The effects of media, growth regulators, low-temperature treatments, culture temperature and light were investigated to improve the callus induction and growth in the anther culture of Comus officinalis Sieb. et Zucco. The frequency of callus induction was more effective on WPM medium than MS medium, and it was highest as 54% in WPM medium supplemented with Img/L NAA. Callus growth was stimulated on MS medium supplemented with 2mg/L NAA. Effect of temperature and light on the callus induction and growth was highest as 62% in the treatment for 16/8 hrs. (light/dark) at $25^{\circ}C$ Effect of low - temperature treatment on callus induction was highest as 19. 5% in the treatment for 36 hrs. at $4^{\circ}C$. For organization, green cells and rootings were promoted on MS medium supplemented with O. 5mg/L 2,4-D and 1 mg/L kinetin. The prevention of callus browning was effective on the medium containing $3{\sim}5mg/L$ ABA or 5mg/L $AgNO_3$. The supplement of ABA or $AgNO_3$,were maintained callus activity for 4-5 weeks and they were promoted the development of green cells.
In maize, immature embryos (IEs) are highly regenerative explants most suitable for producing high frequencies of plantlet regeneration in vitro. Apart from media, explants, and hormones, genotypic variation also influences in vitro characters to a great extent. In the present study, IEs were used to study the distinctive effect of variation of size/stage and hormones in different genotypes on five in vitro characters viz., frequency of callus induction, growth rate of total callus, frequency of E. callus induction, and volume and number of regenerated plantlets. LS medium with different concentrations of 2,4-D (0.5, 1.5, 2.5, 4.0 and 5.0 mg/L) were used to study the former four in vitro characters, and medium with 6-benzylaminopurine and kinetin (0.5 mg/L, each) was used for plantlet regeneration. IEs of 1.0, 1.5, 2.0, 2.5 and 3.0 mm in size were isolated from four inbred lines viz., NM74C, NM81A, NM5883 and NM5884. Two-way ANOVA revealed that explant size and genotypes, as well as hormonal concentrations showed significant effects on in vitro characters. Two millimeter IEs were found to be suitable for in vitro cultures. LS medium with 1.5 mg/L 2,4-D and LS with BAP and Kn (0.5 mg/L, each) were found to be the best hormonal concentrations for callus induction, maintenance, and regeneration, respectively. Among the four genotypes, NM81A and NM5883 yielded more non-embryogenic and Type I E. calli. In contrast, NM74C and NM5884 yielded more highly regenerative Type II calli. Inbred line NM5884 was found to be the best among these four genotypes.

The objective of this study was to optimize the condition for induction and proliferation of callus from Rhus chinensis Mill. and investigate the skin-brightening effect of Rhus chinensis callus (RCC). It was confirmed that the most proper plant growth regulator (PGR) for callus induction is 1.0 mg/L of 2,4-dichlorophenoxyacetic acid (2,4-D). The most optimal condition of PGR, medium and additives for callus proliferation were 2,4-D (1.0 mg/L), MS medium and citric acid, respectively. Inhibitory activities of tyrosinase were higher at 50 and 100 ㎍/mL of RCC extracts (41.86 and 75.56%, respectively) than arbutin (27.32%). As the results of measuring melanin inhibition in B16F1 melanocyte and B16F10 melanoma cell, RCC extracts increased its inhibitory activities concentration-dependently, and were found to have higher whitening effect than arbutin at a concentration of 100 ㎍/mL. Therefore, it is suggested that RCC can be used as an effective material for skin-brightening cosmetics.
In order to optimize tissue culture conditions for genetic transformation of orchardgrass (Dactylis glomerata L.), the effects of culture medium supplements on tissue culture responses were investigated with mature seeds of a cultivar, 'Roughrider', as explant tissues. The optimal concentration of 2,4-D for the induction of embryogenic callus from mature seeds was 3 mg/L. Plant regeneration frequency was 36.3% when embryogenic calli were cultured on the regeneration medium supplemented with 1mg/L 2,4-D and 3mg/L BA. Addition of 1 g/L casein hydrolysate and 300 mg/L L-proline improved frequencies of embryogenic callus induction and plant regeneration up to 57.3 and 60.7%, respectively. Supplementation of the media with 10 mga $\textrm{AgNO}_3$ and 40 mg/L cysteine enhanced frequencies of callus induction and plant regeneration. Efficient regeneration system established in this study will be useful for molecular breeding of orchardgrass through genetic transformation.

