북방수염하늘소(딱정벌레목: 하늘소과)의 성적 성숙에 따른 포획 효율의 변화 (Changes in Catch Rate of Monochamus saltuarius (Coleoptera: Cerambycidae) Relation to Sexual Maturation)

  • 본 연구는 북방수염하늘소 성충이 집합-성 페로몬 트랩에 유인되는 시기를 명확하게 규명하기 위해 수행하였다. 또한 집합-성 페로몬에 유인된 개체의 난황 성숙도를 조사하여 집합-성 페로몬 트랩에 유인되지 않은 개체와 비교하였다. 이를 위해 홍릉시험림 내 망실 안에 유인제인 집합-성 페로몬을 매단 다중깔때기트랩과 집합-성 페로몬을 매달지 않은 다중깔때기 트랩을 각각 1개씩 배치하고, 우화망실에서 채집한 북방수염하늘소 성충을 우화시기별(우화 직후, 후식 1일차, 후식 7일차, 후식 10일 경과 후 교미)로 그룹화한 후 트랩이 배치된 망실 안에 방사하여 트랩에 포획 여부를 조사하였다. 각각의 시기별로 80개체(20개체씩 4반복)를 대상으로 실험하여 총 320개체를 망실 내에 방사하였다. 조사 결과, 우화 후 7일 동안 섭식한 개체들의 유인 포획률이 가장 높았으며, 교미 후에는 유인 포획률이 급격히 낮아지는 경향이었다. 또한 집합-성 페로몬이 있는 경우에만 후식 7일차 그룹의 유인 포획률만 통계적으로 유의미한 차이를 보였다. 성별에 따른 유인 포획률의 경우, 암컷이 수컷에 비해 많이 포획되긴 하였으나, 암컷과 수컷 간의 유인 포획률 차이는 없는 것으로 확인되었다. 우화 직후와 후식 1일차에는 모든 개체에서 난황이 미성숙한 상태였으나, 후식 7일차와 교미 후에는 모든 개체에서 성숙한 난황을 확인할 수 있었다. 따라서 페로몬트랩은 우화 후 초기에 후식을 통한 소나무재선충 전파가 가능하므로 소나무재선충병의 확산 억제를 막기에는 제한적일 가능성이 있다. 반면, 성적으로 성숙한 성충을 포획하고, 야외 발생을 조사하는 등으로는 활용 가능성이 있을 것이다.

Current Status of Phytoplasmas and their Related Diseases in Korea

  • Phytoplasmas have been associated with more than 46 plant species in Korea. Several vegetables, ornamentals, fruit trees and other crop species are affected by phytoplasma diseases. Six 16Sr groups of phytoplasmas have been identified and these phytoplasmas are associated with 63 phytoplasma diseases. Aster yellows phytoplasmas are the most prevalent group and has been associated with more than 25 diseases in Korea. Jujube witches' broom, paulownia witches' broom and mulberry dwarf diseases cause economic losses to host trees throughout the country. So far, Korean phytoplasmas belong to six species of 'Candidatus Phytoplasma'; 'Ca. P. asteris', 'Ca. P. pruni$^*$', 'Ca. P. ziziphi', 'Ca. P. trifolii', 'Ca. P. solani$^*$' and 'Ca. P. castaneae'. The diseases are distributed throughout the country and most of them were observed in Gyeongbuk and Chonbuk provinces. At least four insect vectors; Cyrtopeltis tenuis, Hishimonus sellatus, Macrosteles striifrons and Ophiola flavopicta have been identified for phytoplasma transmission.

  • 꼬리명주나비의 혈구 분화 경로를 투과전자현미경을 이용하여 관찰하였다. 혈구의 분화는 조혈기관의 loose islet내에서 진행되었는데, 간세포로부터 원시혈구, 부정형혈구, 과립혈구, 소구혈구가 독자적 경로를 통해 분화되었으며 편도혈구의 분화양상은 관팔되지 않았다. Brehein과 Zachary(1986)의 기준에 따라 조혈기관 주변의 미성숙한 혈구를 관찰한 결과 원시혈구, 부정형혈구, 과립혈구, 소구혈구, 편도혈구로 구분되었다.

