Removal and Regrowth Inhibition of Microcystis aeruginosa using Artemisia asiatica Extracts (쑥 추출액을 이용한 Microcystis aeruginosa 제거 및 성장억제 연구)

  • Microcystis aeruginosa (M. aeruginosa) is a cyanobacterium species that can form harmful algal blooms in freshwater bodies worldwide. The use of Artemisia asiatica extracts to control M. aeruginosa inhibition will be environmentally friendly and promising. Artemisia asiatica extracts removed successfully upto 88% of M. aeruginosa pH 8 at $25^{\circ}C$ of temperature. These results was indicated that the amount of 2.24 g/L Artemisia asiatica extracts was removed 1g dryweight/L of M. aeruginosa. The kinetic data showed substrate inhibition kinetics and maximum growth rate was obtained when the M. aeruginosa was grown in medium containing 2.5 g/L of initial concentration of Artemisia asiatica extracts. In the various growth control models, Luong model showed the highest correlation coefficient of 0.9916. Therefore, the Luong model was the most suitable control model for the growth control of M. aruginosa using Artemisia asiatica extracts. In conclusion, the growth control of M. aruginosa using Artemisia asiatica extracts can be applied in the field without controlling the temperature and pH of rivers and streams, and it is possible to control the growth of M. aruginosa efficiently in a short time. The natural extract, Artemisia asiatica extracts, can be a promising inhibition due to its high efficiency and low dose requirements.

α-Glucosidase Inhibition Activity of the Extracts of Katsura Tree (Cercidiphyllum japonicum Sieb. Et Zucc) Leaves

  • Katsura tree (Cercidiphyllum japonicum Sieb. Et Zucc) leaves were collected, air-dried and extracted with 70% aqueous acetone, then concentrated and sequentially fractionated using n-hexane, $CH_2Cl_2$, EtOAc, and $H_2O$ to be freeze dried for antioxidant and ${\alpha}$-glucosidase inhibition activity tests. The antioxidant activity of the extracts was evaluated using DPPH (1,1-diphenyl 2-picrylhydrazyl) free radical scavenging assay. The test concentrations were adjusted to 500, 250, 125, 62.5, 31.25, 15.62, 7.81, 3.9, 1.95 and 0.97 ppm. The $H_2O$ and EtOAc fractions showed higher activities compared with the control, ${\alpha}$-tocopherol, at all concentrations. The crude fraction also gave better activity at the concentrations lower than 62.5 ppm. However, the nonpolar n-hexane and $CH_2Cl_2$ fractions gave prominently lower activities compared with the control at all concentrations. The $IC_{50}$ values of the crude, EtOAc, and $H_2O$ fractions exhibited 11.78, 4.29 and $9.80{\mu}g/m{\ell}$, respectively, compared with $12.08{\mu}g/m{\ell}$ of the control. But the n-hexane and $CH_2Cl_2$ fractions indicated 300 and $91.85{\mu}g/m{\ell}$ of $IC_{50}$, respectively. ${\alpha}$-Glucosidase inhibition activity was evaluated at the concentrations of 50, 25, 12.5, 6.3, 3.1, 1.6 and 0.8 ppm. The inhibition activities were increased according to as the increase of sample concentrations. However, the nonpolar n-hexane and $CH_2Cl_2$ fractions indicated very low inhibition activities compared with acarbose, a positive control. The EtOAc fraction showed very good capability as almost 100% compared with the control at the higher concentrations than 12.5 ppm and the crude fraction also indicated good potential as 95% and 100% at 25 and 50 ppm, respectively. The $H_2O$ fraction gave good inhibition value as 90% at 50 ppm although the value was lower than the control. These results showed that the polar fractions had better ${\alpha}$-glucosidase inhibition activities. The $IC_{50}$ values of the nonpolar fractions, n-hexane and $CH_2Cl_2$, showed very lower values as 468 and $103.6{\mu}g/m{\ell}$, respectively, than the control. ${\alpha}$-Glucosidase Inhibition Activity of the Extracts of Katsura Tree (Cercidiphyllum japonicum Sieb. Et Zucc) Leaves However, the polar fractions, crude, EtOAc and $H_2O$, showed 7.1, 3.7 and $13{\mu}g/m{\ell}$, respectively, indicating that these fractions can be used as natural bioresources for treating diabetes mellitus. Also ${\alpha}$-glucosidase inhibition activity had a positive correlation with antioxidant activity of the extracts.

