Immunocytochemical Characteristics of the Short-term Cultured Mesothelial Cells (단기배양한 중피세포의 면역세포화학적 연구)

  • Reactive humsn mesothelial cells were examined by immunocytochemical stain with intermediate filaments (cytokeratin [CK1, CK7, CK8, CK18, CD19), vimentin, desmin, actin), epithelial membrane antigen, carcinoembryonic antigen (CEA), MHC class II antigen (HLA-DR), LeuM-1 (CD15), $\alpha1-antitrypsin$(ACT), $\alpha1-antichymotrypsin$ (ACHT), CD68(KP-1) and FcyRIII(CD16). The mesothelial cells were isolated from patients with liver cirrhosis and pleural effusion, and short-term cultured in RPMI 1640 media containing 10% heat inactivated fetal calf serum and 1% identical supernatant fluid of the patients' transudates. The results obtained are as follows 1. The cultured-reactive mesothelial cells were positive for the protein of cytoskeleton such as cytokeratin and vimentin, but negative for desmin and actin. The resting mesothelial cells showed positive reactions for cylokeratin, but negative for vimentin, desmin and actin. 2. The primary antibodies to the cytokeratin were strongly reactive for CK1, CK8 and CK18 but negative for CK7 and CK19 in both reactive and resting mesothelial cells. 3. Resting mesothelial cells showed negative reactions for CEA, but strong positive reactions in cultured-reactive mesothelial cells. 4. The markers for the monocytes/histiocytes(CD11b, CD14, CD16, CD68, Iysozyme and $\alpha1-antitrypsin$ and $\alpha1-antichymotrypsin$) were nonreactive in resting mesothelial cells, but lysozyme and $\alpha1-antitrypsin$ were weakly reactive in reactive and proliferative mesothelial cells. 5. MHC Class II molecule(HLA-DR antigen) was negative in both resting and reactive mesothelial cells. These results suggest that the short-term cultured, reactive mesothelial cells show a newly aberrant expression of the vimentin and calcine-embryonic antigen. The reason of the aberrant expression of the intermediate filament and oncofetal antigen in reactive and proliferative mesothelial cells should be further evaluated.

  • From the concepts of cellular pathology and of exfoliative cytology, as elucidated by Virchow and Papanicolaou respectively in the late 19th and early 20th century, have evolved the primary methods for the diagnosis of cancer today. From Papanicolaou's concept of exfoliative cytology developed fine needle aspiration biopsy in the early 1960's, this has become a major diagnostic procedure and has contributed to a significant reduction in open biopsies and, therefore, to medical cost-effectiveness immunobiochemical techniques provided us with a supplement to cancer diagnosis in the 1980's. The immunoperoxidase method, using monoclonal antibodies, is applied primarily as an ancillary measure to elucidate the nature of cancers The availability of specific monoclonal antibodies has greatly facilitated the identification of cell products or surface markers. For example, antibodies directed against intermediate filaments have proved to be of value in determining the histogenesis oi poorly differentiated neoplasms. Tumor markers may serve as biochemical indicators of the presence of a neoplasm. They can be detected In plasma and other body fluids. Their concentration can be applied as a diagnostic test, for monitoring the clinical course of known cancer, and as a screening measure to detect certain cancers in a population at risk. Flow cytometry is a useful tool for distinguishing several cell characteristics, such as the immunophenotype of leukemia-lymphoma cells, the DNA content of neoplastic cells, and cell proliferation rate. Molecular biologic techniques provided a giant step for the management of cancer patients encompassing diagnosis, prognostic evaluation, and therapy. Nucleic acid hybridization techniques are utilized as Southern, Northern, and dot blots and in situ hybridization. Molecular biology and its techniques may bring a blight new horizon for understanding cancer biology and in designing therapy on the basis of gene manipulation.

Fine Structure and Histopathological Changes Exposed to Acute High Salinity of the Gill of Japanese Clam, Corbicula japonica (일본재첩, Corbicula japonica 아가미의 정상구조와 고염분 급성노출에 의한 조직병리학적 변화)

  • This study was performed to observe ultrastructure of the gill and to ascertain the effect of salinity on histopathological and ultrastructural changes in the gill of the Japanese clam, Corbicula japonica. Experimental period was 7 days. Experimental groups consisted of control, 5, 10, 20 psu. $LC_{50}$ (96 h.) by the probit was 19.55 psu. Mortality was significantly different from the control (p < 0.05). Inner demibranch of the gill of C. japonica was wider 1.37 times than outer demibranch (p < 0.001). The filament zone on the plica can be distinguished by the six epithelial celll cell; frontal ciliated epithelium ($7{\mu}m$), latero-frontal ciliated epithelium ($5{\mu}m$), postlatero-frontal epithelim ($3{\times}8{\mu}m$), and lateral ciliated epithelium ($5{\mu}m$) in the frontal zone, endothelial cellin the intermediate zone, and abfrontal cell in the abfrontal zone. It had one type of secretory cell that was filled with fibrous substances of low electron density. The gill of C. japonica exposed to 5 psu for 7 days was observed partially disappearance of the cilia, and glycogen granule in the filament. In the 10 psu, gill appeared partially modification of epithelial cell and destruction of the glycocalyx. Gill exposed to 20 psu was extended nuclus of the ciliated epithelial cell, destruction of the organelles, and observed glycogen granules infiltration and numerous vacuoles. Moreover, more than 50% filaments were observed that come out chitinous rod from disappearance of epithelial cell in the filament. Therefore, the destruction of the cilia and epithelial cell induce physiological activity and it may be leading directly to death.
