Evaluation of Chemical Composition and In vitro Digestibility of Appennine Pasture Plants Using Yak (Bos grunniens) Rumen Fluid or Faecal Extract as Inoculum Source

  • Pastures of the Apennines of Central Italy contribute to feed resources of high altitude (above 1,300 m sea level) grazing systems. The objective of this study was to evaluate the effectiveness of faecal extract from the yak (Bos grunnienes) as an alternative microbial inoculum to rumen fluid for estimation of digestibility of several forage species. Forage samples produced at high altitude were tested in this study: four legumes (Lathyrus sativus L., Lotus corniculatus L., Onobrychis viciaefolia L. and Trifolium pratense L.), three forbs (Achillea millefolium L., Potentilla reptans L. and Teucrium flavum L.) and one grass (Brachipodyum pinnatum L.) were incubated with yak rumen fluid or faecal extract. A large variability in chemical composition was observed among the species collected. Rumen liquor and faecal samples were collected from adult healthy yak. The $Daisy^{II}$ incubator was used to evaluate the nutrient digestibility of forages using rumen liquor as control and faecal extract as alternative microbial inoculum sources. Filter bags containing samples of browse species were added to the four digestion vessels along with their respective inoculum and then incubated for 48 h and dry matter (DM), organic matter (OM), crude protein (CP), neutral and detergent fiber (NDF) digestibility was determined. There was a significant relationship between estimates, indicating that faecal liquor has the potential to be used instead of rumen fluid for estimation of in vitro digestibility of plants. It is concluded that the $Daisy^{II}$ incubator results are appropriate for the determination of in vitro digestibility of nutrients using faecal liquor to define the potential for adaptation of yak to new pastures.

  • Number and percentage of diseased area of leaf blast lesions formed on different leaf location were mostly distributed from the flag leaf(n-1) to the 3rd leaf from the top(n-3) in Tongil line rice varieties and on the 2nd leaf from the top(n-2) in Japonica type rice varieties. Especially leaf lesions of Nopung which was more susceptible to leaf blast among Ton1 line rice varieties were mostly distributed on flag leaf. Relation between the degree of lesion distribution and level of fertilizer was more clear with an increase of fertilizer quantity. Leaf blast lesions of rice varieties were generally distributed from the flag leaf to the with leaf from the top but mainly those at flag leaf and the 2nd leaf from the top were found to be most responsible for inoculum source of panicle blast after booting stage. Increase of the conidia formation was resulted from fluctuation of temperature$(24^{\circ}C\~16^{\circ}C)$ in low temperature range after booting stage and many inoculum sources were supplied on panicles until the end of September without impeding dispersal from leaf blast lesions as an inoculum source of panicle blast.

Establishment of suspension culture condition for embryogenic callus proliferation and somatic embryo development of Kalopanax septemlobus (음나무 배발생 캘러스의 증식 및 체세포배 발달을 위한 액체 현탁 배양조건 확립)

  • This study was conducted to establish the optimal suspension culture system for both the propagation of embryogenic cells (ECs) and the induction of somatic embryos (SEs) of Kalopanax septemlobus. The proliferation rate of ECs was reduced as the inoculum density was increased; the highest rate was obtained when 0.1 g/100 ml of cells was initially inoculated. According to the analysis of cell growth pattern and cell growth cycle (G1, Sand G2/M), the cell growth started in 5 days culture initiation, grew rapidly until 15 days and then decreased gradually. Distinctive changes of the cell growth cycle by the culture periods was also observed; the growth cycle was doubled from initial 5.6% to 11.7% of S stage in 5 days culture and then reached in stable stages again. Therefore, the results indicated that a 15-day-cycle was the optimal culture period for the propagation of the ECs through the suspension culture. Furthermore, the cell inoculum density was also important for the induction of SE; more than 65% of SEs at the torpedo stage was induced by using the low level of cell inoculum (0.5 g/L), while the higher inoculum densities were rapidly reduced the proportion of SEs at that stage. Although the higher inoculum density delayed the development of SE, it did not affect the proportion of SEs at the globular and heart stage. In conclusion, this study showed that the suspension culture of the Kalopanax septemlobus ECs through the control of inoculum density was an efficient way for both the propagation of ECs and the induction of SEs, suggesting that the development of this system might help to reduce the culture period for the somatic embryo production.