Effects of Suboccipital Muscle Inhibition and Neck Muscle Stabilization Exercise on Pain and Range of Motion in Patients with Chronic Non-Specific Neck Pain (뒤통수밑근 억제기법과 목 안정화 운동이 만성 비특이적 목 통증 환자의 통증과 관절가동범위에 미치는 영향)

  • Purpose: The aim of this study was to apply suboccipital muscle inhibition combined with neck muscle stabilization exercise to 20~30s IT industry employees who suffer from chronic non-specific neck pain. Methods: This study was designed as single-blind and randomized controlled trial. The study participants were 20~30s IT industry employees with chonic non-specific neck pain (VAS 3/10) who were divided into an experimental group (n= 20) subjected to suboccipital muscle inhibition with neck muscle stabilization exercise, and control group (n= 20); suboccipital muscle inhibition only. The intervention was applied three times per week for eight weeks. The neck pain·pressure pain threshold·range of motion, and disability index were measured at the 1st, 8th, and 10th week at follow up, then analyzed with an analysis of variance(ANOVA) using the SPSS program. Results: The total number of study participants was 37 (experimental group 19, mean age 34.6±5.3, control group 18, mean age 35.7±4.9). The comparison and analysis of change in VAS, the pressure pain threshold, and the range of motion except the extension (p>.05) revealed a statistically significant decrease between groups over eight weeks and follow up measurement (p<.01). Regarding the within the group differences, the right side of the neck pressure pain threshold showed a statistically significant decrease over eight weeks in the control group (p<.01). The right and left lateral flexion, and the right and left rotation were statistically significant for the experimental group over eight weeks and follow up measurement, but only the left lateral flexion (p<.05) for the control group over eight weeks. The neck disability index showed a slight decrease but this was not satistically significant for the between-grop or the within-group differences (p>.05). Conclusion: The intervention of suboccipital muscle inhibition and a neck muscle stabilization exercise are more beneficial for neck pain and the range of motion than the application of suboccipital muscle inhibition alone.

Growth Inhibition Effect of Environment-friendly Agricultural Materials in Botrytis cinerea In Vitro (친환경 유기농자재의 잿빛곰팡이병 병원균의 생장 억제 효과)

  • Inhibition effects on spore germination and mycelia growth for gray mold (Botrytis cinerea) were investigated in vitro using environment-friendly agricultural materials as well as environment-friendly pesticides. The inhibition effect on mycelia growth of gray mold is the highest when the gray mold mycelia were treated with a pesticide (commercial name: Koreayeok, Jihabudea KM, Sootingtan, Sootingstar) that contains a mixture of Bacillus subtilis, resulting in 100% inhibition of the mycelia growth. Meanwhile, the range of less than 20% inhibition effects on the growth of gray mold mycelia was observed with other commercial agricultural materials. The significant inhibition effects on spore germination of gray mold fungus were shown in vitro with two water dispersible pesticides containing sulfur [BTB (97.7%) and SulfurStar (92.3%)], respectively. These in vitro results of inhibiting of the spore germination and mycelia growth together cannot found. It remains to be determined whether the selected environment-friendly agricultural materials in effective control of gray mold in vitro can be used to control gray mold in field.

