Some structural characteristics of hot-water insoluble rice starch [3 varieties of Japonica type and 3 varieties of Tongil type(Japonica-Indica breeding type)]were investigated. The amylose contents of hot-water insoluble starches were 2.7-6.1%. The chain distributions of hot-water insoluble starches were composed of super long chain of above ${\overline{DP}}\;55$ (31.9-38.7%), B chain of ${\overline{DP}}\;40-50$ (12.3-18.0%) and A chain of ${\overline{DP}}\;10-20$ (48.5-50.4%). The chain distributions of hot-water insoluble starches and their ${\beta}-limit$ dextrin were difference between rice varieties and their were differ from those of amylopectin and their ${\beta}-limit$ dextrin. these result suggest that hot-water insolubility of rice starch was showed its characteristic on the varieties and it be responsible for the molecular structure of amylopectin.
To develop high efficiency biofertilizer solubilizing insoluble phosphates, lactate dehydrogenase (ldh) gene was isolated from Staphylococcus sp. LJ2. Genetic constructions were carried out using the pGEM-T-easy vector and pHSG398. Recombinant DNA plasmids containing the ldh gene were transferred to Klebsiella sp. DA71-1 by electroporation. The selected transformant was named as a DA71-1/pLYJ. The insoluble phosphates solubilization activity of DA71-1/pLYJ was higher than that of DA71-1 at various culture conditions. Glucose was the best carbon source for insoluble phosphates solubilization among the used carbon sources. Maximal insoluble phosphates solubilizing was found in sucrose minimal (SM) medium containing 3% glucose. The solubilizing activity of DA71-1/pLYJ against three types of insoluble phosphates, such as tri-calcium phosphate, hydroxyapatite, aluminium phosphate, were quantitatively determined. The optimal temperature and initial pH to solubilize insoluble phosphates in the SM medium was $37^{\circ}C$ and pH 5.0, respectively.

This study was carried out to have the basic and applied informations relating to develop the cultivation methods and to increase the productivity and quality of ginseng. 1 to 6 year old ginsengs of Jakyung cultivar were cultivated and the content and synthetic amount of carbohydrates were investigated with different plant tissues, growth stages, and years old. The concentration of total carbohydrates at six year old ginseng including water soluble and water insoluble carbohydrates was about 18.9%, 42.9%, and 43,6% in leaves, tap roots, and lateral roots, respectively. Water soluble carbohydrate of tap and lateral roots was slightly decreased from August until September, and then increased on November, whereas its water insoluble carbohydrate was increased from August to September and then decreased on November. Comparing with the content of carbohydrates of 1 to 6 year old ginsengs, it was continuously increased from one year old ginseng until five year old ginseng, however it was not increased much in six year old ginseng. The highest content of carbohydrates was at five year-old in all tissues of ginseng. Water soluble and water insoluble carbohydrates were significantly shown different in leaves, stems, tap roots, and lateral root at different growth stages and with different years old. The content of water soluble carbohydrate in the leaves was remarkedly higher compared to that of water insoluble carbohydrate, while in the root the content of water insoluble carbohydrate was clearly higher compared to the water soluble carbohydrate. Comparing with the synthetic amount of carbohydrates, water soluble carbohydrates was higher in the shoot than that in the root, whereas water-insoluble carbohydrates higher in the root than that in the shoot. Carbohydrates which would be utilized in ginseng tissues for short and long-term periods as major energy were appeared differently in between shoot and root, with different growth stages, and years old.

In order to treat electrolysis nickel plating pickling wastewater to meet the effluent limit less than 3.0 mg/L, the electrolysis process by using soluble and insoluble electrode were studied. Electrolysis using soluble electrodes has a characteristic of easy elution from the electrode which the insoluble electrodes close not release metal from the electrode. For these reasons, there exist different characteristics in nickel removal efficiency, purity of nickel sludge. With this connection, the feasibility test were concluded to develop optimal conditions for the treatment of pickling wastewater electrolysis by using soluble electrodes, insoluble electrodes. Optimal condition of current density, pH were derived from the pickling wastewater using insoluble electrodes. It was concluded the highest removal efficiency of nickel at the operation condition of at pH 9, current density of $15mA/cm^2$. At these conditions, 95.3% purity of nickel sludge was achieved, iron content was 2.9%. Optimal condition when using soluble electrodes was derived current density of $10mA/cm^2$, pH 9. Purity of nickel sludge was 77.3%, iron content was 21.0%. 50.7% and 24.2% of operating cost can be saved by the use of soluble electrodes and the use of insoluble electrodes, respectively.
Phenolic acids of white ginseng were extracted and fractionated into free, esterified, and insoluble-bound forms. The contents of individual phenolic acids in different forms were quantified by gas liquid chromatography. Nine different phenolic acids as free, esterified, and insoluble-bound forms were identified in white ginseng. Total phenolic compounds in different forms of extracts was 0.309% (free form), 0.230% (esterified form) and 0.138% (insoluble-bound form), respectively. Total phenolic acid contents in free, esterified and insoluble-bound form were 889.3, 356.8, 1,176.9 mg/100g fraction, respectively. Ferulic acid was the predominant phenolic acid, representing 63.7% and 50.9% of total phenolic acids in esterified fom and insoluble-bound form, respectively. While caffeic acid was only detected in esterified form. At 10 mg/ml insoluble-bound form quenched 95.9% ABTS free radicals generated from 2,2-azobis(2-amidinopropane) dihydrochloride (AAPH). Also, electron donating ability and lipid peroxidation inhibitory activity of insoluble-bound fom were higher than other fraction. All phenolic acid fractions scavenged over 80% of hydroxyl radical at 10 mg/ml.