The Effect of Bacteriocin Produced by Lactobacillus plantarum on the Growth of Listeria monocytogenes

  • The inhibitory effect of Lactobacillus plantarum (Lb. plantarum) which is bacteriocin­producing strain against the growth of Listeria monocytogenes (L. monocytogenes) was examined in trypticase soy broth (TSB). TSB was inoculated with 104 cells/me L. monocytogenes and then with different numbers $(10^6\;10^4\;and\;10^2\;cells/ml)$ of Lb. plantarum. The mixed cultures were incubated at 37, 25 and $4^{\circ}C$. The most effective inhibition of was found at $37^{\circ}C$ and a less inhibition at $25^{\circ}C$. However, there was no significant change in the cell numbers of both L. monocytogenes and Lb. plantarum at $4^{\circ}C$. At same incubation temperature, the higher initial inoculum level of Lb. plantarum, the better inhibitory effect against L. monocytogenes. In addition, TSB was inoculated with L. monocytogenes at different initial inoculum levels of $10^6,\;10^4$ and $10^2$ cells/me and then supplemented with 0, 30, 60 and 100 AU/ml of bacteriocin produced by Lb. plantarum. The mixed cultures were incubated at 37, 25 and $4^{\circ}C$. L. monocytogenes of three different initial inoculum levels began to be inhibited in the presence of more than 60 AU/ml of bacteriocin at $37^{\circ}C$. In TSB containing more than 60 AU/me of bacteriocin and incubated at $25^{\circ}C$, L. monocytogenes decreased by 2 log-units during the period of 12 hrs incubation and thereafter remained steady. At $4^{\circ}C$, L. monocytogenes decreased by 1.5 log-units in the presence of 60 AU/ml bacteriocin during the period of 4 days incubation and dropped to the non-detectable level in TSB with 100 AU/ml bacteriocin.

Optimization of Mass cultivation Media for the Production of Biomass and Natural Colourants from Two Marine Cyanobacteria by a Mathematical Design of Experiments

  • Optimization of chemicals in the large scale sea water medium and inoculum for biomass and natural colourants production in the marine cyanobacteria, Phomidium tenue BDU 46241 (phycoerythrin producer) and P.valderianum BDU 30501 (phycocyanin producer) was carried out by experiments in L8 orthogonal array. Mathematical analysis revealed the significance of these factors. The factor(s) that critically control the yield varied with the organism and the end-product further, the desirable level of these factors between the normal and a higher level tested was identified and improved media were evolved. In both cyanobacteria, higher level of $K_2$$HPO_4$, $NaNO_3$ and inoculum with normal level of ferric ammonium citrate was found to be desirable for biomass production and additionally, higher level of $MgSO_4$ for pigment production. The level of other factors varied with the organism and the end-product. Confirmation experiments showed that the clues obtained based on mathematical experimentation are valid. In P.tenue, the medium optimized for biomass production increased the yield of biomass by 495% and the medium optimized for phycoerythrin production increased the yield of biomass by 408% with 30% increase in phycoerythrin content of the biomass. Similarly in P.valderianum, the medium optimized for biomass production increased the yield of biomass by 224% and the medium optimized for phycocyanin production increased the yield of biomass by 143% with 44% increase in phycocyanin content of the biomass.

Aflatoxin Production by Aspergillus parasiticus NRRL 2999 in Various Varieties of Soybeans (Aspergillus parasiticus에 의한 대두품종별(大豆品種別) Aflatoxin 생성)

  • Aflatoxin production by Aspergillus parasiticus NRRL 2999 with two different levels of inoculum sizes ($10^2$ and $10^6$ spores) was studied on such varieties of soybeans produced in Korea as Bongeui, Hwang Keum, Jangbaek, Danyeop, Jangyeop and Namcheon. With the inoculum size of $10^2$ spores/10 g of soybeans, there were statistically difference (p<0.05) on the production of aflatoxins in the varieties of soybeans. The variety of Namcheon produced 7 times higher amount of aflatoxin $B_1$ than that of Bongeui. The variety of Jangyeop produced the highest level of aflatoxin $G_1$ among the varieties employed. Namcheon and Jangyeop were the varieties which supported high amounts of aflatoxins. Much higher levels of aflatoxins were obtained on the soybeans than on rice in general with the inoculum size of $10^2$ spores. With the inoculum size of $10^6$ spores/10 g of soybeans, significantly lower levels of aflatoxins were synthesized by the mold than those with $10^2$ spores. The variety of Namcheon resulted in the lowest production of aflatoxin $B_1$ while Hwangkeum was the variety that produced highest amount of aflatoxin $B_1$. There were no significant differences within the varieties on the production of aflatoxin $G_1$ by the mold except the variety of Hwangkeum. Lower quantities of aflatoxin $B_1$ were produced by the mold en the soybeans than on rice with the inoculum size of $10^6$ spores.

A Mechanism of Density-Dependent Population Change in Heterodera glycines (콩시스트선충의 밀도변화 기작)

  • Penetration level, female development and histological changes in infected root tissues were investigated following inoculation with different inoculum levels (110,440 and 1760 juveniles/plant) of Heterodera glycines (SCN) race 3 on susceptible 'Lee' and resistant 'Pickett' soybean cultivars. Penetration level was lower in Pickett at the higher inoculum levels but no differences were detected in Lee. However, the lower penetration level in the resistant soybean cultivar appeared not to be directly related to plant resistance (female maturation). The number of females recovered from Lee was lower at the highest inoculum level. The number of females maturing on Pickett was much less than that on Lee, showing that changes of SCN population is associated with the number of SCN maturing rather than nematode penetration. In Lee mono-infection sites (a single nematode per site) syncytia had dense cytoplasm and no central vacuoles. while multi-infected sites (many overlapping nematodes per site) had syncytia with a large central vacuole and many small vacuoles. Resistant responses in mono-and multi-infected root tissues of Pickett were delayed and rapid necrosis, respectively. The differences in tissue response are suggested as a mechanism that controls density-dependent population changes in resistant and/or susceptible soybean cultivars.

