of the ciliated epithelium and the mucus secretion of the respiratory tract were decreased compared with Control and Sample I group. conclusions : According to the above results, Scrophulariae Radix has significant effects on the injuried tracheal tissue caused by $SO_2$ in rats.

Human Pluripotent Stem Cell-Derived Alveolar Epithelial Cells as a Tool to Assess Cytotoxicity of Particulate Matter and Cigarette Smoke Extract

  • Human pluripotent stem cells (hPSCs) can give rise to a vast array of differentiated derivatives, which have gained great attention in the field of in vitro toxicity evaluation. We have previously demonstrated that hPSC-derived alveolar epithelial cells (AECs) are phenotypically and functionally similar to primary AECs and could be more biologically relevant alternatives for assessing the potential toxic materials including in fine dust and cigarette smoking. Therefore, in this study, we employed hPSC-AECs to evaluate their responses to exposure of various concentrations of diesel particulate matter (dPM), cigarette smoke extract (CSE) and nicotine for 48 hrs in terms of cell death, inflammation, and oxidative stress. We found that all of these toxic materials significantly upregulated the transcription of pro-inflammatory cytokines such as IL-1α, IL-β, IL-6, and TNF-α. Furthermore, the exposure of dPM (100 ㎍/mL) strongly induced upregulation of genes related with cell death, inflammation, and oxidative stress compared with other concentrations of CSE and nicotine. These results suggest that hPSC-AECs could be a robust in vitro platform to evaluate pulmotoxicity of various air pollutants and harmful chemicals.

Opportunistic investigation of vascular calcification using 3-dimensional dental imaging

  • Purpose: Given the growing use of cone-beam computed tomography (CBCT) scans, this study assessed radiation exposure from these scans in the context of national guidelines and recommended dose limits. Materials and Methods: The current literature was reviewed to quantify the benefit of opportunistic diagnosis of carotid artery calcification relative to the potential risk of radiation-induced cancer. Results: The average radiation from CBCT at its largest field of view and highest resolution possible amounts to a reasonable but still low ionizing radiation exposure. This exposure is comparable to 22 days of background radiation and is notably lower than the radiation exposure from medical CT scans. According to the risk assessment analysis, the risk of stroke events involving internal and external carotid artery calcification (CAC) was 202 and 67 per 100,000 individuals, respectively. In contrast, the estimated risk of radiation-induced cancer associated with CBCT was notably lower, at 0.6 per 100,000. Conclusion: The present study advocates for a comprehensive assessment of CBCT scans encompassing the areas of the internal and external carotid arteries by a knowledgeable professional, given the potential advantages of early detection of vascular abnormalities. Dental professionals who take scans involving these areas need to be mindful of reporting these findings and refer patients to their primary care physician for further investigation.

Analysis of Domestic and Overseas Radioactive Waste Maritime Transportation and Dose Assessment for the Public by Sinking Accident (국내·외 방사성폐기물 해상운반 현황 및 침몰사고 시 일반인 선량평가 사례 분석)

  • Demand for RW transportation is expected to increase due to the continuous generation of RW from nuclear power plants and facilities, decommissioning of plants, and saturation of spent fuel temporary storage facilities. The locational aspect of plants and radiation protection optimization for the public have led to an increasing demand for maritime transportation, necessitating to apprehend the overseas and domestic current status. Given the potential long-term radiological impact on the public in the event of a sinking accident, a pre-transportation exposure assessment is necessary. The objective of this study is to investigate the overseas and domestic RW maritime transportation current status and overseas dose assessment cases for the public in sinking accident. Selected countries, including Japan, UK, Sweden, and Korea, were examined for transport cases, Japan and the U.S were chosen for dose assessment case in sinking accidents. As a result of the maritime transportation case analysis, it was performed between nuclear power plants and reprocessing facilities, from plants to disposal or intermediate storage facilities. HLW and MOX fuel were transported using INF 3 shipments, and all transports were performed low speed of 13 kn or less. As a result of the dose assessment for the public in sinking accident, japan conducted an assessment for the sinking of spent fuel and vitrified HLW, and the U.S conducted for the sinking of spent fuel. Both countries considered external exposure through swimming and working at seashore, and internal exposure through seafood ingestion as exposure pathway. Additionally, Japan considered external exposure through working on board and fishing, and the U.S considered internal exposure through spray inhalation and desalinized water and salt ingestion. Internal exposure through seafood ingestion had the largest dose contribution. The average public exposure dose was 20 years after the sinking, 0.04 mSv yr-1 for spent fuel and 5 years after the sinking, 0.03 mSv yr-1 for vitrified HLW in Japan. In the U.S, it was 1.81 mSv yr-1 5 years after the sinking of spent fuel. The results of this study will be used as fundamental data for maritime transportation of domestic RW in the future.

