Antioxidant Activities of Rumex crispus Extracts and Effects on Quality Characteristics of Seasoned Pork (소리쟁이 추출물의 항산화 활성과 이를 첨가한 양념돈육의 품질특성)

  • Antioxidant activities of ethanol extracts of Rumex crispus (RC) were investigated in vitro. Contents of total polyphenols and total flavonoids in RC ethanol extracts were $108.24{\pm}3.29\;{\mu}g/mg$ and $33.06{\pm}2.24\;{\mu}g/mg$, respectively. The antioxidant capacities of RC extracts were high. The RC extracts were capable of directly scavenging DPPH free radicals by acting as reducing agents. This study was also conducted to evaluate the quality characteristics of seasoned pork treated with RC extracts during storage at $4^{\circ}C$ for 21 days. Seasoned pork was produced containing 0, 1, and 3% RC extracts and 0.1% BHA. The DPPH scavenging effect of the seasoning sauces was increased by the addition of RC extracts. The total bacteria and TBARS values of seasoned pork with RC extracts were decreased compared to control, whereas coliform bacteria were not detected after longer storage periods. With regards to sensory evaluation, seasoned pork treated with 1% RC had stronger odor, juiciness, and tenderness compared to seasoned pork. These results suggest that it is possible to manufacture seasoned pork with RC extracts for the improvement of shelf-life and quality.

The Effect of Long-term Application of different Organic Material Sources on Soil Physical Property and Microflora of Upland Soil (유기물원이 다른 퇴비연용이 밭토양의 물리성 및 미생물상 변화에 미치는 영향)

  • The objective of this study was to determine the effects of various kinds of composts on the change of soil physical properties and microorganism in upland soils. Field experiments were conducted in the loam and sandy loam soils, while the clay loam and sandy loam soils were used for laboratory experiments. Various kinds of composts such as poultry manure compost(PMC). cow manure compost(CMC). human excrement sludge(HES), and food industrial sludge compost(FISE) were applied annually at rates of 0, 40, and $80Mg\;ha^{-1}$ to soils grown with soybean and maize plants for 4 years during 1994 to 1997. The results of this study were as follows : Bulk density of loam soil decreased with compost application to $1.07{\sim}1.32Mg\;m^{-3}$ compared with $1.49Mg\;m^{-3}$ of control plot, while in sandy loam soil it decreased to $1.00{\sim}1.20Mg\;m^{-3}$ compared with $1.25Mg\;m^{-3}$ of control plot. Bulk density of soil was decreased according to maize cultivation compared with bare control, but soybean cultivation was similar. Population of organic material decomposing microorganisms was increased rapidly at the initial incubation stage at $25^{\circ}C$, and increased more sensitively at the loam soil than sandy loam soil. In the case of the change of microorganisms associated with nitrogen circulation, ammonia oxidizing bacteria was more at the initial incubation stage, and denitrifying bacteria was more at the initial incubation stage, and denitrifying bacteria increased until 1~4 weeks after incubation and increased more at the loam soil than sandy loam soil.

  • Thermal treatment and ozone oxidation methods were examined to reuse waste activated sludge (WAS) produced from a livestock wastewater treatment plant. Analysis of WAS property was made to study usefulness of the recycled waste as fertilizer. From the results of quantitative analysis, WAS particles were found to be composed of 44.25 wt% carbon, 8.43 wt% nitrogen, and 1.35 wt% phosphorus. It was confirmed that the inactivation of pathogenic microorganism was required from the quantitative analysis of microbes. From the results of TSS, COD, SCOD, and pathogenic microorganism measurement, the optimal operating conditions of thermal treatment and ozone oxidation were determined to be 70, 10 min and $0.6L\;O_3/L\;solution{\cdot}min$, 60 min, respectively. The optimized thermal treatment and ozone oxidation represented the efficient pathogen inactivation and particle dissolution, respectively. However, the two methods examined were not themselves sufficient but they need to combine with another treatment for the effective reuse of wastes.

Effect of Chitosan-Ascorbate and Morea(roasted of oyster shell at $1300^{\circ}C$) on Growth of Contaminating Bacteria in Dombaeki(traditional shark dish) during Storage (돔배기의 저장 중 오염미생물의 생육에 미치는 키토산-아스코베이트 및 모려의 처리효과)