The quality inspections items such as loss on drying, total ash, and acid-insoluble ash contents in herbal medicines, have a correlation with external sources of pollution, but are not themselves hazardous factors. Z-scores for standard normal distribution were used to investigate herbal medicines requiring quality control, which exceeds the regulatory limits of quality inspection. In total, 7,773 samples were analyzed based on plant parts. For the loss on drying, the numbers of items of herbal medicines requiring quality control are like these; 15 items for above-ground parts and 5 items for underground parts. For the total ash, 21 items for above-ground parts and 4 items for underground parts. For the acid-insoluble ash, 8 items for above-ground parts and 1 item for underground parts.

The glutathione S-transferase (GST) system is useful for increasing protein solubility and purifying soluble GST fusion proteins. However, purifying half of the GST fusion proteins is still difficult, because they are virtually insoluble under non-denaturing conditions. To optimize a simple and rapid purification condition for GST-pyruvate kinase muscle 2 (GST-PKM2) protein, we used 1% sarkosyl for lysis and a 1 : 200 ratio of sarkosyl to Triton X-100 (S-T) for purification. We purified the GST-PKM2 protein with a high yield, approximately 5 mg/L culture, which was 33 times higher than that prepared using a conventional method. Notably, the GST-high-temperature requirement A2 (GST-HtrA2) protein, used as a model protein for functional activity, fully maintained its proteolytic activity, even when purified under our S-T condition. This method may be useful to apply to other biologically important proteins that become highly insoluble in the prokaryotic expression system.
The electrophoretic and innunological cnalyses of organic matrices in the shells of freshwater bivalves were made in order to dlucidate the biochemical characteristics and species-specific differinces of the applied shells, The water-soluble and insoluble matrices of four species of freshwater bivalves, Andodonta fukudai, Unio douglasiae, Lanceolaria and Chrbicula fluminea, were used as analytical materials, There was non-identity in immuno affinity between anti soluble matrix(anti-Sm) and anti insoluble matrix(anti-ISM)sera against the organic matrix of Andodnta fukudai. The SMs of four species (S. fukudai, U.douglasiae, L. acrorhyncha, and C. fluminea) showed the differences in the precipitate arcs at the level of family, though ISMs did mot show differences. In the electrophoretic analysis, all foru species had two SDS-electrophoretic bands of SM, of which molecular wights appeared to be lower than 55,000, shereas the native organic matrices of foru speceis had higher molecular weighrs than those from SDS-dldctrophoresis. Only calcium ion among many ions in extrapallial fluid(EPF) caused SM to change into insoluble molecules, thus the EPF pretreated with Ca++did not form the precipitate arc when did the immuno diffusion whth anti SM serum. ISM precursor may be polymerized into macromolecules like periostracin, a precusor of periostracum, judging from the similat polymerization patterns in 0.1M Tris formate buffer(pH 3); they may be made insoluble macromolecules due to their strong natrue of hydrophobicity.
Desmutagenic effects of water-soluble and water-insoluble fractions from Aloe vera and Aloe avoresence with different solvents against the mutagenicity of 4-nitro-o-phenylenediamine(NPD) were investigated. Desmutagenic effects of water-insoluble fractions from Aloe vera and Aloe avoresence against NPD-induced mutations in TA97 and TA98 strains were very high compared with water-soluble fractions: whereas water-soluble fractions from two kinds of Aloes showed the inhibition effects of 4∼56% water-insoluble fractions showed the activities of 54∼100% for Aloe vera and 74∼100% for Aloe avoresence. Especially, the addition of 20 $\mu$ι of water-insoluble fractions per plate showed high activities up to 90% in TA97 and TA98 strains.
To provide the basic information of buckwheat insoluble dietary fiber (IDF) and efficient material for the buckwheat processed foodstuffs, buckwheat (Fagopyrum egculentum Moench) was germinated at 1$0^{\circ}C$ for 7 days and the contents and composition of the insoluble dietary fiber were examined at 24 hour intervals. The NDF and ADF contents in ungerminated seeds were 20.27%, 18.95% on dry weight basis, respectively. During the germination period, the NDF and ADF contents increased gradually. The cellulose, hemicellulose and lignin contents in ungerminated seeds were 11.10%, 1.32% and 7.85%, respectively. During the germination period, the cellulose and lignin contents increased. However, there was no significant change in the hemicellulose contents. The composition of cellulose and hemicellulose in the NDF decreased with germination time, whereas that of lignin increased. The IDF contents obtained by Prosky method were higher than the NDF contents obtained by Van Soest method. However, the IDF and NDF contents were closely correlated(r=0.9785, p<0.01). The cellulose(Xl), hemicellulose(X2), lignin(X3) and soluble dietary fiber (SDF) (BL) showed the significant regression equation(p<0.001) with the root length(Y). The multiple regression equation was Y = -12.1306 + 0.9213xl - 0.1615$\times$2 + 0.1071$\times$3 + 0.7397$\times$4 and R2(coefficient of determination) was 0.942.
