Kinetic Characterization of an Iron-sulfur Containing Enzyme, L-serine Dehydratase from Mycobacterium tuberculosis H37Rv (Mycobacterium tuberculosis H37Rv로부터 유래된 철-황 함유 효소인 L-세린 탈수화효소의 동력학적 특성)

  • L-Serine dehydratase (LSD) is an iron-sulfur containing enzyme that catalyzes the conversion of L-serine to pyruvate and ammonia. Among the bacterial amino acid dehydratases, it appears that only the L-serine specific enzymes utilize an iron-sulfur cluster at their catalytic site. Moreover, bacterial LSDs are classified into four types based on structural characteristics and domain arrangement. To date, only the LSD enzymes from a few bacterial strains have been studied, but more detailed investigations are required to understand the catalytic mechanism of various bacterial LSDs. In this study, LSD type II from Mycobacterium tuberculosis (MtLSD) H37Rv was expressed and purified to elucidate the biochemical and catalytic properties using the enzyme kinetic method. The L-serine saturation curve of MtLSD exhibited a typically sigmoid character, indicating an allosteric cooperativity. The values of $K_m$ and $k_{cat}$ were estimated to be $59.35{\pm}1.23mM$ and $18.12{\pm}0.20s^{-1}$, respectively. Moreover, the plot of initial velocity versus D-serine concentration at fixed L-serine concentrations showed a non-linear hyperbola decay shape and exhibited a competitive inhibition for D-serine with an apparent $K_i$ value of $30.46{\pm}5.93mM$ and with no change in the $k_{cat}$ value. These results provide insightful biochemical information regarding the catalytic properties and the substrate specificity of MtLSD.

Clinical Evaluation of Abdominal Stab Wound Patients in the ED (복부 자상환자의 처치에 대한 응급의학에서의 고찰)

  • Purpose: In Korea, most abdominal penetrating wounds are caused by stab wounds rather than gun-shot wounds. However, not many studies have been performed on stab injuries and their epidemiologic characteristics. Disagreements of opinions of obligatory surgical exploration and conservative treatment exist, and this subject is still being debated. Therefore, the authors studied the epidemiologic characteristics of abdominal stab wound patients visiting the emergency department and reviewed the proportion of patients that received nontherapeutic surgery and conservative treatment. Methods: This study included patients visiting the emergency department with abdominal stab wounds. A retrospective chart review was done on the abdominal stab wound patients. Sex, age, cause of injury, location of wound, initial vital signs, operation results, injured organs and CT & LWE results were reviewed. Results: The median age of the 121 patients was 40.9 years, of these patients, 88 were males, of which 52 (43.0%) were drunken. Of the patients that received non-therapeutic operations, only 3 patients (15.0%) were drunken, significantly lower than the therapeutic operation group. For the location of the wound, most patients were injured in the right and left upper quadrants, 27 patients each. The most common injured organ was the small bowel; 13 patients were injured in the small bowel. With abdominal CT scans and local wound explorations together, the results exhibited a sensitivity of 97%, a specificity of 44%, a positive predictive value of 56%, and a negative predictive value of 95%. Conclusion: In our study, the sensitivity was 97% when CT & LWE were performed together; thus we can conclude that CT and LWE can be used together to select the treatment method. Although in our study, the surgical indications in abdominal stab wound patients is not sufficient, our results showed a higher rate of nontherapeutic surgery compared to previous studies. Therefore, more research is needed to prevent unnecessary laparotomies in hemodynamically-stable patients without symptoms.

The effect of finishing diet supplemented with methionine/lysine and methionine/α-tocopherol on performance, carcass traits and meat quality of Hanwoo steers

