Potential Suppression of Dental Caries by Maltosyl-Mannitol Produced by Bacillus stearothermophilus Maltogenic Amylase

  • Maltosyl (G2)-mannitol, produced by the transglycosylation of mannitol with maltotriose by Bacillus stearothermophilus maltogenic amylase, was not found to support lactic acid production by Streptococcus sobrinus NRRL 14555. Furthermore, the synthesis of water-insoluble glucans from maltosyl-mannitol by S. sobrinus NRRL 14555 was much lower than that from xylitol or mannitol. Consequently, these results suggest that maltosyl-mannitol could be used as a noncariogenic sugar substitute in food products.

오배자(Rhus japonica Linne) Methanol 추출물의 황산화효과 (Antioxidative Effectiveness of Methanol Extract in Galla Rhois)

  • Free-, soluble- and insoluble phenolic acids were extracted from defatted Galla Rhois. The extracts were then dissolved in equal amounts of an soybean oil, and POV (peroxide value) of the resulting substrates, portion of the soybean oil (control) and 0.02% BHT were measured by AOM (active oxygen method) test at 97.8$^{\circ}C$ for 40 hours through Rancimat method. Induction period of control, BHT, free phenolic acids, soluble phenolic acids and insoluble phenolic acids by the Rancimat method were 4.8, 10.5, 23.9 and 30.5hr. The phenolic acids separated and tentati-vely identified by gas chromatography were catechol, gallic acid, vanillin, protocatechuic acid, syri-ngic acid, ferulic acid.

A Novel Drug Delivery System Design for Meloxicam

  • A drug delivery system(DDS) for practically insoluble meloxicam was developed and evaluated by dissolution study. A novel DDS is two layered system, where the first layer is consisted of gas-forming agent for an immediate release and the second layer is composed of metolose SR(HPMC) for sustained release. This bilayered tablets were manufactured by using manual single punch machine. The results of dissolution study showed an initial burst release followed by sustained release for the experimental period time. From a pharmaceutical point of view, the designed DDS for meloxicam would be informative system in terms of poorly soluble analgesic medicines.

Experimental Studies on Wet Scavenging of Atmospheric Aerosols by Rain Drops

  • Wet scavenging by rain drops is a most important removal process of air pollutants. In order to study the scavenging mechanisms of aerosol particles, the characteristics of chemical components in the rain water were examined as a function of the amount of rainfall. Rain water were collected continuously and separated into the soluble and insoluble components. The elemental concentrations in both components were determined by a PIXE analysis. The physical and chemical characteristics of atmospheric aerosols during the rainfall events were measured simultaneously. The elemental concentrations in rain water decreased substantially just after rain started and then gradually declined in subsequential rain fall exceeding 1.0 mm. The large particles were scavenged more easily than the fine particles. Fe, Ti and Si in rain water were in high insoluble state. Contrarily, almost whole of S was dissolved in rain water.

  • Porcine transforming growth factor-$\beta$1 (TGF-$\beta$1) was expressed in Escherichia coli using cDNA of TGF-$\beta$1 and glutathione S-transferase (GST) fusion vector pGEX-1$\lambda$T. An ApoI-Tth111I fragment of cDNA which correspond to the amino acid residues from 123 to 390 of the precursor TGF-$\beta$1 was inserted into EcoRI-Tth111I digested pGEM#-l$\lambda$T and the recombined plasmid was named pGET-12. Gene products from the cloned regions of the recombinant plasmids pGET-12 was not detected in soluble fraction of cell free extract but detected in insoluble fraction. The solubilization of insoluble gene product was achieved by the treatment of N-laurylsarcosine. Molecular weight of partially purified proteins determined by electrophoresis was same as expected from cloned fragment. The ELISA test results of the purified proteins showed that immunologically detectable epitope was preserved in recombinant protein.

