향부자 메탄올 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성 억제에 미치는 영향 (Effects of methanol extract of Cyperus rotundus on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans)

  • Streptococcus mutans (S. mutans) is known as the causative bacterial playing the most important role informing plaque and it is being noticed as major causative bacteria of dental caries. Therefore, development of more effective, substantial and safe preventive agent against dental caries and periodontal disease is honestly required. The present study was designed to investigate the effect of Cyperus rotundus (Cyperaceae) methanol extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of C. rotundus showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration of 0.5 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of C. rotundus showed more than 10% inhibition over the concentration of 2 mg/ml. Thus, the application of C. rotundus can be considered a useful and a practical method for the prevention of dental caries.

삼백초 추출물의 항치아우식 효과 (Anticariogenic Properties of the Extract of Saururus chinensis)

  • It has been well established that S. mutans is the major etiological agent in dental caries, one of the most common oral diseases worldwide. The present study was designed to investigate the effect of Saururus chinensis (S. chinensis) ethanol extracts on the growth, acid production, biofilm formation, adhesion, and insoluble glucan synthesis of S. mutans. The ethanol extracts of S. chinensis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2 and 0.4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 80% at the concentration of 0.05 mg/ml and complete inhibition was observed at the concentration of 0.4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of S. chinensis showed more than 10% inhibition over the concentration of 0.025 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.025 ${\sim}$ 0.4 mg/ml of the ethanol extract of S. chinensis. Our research strongly suggested S. chinensis was a promising natural product for the prevention of dental caries.

  • 본 연구의 목적은 우수한 미생물 제제 개발용 인산염 가용화 균주의 개발이다. 경작지 토양에서 분리한 인산염 가용화 균주의 특성 및 16S rDNA 염기서열을 조사한 결과 Bacillus subtilis HR-1019로 동정되었다. Bacillus subtilis HR-1019는 hydroxyapatite, tri-calcium phosphate 및 aluminum phosphate 3가지의 난용성 인산염을 모두 가용화하였다. 난용성 인산염의 분해능이 최대가 되는 배양온도는 $37^{\circ}C$이었으며, 배양초기 pH가 5.0이었다. 탄소원으로 glucose를 5% 첨가시 가용화능이 높았으며, 가용화된 유리인산의 함량이 증가함에 따라 pH가 크게 감소하였다. 분리균주 HR-1019가 식물병원균에 대하여 균의 생육을 저지하는 clear zone 확인으로 항균효과를 확인하였다.

오이의 Pectinesterase 에 관한 연구 (Characteristics of Pectinesterase (PE) in Cucumbers)

  • This study was attempted to investigate the occurrence and the partial characteristics of pectinesterase (PE) in cucumbers. And the involvement of endogenous cucumber PE with the formation of insoluble pectic acid, in the presence of endogenous or added calcium ions, was also studied. The results of this study are as follows: 1) PE activity was detected in whole cucumber homogenate. 2) The optimum pH and temperature of this enzyme were found to be 8.5 and $50^{\circ}C$, respectively 3) When PE extract was added to 0.25% pectin solution, pectin gel was formed. The time required for the formation of pectin gel was reduced when the pectin solution was adjusted to the optimum conditions for PE activity(pH 8.5, $50^{\circ}C$) and $CaCl_2$ was added. 4) Cucumber juice which was heated to $100^{\circ}C$ for 10 minutes fatted to form any insoluble pectate precipitate. The formation of precipitate in cucumber juice was accelerated by preheating to $50^{\circ}C$, adjusting to pH 8.5 and addition of $CaCl_2$. 5) These results can be interpreted that endogenous PE in cucumber juice demethylates pectin, allowing interlinking of pectin molecules via divalent cation $(Ca^{++})$ and thus forming insoluble Ca-pectate. Therefore additional firming effects of cucumber can be expected to be obtained through activation of PE in conjunction with calcium ions.