  • The effects of 0.01%(w/v) chitosan-ascorbate(CA) and 10 ppm morea on the number of total microbes, Escherichia coli levels, and growth of food poisoning bacteria in dombaeki during storage at $10^{\circ}C$ over 6 days were investigated. Total microbes in meat, cartilage, and skin of untreated samples increased by 4.24, 3.81, and 2.20 logs compared to the zero timepoint, respectively, but, in CA-treated samples, counts fell by 2.66, 2.37, and 1.24 logs. Total microbial levels in morea-treated meat, cartilage, and skin showed similar tendencies but the effects were slightly less than seen in CA-treated samples. E. coli numbers in CA-treated meat, cartilage, and skin stored for 6 days decreased by 1.69, 1.25, and 1.52 logs respectively, compared with control samples. Morea-treated samples showed similar falls, but the effects were again slightly less than seen after CA-treatment. Both Salmonella and Vibrio parahaemolyticus were detected in untreated meat stored for 3 or 6 days. Food poisoning bacteria were found in both untreated and morea-treated samples stored over 6 days. However, no such bacteria were detected in CA-treated samples. Also, CA-treated meat, cartilage, and skin showed low degrees of degeneration. Thus, CA treatment enhanced shelf-life and dombaeki quality by inhibiting microorganism growth and tissue breakdown during storage.

  • As the concerns about environmental problems, climate change and limited fossil resources increase, bio-based production of chemicals and polymers from renewable resources gains much attention as one of the promising solutions to deal with these problems. To solve these problems, much effort has been devoted to the development of sustainable process using renewable resources. Recently, many chemicals and polymers have been synthesized by biorefinery process and these bio-based chemicals and plastics have been suggested as strong candidates to substitute petroleum-based products. In this review, we discuss current advances on the development of metabolically engineered microorganisms for the efficient production of bio-based chemicals and polymers.

Characterization of Double-layer Optical Sensing Membranes for Dual Sensing of Dissolved Oxygen Concentrations and pH and Their Application to the On-line Monitoring of Fermentation Processes (용존산소농도와 pH의 동시 검출용 이중층 광학 센서막의 특성 및 발효공정의 온라인 모니터링에의 응용)

  • A double-layer optical sensing membrane has been fabricated to measure the concentration of dissolved oxygen (DO) and pH value simultaneously. (tris(4,7-diphenyl-1,10-phenanthroline (Rudpp) ruthenium(II)) as a DO sensitive dye has been mixed in the methyl trimethoxy silane (MTMS) sol-gel solution and coated onto one well in a 24-well microtiter plate. On the DO-sensing layer the GA (3-glycidoxypropyl trimethoxy silane (GPTMS), 3-aminopropyl trimethoxy silane (APTMS)) sol-gel solution mixed with 8-hydroxypyrene-1,3,6-trisulfonic acid trisodium salt (HPTS) has been coated and used to measure pH values. The double-layer sensing membrane was affected by ionic strength and temperature. The double-layer sensing membrane for DO and pH has been applied to online monitor in microorganism cultivation processes and showed a good performance.

Effect of Replacing Concentrate with Mugwort Pellet on Nutrient Utilization and Ruminal Fermentation Characteristics in Sheep (쑥 Pellet이 면양의 영양소 이용율과 반추위내 발효특성에 미치는 영향)

  • This study was carried out to investigate the effect of replacing dried mugwort (Artemisia montana Pampan) in concentrate a time of preparation pellet. The treatments, arranged in a 4${\times}$4 Latin square design, were four additional levels of mugwort at 0, 3, 5 and 10% in concentrate. Four crossbred (CorriedalexPolwarth, t) sheep with a mean body weight of 41.3 kg were used to evaluate nutrients digestibility, palatability, fermentation characteristics and microbial protein synthesis in the rumen. The digestibility of crude protein was improved (p < 0.05) to 6.1 % - 8.6 % in sheep fed 3, 5 and 10% mugwort pellet treatments compared with that of control. That of crude fat and NDF was improved (p < 0.05) to 5.8 % - 7.3 % in sheep fed 3 % compared to other treatments. The ruminal pH was significantly (p < 0.05) decreased in sheep fed 3 % mugwort pellet compared to other treatments when observed at 0.5 hour after feeding. The ammonia nitrogen concentrations were the highest in sheep fed all treatments at 1 hour after feeding. The ruminal concentrations of acetic acid and propionic acids were an improvement (p < 0.05) at the 3% and 5% treatments. Retained nitrogen of 3, 5 and 10% treatment with the value of 2.24 - 2.82 g was higher (p < 0.05) than that of the control with 0.78 g and microbial protein production of 10% treatment was higher (p < 0.05) than that of control. This study suggested that the replacing with 3% dry mugwort (Artemisia montana Pampan) in concentrate a time of preparation pellet will improve nutrient digestibility, palatability, ruminal fermentation characteristics and feed value.