Ultrastructural Changes in Midgut of CPV infected Tropical Tasar Silkworm, Antheraea mylitta (D) (Lepidoptera : Saturniidae)

  • The tropical tasar silkworms, Antheraea mylitta (D) produce famous silk 'Kosa' in central part of India. Due to outdoor rearing it became susceptible to viral infection including cytoplasmic polyhedrosis virus (CPV). The common mode of entry of cytoplasmic polyhedrosis virus is per os and cause gresserie disease to the larvae. Histopathological studies elucidated the insect CPV virus produces infective polyhedral inclusion bodies (PIBs) in the midgut cell cytoplasm of virus infected fifth instar larvae. The PIBs multiply enormously in the cytoplasm without invading the nucleus. Ultrastructural studies confirmed the pathological effects of CPV on in midgut cell cytoplasm. The multiplication of polyhedral inclusion bodies took place into the vacuoles and form virogenic stromata in the cytoplasm of cells. However, the encapsulations of polyhedral inclusion bodies into the polyhedrin protein occurred and polyhedra were released into the lumen. At the late stage of infection, cells showed the regressed cytoplasmic organelles with large vacuoles and elongated mitochondria. Hence, the horizontal transmission of CPV causing the midgut cells disintegration in the tasar silkworm, Antheraea mylitta (D) confirmed during infection.

Suppression of Rice Stripe Virus Replication in Laodelphax striatellus Using Vector Insect-Derived Double-Stranded RNAs

  • RNA interference (RNAi) has attracted attention as a promising approach to control plant viruses in their insect vectors. In the present study, to suppress replication of the rice stripe virus (RSV) in its vector, Laodelphax striatellus, using RNAi, dsRNAs against L. striatellus genes that are strongly upregulated upon RSV infection were delivered through a rice leaf-mediated method. RNAi-based silencing of peroxiredoxin, cathepsin B, and cytochrome P450 resulted in significant down regulation of the NS3 gene of RSV, achieving a transcriptional reduction greater than 73.6% at a concentration of 100 ng/μl and, possibly compromising viral replication. L. striatellus genes might play crucial roles in the transmission of RSV; transcriptional silencing of these genes could suppress viral replication in L. striatellus. These results suggest effective RNAi-based approaches for controlling RSV and provide insight into RSV-L. striatellus interactions.

젖소 목장에서 분리된 황색포도상구균의 아형 분포와 특성 (Distribution and characteristics of Staphylococcus aureus subtypes isolated from dairy herds)

  • Staphylococcus aureus is one of most prevalent intramammary pathogens and have characteristics which are not easily eradicated. Recently, to understand the sources and transmission of S aureus, many studies have focused on the subtyping of field isolate. This study was preformed to investigate the distribution pattern and characteristics of the isolates using phenotyping and genotyping. Samples were collected from milk of each udder, cow bodies (perianal region, vagina, tail, udder skin, sole) and environment (floor, liner, milker's hands, water, towel, insect) from 6 herds located in Kyung-gi province. Forty five strains of S aureus were isolated from 3 dairy herds (A, B, C) and were typed by hemolytic pattern, antibiotic resistant pattern, enterotoxin typing and PCR-based DNA fingerprinting. Slime productivity was also compared by each subtype to examine potential infectiousness. Of 45 strains, 41 were isolated from milk samples and 4 were isolated from liners. No strains isolated in the bodies and environment. Forty five strains isolated were classified as 18 subtypes by phenotyping and genotyping. There was common subtype between A and B herd, but the subtype of C herd showed different pattern. Among predominant subtypes, 60% of S aureus strain isolated from A and B herd showed subtype I and 50% of S aureus strain isolated from C herd belonged to subtype VI and X II. Neither somatic cell count (SCC) nor slime production was significantly different between predominant and minor subtypes. In summary, the study revealed that liners play more important roles in the mode of transmission than environmental sources. Several subtypes can be found in a herd, only a few subtype, however, was largely associated with the majority of infection.

