Rosmarinus officinalis 정유의 수종 항생제 감수성 및 내성 균주에 대한 억제효과 (In Vitro Inhibitory Activities of Essential Oils from Rosmarinus officinalis L. Against Antibiotic-Susceptible and Resistant Strains of Some Pathogenic Bacteria)

  • The in visto inhibitory activities of essential oils of the Rosmarinus officinalis as well as its main constituents were evaluated against antibiotic-susceptible and -resistant strains of Staphylococcus aureus, streptococcus pneumoniae, Salmonella enteritidis and S. typhimurium. The essential oil fraction of R. officinalis and its main components, 1,8-cineole and camphor, exhibited significant inhibitory activities against most of the tested strains in this study, with MICs(minimum inhibitory concentrations) racing from 0.5mg/ml to 16mg/ml. The total oil fraction showed higher activity than its main components, 1,8-cineole and camphor against S. aureus strains. No remarkable differences were evident between MICs of the susceptible and resistant strains of S. aureus. Among the tested strains, S. pneumoniae CCARM 3523, the resistant strain to norfloxacin, oxacillin and erythromycin exhibited significantly lower sensitivity to the tested oils than antibiotic-susceptible strain. The oils revealed mostly higher inhibitory activity against S. typhimurium than against S. enteritidis.

Antimicrobial Effects of 8-Quinolinol

  • 8-Quinolinol and other quinolinol derivatives were evaluated with regard to their growth-inhibitory effects against intestinal bacteria, using the paper disk-agar diffusion method. The observed growth responses varied according to the chemicals and dosages used, as well as the bacterial species tested. 8-Quinolinol showed a significant inhibitory effect against Clostridium difficile, C. perfringens, and Escherichia coli, at 5, 2, 1, and 0.5 mg/disk, and also exhibited a very strong inhibitory effect at 0.25 mg/disk. At low concentrations, 8-quinolinol had strong inhibitory effects against C. perfringens at 0.1 and 0.05 mg/disk; 8-quinolinol also manifested a moderate inhibitory effect against C. perfringens at 0.025 mg/disk. Furthermore, 8-quinolinol revealed moderate and weak growth inhibition against C. difficile and E. coli at concentrations of 0.1 and 0.05 mg/disk, respectively, but 2-quinolinol, 4-quinolinol, and 6-quinolinol evidenced no growth inhibition against B. bifidum, B. longum, C. difficile, C. perfringens, E. coli, or L. casei. The inhibitory effects of 8-quinolinol against C. difficile, C. perfringens, and E. coli lead to its consideration as a possible therapeutic modality for the treatment of diseases associated with harmful intestinal bacteria.

Characterization of a New Antidementia $\beta$-Secretase Inhibitory Peptide from Rubus coreanus

  • In order to develop a potent antidementia $\beta$-secretase inhibitor from phytochemicals, $\beta$-secretase inhibitory activities of extracts from many medicinal plants and herbs were determined. Water extracts from Rubus coreanus showed the highest $\beta$-secretase inhibitory activity of 84.5%. After purification of the $\beta$-secretase inhibitor from R. coreanus using systematic solvent extraction, ultrafiltration, Sephadex G-10 column chromatography, and reverse-phase high performance liquid chromatography (HPLC), a purified $\beta$-secretase inhibitor with $IC_{50}$ inhibitory activity of $6.3{\times}10^3\;ng/mL$ ($1.56{\times}10^{-6}\;M)$ was obtained with a 0.08% solid yield. The molecular mass of the purified $\beta$-secretase inhibitor was estimated to be 576 Da by liquid chromatography-mass spectrometry (LC-MS) and $\beta$-secretase inhibitor also is a new tetrapeptide with the sequence Gly-Trp-Trp-Glu. The purified $\beta$-secretase inhibitory peptide inhibited $\beta$-secretase non-competitively and also show less inhibition on trypsin, however no inhibition on other proteases such as $\alpha$-secretase, chymotrypsin, and elastase.