  • Recently, bathes have been suspected to an Important source of indoor exposure to volatile organic compounds(VOCs). Two experiments were conducted to evaluate chloroform exposure and corresponding body burden by exposure routes while bathing. Another experiment was conducted to ekamine the chloro- form dose during dermal exposure and the chloroform decay In breath after dermal exposure. The chioroform dose was determined based on exhaled breath analysis. The ekamine breath concentration measured after normal baths (2.8 Vg/$m^3$) was approxidmately 13 tomes higher that measured prior to normal bathes (0.2 ug/$m^3$). Based on the means of the normalized post exposure chloroform breath concentration. the dermal exposure was estimated to contribute to 74% of total chloroform body burden while bathing. The Internal dose from bathing (Inhalation plus dermal) was comparable to the dose ostimated Srom dally water Ingestion. The rusk associated 10 a weekly, 30-min bath was estimated to be 1 x 10.5, while the rusk firom dally Ingestion of tap water was to be $0.5{\times}0^{-5} for 0.151 and 6.5{\times}10^{-5}$ for 2. 0 1. Chloroform breath concentration Increased gradually during the 60 minute dermal exposure. The breath decay after the dermal exposure showed two-phase mechanism, with early raped decay and the second slow decay. The mathematical model was developed to describe the relationship between water and air chloroform concentrations, with $R^2$ : 0.4 and p<0.02.

EphA2 Receptor Signaling Mediates Inflammatory Responses in Lipopolysaccharide-Induced Lung Injury

  • Background: Eph receptors and ephrin ligands have several functions including angiogenesis, cell migration, axon guidance, fluid homeostasis, oncogenesis, inflammation and injury repair. The EphA2 receptor potentially mediates the regulation of vascular permeability and inflammation in response to lung injury. Methods: Mice were divided into 3 experimental groups to study the role of EphA2 signaling in the lipopolysaccharide (LPS)-induced lung injury model i.e., IgG+phosphate-buffered saline (PBS) group (IgG instillation before PBS exposure), IgG+LPS group (IgG instillation before LPS exposure) and EphA2 monoclonal antibody (mAb)+LPS group (EphA2 mAb pretreatment before LPS exposure). Results: EphA2 and ephrinA1 were upregulated in LPS-induced lung injury. The lung injury score of the EphA2 mAb+LPS group was lower than that of the IgG+LPS group ($4.30{\pm}2.93$ vs. $11.45{\pm}1.20$, respectively; p=0.004). Cell counts (EphA2 mAb+LPS: $11.33{\times}10^4{\pm}8.84{\times}10^4$ vs. IgG+LPS: $208.0{\times}10^4{\pm}122.6{\times}10^4$; p=0.018) and total protein concentrations (EphA2 mAb+LPS: $0.52{\pm}0.41mg/mL$ vs. IgG+LPS: $1.38{\pm}1.08mg/mL$; p=0.192) were decreased in EphA2 mAb+LPS group, as compared to the IgG+LPS group. In addition, EphA2 antagonism reduced the expression of phospho-p85, phosphoinositide 3-kinase $110{\gamma}$, phospho-Akt, nuclear factor ${\kappa}B$, and proinflammatory cytokines. Conclusion: This results of the study indicated a role for EphA2-ephrinA1 signaling in the pathogenesis of LPS-induced lung injury. Furthermore, EphA2 antagonism inhibits the phosphoinositide 3-kinase-Akt pathway and attenuates inflammation.

Lack of Sunlight Exposure Influence on Primary Glioblastoma Survival

  • Background: The prognosis of primary glioblastoma (GBM) is poor. Approximately 2/3 of primary brain tumor diagnoses are GBM, of which 95% are primary lesions. In this study, we aimed to evaluate whether more sunlight exposure has an effect on survival of patients with primary GBM. Materials and Methods: A total of 111 patients with primary GBM were enrolled from Kayseri in inner Anatolia which has a cold climate (n: 40) and Mersin in Mediterranean region with a warm climate and more sunlight exposure (n: 71). The patients with primary GBM were divided into two groups as Kayseri and Mersin and compared for progression free survival (PFS) and overall survival (OS).Results: The PFS values were 7.0 and 4.7 months for Kayseri and Mersin groups, respectively (p=0.10) and the repsective OS values were 13.3 and 9.4 months (p=0.13). We did not found any significant difference regarding age, sex, comorbidity, smoking, surgery, resurgery, adjuvant chemoradiotherapy and palliative chemotherapy between the groups. Conclusions: We found that more sunlight exposure had no impact on prognosis of patients with primary GBM, adding inconsistency to the literature about the relationship between sunlight and GBM.