Growth Ingibiton Effect of E. coli O157:H7 and Salmonella typhimurium by Lactic Fermented Milk Products Administrated Orally in Rabbit (토끼에서 유산 발효유제품 급여에 의한 Escherichia coli O157:H7 및 Salmonella typhimurium의 증균억제효과)

  • The growth inhibition effect of Orally administrated yogurt ACE and Metchnikoffupon E. coli O157:H7 and S. typhimurium inoculated into gastric lumen of rabbits was in vestigated. The rabbits challenged with each 1 $m\ell$ of suspension containing 108 CFU/$m\ell$ of the pathogens were divided into 4 groups by the interval of yogurt administration: A group; preadministrated 7 days before inoculation of the pathogens and fed daily; B group; administrated daily after inocjlation of the pathogens, C group; administrated every 3 days after inoculation of the pathogens; Control group, not fed after inoculation of the pathogens. Each 3 $m\ell$ of yogurt containing 109 CFU/$m\ell$ was orally administrated into rabbits. All yogurt administrated groups (A, B, c) chowed growth ingibition effect on E. coli O157:H7 in one day after inoculation of the pathogen by the level of 0.8~1.0 log CFU/g, compared with the result differences between the control group and the yogurt administrated groups. In the control group after 5 days of inoculation, the number of colonized pathogens was 105~106 CFU/g, whereas 103~104 CFU/g was detected in the yogurt administrated groups. After 10 days of inoculation, the viable pathogen number per gram (g) of the rabbit feces was 103 CFU/g in the control group, whereas the number below 101 CFU/g was detected in the group A, and 102 CFU/g in the control group, B and C. The growth inhibition effect of yogurt administration on E. coli O157:H7 was highly increased in the order of A, B, and C group. The same effect on S. typhimurium was observed at the level of 2 log CFU/g in the Metchnikoff yogurt administrated groups, compared with the control group result in one day after inoculation of the pathogen. In 7 days after inoculation of the pathogen, the viable number was increasingly decreased, and finally after 15 days no viable cell of S. typhimurium was discharged into the fecal samples in the group A, and the mean level of 10* CFU/g was detected in the group B, but there was no growth inhibition effect in the group C. The growth inhibition effect on S. typhimurium was observed at the same level of viable cell number between the yogurt ACE administrated groups and the control group in 5 days after inoculation. But, after 10 days of inoclation the viable cell number was started to decrease, and the viable cell of S. typhimurium was not discharged from rabbit intestinal contents after 15 days of inoculation in the yogurt ACE administrated groups. In such a case that yogurt was administrated in order to prevent the pathogens, pre-administration on a daily basis one week before inoculation of the pathogens exerted considerable effect in growth inhibition. In comparison with two kinds of yogurt tested in this study, the growth inhibition effect on two kinds of pathogens was observed more highly in the Metchnikoff administated group than the ACE administrated group.

INHIBITION, PLANNING, AND WORKING MEMORY IN CHILDREN WITH ATTENTION DEFICIT HYPERACTIVITY DISORDER (주의력 결핍 과잉행동 장애 아동의 억제능력, 계획능력, 그리고 작업기억 능력)

  • Objectives:The study was performed to compare the executive function(EF;Inhibition, Planning and Working memory) between ADHD and normal group, and to controll the effect of age and intelligence. And the function of inhibition was assessed in two dimensions(cognitive inhibition and motor inhibition). Methods:K-WISC III and EF test(Go-No-Go, Stroop test, Tower of Hanoi, Digit) was administered to both 25 children with ADHD and 25 normal control participants, all aged between 7 and 12. The results were analyzed after statistically controlled for age and intelligence. Results:Children in the ADHD group had significantly lower IQ score than those in the control group and consistent relations were found between the child's age and the study's major variables. Once IQ and age were controlled, results indicated that children with ADHD had deficit only cognitive inhibition and motor inhibition. There was no significant difference in planning and working memory. Conclusion:These results suggested that specific deficits in inhibition control rather than general EF deficits are associated with ADHD. So inhibition is the core problem of children with ADHD. Thus, the therapeutic approach focused on cognitive inhibition and motor inhibition is required.