Inhibitiory Activity of Lactic Acid Bacteria against Hazardous Microbes

  • One hundred of lactic cultures were evaluated for their ability to inhibit hazardous microbes, such as Salmonella enteritidis, Salmonella typhimurium, Escherichia coli, Listeria monocytogenes, and Bacillus cereus by agar well diffusion method. None of them showed inhibitory halo against S. enteritidis, while 27 strains showed inhibitory activity against S. typhimurium, 6 against E. coli, 9 against ampicillin resistant E. coli, 31 against L. monocytogens, 10 against B. cereus. pH of the culture does not explain for the inhibitory activity except against B. cereus. A neutralized culture from corn silage showed highest inhibitory activity against S. typhimurium, and the size of inhibitory halo was same as 10 ug/mL of ampicillin. The culture was identified to be Lactobacillus buchneri on the basis of biochemical characteristics and utilization of substrates. Using the culture as probiotics could be expected to reduce antibiotics for animal feeding.

미숙과 건여주(Momordica charantia L.) 열수 및 에탄올 추출물의 탄수화물 분해효소 저해 활성 및 항염증 활성에 대한 연구 (A Study on Inhibitory Activities on Carbohydrase and Anti-Inflammatory Activities of Hot-Water and Ethanol Extracts from Immature Dried Bitter Melon (Momordica charantia L.))

  • Bitter melon (Momordica charantia L.) has been used as a medicinal plant due to its biological activity. This study investigated the inhibitory effects on carbohydrase and anti-inflammatory effects of hot-water extract (WEM) and ethanol extract (EEM) from dried bitter melon cultivated in Korea. The inhibitory activities of EEM against ${\alpha}$-amylase and ${\alpha}$-glucosidase were over 70% at 4,000 ppm. The inhibitory activities of EEM were significantly higher than those of WEM at all concentrations. The anti-inflammatory effects were evaluated by measuring nitric oxide (NO) production in LPS-stimulated RAW264.7 cells. EEM did not show cytotoxicity on RAW264.7 cells at the test concentrations. Moreover, the inhibitory effect on NO production in LPS-stimulated RAW264.7 cells was significantly increased by the addition of EEM. These findings suggest that EEM has good inhibitory effect on carbohydrase and anti-inflammatory activity and it has potential as a source of natural functional material.

전염성기관지염(傳染性氣管支炎) 바이러스에 대한 가토혈청억제물질(家兎血淸抑制物質)에 관한 연구(硏究) (Studies on Rabbit Serum Inhibitor of Avian Infectious Bronchitis Virus)

  • It has been-reported that rabbit serum exhibit an inhibitory action on avian infectious bronchitis virus in embryonating chicken embryo. In this thesis, the biological, serological, physical and chemical properties of normal rabbit serum on the effect of the virus propagation were studied. Throughout the studies, the following experimental results 'were obtained and summarized here. 1. An inhibitory action of rabbit serum on avian infectious bronchitis vrius is due to the normal serum constituents. 2. The nature of the neutralization between normal rabbit serum and the virus is similar to that of the specific antiserum and the virus. 3. Rabbit serum, heat inactivated at $56^{\circ}C$, for 30 minutes, showed its average $log_{10}El,D_{50}Nl$ of 3.7. 4. The inhibitory compound present in the normal rabbit serum is inactivated by means of 5 per cent trypsin, 0.01 M potassium periodate, and absorbed to zymosan. 5. The inhibitory compound was not affected by 0.05 M trichloroacetic acid and 0.005M $KH_2PO_4$. 6. The higher the temperature of heat inactivation of rabbit serum caused the lesser the neutralizing effect on the virus. Heating the serum at $66^{\circ}C$, for 30 minutes brought about a complete loss of the neutralizing index of the serum. 7. No ions, as a cofactor, was incorporated to the inhibitory action of rabbit serum on the virus. 8. The inhibitory compound amays be found in a fraction of serum globulin.

Epigallocatechin gallate의 인체 피부흑색종세포와 인체 구강유상피암종세포에 대한 성장억제효과 (The Growth Inhibitory Effects of Epigallocatechin Gallate Against Human Skin Melanoma Cells and Human Oral Epitheloid Carcinoma Cells)