Amplified fragment length polymorphism analysis and genetic variation of the pinewood nematode Bursaphelenchus xylophilus in South Korea

  • The pinewood nematode Bursaphelenchus xylophilus causes pine wilt disease and is a serious economic concern for the forest industry of South Korea. To achieve effective control with limited resources, it is necessary to clarify the transmission routes and mechanisms of dispersal of this organism. Highly polymorphic and easy-to-use molecular markers can be used for investigating this aspect. In this study, we evaluated the usefulness of amplified fragment length polymorphisms (AFLPs) for investigating the genetic variations of B. xylophilus and related individuals from China, Japan, and South Korea. The AFLP patterns obtained in our study were similar to the microsatellite patterns reported in a previous study; our AFLP patterns indicated high genetic variability and cryptic genetic structure, but did not indicate any peculiar geographic structure. Moreover, the genetic distances between individuals suggested that the Korean population was affected to a greater extent by the Chinese population than the Japanese population. Further, the gene flow among the related species appeared to be limited; however, there may be also the possibility of genetic introgression among species. These results confirm the usefulness of AFLPs for understanding the epidemiology of pine wilt disease, thereby contributing to the effective control of this disease.

Role of Metcalfa pruinosa as a Vector for Pseudomonas syringae pv. actinidiae

  • After 20 years of steady increase, kiwifruit industry faced a severe arrest due to the pandemic spread of the bacterial canker, caused by Pseudomonas syringae pv. actinidiae (Psa). The bacterium penetrates the host plant primarily via natural openings or wounds, and its spread is mainly mediated by atmospheric events and cultural activities. Since the role of sucking insects as vectors of bacterial pathogens is widely documented, we investigated the ability of Metcalfa pruinosa Say (1830), one of the most common kiwifruit pests, to transmit Psa to healthy plants in laboratory conditions. Psa could be isolated both from insects feeding over experimentally inoculated plants, and from insects captured in Psa-infected orchards. Furthermore, insects were able to transmit Psa from experimentally inoculated plants to healthy ones. In conclusion, the control of M. pruinosa is recommended in the framework of protection strategies against Psa.

RNA Interference in Infectious Tropical Diseases

  • Introduction of double-stranded RNA (dsRNA) into some cells or organisms results in degradation of its homologous mRNA, a process called RNA interference (RNAi). The dsRNAs are processed into short interfering RNAs (siRNAs) that subsequently bind to the RNA-induced silencing complex (RISC), causing degradation of target mRNAs. Because of this sequence-specific ability to silence target genes, RNAi has been extensively used to study gene functions and has the potential to control disease pathogens or vectors. With this promise of RNAi to control pathogens and vectors, this paper reviews the current status of RNAi in protozoans, animal parasitic helminths and disease-transmitting vectors, such as insects. Many pathogens and vectors cause severe parasitic diseases in tropical regions and it is difficult to control once the host has been invaded. Intracellularly, RNAi can be highly effective in impeding parasitic development and proliferation within the host. To fully realize its potential as a means to control tropical diseases, appropriate delivery methods for RNAi should be developed, and possible off-target effects should be minimized for specific gene suppression. RNAi can also be utilized to reduce vector competence to interfere with disease transmission, as genes critical for pathogenesis of tropical diseases are knockdowned via RNAi.

메뚜기 (Euprepocnemis shirakii) 성충의 혈구유형 (Hemocyte Types in Adult Grasshpper, Euprepocnemis shirakii Bolivar)

  • Six types of hemocytes were identified in adult grasshopper, Euprepocnemis shirakii Bolivar. The morphological and ultrastructural characteristics of these cells were characterized by phase contrast, scanning and transmission electron microscope. The prohemocytes are small and spherical cells with large nucleus. The plasmatocytes are spindle shaped or polymorphic cells which show numerous cytoplasmic processes on the cell surface and they have lysosomes and vesicles that may be involved in phagocytic function. Especially, multivesicular bodies are observed in the polymorphic cells. The granulocytes I are spherical shaped cells. They are characterized by a number of electron dense granules measuring $0.2-0.3{\mu}m$ in average diameter and marginal band of microtubules which are always in close proximity to the cell membrane. The granulocytes II are oval or spindle-shaped cells. They contain large electron dense granules measuring $0.5-0.8{\mu}m$ in average diameter. Their cytoplasm is filled with numerous granules. The spherulocytes contain large amounts of spherules that most of their cytoplasm. Spherules filled with fine granules or flocculent materials. The oenocytoids are large spherical cells with few cytoplasmic organelles. Their cytoplasm contains peculiar aspect of motochondria and numerous polyrobosome.