  • Objective: This study aimed to compare the effects of diets with and without supplements of methionine/lysine (met/lys) and methionine/α-tocopherol (met/α-tocopherol) on the performance, carcass traits and meat quality of Hanwoo steers. Methods: A total of 21 Hanwoo steers were divided into three different groups. Each group consisted of 7 animals that received different dietary treatments for 120 days as follows: a control (C) diet composed of a basal diet with 74% total digestible nutrient and 12% crude protein; treatment 1 (T1) composed of a basal diet enriched with methionine 69%+lysine 31%; and treatment (T2) composed of a basal diet enriched with methionine 84.65%+α-tocopherol 15.35%. Daily supplementation was given using the top-dressing method (20 g/animal). Results: The met/lys groups yielded a longissimus lumborum with increased water holding capacity (p<0.01) and lower shear force value (p<0.05). Dietary met/lys did not have an adverse effect on the animal performance and carcass traits. Instead, these supplements contributed significantly to the higher protein content compared to the control diet (p<0.05). Myristic acid (C14:0) was the only fatty acid affected by the supplementation. While the met/α-tocopherol group led to significantly higher protein and oxymyoglobin contents during storage (p<0.05). It also contributed significantly to the lower shear force value and 2-thiobarbituric acid reactive substances score during storage (p<0.05) compared to the control and treatment 1 since the initial storage day. The met/α-tocopherol diet also yielded meat with a redder color (p<0.05) after 3 days of storage. However, it did not significantly contribute to the fatty acid profiles of Hanwoo steers. Conclusion: Met/lys supplementation resulted in higher protein scores, water holding capacity and lower shear force scores. While met/α-tocopherol supplementation contributes to the production of redder meat, reduces lipid oxidation, production of more tender meat and increases the content of protein and oxymyoglobin percentage.

  • Introduction: The utility of the C-terminal cross-linking telopeptide test (CTX) as a method for staging Bisphosphonate-related osteonecrosis of the jaws (BRONJ) and its healing process was examined. Materials and Methods: A total 19 patients who were diagnosed with BRONJ underwent a fasted morning CTX test, were enrolled in this study. The serum CTX values ranged from 50 to 630 pg/mL (mean 60). The risk assessment was rated according to the CTX values of the individual patient (minimal risk, ${\geq}$ 150 pg/mL, moderate, 100 to 150 pg/mL, high, ${\leq}$100 pg/mL). The BRONJ scores were then calculated according to the number of BRONJ lesions and their stage. The operation was done as soon as possible, regardless of BORNJ stage. Results: The mean duration of bisphosphonate therapy was 4.1 years. Of the 19 patients, 15, 2 ans 2 received alendronate, risedronate and zoledronate, respecively. Of the 19 patients who underwent a sequestrectomy, saucerization and smoothing, 15 healed after the initial surgery, 1 patient healed after one more surgical procedure, 3 patients did not heal completely but showed improvement in symptoms. Therefore, 17 out of the 19 patients healed completely with complete mucosal coverage and the elimination of pain. The risk assessment using the CTX value and disease severity were not correlated (r=-0.264, P=0.275). In addition, the risk assessment using CTX value and healing after surgery were not correlated (r=-0.147, P=0.547). Conclusion: The serum CTX should be considered carefully by clinicians as part of overall management. Early surgical intervention is of benefit in the treatment of stage II BRONJ.

The study about the cause of the Korean professional pitchers' injury and its classification (한국 프로야구투수들의 부상 발생원인 및 분류에 관한 연구)

  • Objectives: We did research in the cause of the Pitchers' injury and their recovery process to make a detailed injury list for the purpose of finding the cause of the Korean professional pitchers' injury and its classification. We drew the conclusion through the results as following. Methods: We posed a question to the 80 pitchers playing in the first team of the eight Korean professional baseball team and analyzed the 62 pieces of question paper except the paper having a mistake. We used SAS/PC statistical package in analyzing the data. Results: In the frequency of the pitchers' shoulder injury in the last three years, the injured of all the players were 61.3$\%$ and the injury free players were 28.7$\%$. The cause of the injury was 45.2$\%$ wrong pitching motion, which was the highest value. For the shape of a pain when injured, the reverberation ache feeling when he is hit in the weight commanded an absolute majority as 19.4$\%$. Those who had muscular pain were 17.7$\%$, which was felt mostly at the pitching motion. The most trouble name of the injured shoulder was bicepstendinitis as 16.1$\%$ while the injury of shoulder joint was the lowest as 1.6$\%$. As the most widely used treatment, 25.8 percent of all the players had taken an electronical thraphy after injury. 14.5 percent of the players who had an injury to the shoulder told that they have an operation and 85 percent of them didn't. As a sort of the operation, a repairing of labrum was 44.4 percent, which is the highest value and the 77.8 percent pitchers are performing a normal pitching through rehabilitation after the operation and 22.2 percent of them are undergoing rehabilitation training. Conclusion: The research have shown that the main cause of the injury, concerning the Korean professional pitchers throwing lots of ball in both matches and practices, is overuse syndrome, bad mechanism, muscle weakness and instability of balance. I think that the role of trainer, physical therapy, and team physician taking charge of the players' injury must learn physical test method by heart exactly to check up the state of the injury definitely at the initial phase. Moreover, when the cause of the injury part after a close examination is discovered, the scientific and good surgery is essential to the rehabilitation success and making a classification of shoulder instability is useful to make a operation plan as well as the players' rehabilitation, treatment.