  • Epigallocatechin gallate (EGCG) was reported to exert weak cytotoxicity against normal healthy cells such as C3H10T1/2 cells, but profound inhibitory effects on the initiation or promotion stage of chemical carcinogenesis in mammary gland, blood and mouse skin. This study was carried out to develop antitumor agents with weak side effects and strong antitumor activity. Human skin melanoma cells (HBT 69) and human oral epitheloid carcinoma cells (OCL 17) were cultured in RPMI-1640 media containing 10% fetal bovine serum, antibiotic, and fungizone. After incubation for 24 hrs, the cells were treated with various amounts of (EGCG) for 48 hrs. The growth inhibitory effects of EGCG in human oral epitheloid carcinoma cells were evaluated by the 3- (4,5-djmethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT), neutral red (NR), and sulforhodamine B protein (SRB) assays of colorimetric methods. The light microscopic study was also carried out to observe morphological changes of the treated cells. These results obtained were as follows; 1. Significantly inhibitory effects of EGCG against cultured human oral epithelioid carcinoma cells. 2. Significantly inhibitory effects against cultured human skin melanoma cells treated with 50 $\mu$M EGCG, but decreased inhibitory effects in 100 $\mu$M EGCG. 3. Degenerative changes against cultured human oral epitheloid carcinoma cells. 4. Degenerative changes against human skin melanoma cells treated with 50 UM EGCG, but recovered degenerative changes in 100 $\mu$M EGCG.

김 단백질 가수분해물의 Angiotensin Ⅰ 전환효소 저해 활성 (Angiotensin Ⅰ Converting Enzyme(ACE) Inhibitory Activities of Laver(Porphyra tenera) Protein Hydrolysates)

  • Angiotensin Ⅰ converting enzyme(ACE) inhibitory activities of laver(Porphyra tenera) protein hydrolysates were investigated by enzymes used for hydrolysis, molecular fractions and drying methods. For the enzymatic hydrolysis, crude laver protein, separated by filtration of water extract of dried laver extracted with 20 times(w/v) water for 3 hours at boiling temperature, were hydrolyzed with three commercial protease, Pepsin, alcalase and maxazyme NNP at optimal conditions. The yield of hydrolysis and ACE inhibitory activities of which were high in order of pepsin, alcalase and maxazyme NNP. ACE inhibitory activities of laver hydrolysates by molecular levels were high in order of 3 kDa > 10 kDa > 3∼10 kDa, and the IC/sub 50/ ACE inhibitory activities by molecular lebels were 4 mg/mL(3 kDa), 5 mg/mL(total hydrolysate), and 20 mg/mL(10 kDa), respectively. The storage stability of dried laver hydrolysates at 20℃ were strongly affected by drying methods, hot air dried of which were much stabler than freeze-dried one.

  • Experimental studies were performed to investigate the effect of Houttuyniae herba Aqua-acupuncture on the growth of Melanoma 816 tissue and Mononuclear cell subsets in mice. The results are as follows : 1. In the study of lymphocytes numbers in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 2. In the study of $CD4^+$ T cell percentage in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 3. In the study of $CD8^+$ T cell percentage in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, with stastistical significance. 4. in the study of lymphocytes numbers in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 5. In the study of $CD4^+$ T cell percentage in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 6. In the study of $CD8^+$ T cell percentage in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 7. In the measurement of melanoma tissue weight, Houttuyniae herba Aqua-acupuncture showed increasing effect, but without stastistical significance. According to above results, it is concluded that Houttuyniae herba Aqua-acupuncture is effective upon immune responses in Melanoma 816 inoculated mice.

작약 메탄올 추출물 및 분획물의 Tyrosinase 저해 및 Melanin 생성 억제활성 (Tyrosinase Inhibitory Activity and Melanin Production Inhibitory Activity of the Extract and Fractions from Paeoniae Radix)

  • We investigated antioxidative activity, tyrosinase inhibitory activity, and melanin production inhibitory activity of the methanol extract of Paeoniae Radix and its fractions. The total polyphenol content of the extract was 73.45 mg/g, and content of the ethyl acetate fraction was 514.50 mg/g as the highest content of fractions. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate was 3.86 ${\mu}g/ml$ as a result of greater activity in the positive control (ascorbic acid). Moreover, tyrosinase inhibitory activity of the ethyl acetate fraction showed higher activity than arbutin used as a positive control. In the cytotoxicity measurement by MTT assay, the extract and fractions were exhibited cell viabilities of 76.96~157.26% against Raw 264.7 and B16F10 cell in concentration of 10~100 ${\mu}g/ml$. In nontoxic concentration range, the ethyl acetate fraction showed strong melanin production inhibitory effect in ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-stimulated B16F10 cell. As a result, the ethyl acetate fraction of the methanol extract from Paeoniae Radix could be applicable to functional materials for skin-whitening agents.
