한국산 마늘에 의한 Aspergillus parasiticus의 성장 억제 효과 (Inhibitory Effect of Garlic on the Growth of Aspergillus parasiticus)

  • This study was performed to investigate the possible effect of garlic produced in Korea on the inhibition/reduction of growth of A. parasiticus, a toxigenic strain. The effect was studied using different concentrations of freeze-dried garlic in potato-dextrose agar (PDA) and/or in yeast-extract sucrose (YES) broth at $25^{\circ}C$ for 15 days. While inhibition of the fungal growth due to increasing the concentration of garlic was observed, the more remarkable effect was observed on the ninth day. Reduction of fungal diameter as a result of addition of garlic on PDA was observed to range between 3.4% to 20.1 % while reduction of mycelial weight in YES broth ranged from 9.9% to 30.5%. The 0.5% and 1.0% concentrations of garlic significantly reduced fungal diameter in PDA on the 9th day, while 0.1 %, 0.5%, and 1.0% concentrations of garlic significantly reduced the mycelial weight in YES broth (p<0.05). Dose-response relationships were observed between the concentration of garlic and inhibition of growth both in solid culture and in liquid culture. This study indicates that garlic could be an effective inhibitor at a human consumption level of the growth of A. parasiticus. More research is needed to study the inhibitory effects of the main active component of garlic.

Effect of 2-NBDG, a Fluorescent Derivative of Glucose, on Microbial Cell Growth

  • A fluorescent glucose analogue,2-[N-(7-nitrobenz-2-ox a-1,3-diazol-4-yl) amino] -2- deoxy-D-glucose (2-NBDG), which had previously been developed for the analysis of glucose uptake in living cells, was investigated to determine its biological activity on microorganisms.2-NBDG did not show any inhibitory effect on growth of yeast cells and bacteria. In contrast, 2-NBDG exhibited strong inhibitory effects on filamentous fungal growth. The growth of filamentous fungi was completely inhibited, when 2-NBDG was supplemented as sole carbon source. The inhibitory effect was decreased by the addition of glucose in the test medium. Furthermore, 2-NBDC inhibited chitinase activity of Trichoderma sp. These results suggested that the inhibitory effects of 2-NBDG on filamentous fungi might be partially due to the inhibition of chitinase.

Inhibitory Effect of Algal Extracts on Mycelial Growth of the Tomato-Wilt Pathogen, Fusarium oxysporum f. sp. lycopersici

  • The present study was undertaken to explore the inhibitory effect of cyanobacterial extracts of Nostoc commune FA-103 against the tomato-wilt pathogen, Fusarium oxysporum f. sp. lycopersici. In an optimal medium, cell growth, antifungal activity, and antifungal compound production could be increased 2.7-fold, 4.1-fold, and 13.4-fold, respectively. A crude algal extract had a similar effect as mancozeb at the recommended dose, both in laboratory and pot tests. In vitro and in vivo fungal growth, spore sporulation and fungal infection of wilt pathogen in tomato seeds were significantly inhibited by cyanobacterial extracts. Nostoc commune FA-103 extracts have potential for the suppression of Fusarium oxysporum f. sp. lycopersici.

영지 노랑병 방제에 효과적인 살균제의 선발 (Selection of Effective Fungicides Against Xylogone sphaerospora, a Fungal Pathogen of Cultivated Mushroom, Ganoderma lucidum)

  • A fungal disease of the cultivated mushroom, Ganoderma lucidum, caused by Xylogone sphaerospora was epidemic throughout all cultivation areas in Korea which caused a lot of yield losses in the mushroom production. For controlling the disease, the screening of effective fungicides against the pathogenic fungus were conducted. Thirty seven commercially available fungicides were tested for their inhibitory activities on potato dextrose agar media supplemented with these fungicides at various concentrations. Twenty one fungicides significantly inhibited mycelial growth of the pathogen, Xylogone sphaerospora, but 16 fungicides had no inhibitory effect. Among these 21 fungicides, 17 fungicides also inhibited mycelial growth of Ganoderma lucidum as well, but imazalil, procymidone, triforine, and vinclozolin had no inhibitory effects. However, vinclozolin showed no inhibitory effect on mycelial growth of the mushroom even at the concentration of 50 $\mu\textrm{g}$/ml vinclozolin solution for 2 hours, and then the pathogen was inoculated. After two month-cultivation of the mushroom, over 90% of logs treated with vinclozolin without pathogen inoculation produced fruiting bodies. However, fruiting bodies were not produced form the logs inoculated with the pathogen, but not treated with vinclozolin. Fifty seven percent of logs. which were pre-treated with vinclozolin and then inoculated with the pathogen produced fruiting bodies. Based on the results, vinclozolin is effective for the control of yellow disease of the Ganoderma lucidum caused by Xylogone sphaerospora.

Polyphosphate의 진균 성장 억제 작용에 관한 연구 (Studies on Antifungal Effect of Polyphosphate)

  • Polyphosphate의 Candida albicans와 Trichophyton mentagrophytes에 대한 항진균 작용에 대하여 연구하였다. 평균 사슬길이가 15, 45, 75인 polyphosphate는 진균에 대하여 성장억제 효과를 나타낸 반면, pyrophosphate는 성장억제 효과가 없었다. 사슬길이가 증가함에 따라서 진균 성장억제 효과가 증가하는 것으로 관찰되었으며 $800\;{\mu}g/ml$ 농도에서는 균주의 성장을 완전히 억제시켰다. $Mg^{2+}$$Ca^{2+}$의 첨가는 polyphosphate의 성장억제 효과를 감소시켰다. Polyphosphate를 처리한 C. albicans에서는 핵산물질의 유리가 관찰되었으며, 세포 swelling이나 표면돌출과 같은 세포형태 변화도 관찰되었다. 세포막 불투과성 물질인 propidium iodide는 Polyphosphate를 처리한 C. albicans의 핵을 염색하는 것이 관찰되었다. Polyphosphate의 항진균 작용은 polyphosphate가 진균 세포벽이나 세포막의 필수 양이온에 대한 chelation 효과에 의한 것으로 사려된다.

