Isolation of Insoluble Phosphate-Solubilizing Bacteria and Optimum Condition for Solubilization (인광석 가용화 세균의 분리 및 가용화 최적조건)

  • 850 strains of phosphate-solubilizing bacteria were isolated from soil of Chung-nam and Daejeon region using 0.5% calcium phosphate containing medium. The HS-2 strain with the highest rock phosphate-solubilizing activity was selected and identified as Azotobacter sp. HS-2 based on the microbiological characteristics. The optimum culture temperature and initial pH of medium for solubilization of rock phosphate were $30^{\circ}C$ and pH 6.0-7.0, respectively. Addition of oxalic acid(0.5M) into the PDB-rock phosphate medium increased 50% solubilization of rock phosphate.

Effectiveness of continuous hypertonic saline infusion with an automated infusion pump for decompressive neuroplasty: a randomized clinical trial

  • Background: Hypertonic saline (HS) injections for decompressive neuroplasty (DN) can cause pain. We assessed whether a continuous infusion of HS through an infusion pump would reduce injection-related pain compared with repeated bolus administrations. Methods: Fifty patients scheduled for DN were randomized to either the bolus injection or the continuous infusion group. After appropriately placing the epidural catheter, 4 mL of 5% NaCl was injected as four boluses of 1 mL each at 15-minute intervals or infused over 1 hour using an infusion pump. The severity of pain induced by HS injection, as measured by the 11-point numerical rating scale (NRS), was the primary outcome. The severity of low back or lower extremity pain, as measured by the 11-point NRS and Oswestry Disability Index (ODI), 3 months following the procedure, was the secondary outcome. Results: Data from 21 patients in the bolus group and 23 in the continuous infusion group were analyzed. No statistically significant difference in injection-related pain was identified between the two groups during the initial HS administration (P = 0.846). However, there was a statistically significant reduction in injection-related pain in the continuous infusion group compared to the bolus injection group from the second assessment onwards (P = 0.001, < 0.001, and < 0.001, respectively). No significant between-group differences in the NRS and ODI scores 3 months post-procedure were noted (P = 0.614 and 0.949, respectively). Conclusions: Our study suggests that administering HS through a continuous infusion is a useful modality for reducing HS injection-related pain during DN.

Effect of different short-term high ambient temperature on chicken meat quality and ultra-structure

  • Objective: This study investigated the effect of different acute heat stress (HS) levels on chicken meat quality and ultra-structure. Methods: Chickens were randomly divided into 7 groups to receive different HS treatments: i) $36^{\circ}C$ for 1 h, ii) $36^{\circ}C$ for 2 h, iii) $38^{\circ}C$ for 1 h, iv) $38^{\circ}C$ for 2 h, v) $40^{\circ}C$ for 1 h, vi) $40^{\circ}C$ for 2 h, and vii) un-stressed control group ($25^{\circ}C$). Blood cortisol level, breasts initial temperature, color, pH, water holding capacity (WHC), protein solubility and ultra-structure were analyzed. Results: HS temperatures had significant effects on breast meat temperature, lightness ($L^*$), redness ($a^*$), cooking loss and protein solubility (p<0.05). The HS at $36^{\circ}C$ increased $L^*{_{24h}}$ value (p<0.01) and increased the cooking loss (p<0.05), but decreased $a^*{_{24h}}$ value (p<0.05). However, as the temperature increased to $38^{\circ}C$ and $40^{\circ}C$, all the values of $L^*{_{24h}}$, cooking loss and protein denaturation level decreased, and the differences disappeared compared to control group (p>0.05). Only the ultimate $pH_{24h}$ at $40^{\circ}C$ decreased compared to the control group (p<0.01). The pH in $36^{\circ}C$ group declined greater than other heat-stressed group in the first hour postmortem, which contributed breast muscle protein degeneration combining with high body temperature, and these variations reflected on poor meat quality parameters. The muscle fiber integrity level in group $40^{\circ}C$ was much better than those in $36^{\circ}C$ with the denatured position mainly focused on the interval of muscle fibers which probably contributes WHC and light reflection. Conclusion: HS at higher temperature (above $38^{\circ}C$) before slaughter did not always lead to more pale and lower WHC breast meat. Breast meat quality parameters had a regression trend as HS temperature raised from $36^{\circ}C$. The interval of muscle fibers at 24 h postmortem and greater pH decline rate with high body temperature in early postmortem period could be a reasonable explanation for the variation of meat quality parameters.

  • The morphological and cultural characteristics of alkalophilic Coryneform bacteria TU-19 were investigated at various pHs. This bacterium showed normal growth pattern at $pH\;9.0{\sim}10.0$, but the cell growth was completely inhibited at extreme pH (12.0 or more). Interestingly, at pH 8.0 the morphology of the bacterial cells seems to form convoluted filaments during the exponential growth phase while at pH 10.0, the optimal pH for the growth of this organism, the bacteria grew with variable paired or single forms, and straight rods during growth stages. Growing in alkaline media $(pH\;9.0{\sim}11.0)$, it adjusted the pH of the culture media to around pH 8.5 by itself.

