Expression and Purification of Human Farnesoid X Receptor-Ligand Binding Domain as Soluble Form Using a Dual Cistronic Expression Vector

  • In this study, we show the expression and purification of the human recombinant farnesoid X receptor (FXR)- ligand binding domain (LBD) protein in E. coli using a double cistronic vector, pACYCDuet-1, as a soluble form. We describe here the expression and characterization of a biologically active $FXR-LBD_{(248-476)}$. When expressed in the influence of bacterial promoters ($P_{T7}$ and $P_{Tac}$) of the single cistronic expression vectors, the human recombinant $FXR-LBD_{(248-476)}$ was found to be totally insoluble. However, by using a double cistronic expression vector, we were able to obtain the human recombinant $FXR-LBD_{(248-476)}$ in a soluble form. To allow for biological activities, we have subcloned into the pACYCDuet-1 vector, expressed in E. coli cells at some optimized conditions, and purified and characterized the human recombinant active $FXR-LBD_{(248-476)}$ proteins using the fluorescence polarization assay. This suggests that the expression of FXR-LBD in a double cistronic vector improves its solubility and probably assists its correct folding for the biologically active form of the proteins. We suggest that this may represent a new approach to high expression of other nuclear receptors and may be useful as well for other classes of heterodimeric protein partners.

Analysis of Influence by soil water-characteristic curve and permeability-suction relationship for the water flow in unsaturated soil (함수비 특성곡선과 투수계수 특성곡선이 불포화토내에서의 물의 흐름에 미치는 영향분석)

  • Water flow in unsaturated soils is affected by two mathematical equations called soil water-characteristic curve and permeability-suction relationship. Soil water-characteristic curve is an equation showing volumetric water content-suction relationship. Many researchers have presented equations for the relationships. This paper illustrates the importance of correctly determining the two relationships when analyzing unsaturated water flows. Results from two methods, Gardner (1958) and Fredlund et al. (1994), are used for comparison purposes. Numerical simulations of water flow by finite element method are performed using the two methods. The results by the numerical simulations are compared with the field data which was obtained from time-domain reflectometry (TDR) probes in Delaware County, Ohio. This data was obtained by the Seasonal Instrumentation Program which is included as a part of the Strategic Highway Research Program (SHRP).

A Study on the Analysis of the Microstrip Line by Using Inner Source at the FDTD Method (유한차분 시간영역 해석법에 내부전원을 이용한 마이크로스트립 선로 해석에 관한 연구)

  • When continuous and discontinuous microstrip is analyzed with FDTD(Finite Difference Time Domain) method, we used Berenger's 3D-PML as absorbing boundary condition, and IST(Inner Source Technique) was used for source excitation instead of front excitation that is existing method. In the case using IST, we have observed that analyzed characteristic is not affected by the reduced computational domain of the side and top face in which evanescent field and radiation field is exist. Also, if we control the position of the inner source, we could effectively reject the influence of the reflective wave by mean of imperfective boundary condition. In this paper, by using IST, we have calculated dispersive characteristic and characteristic impedance of the microstrip. And we have calculated magnitude and phase of the scattering coefficient, and obtained equivalent circuit of the open microstrip end.

  • As development of personal devices have made everyday use of internet much easier than before, it is getting generalized to find information and share it through the social media. In particular, communities specialized in each field have become so powerful that they can significantly influence our society. Finally, businesses and governments pay attentions to reflecting their opinions in their strategies. The stock market fluctuates with various factors of society. In order to consider social trends, many studies have tried making use of bigdata analysis on stock market researches as well as traditional approaches using buzz amount. In the example at the top, the studies using text data such as newspaper articles are being published. In this paper, we analyzed the post of 'Paxnet', a securities specialists' site, to supplement the limitation of the news. Based on this, we help researchers analyze the sentiment of investors by generating a domain-specific sentiment lexicon for the stock market.

Performance Analysis of Cost-Effective Handoff Scheme in PMIPv6 Networks with DNS Supporting (PMIPv6 네트워크에서 DNS기반의 비용효과적인 핸드오프 기법의 성능분석)

  • Proxy Mobile IPv6 (PMIPv6) is designed to provide a network-based localized mobility management protocol, but it does not handle the global mobility of hosts. In this paper, we propose a location management scheme based on Domain Name System (DNS) for PMIPv6. In this proposed scheme, DNS as a location manager provides PMIPv6 for global mobility. In addition, a paging extension scheme is introduced to PMIPv6 in order to support large numbers of mobile terminals and enhance network scalability. To evaluate the proposed location management scheme, we establish an analytical model, formulate the location update and the paging cost, and analyse the influence of the different factors on the total signalling cost. The performance results show how the total signal cost changes under various parameters.