Analysis of the Inhibitory Effect of two Bacterial Strains on Metarhizium anisopliae Induced Fatality Rates in Protaetia Brevitarsis

  • Bacterial species, Bacillus amyloliquefaciens and Lactobacillus species (L. sp.5-1), are known to inhibit the growth of pathogenic bacteria and fungi. Metarhizium anisopliae is a pathogenic fungal species which causes fatal damage to P. brevitarsis populations. Therefore, we investigated the inhibitory effect of B. amyloliquefaciens and L. sp. 5-1 on M. anisopliae induced fatality rates in P. brevitarsis. Samples of M. anisopliae-infected sawdust were treated with strain B. amyloliquefaciens KACC10116, strain L. sp. 5-1 KACC19351, and a combination of the two. P. brevitarsis were fed treated sawdust samples, and their subsequent fatality rate was monitored. The fatality rate fell below 1.5% after 10 days and decreased by approximately 40% after 15 days. On average, the fatality rate decreased by 20%, compared to the control. The difference in the decrease in fatality rate between B. amyloliquefaciens treatment and L. sp. 5-1 treatment was not significant. Results indicate that both strains exhibit high anti-fungal activity, which may be useful in environmental purification efforts. These strains may be used for effective prevention of fungal infection in P. brevitarsis.

Antifungal Activity of Thymol against Aspergillus awamori and Botrytis aclada Isolated from Stored Onion Bulbs

  • The antifungal activity of thymol against Aspergillus awamori F23 and Botrytis aclada F15 in onions was examined through direct treatment with amended media and gaseous treatment with I-plates (plastic plates containing central partitions). The protective and curative control efficacy of thymol was examined 24 h before and after the inoculation of onion bulbs with the fungal isolates. Mycelial growth, sporulation, and spore germination of the isolates were inhibited on potato dextrose agar amended with various concentrations of thymol or acetic acid (positive control). Overall, thymol produced a stronger inhibitory effect on the mycelial growth and development of the isolates than acetic acid. Following gaseous treatment in I-plates, mycelial growth, sporulation, and spore germination of the isolates were inhibited at higher concentrations of thymol or acetic acid; however, acetic acid showed a little effect on the sporulation and spore germination of the isolates. Following the treatment of onion bulbs with 1000 mg L-1 of thymol 24 h before and after fungal inoculation, lesion diameter was greatly reduced compared with that following treatment with 0.5% ethanol (solvent control). Onion bulbs sprayed with thymol 24 h before fungal inoculation generally showed reduced lesion diameters by isolate F23 but not in isolate F15 compared with those sprayed 24 h after fungal inoculation. Collectively, thymol effectively inhibited the growth and development of A. awamori and B. aclada on amended media and in I-plates. In addition, spraying or fumigation of thymol is more desirable for effectively controlling these postharvest fungal pathogens during long-term storage conditions.

Effect of Acaromyces Ingoldii Secondary Metabolites on the Growth of Brown-Rot (Gloeophyllum Trabeum) and White-Rot (Trametes Versicolor) Fungi

  • We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brownrot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems.

  • The growth and biochemical activities of Chlorella fusca were studied in the presence of different concentrations of either filtrates or mycelial mats of Saprolegnia ferax and Pythium graminicola. Low concentrations of both fungal filtrates exerted increase in total count, dry weight and in the biosynthesis of photosynthetic pigments, carbohydrates and nitrogen content. High concentrations showed inhibitory effect on both growth and biochemical activities of Chlorella fusca. Supplementation with different concentrations of dry mycelial mats of either fungi the culture of Chlorella showed elevation in biomass, dry weight, and biosynthesis of carbohydrates and nitrogen content especially at low concentrations. The contents of photosynthetic pigment were inhibited only at low concentrations. Neither the culture filtrate of Pythium nor Saprolegnia had cellulolytic activity, although polygalacturonase enzymes were detected, whereas chloroform-extract of both fungal filtrates showed blue spots under long wave light (366 nm).

Effect of Grass Lipids and Long Chain Fatty Acids on Cellulose Digestion by Pure Cultures of Rumen Anaerobic Fungi, Piromyces rhizinflata B157 and Orpinomyces joyonii SG4

  • The effects of grass lipids and long chain fatty acids (LCFA; palmitic, stearic and oleic acids), at low concentrations (0.001~0.02%), on the growth and enzyme activity of two strains of anaerobic fungi, monocentric strain Piromyces rhizinflata B157 and polycentric strain Orpinomyces joyonii SG4, were investigated. The addition of grass lipids to the medium significantly (p<0.05) decreased filter paper (FP) cellulose digestion, cellulase activity and fungal growth compared to control treatment. However, LCFA did not have any significant inhibitory effects on fungal growth and enzyme activity, which, however, were significantly (p<0.05) stimulated by the addition of oleic acid as have been observed in rumen bacteria and protozoa. This is the first report to our knowledge on the effects of LCFA on the rumen anaerobic fungi. Continued work is needed to identify the mode of action of LCFA in different fungal strains and to verify whether these microorganisms have ability to hydrogenate unsaturated fatty acids to saturated fatty acids.