Effect of Alcohol Insoluble Residues from Stem and Root Barks of Elm (Ulmus davidiana) on Intestinal Characteristics in Rats

  • Sprague-Dawley rats (n=32) were fed a diet containing basal (control), cellulose (5%), or alcohol insoluble residue (AIR) (5%) extracted from the stem and root barks of elm (Ulmus davidiana var. japonica Nakai) for 4 weeks. The effects of the diets, on gastrointestinal functions and morphology were evaluated. The weight gains, food intake, and food efficiencies for the cellulose and AIR diet-fed groups were not significantly different from those of the AIR-free (basal) diet. The gastrointestinal transit times of the stem and root bark AIR diets were significantly reduced (p<0.01) compared to the basal diet, and were slower than those of the cellulose diet. The fecal weights of the stem and root bark AIR diets were significantly increased (p<0.01) up to 4-fold compared to those of the basal diet. The height of the mucosal villi, and mucosal and muscle layer thicknesses of the colon were greater and more developed in the stem and root bark AIR diets (p<0.01) than in the basal diet. The villus heights in the jejunum and the colon mucosal goblet cells were more developed in the order of cellulose > stem bark AIR > root bark AIR diets.

Physicochemical Properties and Intestinal Bacterial Growth-Promoting Effect of Cell-Wall Polysaccharides from Cucumber Peel

  • Physicochemical properties, intestinal microbial growth, and inhibitory effects of alcohol-insoluble polysaccharide (AIP) extracted from cucumber peel were investigated. AIP was composed of 14.54% crude protein, 1.04% crude lipid, 13.74 % crude ash, 9.1% soluble dietary fiber, and 41.2% insoluble dietary fiber. AIP showed low bulk density (0.18 g/mL) and water-holding capacity (6.39 g/g), and high oil-holding capacity (3.96 g/g). Pectic substance fractions [water-soluble pectic substance (WSP), ethylenediaminetetraacetic acid-soluble pectic substance (ESP), and alkali-soluble pectic substances (ASP)] and hemicellulose fractions [1 M KOH-soluble hemicellulose (KHP1) and 4 M KOH-soluble hemicellulose (KHP4)] were obtained from sequential chemical fractionation of AIP. WSP showed higher total sugar contents than total uronic acid contents, whereas opposite results were observed in ESP and ASP. Molecular weight distributions of three pectic substance fractions were in order of ASP>ESP>WSP. Ion exchange chromatogram pattern of WSP was different from those of ESP and ASP. Major component of WSP was fraction eluted by 0.05 M ammonium acetate buffer, whereas that of ESP and ASP was fraction eluted by 0.2 M NaOH. WSP and ASP showed growth-promoting activities against Lactobacillus brevis, Bifidobacterium bifidum, and B. longum, whereas B. bifidum and B. longum for ESP. KHP1 and KHP4 fractions had significant growth-promoting activities against B. bifidum.

Comparison of the Nutritional Composition of Bullfrog Meat from Different Parts of the Animal

  • The objective of this study was to evaluate the nutritional content of bullfrog meat from different parts of the animal, including fore-chest, thigh and calf. Bullfrog meat was found to be a rich source of proteins, essential amino acids and minerals, but with a low fat content, compared with other aquatic meat products. There was no significant difference (p>0.05) between thigh and calf in mineral content (K, P, Na, Mg, Ca, Zn, Fe, Cu, and Mn), but the contents of K, P, and Mg were higher in thigh and calf than in the fore-chest (p<0.05). The salt-soluble, water-soluble and insoluble protein bands in sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) electrophoresis, from fore-chest, thigh and calf were similar, with the most abundant bands being 35 kDa (salt-soluble protein), 35-48 kDa (water-soluble protein) and 48 kDa (insoluble protein). The results showed that the insoluble protein content in the fore-chest meat was higher than that in the thigh meat and calf meat, but the salt-soluble protein fraction was the most abundant in thigh meat. These results showed that the nutrients in different parts of bullfrog meat were different.