Thermal Stability and Critical Ignition Temperature of RPF (RPF의 열적 안정성과 한계발화온도)

  • It is important to understand thermal characteristic as a method to estimate the new materials, because spontaneous ignition characterized by causing combustion in the low temperature without ignition source. If can not find out the thermal characteristics of materials, it is frequent that causes of fires could not be found. The danger level of spontaneous ignition material should be estimated and by closely studying its thermal characteristic. However, RPF(Refuse Paper & Plastic Fuel) is a solid matter and getting increasesa year by year because it is an economy profit as alternative energy for limited fossil fuels. Some time RPF occur a fire in the cases of its production process and conservation. Therefore study for thermal stability and critical ignition temperature of RPF was so imperative that the experiment by means of Bombe Calorimeter, TG-DTA, MS80, SIT-II, and Wire Basket Test was implemented. As a result, RPF had a caloric value 26.4-28.3 MJ/kg, and its initial pyrolysis temperature was $192^{\circ}C$ at heating rate 2 K/min. With the result of analysis by MS 80 which is an instrument measuring microscopic calory, pure RPF not containing water has higher caloric value than RPF containing 20% water. Also, SIT-II which is an instrument of insulated auto-ignition was ignited by $118.5^{\circ}C$. This temperature is lower than that of Wire Basket Test. The critical ignition temperature was calculated by Frank-Kamenetskii equation can cause ignition at $80^{\circ}C$ when conserved in the height of 10 m by the standard of infinity slab.

Preparation of Fe4[Fe(CN)6]3 Coated on Mica or TiO2/Mica for Infrared Reflective Blue Pigments and Isolation-heat Properties of These Paints (Fe4[Fe(CN)6]3가 코팅된 Mica 또는 TiO2/Mica 적외선 반사용 청색안료 제조 및 이 도료의 차열 특성 평가)

  • $Fe_4[Fe(CN)_6]_3$ coated on a mica or $TiO_2$/mica surface as infrared reflective blue pigment was prepared by a hydrothermal method. $Fe_4[Fe(CN)_6]_3$, used as coloring agent, was uniformly coated on mica or $TiO_2$/mica under the optimized condition of a 1.2 : 1 weight ratio between iron(III) chloride hexahydrate and potassium ferrocyanidetrihydrate at the initial pH level of 4.5 at $70^{\circ}C$. The infrared (IR)-reflective pigments were characterized by SEM, Zeta-potenial, FT-IR, and UV-VIS NIR spectrophotometry. Especially the CIE color coordinate and total solar reflectance(TSR) properties of the pigments were investigated in relation to variation of the coating and coated substrate thicknesses. Isolation-heat paint was prepared with 20 wt% blue pigments fully dispersed in acryl-urethane resin and several additives to coat the film uniformly. The films were also measured with CIE color coordinate, TSR, and the surface temperature was recorded by an isolation-heat measuring system. The pigments and films of $Fe_4[Fe(CN)_6]_3$ coated on mica and $TiO_2$/mica showed high TSR values compared with the TSR value of $Fe_4[Fe(CN)_6]_3$ itself. According to the increase of TSR value, the property of isolation-heat is effective. To realize the optimal blue color, we applied the the pigment to $TiO_2$ coated mica(TM(b)) which has blueish interference color. The pigment of $Fe_4[Fe(CN)_6]_3$ coated on TM(b) shows a strong blue color compared with that of $Fe_4[Fe(CN)_6]_3$ coated on $TiO_2$/Mmca(TM(w)), which has a whitish interference color.