  • [ ${\beta}-1,3-Glucan$ ] (curdlan) is a water-insoluble polysaccharide composed exclusively of ${\beta}-1,3\;linked$ glucose residues. Extracellular curdlan was mostly synthesized by Agrobacterium species and Alcaligenes faecalis under nitrogen-limiting conditions. In this study, we screened the microorganisms capable of producing extracellular curdlan from soil samples. For the first time, we reported Gram-positive bacterium Bacillus sp. SNC 107 capable of producing extracellular curdlan in appreciable amounts. The effect of different carbon sources on curdlan production was studied and found that the yield of curdlan was more when glucose was used as carbon source. It was also found that maximum production was achieved when the initial concentration of ammonium and phosphate in the medium was 0.5 and 1.9 g/L respectively. In this study the curdlan production was increased from 3 to 7g/L in shake flask cultures.

냉동 저장 중 농축 딸기 펄프의 품질 변화 (Qualify Changes of Concentrated Strawberry Pulp during Frozen Storage)

  • Strawberry pulp was separated into serum and insoluble pulp by centrifugation and the serum was concentrated in vacuo to five folds at 55~58$^{\circ}C$ and 30~60mmHg. Concentrated strawberry pulp was prepared by mixing of concentrated serum and insoluble pulp. To evaluate the quality changes of straw-berry pulp, color, physicochemical and sensory properties were analyzed after concentration and during 18 weeks storage at -18'2. The results obtained from the study were as follows : during 18 weeks storage at - l8$^{\circ}C$, soluble solid, browning, reducing sugar content were increased and total anthocyanin, vitamin C, apparent viscosity were decreased. The result of sensory evaluation indicated that texture of concentrated pulp was inferior to that of control but flavor, color intensity and overall acceptance were not different from each other.

  • Phellinus linteus (Hymenocaetaceae) has been used for the treatment of gastric cancer, noninsulin dependant diabetes, diarrhea, and menstrual irregularity. The antioxidative effect of cultivated fruit body of Phellinus linteus on low density lipoprotein oxidation was investigated. The MeOH and water extracts were examined by TEARS assay on human plasma LDL in vitro system. The MeOH ex. showed antioxidative activity, and then was fractionated into Precipitates, Water Insoluble fr., Water fro and ether fro The results showed that the lipophilic fractions, Water Insoluble fr. and Ether fr. of cu 1-tivated fruit body of Phellinus linteus, inhibited the oxidative modification of LDL.

Changes in the Mineral Components in Cheese Juice (Aqueous Phase of Cheese) during Ripening

  • Four different types of cheeses were measure detailed changes in the mineral concentrations of cheese-serum during ripening. Concentrations of minerals in cheese juice were measured. The pH value using the low pH method (LPM) cheese was significantly (p<0.05) lower than that of other cheeses. Similarly the total Ca, S, Mg, and P contents of LPM cheese were significantly lower in than those of other cheeses. Ca, S, Mg, and P remained in colloidal form, while other minerals were mostly in soluble forms after 1 day. The minerals associated with the structure of cheese (i.e., casein or colloidal calcium phosphate) remained largely insoluble even after 1 month of ripening.

Modification of Acetobacter xylinum Bacterial Cellulose Using Dextransucrase and Alternansucrase

  • In addition to catalyzing the synthesis of glucan from sucrose as a primary reaction, glucansucrase also catalyzes the transfer of glucose from sucrose to other carbohydrates that are present or are added to the reaction digest. Using dextransucrase and altemansucrase, prepared from Leuconostoc mesenteroides B-742CBM and B-1355C, respectively, we modified the bacterial cellulose in Acetobacter xylinum ATCC10821 culture, and then produced a characteristic cellulose that is soluble and has a new structure. There were also some partially modified insoluble cellulose and oligosaccharides in the modification culture. After methylation and following acid hydrolysis of both the soluble and insoluble glucans, there were ($1{\rightarrow}4$) as well as ($1{\rightarrow}6$) and ($1{\rightarrow}3$) glycosidic linkages in the soluble glucan.
