Biofilm 생성이 토양흡착 나프탈렌 분해에 미치는 영향 (Effect of Biofilm Formation on Soil Sorbed Naphthalene Degradation)

  • 나프탈렌 분해균주인 Pseudomonas aeruginosa CZ6을 오염된 토양에서 분리하였으며 분리된 균주는 결정상태의 나프탈렌에 부착하고 그 주변에 extracellular polymeric substance를 분비하는 특성을 가졌다. LB, YM과 MSM 배지를 사용하여 배지의 종류에 따른 biofilm 생성량을 측정한 결과, LB 배지에서 biofilm이 가장 많이 생성되는 것으로 나타났다. 나프탈렌을 기질로 한 배양조건에서 균주는 기질의 농도 영향을 크게 받지 않고 0.10% 기질, 150 rpm 조건에서 최적 배양조건은 $30^{\circ}C$, pH 7로 나타났다. 두 가지 토양에서 배지의 종류에 따른 나프탈렌의 분해특성을 관찰한 결과 초기에는 MSM 배지에서 나프탈렌이 가장 많이 분해가 되었다. 그러나 생물이용성이 제한을 받는 조건에서 LB 배지의 나프탈렌이 가장 빨리 제거가 되었다. 이런 결과는 biofilm의 형성과 extracellular polymeric substance 생성이 토양에 흡착된 잔류 나프탈렌의 생물학적 이용성을 향상시키기 때문인 것으로 사료된다.

  • Monascus purpureus MMK2를 이용하여 수용성 적색색소 생산을 위한 배양조건의 최적화에 관한 결과는 다음과 같다. 실험균주가 생산하는 적색색소의 최적 배양조건은 탄소원으로 wheat flour 3.0% 첨가, 질소원으로 $NaNO_3\;0.15%$, 인산염으로 $Na_2HPO_4\;12H_2O$의 농도가 0.25% 및 $MgSO_4$의 농도가 0.15%일 때 가장 높은 적색색소 생성을 나타내었다. 배양온도는 $30^{\circ}C$, 초기 pH가 6.5 및 배양시간 7일일 때 적색색소의 생성능은 모 균주의 생성능인 1.77unit에 비해 23.38배 높은 41.39 unit로 가장 우수한 특성을 조사하였다.

  • Agorobacterium rhizogenes에 의하여 유도된 당근(Daucus carota L. )의 hairy root를 배양하였으며 opine 유무에 대한 형질전환 확인 및 배지조성을 달리하여 성장률에 따른 색소함량을 비교하였고, 재분화된 식물체의 형태적 차이를 관찰하여 몇가지 결론을 얻었다. Hairy root는 균 접종 2-4주 후에 형성층 부위를 중심으로 유도되었다. 유도된 hairy root의 초기 배양에는 R.C.M 배지가 적합하였으며 MSO(2, 4-D $10^{-4} ml/ l, pH6, sucrose 5%, 질소원 0.03M 등)에서 최대 성장을 보여주었고 성장의 증가에 따른 색소의 형성은 비교적 안정하였다. 재분화된 식물체는 정상 식물체에 비하여 형태적으로 차이를 나타내었으며 형질전환된 hairy root 및 재분화된 식물체에서 mannopine 분석으로 Ri-plasmid에 의한 형질전환이 이루어졌음을 확인하였다.

지황의 액체배양에서 탄소원.질소원 및 pH가 직접 체세포배 형성에 미치는 영향 (Effects of Carbon, Nitrogen Sources and pH on Direct Somatic Embryogenesis in Liquid Culture of Rehmannia glutinosa Lib.)

  • 지황(Rehmannja glutjnosa)에서 종근 저장시 나타나는 병원균의 오염을 배제하면서 종묘를 대량 증식시키는 방법을 개발하기 위하여 액체 배양을 통해 직접 체세포배를 형성하고자 할 때 요구되는 배지, 질소원, 탄소원 및 pH와 배양 기간 중 배지의 성분 변화를 조사하였다. 줄기나 엽병을 접종조직으로 할 경우, 배지내 질소원인 $NH_4NO_3$$KNO_3$의 적정비율은 825 mg/1 : 1900 mg/l 이었고, 탄소원으로는 자당이 적합하였으며, 적정 농도는 3% 이었다. 배지로는 1X MS 배지가 적합하였고, 적정 BA 농도는 2.0 mg/l 였는데, NAA와 조합하였을 경우에는 BA 1 mg/l에 NAA를 0.5 mg/l 를 조합한 배지 와 BA 2.0 mg/l 에 NAA를 0.1 mg/l로 첨가한 배지가 체세포배 발생에 효과적이었다. 멸균 전에 pH를 5.7로 조정한 경우에 직접 체세포배의 형성에 효과적이었다. 배양기간 중 배지 내 $Ca^{++}$$Mg^{++}$이온은 거의 변화가 없었으나, $K^{+}$이온은 많이 흡수되었다. 배양 3주 째에 자당은 거의 고갈되었고, 과당과 포도당은 3주 째부터 흡수되기 시작하였다.