  • 한국산의 참깨를 탈피(脫皮) 한 것과 탈피(脫皮) 하지 않은것, 이들을 각각 채유(採油)한 기름 및 박(粕)에 대하여 일반성분, 기름의 성상(性狀), 지방 조성, 아미노산 조성, 특수 미량성분 및 oxalate 의 함량 들을 비교 분석한 결과 다음과 같은 결론을 얻었다. 1) 탈피(脫皮)참깨 및 그 박(粕)은 탈피(脫皮)하지 않은 전(全)참깨 및 박(粕)보다 조섬유, 조회분, 산불용성회분의 함량이 더 적었다. 2) 탈피(脫皮)참깨기름은 전(全)참기름 보다 비중, 굴절률, 옥소가, 비검화물, 불용성잡물의 함량이 더 컸었다. 3) 탈피(脫皮)참기름과 전(全)참기름의 지방상 조성은 서로 비슷하였으나 전자는 후자보다 불포화지방산의 함량이 다소 적었다. 4) 탈피(脫皮)참깨박(粕)은 전(全)참깨박(粕)보다 단백질 함량이 매우 높았다. 그리고 아미노산 조성은 서로 거의 비슷하였으며 이들의 박(粕)은 모두 methionine, Cystine, tryptophan 은 풍부하나 lysine 의 함량이 적었다. 5) 탈피(脫皮)참깨기름과 전(全)참깨기름중의 sesamol, sesamolin 및 sesamin 의 함량은 서로 차이점이 없었다. 6) 탈피(脫皮)참깨 및 그박(粕)은 전(全)참깨 및 그박(粕)보다 oxalate 및 calcium 의 함량이 적었다.

  • Dental plaque is a film of microorganisms on the tooth surface that plays an important part in the development of caries and periodontal diseases. Streptococcus mutans (S. mutans) is present in almost all types of dental plaque. Teeth and their supporting structure, the gums (gingiva) are subjected to infection by S. mutans that causes cavities and pyorrhea which, if left untreated, can eventually lead to gingivitis. Various chemical agents have been evaluated over the years with respect to their antimicrobial effects in the oral cavity; however, all are associated with side effects that prohibit regular long-term use. The present study was designed to investigate the effect of Aralia continentalis (Araliaceae) extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of A. continentalis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.25, 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 60% at the concentration of 0.25 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of A. continentalis showed more than 10% inhibition over the concentration of 0.25 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.25 - 4 mg/ml of the methanol extract of A. continentalis. Hence, we conclude that A. continentalis might be a candidate of anticaries agent.

불용성 폴리머가 탄소섬유 보강 Polymer-MDF 시멘트 복합재료의 기계적 특성에 미치는 영향 (The Effects of Insoluble Polymers on Water Stability of Carbon Fiber Reinforced Polymer-MDF Cementitious Composites)

  • HAC/PVA계 MDF시멘트 복합재료의 수분안정성 및 기계적 강도향상을 위하여 불용성 폴리머류(폴리우레탄, 페놀, 에폭시수지)와 탄소섬유(길이3mm)를 1-4wt% 보강하였다. 그리고 가압성형법으로 제조한 복합체의 수분침적 기간별 강도특성(수분안정성)과 계면 및 기계적 특성에 영향을 미치는 폴리머와 시멘트의 가교반응에 대하여 SEM 및 TEM 분석을 통하여 관찰하였다. HAC/PVA계 MDF 시멘트 복합체의 섬유함량에 따른 건조 굽힘강도는 섬유함량이 증가할수록 치밀화 구조가 저하되어 비례적으로 감소되었다. 또한 불용성 폴리머류를 사용한 경우에 건조 굽힘강도는 섬유함량이 증가될수록 저하되는 반면, 수분안정성은 크게 향상되었다. 에폭시 수지를 첨가한 경우에 수분안정성이 가장 우수하였으며, 섬유함량 4% 첨가의 경우에 3일 침적강도가 95%, 7일침적시 87%강도를 유지하였다. 이 점은 폴리머와 시멘트의 금속이온이 가교반응을 일으켜 섬유-메트릭스간 계면 부착강도를 크게 개선되었기 때문으로 추정된다. 반면 인장강도 특성은 모든 불용성 폴리머류 첨가 수준에서 섬유 함량이 증가할수록 비례적으로 증가되었으며, 역시 에폭시 수지 첨가의 경우에 강도특성이 가장 우수하였다. 그리고 섬유함량 4% 첨가된 경우에 있어서 인장강도는 섬유함량 0% 대비 약 30~80% 높게 나타났다.