Biological Control of Powdery Mildew by Antibiotic-producing Microorganisms Antagonistic to Erysiphe graminis

  • Seventy four microorganisms, which have antagonistic activity against to Fusarium culmorum, were tested for their inhibitory effect on colony development of obligate biotroph Erysiphe graminis f. sp. hordei Marchal, the causal agent of powdery mildew on barley plants. Of these, 13 actinomycetes isolates were shown to reduce the colony development of mildew completely by application of their 10% cell-free culture filtrates on barley leaves. An Isolate, A252, was the most powerful antagonist and its antifungal activity was further assessed. The colony development of mildew was significantly reduced by application of the 1% cell-free culture filtrate of isolate A252. In comparison to the control, the protective and curative application of 10% cell-free culture filtrate from A252 showed 88.5% and 96.1% reduction of colony numbers respectively. By the protective application, 68.3% of the inhibition was observed after 9 days of treatment, thus showed prolonged inhibitory effect. In vitro test, complete inhibition of the mycelial growth of Microdochium nivale was achieved by the treatment of 1% A252 culture filtrate and 80.2% of inhibition was observed by the 0.1% treatment.

Fermentation Characteristics and anti-Helicobacter pylori Activity of Aqueous Broccoli Fermented by Lactobacillus plantarum MG208

  • Helicobacter pylori infection causes gastrointestinal diseases such as chronic gastritis, peptic ulcers, and may lead to gastric cancer. Several studies have reported that lactobacilli present on broccoli show inhibitory activity against H. pylori. Here, we evaluated aqueous broccoli, fermented by Lactobacillus plantarum MG208, for its fermentation characteristics and anti-H. pylori activities including antibacterial activity, growth inhibition, anti-adhesion, and urease inhibition. The results indicated that the fermentation characteristics changed significantly depending on the amount of aqueous broccoli used for fermentation (p <0.05). There was no significant difference between the samples before fermentation (p >0.05). However, a significant concentration-dependent difference was noted in antibacterial activity and urease inhibition (p <0.05) following the addition of aqueous broccoli. Growth inhibition in the 10 mg/mL sample was significantly higher as compared to the negative control and similar to that with amoxicillin (positive control) (p <0.05). Anti-adhesion activity of aqueous broccoli was also significantly different (p <0.05) from the negative control. Therefore, aqueous broccoli fermented by L. plantarum MG208 could prove useful as a functional diet for protection of the gastric environment against H. pylori infection.

Antioxidative Properties of Amaranth Cauline Leaf and Suppressive Effect against CT-26 Cell Proliferation of the Sausage Containing the Leaf

  • The study investigated antioxidative properties and rectal cancer cell inhibition effect of amaranth (Amaranthus cruentus L.) cauline leaves (ACL) to produce the sausage with ACL powder (ACLP). Antioxidative effects of ACLP prepared with different stem lengths (10-45 cm) were evaluated through DPPH, ABTS, reducing power, total phenol, and total flavonoid. Inhibition effect on rectal cancer cells growth was also examined with CT-26 cell. To determine appropriate ACL amounts in sausage formula, response surface methodology was used. The sausages without ACL (control) and the sausage with ACL (ACLP sausage) were the subjected to the examinations of antioxidation, growth inhibition on CT-26, and physicochemical properties (pH and water content). ACLP made from the leaf with 15 cm length stem generally showed the highest antioxidative effect through results of DPPH, ABTS, reducing power, total phenol, and total flavonoid. ACLP also showed inhibition effect on the proliferation of CT-26, depending on concentration of ACLP. The surface response model showed that 4.87 g of ACLP was optimized amount for sausage production. Physicochemical properties between optimized ACLP and control sausages were not significantly different. Higher antioxidative effect of optimized ACLP sausage extract was observed (p<0.05) in antioxidation tests than control sausage extract except for DPPH. Cell viability of CT-26 cells were higher (p<0.05) in ACLP than in control sausage extracts. These results indicate that ACLP has functional effects on antioxidation activity and growth inhibition on CT-26 cell, and thus, it should be useful as a supplement in sausage, which may some effect as ACLP itself.

Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs (한약재 복합 추출물이 항산화 및 항노화에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.