  • A bacteium, KP-6408, capable of hydrolyzing fibrin was isolated from Chungkook-jang, which was possibly identified as a strain of Bacillus sp. The effects of culture condition and medium composition on the enzyme production were investigated. Among nitrogen sources tested, yeast extract was the most effective for the enzyme production, and the level of the concentration for the optimal enzyme production was 0.2%(w/v). For carbon sources, glucose was the best for the enzyme production with the level of 2.0%(w/v). The enzyme was maximally produced by cultivating the enzyme production with the level of 2.0%(w/v). The enzyme was maximally produced by cultivating the organism at the liquid medium of the initial pH 8.0 and temperature of 4$0^{\circ}C$. In Chungkook-jang fermentation, the enzyme was maximally produced when incubated at 35$^{\circ}C$ for 24 hrs using soybean as a solid medium. The addition of various rice starch to the soybean in Chungkook-jang fermentation lowered the enzyme production.

해양세균 Zoogloea sp.로부터 유용 다당류의 생산에 미치는 발효조건의 영향 (Effects of Fermentation Conditions on the Production of the Useful Polysaccharides from Marine Bacterium Zoogloea sp.)

  • The fermentation conditions for the maximal production of the useful polysaccharides(water soluble polysaccharide and cell bound polysaccharide) from marine bacterium Zoogloea sp.(KCCM 10036) were investigated with a 5 L jar fermentor. The maximal production of these polysaccharides was obtained under the conditions of initial pH 7.8, 30$^{\circ}C$, 400 rpm of agitation speed, 2 Wm of aeration rate, 10%(w/v) of inoculum size and 2.5%(w/v) of glucose substrate and 10.38 g/L of total polysaccharide was produced. Apparent viscosity of the culture broth was increased with the production of these polysaccharides and the maximum value was reached to 22,500 cp.

플라스크 배양에서 Bacillus subtilis BK-17의 혈전용해효소 생산에 대한 환경 및 영양 조건의 영향 (Effects of Environmental and Nutritional Conditions on Fibrinolytic enzyme Production from Bacillus subtilis BK-17 in Flask Culture)

  • The production of fibrinolytic enzyme from Bacillus subtilis BK-17 was studied in the shake flask cultures. The important medium components studied include nitrogen source, carbon source and inorganic salts. The environmental conditions include initial pH, temperature, shaking speed and working volume. Among various N-sources, C-sources and inorganic salts tested, soybean flour, D-glucose and Na2HPO4 gave the best results, and their optimal concentrations were 1.5%, 0.5% and 0.05%, respectively. The optimal pH and temperature were 9.0 and 37$^{\circ}C$. With decreasing working volume in the range of 25∼100ml in the 250ml flask or increasing shaking speed in the range of 100∼300rpm, the enzyme production was greatly enhanced. The enzyme activity under the optimal conditions was about 1400I.U./ml with urokinase as a standard.

음식폐기물 추출물을 이용한 느타리버섯균의 최적성장조건 (Optimal Growth Condition of Pleurotus ostreatus Cultured in the Foodwastes Extracts)

  • For the purpose of the methodological development to convert the food wastes into resources, we have attempted to culture the mushroom hypha, Pleurotus ostreatus. The food-wastes were mixed with distilled water, and the mixture was autoclaved to produce fluid, which was centrifugated and used as the growth media. Concentrations of the food wastes extracts were prepared with 10, 20, 30, 40, and 50%(W/V), and the initial pH were set variously with 4, 5, 6, and 7. These were cultured for 9 days at the temperature of $25^{\circ}C$ and the rotation rate of 120 rpm. The result is that the fluid form of the mushroom hypha have been grown best at the concentration of 30% and the optimal pH was 5 and 6.

  • Several bacterial strains utilizing crude oil as their sole carbon and energy sources were isolated from marine. One of the strains named KCL-1 showed the highest degradative activity for crude oil and the best growth rate. This strain was identified as a Klebsiella sp. based on the morphological, biochemical, and physiological characteristics. The optimum cultural conditions were as follows; $32^{\circ}C$~$37^{\circ}C$ for temperature and 7.0 for initial pH. Additionally, the optimal concentration of sodium chloride was 3.0%, indicating that this strain was derived from seawater. KCL-1 could use several kinds of n-alkane hydrocarbons from octadecane to hexacosane as a sole carbon source. The degradation of crude oil by KCL-1 was stimulated by addition of octadecane in the culture. The emulsifying activity by KCL-1 was highest after 3 days of cultivation under the condition of 3.0% sodium chloride, pH 7.0 and $37^{\circ}C$.

  • The conditions for the $\beta$-fructofuranosidase production from alkalophilic, thermophilic Bacillus sp. TA-11 were investigated. The maximal enzyme production was obtained when the strain was cultured at 50$\circ$C for 36 hrs with batch culture in the optimal medium containing 1.0% sucrose, 0.6% yeast extract, 0.1% each of KH$_{2}$PO$_{4}$, K$_{2}$HP0$_{4}$, NaH$_{2}$PO$_{4}$, and initial pH 9.5, and the final enzyme activity under the above condition was 102 units per ml of cell free extract.