Effect of Mugwort Processing Types on in vivo Digestibility and Ruminal Fermentation Characteristics in Sheep (쑥(Artemisia sp.)의 가공방법이 면양의 소화율과 반추위내 발효특성에 미치는 영향)

  • This study was conducted to examine the nutrient digestibility and ruminal fermentation characteristics in sheep fed dried mugwort and mugwort silage for 5% levels of rice straw in the basal diet, and mugwort pellet for 5% levels of concentrate in the basal diet. For the experiment, they were given a basal diet containing of rice straw and concentrate mixed at a 3: 7 ratio (DM basis). The treatments were designed as a 4 ${\times}$ 4 Latin square design with four sheep (50.2 kg body weight). The digestibility of crude protein was increased (p < 0.05) to 4.6 - 6.2 % in sheep fed mugwort silage treatments (60.23 %) compared with those of control (54.08 %) and dried mugwort treatment (55.67 %). That of ether extract was iicreased (p < 0.05) to 4.8 - 8.8 % in sheep fed mugwort silage treatments (80.22 %) compared with those of control (71.47 %) and dried mugwort treatment (75.46 %). In the dry matter intake, mugwort silage treatment (904.44 g) was the hightest and mugwort pellet treatment, dried mugwort treatment and control were 810.66 g, 780.66 g and 742.18 g, respectively. The ruminal pH in all treatments were rapidly decreased (p < 0.05) at 0.5 and 1 hour after feeding and slowly increased at 2, 4 and 8 hours after feeding, especially mugwort silage treatment. The ammonia nitrogen concentrations were the highest (p < 0.05) in sheep fed mugwort silage treatment (11.24 - 12.05mg / 100 rnz) at 0.5 and 2 hours after feeding. The ruminal concentrations of acetic acid (6.06 mmol /100 $m\ell$) and propionic acid (2.35 mmol/ 100 mz) were an increased (p < 0.05) at the mugwort silage treatments at 1 and 2 hours after feeding. Purine derivatives out put (13.41 mmol / d) and microbial protein production (11.61 mmol / d) were increased (p < 0.05) compared with those of control (5.42 and 4.93 mmol / d).

A Study on Airborne Microorganism in Hospital (일부 병원 실내에서의 공기중 미생물 오염에 관한 연구)

  • To assess biological indoor air quality in hospital, concentrations of viable airborne microbes were determined at intensive care unit(ICU), patient room (PR), outpatient waiting room(OPWR) in hospitals of large(1000 beds), middle(500 beds), small(100 beds) hospitals, respectively. Gram positive bacteria, gram negative bacteria, fungi were sampled using suctional sampling method by RCS sampler (Reuter centrifugal air sampler) and RCS GK-A agar plate. In gram positive bacteria groups, CNS(Coagulase Negative Staphylococcus), Micrococcus, Lactobacillus, S. aureus, Enterococcus, St. viridans identified. In gram negative bacteria groups, A. baumannii, Kl. peumoniae and E. coli were identified, and Penicillium was identified in fugi groups. Results of the study were as follows. 1. The highest concentrations of airborne microbes was $971CFU/m^3$ at 5:00 PM in small hospital patient room, and average concentrations of airborne microbes in large, middle and small hospitals were $282CFU/m^3$, $289CFU/m^3$ and $625CFU/m^3$, respectively. Average concentrations of airborne microbes in office(control) was $90CFU/m^3$. Thus, the small hospital showed the worst condition. 2. Representatives of 8 different genera were identified in 150 samples. The most frequently isolated organisms were Staphylococcus (73.0%), Micrococcus (20.7%) and Lactobacillus (4.7%), respectively. Pathogenic microbes isolated were A. baumannii, E. coli, Enterococcus, Kl. peumoniae, S. aureus, St. viridans and Penicillium as fungi. In office, no pathogenic microbes were identified. Average concentrations of airborne pathogenic microbes in large, middle and small hospital were $5CFU/m^3$ (2%), $11CFU/m^3$ (4%) and $12CFU/m^3$ (2%), respectively. Thus, condition in a large hospital was better than those in a middle and a small hospital.

A Study on the Development of Microorganism Test Strips (대장균군 검사용 간이 시험지 개발)

  • The objective of this study was to develop a paper strip which could determine E. coli qualitatively and quantitatively in water, wastewater, drinks, or food. This paper strip method was a simple and rapid test method that determine E. coli by visual identification. In this study, nutrient culture media were formulated and characterized for optimum conditions. Paper strips were then prepared by impregnating into the media and dried at $60^\circ$C. The test procedure is quite simple to use. The paper strip was dipped into a sample, and excess sample was removed. The strip was then incubated at $37^\circ$C for 16 to 20 hours and the number of colonies on the strip was counted. The color of the colony spots produced by microorganisms varied depending on the media formulation. Violet-red spots were produced by E. coli. The test method was simple, rapid and no special laboratory equipment was necessary for visual identification. Therefore, this test method is applicable to on-site tests such as field tests or home tests. The paper strip method was compared with the standard agar plate method and Japanese commercial product. The method of the economical preparation of test strips was studied for production on industrial scale.