The Dyeability of Indigo Depending on the pH of Reduction Bath (환원액의 pH에 따른 인디고의 염색성)

  • Cotton fabric was dyed with indigo at different pHs, dye concentrations, and repeated dyeings. And the K/S value and dye uptake of the dyed materials at those conditions were determined. The decrease of pH during the reduction process of indigo stock solution was observed. As a result the decrease of pH of the dye stock solution depended on the dye concentration and initial pH. The highest K/S value was shown at fabric dyed at pH 11 and the relationship between K/S value and dye uptake depended on initial pH of reduction bath and the slope was lower as the pH increased to 13.

  • pH has been regarded as a master variable governing the heavy metal sorption on fly ash. However, the chemical constituents in the fly ash could also suggest a potential sorption site for heavy metals. So, in this study sequential extraction method is employed to evaluate the sorption behavior of fly ash for cadmium. Two different fly ashes (S-fly ash, T-fly ash) were obtained from different power plants in Korea. First, cadmium is adsorbed under four different initial pHs. And, Cd sorbed in fly ash was sequentially desorbed following the sequential extraction method suggested by Tessier. In test results, the effect of pH increase was differently exerted in two fly ash. In S-fly ash, exchangeable fraction was dominated in low initial pH, however, as increasing initial pH, the fraction bound to carbonate increased. In the T-fly ash, regardless of initial pH the fraction bound to carbonate was major part of sorption estimated. The fraction bound to Fe/Mn oxide was about 10% in T-fly ash, and 5% in S-fly ash at high pH.

Changes of Brine Characteristics during the Salting Process of Winter, Spring, and Summer Chinese Cabbage (계절별 배추 절임염수의 특성변화)

  • Major characteristics of brines produced from the salting process were compared among winter, spring, and summer (highland) Chinese cabbages. The soluble-solids contents of final brine were decreased to 87~90% of initial brine during salting process, and showed 16.3, 15.8, and 14.4$^{\circ}$Brix for winter, spring, and highland Chinese cabbages, respectively. The pHs showed similar changes during salting process from pH 8.40~8.63 for initial to pH 6.03~6.24 for final. The high salting(12.4~14%) of final brine needs dilution or reuse treatemnt before discard. The COD of final brine were increased to 39.6 ppm, 52.1 ppm, 37.7 ppm, respectively. During salting total microbial counts of final brine were increased ten times from those of the initial brines for all samples.

  • The aim of this study was to investigate the antimicrobial characteristics of citral against Salmonella Typhimurium and Staphylococcus aureus. Antimicrobial activities were determined according to the citral concentration and initial pH. The tested citral concentrations were 0-1,000 ppm in tryptic soy broth (TSB) and 0-5,000 ppm in Angelica keiskei juice (NokJeup). The initial pHs tested were 4-7. Antimicrobial activities increased as citral concentration increased. S. aureus was more susceptible than S. Typhimurium during culture in TSB. But S. aureus was less susceptible to pH changes. Citral caused about 1-2 log reduction of S. aureus and 2-5 log reduction of S. Typhimurium after 10 min exposure at different pHs. As the citral concentration in the Algelica keiskei juice increased, S. aureus was easily inactivated but S. Typhimurium was not inactivated.

Effect of Culture Conditions on Canthaxanthin Production by Dietzia natronolimnaea HS-1

  • This study investigated the effects of various culture parameters (carbon sources, temperature, initial pH of culture, NaCl concentration, and light) on the growth and canthaxanthin production by Dietzia natronolimnaea HS-1. The results showed that the most effective carbon source for growth and canthaxantin production was glucose, and the best pH and temperature were 7 and $31^{\circ}C$, respectively. In addition, the biomass and canthaxanthin production increased in a medium without NaCl and in the presence of light. Under the optimized conditions, the maximum biomass, total carotenoid, and canthaxanthin production were $6.12{\pm}0.21g/l,\;4.51{\pm}0.20mg/l,\;and\;4.28{\pm}0.15mg/l$, respectively, in an Erlenmeyer flask system, yet increased to 7.25 g/l, 5.48 mg/l, and 5.29 mg/l, respectively, in a batch fermenter system.

A Study on the Degradation Characteristics of 1,4-dioxane at Different Initial pHs with Advanced Oxidation Process Using $O_3/H_2O_2$ ($O_3/H_2O_2$ 고급산화공정에서 초기 pH 변화에 따른 1,4-dioxane의 제거 특성 연구)

  • The pH efforts on the removal of 1,4-dioxane and the biodegradobility enhancement of dioxane contaminated water were investigated using $O_3/H_2O_2$ baled advanced oxidation process. Experiments were conducted using a bubble column reactor under different initial pH. The $O_3/H_2O_2$ process effectively converted 1,4-dioxane to more biodegradable intermediates which had a maximum $BOD_5$ enhancement at pH 11 within the experimental range, precisely, when the initial pH increased, $BOD_5$ enhanced. However, in case of removal efficiencies of 1,4-dioxane during $O_3/H_2O_2$ oxidation the optimum condition was shown at pH 9 compared with pH 6 and 11. TOC and COD values were not largely changed for all reaction time. From the results of 1,4-dioxane removal efficiency, TOC, COD, and $BOD_5$ enhancement with reaction time, it was surely observed that 1,4-dioxane was just converted to biodegradable materials, not completely oxidized to carbon dioxide.