  • Similar to the essential components of many mechanical systems, the geometrical properties of the teeth of spiral bevel gears greatly influence the kinematic and dynamic behaviors of mechanical systems. Logarithmic spiral bevel gears show a unique advantage in transmission due to their constant spiral angle property. However, a mathematical model suitable for accurate digital modeling, differential geometrical characteristics, and related contact analysis methods for tooth surfaces have not been deeply investigated, since such gears are not convenient in traditional cutting manufacturing in the gear industry. Accurate mathematical modeling of the tooth surface geometry for logarithmic spiral bevel gears is developed in this study, based on the basic gearing kinematics and spherical involute geometry along with the tangent planes geometry; actually, the tooth surface is a parametric surface defined on a parallelogrammic domain. Equivalence proof of the tooth surface geometry is then given in order to greatly simplify the mathematical model. As major factors affecting the lubrication, surface fatigue, contact stress, wear, and manufacturability of gear teeth, the differential geometrical characteristics of the tooth surface are summarized using classical fundamental forms. By using the geometrical properties mentioned, manufacturability (and its limitation in logarithmic spiral bevel gears) is analyzed using precision forging and multiaxis freeform milling, rather than classical cradle-type machine tool based milling or hobbing. Geometry and manufacturability analysis results show that logarithmic spiral gears have many application advantages, but many urgent issues such as contact tooth analysis for precision plastic forming and multiaxis freeform milling also need to be solved in a further study.

Analysis of the monopole antenna characteristcs of handy phone using Finite Difference Time Domain(FDTD) Method (시간영역 유한차분법을 이용한 휴대용 전화기의 모노폴 안테나 특성해석)

  • The broadband input impedance, the input power and the radiation pattern of the monopole antenna attached to the handy phone operated at 800MHz are calculated by using the Finite Difference Time Domain(FDTD) Method. For the FDTD analysis of frequency characteristics of monopole antenna, the handy phone is modeled with the geometry that the monopole antenna is connected to a conducting box, and the modified FDTD algorithm[11] used the thin wire appproximation method and the Maxwell's integral equation from the original Yee algorithm is applied for the analysis of the wire structure. Also, by means of finding the current distribution directly from circumferencial magnetic filelds around the monopole antenna and the conducting box, the radiation pattern is calculated to observe the influence of the conducting box, and is compared with the results of the known mothod for the FDTD calculation of radiation pattern, For the experiments, the handy phone of which full length including antenna is .lambda. $\lambda$/2 is manufactured and we confirm that all computation results are agree well with the mea- sured values.

  • With the rapid and wide-spread use of mobile communications much attention has been focussed on propagation in the urban area crowed with buildings. It is often surrounded by hills, forests, and mountains. The importance of surface scattering interference between transmitters and receivers on the rough surfaces has been interested and investigated. Therefore, a prediction method is necessary to estimate the influence of rough surfaces on microwave radio propagation. Moreover, most of the mobile communications are performed based on the digital communication system rather than the analog one. In this case, we must pay more careful attention to the signal delay caused by the phase delay due to the multi-path propagation. In this paper we have analyzed numerically scattering of electromagnetic waves from Composite structures by using FVTD (Finite Volume Time Domain) method. We consider two different types of rough surfaces such as periodic and composite structures.

  • Nowadays, industry of smart grid is important for practical use of the renewable energy. In this situation, it is important to use the energy storage to make more stable and efficient renewable energy sources. The power conditioning systems consist in a boost converter which makes renewable energy source connected with the grid-connected inverter and the charger/discharger which takes the energy transfer between the boost converter and an energy storage. The effects on the controller design of each converter must be investigated to avoid the instability of the entire system. small-signal modelling of the boost converter and charger/discharger have been done and a controller design example is also presented. In this paper, effects on the controller design of the boost converter and the charger/discharger are investigated according to the existence of the parasitic resistance of the boost converter. In conclusion, the parasitic resistance of the inductor should be considered from the aspect of both the frequency domain analysis and time domain simulation using both MATLAB and PSIM.

Domain decomposition technique to simulate crack in nonlinear analysis of initially imperfect laminates

  • In this research, an effective computational technique is carried out for nonlinear and post-buckling analyses of cracked imperfect composite plates. The laminated plates are assumed to be moderately thick so that the analysis can be carried out based on the first-order shear deformation theory. Geometric non-linearity is introduced in the way of von-Karman assumptions for the strain-displacement equations. The Ritz technique is applied using Legendre polynomials for the primary variable approximations. The crack is modeled by partitioning the entire domain of the plates into several sub-plates and therefore the plate decomposition technique is implemented in this research. The penalty technique is used for imposing the interface continuity between the sub-plates. Different out-of-plane essential boundary conditions such as clamp, simply support or free conditions will be assumed in this research by defining the relevant displacement functions. For in-plane boundary conditions, lateral expansions of the unloaded edges are completely free while the loaded edges are assumed to move straight but restricted to move laterally. With the formulation presented here, the plates can be subjected to biaxial compressive loads, therefore a sensitivity analysis is performed with respect to the applied load direction, along the parallel or perpendicular to the crack axis. The integrals of potential energy are numerically computed using Gauss-Lobatto quadrature formulas to get adequate accuracy. Then, the obtained non-linear system of equations is solved by the Newton-Raphson method. Finally, the results are presented to show the influence of crack length, various locations of crack, load direction, boundary conditions and different values of initial imperfection on nonlinear and post-buckling behavior of laminates.