In order to develop an efficient, reliable and reproducible tissue culture system for reed (Phragmites communis Trinius), an efficient plant regeneration system via callus induction was established using mature seeds as explants. MS medium containing 1 mg/L 2,4-D and 0.5 mg/L BA was optimal for callus induction from mature seeds. The highest frequency (88.7%) of callus formation was obtained in 1.0 mg/L 2,4-D. The highest plant regeneration frequency (59.6%) was found when the embryogenic calli were subcultured on MS medium supplemented with 100 mg/L myo-inositol, whereas, adding of plant growth regulators was not so promising in this case. Our result would be useful for development of transgenic reed plants.
The internal structures of Arabidopsis thaliana infected with beet curly top virus (BCTV) were studied by light microscopy. Hyperplasia was observed in the inflorescence stems of Arabidopsis thaliana ecotype Sei-O at 2 weeks after BCTV-Logan inoculation and callus was induced on symptomatic tissues at 4 weeks after virus inoculation. The infection processes were revealed as follows: hyperplasia of phloem tissue, necrosis of hyperplastic phloems, lacuna formation of necrotic tissues, elongation and enlargement of cortex and epidermal cells surrounding the lacuna formed phloem tissues, induction of cell division in the enlarged cortex and epidermal cells, and induction of callus tissue. Callus formation on Arabidopsis was caused by the virus infection, and virus inclusion body was observed in both phloem and callus tissue by azure-A staining.
In spite of potential benefits of anther culture, low productivity of plant regeneration in some genotypes; e.g. tonsil and indica rice, is one of the major obstacles for practical use of anther culture. This study was conducted to improve cold shock method and carbohydrate source for increasing the efficiency of anther culture in rice. The most common carbon source, sucrose was replaced to maltose, which has two molecules of glucose. Maltose increased callus induction 1.4-to 1.8-fold higher in japonica rice, 3.2-to 11.6-fold in tongil types and 2.7-fold in indica rice IR50. Callus induction was increased from 0.2% to 12.5% in maltose medium compared to the medium supplemented with sucrose plus glucose in indica rice "Tetep". A simple procedure of vacuum packaging of panicles during cold shock treatment prolonged not only anther viability more than 15 days but also increased callus induction more than 2-fold compared to open-air storage (conventional method). Combining of above two methods, callus induction was increased 28 to 56% in japonica, 13 to 33% in tonsil type and 12 to 31% in indica rice. Plant regeneration was increased 14 to 35% in japonica, 10 to 20% in tonsil and 4 to 15% in indica rice, respectively.

To obtain regeneration system of onion, we analyzed the effects of 2,4-D and BA concentration on the embryogenic callus induction from mature embryos. The highest embryogenic callus induction ratio was shown on MS medium (Murashie and Skoog 1962) containing $2.5\;\cal{mg/L}\;or\;5\;\cal{mg/L}$ picloram after mature embryos were placed on medium. When induced callus were cultured on half strength of MS medium containing $1\;\cal{mg/L}$ Kinetin, the highest shoot formation ratio was observed on MS medium containing $1\;{mg/L}$ 2,4-D and $1\;{mg/L}$ BA. Embryogenic callus were cultured in MS liquid medium containing $1\;\ccal{mg/L}$ of 2,4-D and $1\;\cal{mg/L}$ BA. The suspension cultured cell clumps could be mass propagated. Embryogenic callus were friable, but non-embryogenic callus included a lot of moisture, hence the identification between embryogenic and non-embryogenic callus as easily achieved. When embryogenic callus as cultured on half strength of MS medium containing $1\;\cal{mg/L}$ Kinetin, shoots were induced. The whole plantlet was obtained on rooting medium containing $0.5\;\cal{mg/}$ of NAA.
This study was conducted to determine the appropriate culture conditions for developing photoautotrophic cells from tobacco using plant growth regulators, sucrose and light condition in LS medium. The greatest callus induction was observed in the LS medium supplemented with $10^{-5}$ M NAA and $10^{-4}$ M BA, showing 3.08 g of callus fresh weight determined at 30 days after incubation. The highest chlorophyll content of callus was obtained in the LS medium supplemented with $10^{-5}$M NAA and $10^{-6}$ M BA, showing 28.42${\mu}g/g$. In both light and dark conditions, callus induction increased as the concentration of sucrose increased from 0.5%to 3.0%. In particular, in the light condition, the greatest callus induction was made in the LS medium supplemented with 2% sucrose. On the other hand, the heighest chlorophyll content was observed at 0.5% sucrose and chlorophyll wasn't induced in dark condition. The chlorophyll content of callus cultured in LS medium containing 0.75% sucrose was similar to 1% sucrose, as far as the chlorophyll content of callus was concerned.
Rhodiola sachalinensis has been used as a traditional medicine in Asia. We were germination in vitro seedling of grow naturally in Chang bai Moutain. And callus induction from leaf segments, treatmented plant regeneration in plant growth regulators (Auxins and cytokinins). We investigated optimal conditions for efficient plant regeneration through callus induction and shoots formation on medium with various kinds of growth regulators. Callus induction and adventitious shoots formation was achieved when cytokinin and auxin combinated to this experiment. Especially, there was the highest callus induction rates when we were used to 1 mg/L kinetin and 2 mg/L NAA $(98\%)$, Adventitious shoots formation wear obtained difference rate when cytokinin alone 1 mg/L BA $(96.6\%)$. And regenerated plantlet was acclimatized and transplanted to the soil, showed $100\%$ survival.