Screening Procedure of Tobacco Cultivars for Resistant to Bacterial Wilt Caused by Ralstonia solanacearum (담배세균성마름병[립고병(立枯病)]에 대한 담배품종의 저항성 검정법)

  • Bacterial wilt caused by Ralstonia solanacearum has become a severe problem on tobacco in Korea. No effective single control measure is available at present time. One of the most potential way for controlling the bacterial wilt on tobacco is growing tobacco cultivars resistant to the bacterial wilt. In this study, optimal conditions for screening tobacco cultivars resistant to the bacterial wilt were examined to provide reproducible and efficient methods in growth chamber testing and field experiments for evaluating plant disease resistance. For this, already-known inoculation methods, inoculum densities, and incubation temperature, and plant growth stages at the time of inoculation were compared using tobacco cultivars resistant (Nicotiana tabacum cv, NC95), moderately resistant (N. tabacum cv. SPG70), and susceptible (N. tabacum BY4) to the bacterial disease. It was determined that root-dipping of tobacco seedlings at six true leaf stage into the bacterial suspension with inoculum level of $10^8$ colony-forming units (CFU)/ml for 20 min before transplanting was simple and most efficient in testing for resistance to the bacterial wilt of tobacco caused by R. solanacearum, for which disease incidences and severities were examined at 2 weeks of plant growth after inoculation at $20{\sim}25^{\circ}C$ in a growth chamber. These experimental conditions could discriminate one tobacco cultivar from the others by disease severity better than any other experimental conditions. In field testing, the optimum time for examining the disease occurrence was late June through early July. These results can be applied to establishing a technical manual for the screening of resistant tobacco cultivars against the bacterial wilt caused by R. solanacearum.

Production of Biomass and Bioactive Compounds from Cell Suspension Cultures of Eurycoma longifolia in Balloon Type Bubble Bioreactors

  • Eurycoma longifolia is an important rare medicinal plant that contains valuable bioactive compounds. In the present study, cell suspension culture of E. longifolia was established for the production of biomass and phenolic compounds. Various medium parameters, such as concentration of auxin, salt strength of the medium, and sucrose and nitrogen concentrations, were optimized for the production of biomass at the flask-scale level. Full strength Murashige and Skoog (MS) medium supplemented with $3.0mg{\cdot}L^{-1}$ naphthaleneacetic acid (NAA), 3% (w/v) sucrose, 0:60 $NH{_4}^+:NO{_3}^-$ was found suitable for biomass accumulation. Based on the optimized flask-scale parameters, cell suspension cultures were established in balloon-type bubble bioreactors, and bioprocess parameters such as inoculum density and aeration rate were optimized. Inoculum density of $50g{\cdot}L^{-1}$ and increasing aeration rate from 0.05 to 0.3 vvm, with increases every 7 days, were suitable for the accumulation of both biomass and phenolic compounds. With the optimized conditions, $14.70g{\cdot}L^{-1}$ dry biomass, $10.33mg{\cdot}g^{-1}$ DW of phenolics and $3.89mg{\cdot}g^{-1}$ DW of flavonoids could be achieved. Phenolics isolated from the cell biomass showed optimal free radical scavenging activity.

A Detail Investigation of Major Diseases Occurrence and Pathogen Population on Turfgrass Cultivation in Nationwide (국내 잔디 재배지 주요 병해 발생 및 병원균 밀도 조사)

  • We investigated turfgrass diseases and inoculum density at nationwide turfgrass cultivation sites in year of 2013. Occurrences of large patch and rust disease appeared in September. Brown patch recorded in September to October at Namhea and Pythium blight disease occurred outbreaks in early July at Namhea site. Some sites in Namhea damaged 3% area of total cultivation field by Sclerotinia homoeocarp. In Daepyeong (Gyeongnam), Fairy ring and large patch were recorded. Severe takeall and fairy ring have been observed in Gochang-si. Multi-site in Cheongju-si, brown patch was observed in pandemic level. Interesting enough, a cool-season turfgrass cultivate sites in Pyeongtaek-si brown patch, leaf blast, summer patch, and Curvularia leaf spot were investigated during the surveys period. Inoculum densities (Rhizoctonia spp.) at turfgrass cultivations sites were increased as cumulatively in all survey sites. The investigation result indicated that the disease occurrence and pathogens are similar as diseases in golf courses.