  • Nuclear reaction which occurs in the cyclotron generate unnecessary neutrons. The results of this happening can radioactivate surrounding materials and radioactive materials cause radiation exposure. When people take radioactive air, it makes internal exposure. The purpose of this study was to analyze the radioactive air inside of the ultra-compact 16.5 MeV cyclotron in operation. As a result of study, the radio activation occurred by compact cyclotron generates a very low internal exposure to workers. Comparing the radioactivity from radioactive nuclide with legal standard, that was under reference value. However, it could be at risk for internal exposure in case of higher energy cyclotron. Therefore, legal standard is needed for ventilation equipment of radiation facilities.

Characteristics of Internal and External Exposure of Radon and Thoron in Process Handling Monazite (모나자이트 취급공정에서의 라돈 및 토론 노출 특성)

  • Objectives: The purpose of this study was to evaluate airborne radon and thoron levels and estimate the effective doses of workers who made household goods and mattresses using monazite. Methods: Airborne radon and thoron concentrations were measured using continuous monitors (Rad7, Durridge Company Inc., USA). Radon and thoron concentrations in the air were converted to radon doses using the dose conversion factor recommended by the Nuclear Safety and Security Commission in Korea. External exposure to gamma rays was measured at the chest height of a worker from the source using real-time radiation instruments, a survey meter (RadiagemTM 2000, Canberra Industries, Inc., USA), and an ion chamber (OD-01 Hx, STEP Co., Germany). Results: When using monazite, the average concentration range of radon was $13.1-97.8Bq/m^3$ and thoron was $210.1-841.4Bq/m^3$. When monazite was not used, the average concentration range of radon was $2.6-10.8Bq/m^3$ and the maximum was $1.7-66.2Bq/m^3$. Since monazite has a higher content of thorium than uranium, the effects of thoron should be considered. The effective doses of radon and thoron as calculated by the dose conversion factor based on ICRP 115 were 0.26 mSv/yr and 0.76 mSv/yr, respectively, at their maximum values. The external radiation dose rate was $6.7{\mu}Sv/hr$ at chest height and the effective dose was 4.3 mSv/yr at the maximum. Conclusions: Regardless of the use of monazite, the total annual effective doses due to internal and external exposure were 0.03-4.42 mSv/yr. Exposures to levels higher than this value are indicated if dose conversion factors based on the recently published ICRP 137 are applied.

Study of correlation between airborne benzene and urinary trans,trans-muconic acid in Petrochemical industry processes (공기 중 벤젠과 소변 중 뮤콘산과의 상관성 연구)

  • To investigate the exposure effect of benzene, we measured airborne benzene as external doses, uninary tt-muconic acid as an internal dose of benzene exposure and analyzed the relationship between tt-muconic acid concentration and benzene exposure. The study population of eight businesses included 157 workers(87 workers in field; exposure group, 70 workers in board; control group) who produce or use benzene in petrochemical industry. The concentrations of airborne benzene were evaluated by personal samples and urine was sampled at the pre and end shift. Urinary t,t-muconic acid as internal dose was to analyze the relationship with airborne benzene. The geometric means(GM) of airborne benzene was 0.0231 ppm(range ND-1.0471 ppm) in exposure group and 0.0147 ppm(range ND-0.3162 ppm) in control group. The geometric means(GM) of urinary t,t-muconic acid at end-shift was $196.8{\pm}2.23{\mu}g/g$ creatinine in exposure group and $149.2{\pm}2.08{\mu}g/g$ creatinine in control group. There was significant correlation between the airborne concentration of benzene and the urinary concentration of t,t-muconic acid( r=0.711, p<0.01). From the results of stepwise multiple regression analysis about t,t-muconic acid at end-shift, significant independents was airborne benzene. In this study, there were significant correlation between the urinary concentration of t,t-muconic acid and the airborne concentration of benzene. More extensive studies ruling out healthy worker effect is needed.