EFFECT OF INCREMENTAL FILLING TECHNIQUE ON THE POLYMERIZATION SHRINKAGE OF COMPOSITE RESIN (적층충전법이 복합레진의 중합수축에 미치는 영향)

  • The aim of this study was to investigate the relationship between the C-factor and shrinkage strain values of composite resin and examine the strain values in different incremental filling techniques. The strain gauge method was used for measurement of polymerization shrinkge strain. Experiment was divided two step. In a first experiment, we compared with strain value in three different depth (2mm, 3mm, 4mm) and microhardness of each samples after 24hours were measured. In a second experiment, we examined the strain values in five different filling techniques(Group 1: bulk filling, Group 2: oblique incremental filling, Group 3: horizontal incremental filling, Group 4: vertical incremental filling, Group 5: lining of flowable resin and bulk filling) The results of the present study can be summarized as follows: 1. Composite resin in acrylic molds showed the initial expansion at the early phase of polymerization. 2. Contraction stress was not revealed significant difference between depth of 2mm and 3mm(P>0.05). 3. Contraction stress in sample of 4mm was showed the lowest value(P<0.05). 4. Microhardness of specimen was revealed more difference between upper and lower surface in depth of 4mm than 2 and 3mm(P<0.05). 5. Lining of flowable resin and bulk filling (Group 5) was showed the lowest contraction stress, Group 2 and 3 was showed the highest contraction stress(P<0.05). On the basis above results, the stress that result from the polymerization shrinkage, when incremental curing techniques are used, showed that there is no advantage in incremental placement and curing.

The Development of the Simple SHGC Calculation Method in Case of a Exterior Venetian Blind Using the Simulation (시뮬레이션을 이용한 외부 베네시안 블라인드의 약식 SHGC 계산법 개발)

  • When it comes to these buildings for business use, cooling load during summertime was reported to have great importance which, as a result, impressively increased interest in Solar Heat Gain Coefficient (SHGC). Such SHGC is considered to be lowered with the help of colors and functions of glass itself, internal shading devices, insulation films and others but basically, these external shading devices for initial blocking that would not allow solar heat to come in from outside the buildings are determined to be most effective. Of many different external shading devices, this thesis conducted an analysis on Exterior Venetian Blind. As for vertical shading devices, previous researches already calculated SHGC conveniently using concepts of sky-opening ratios. However in terms of the Venetian Blind, such correlation is not possibly applied. In light of that, in order to extract a valid correlation, this study first introduced a concept called shape factor, which would use the breadth and a space of a shade, before carrying out the analysis. As a consequence, the concept helped this study to find a very similar correlation. Results of the analysis are summarized as follows. (1) Regarding SHGC depending on the surface reflectance of a shade, an average of 2% error is observed and yet, the figure can always be ignored when it comes to a simple calculation. (2) As for SHGC of each bearing, this study noticed deviations of 4% or less and in the end, it is confirmed that extraction can be achieved with no more than one correlation formula. (3) When only the shape factor and nothing else is used for finding a correlation formula, the formula with a deviation of approximately 5% or less is what one would expect. (4) Since the study observed slight differences in bearings depending on ranges of the shape factors, it needed to extract a weighted value of each bearing, and learned that the smaller the shape factor, the wider the range of a weighted value. The study now suggests that a follow-up research to extract a simple calculation formula by dealing with all these various inclined angles of shade, solar radiation conditions of each region (the ratio of diffuse radiation to direct radiation and others) as well as seasonal features should be carried out.

  • In this paper, an automated segmentation algorithm is proposed for MR brain images using T1-weighted, T2-weighted, and PD images complementarily. The proposed segmentation algorithm is composed of 3 step. In the first step, cerebrum images are extracted by putting a cerebrum mask upon the three input images. In the second step, outstanding clusters that represent inner tissues of the cerebrum are chosen among 3-dimensional(3D) clusters. 3D clusters are determined by intersecting densely distributed parts of 2D histogram in the 3D space formed with three optimal scale images. Optimal scale image is made up of applying scale space filtering to each 2D histogram and searching graph structure. Optimal scale image best describes the shape of densely distributed parts of pixels in 2D histogram and searching graph structure. In the final step, cerebrum images are segmented using FCM algorithm with its initial centroid value as the outstanding clusters centroid value. The proposed cluster's centroid accurately. And also can get better segmentation results from the proposed segmentation algorithm with multi spectral analysis than the method of single spectral analysis.
