Callus Induction and Somatic Embryogenesis from Sicyos angulatus L. (야생식물 Sicyos angulatus L.로부터 캘러스 유도 및 체세포배 발생)

  • In order to investigate the possibility of in vitro mass propagation via somatic embryogenesis from Sicyos angulatus L., effects of plant growth regulators and carbon sources on callus induction and somatic embryogenesis were evaluated. Optimal combinations of plant growth regulator for callus induction from cotyledon and inflorescence explants were 2,4-D 2.0 mg/L + BA 0.1 mg/L and 2,4-D 1.0 mg/L + BA 0.1 mg/L in MS basal medium supplemented with sucrose 30 g/L,, respectively. Somatic embryogenesis was observed from cultured inflorescence explants, but it could not be achieved from leaf or cotyledon explants. The most effective plant growth regulators for somatic embryogenesis from callus was NAA 1.0 mg/L + kinetin 10 mg/L in the half strength of MS basal medium supplemented with 20 g/L sucrose.

Callus Induction and Plant Regeneration from Leaf Tissue. Culture of $\emph{Aralia elata}$ S. (두릅의 엽조직배양에 의한 Callus유기 및 식물체 재분화)

  • This study was conducted to determine the optimum culture conditions for inducing callus and regenerating plantlets from cultured leaf tissues of Aralia elata. Young leaf tissues(1cm) of A. elata plant were cultured on MS medium supplemented with 2, 4-D and Thidiazuron. Embryogenic callus was induced along the leaf veins, more efficiently on the medium containing 1.0mg /1 Thidiazuron in 4 weeks after culture initiation. Calli were subcultured to proliferate on MS media containing 2, 4-D, Dicamba, Picloram, and Thidiazuron. Callus was better proliferated on the medium containing Dicamba than on the others.. However, callus subcultured on the medium containing Thidiazuron was more embryogenic and light green-colored, of which some showed embryoid-like structure on the surface. Hormone-free medium was more efficient to regenerate plantlets than media supplemented with Kinetin, BA, and Thidiazuron.

  • In order to optimize tissue culture conditions for genetic transformation of zoysiagrass (Zoysia japonica Stued.), the effect of plant growth regulators and culture medium supplements on embryogenic callus induction from mature seeds of a cultivar 'Zenith' were investigated. The optimal concentration and treatment period of NaOCl is 30% (v/v) for 60 minutes. Cultivation of mature seed on the callus Induction medium containing 3 mg/L 2,4-D and 3 mg/L dicamba showed 17.5% of embryogenic callus formation frequency. Supplementation of 1 g/L casein hydrolysate and 500 mg/L L-proline improved frequency of embryogenic callus induction. Audition of the medium with 5 mg/L $AgNO_3$ and 20 mg/L cysteine enhanced frequencies of embryogenic callus induction. Efficient callus induction system established in this study will be useful for molecular breeding of Boysiagrass through genetic transformation.

Increase of isoflavones in soybean callus by Agrobacterium-mediated transformation

  • Plant secondary metabolites have always been a focus of study due to their important roles in human medicine and nutrition. We transferred the isoflavone synthase (IFS) gene into soybean [Glycine max (L.) Merr.] using the Agrobacterium-mediated transformation method in an attempt to produce transformed soybean plants which produced increased levels of the secondary metabolite, isoflavone. Although the trial to produce transgenic plant failed due to unestablished hygromycin selection, transformed callus cell lines were obtained. The induction rate and degree of callus were similar among the three cultivars tested, but light illumination positively influenced the frequency of callus formation, resulting in a callus induction rate of 74% for Kwangan, 67% for Sojin, and 73% for Duyou. Following seven to eight subcultures on selection media, the isoflavone content of the transformed callus lines were analyzed by high-performance liquid chromatography. The total amount of isoflavone in the transformed callus cell lines was three- to sixfold higher than that in control callus or seeds. Given the many positive effects of isoflavone on human health, it may be possible to adapt our transformed callus lines for industrialization through an alternative cell culture system to produce high concentrations of isoflavones.

Studies on the Tissue Culture of Korean-Ginseng III. Effects of NAA on the Callus Induction and Organ Differentiation trom Korean-ginseng Explants (고려인삼의 조직배양에 관한 연구 제3보 NAA가 인삼 Callus의 유기 및 기관의 변화에 미치는 영향)

  • These experiments were carried out to define the effects of NAA, 2, 4-D and Benzyladenine on the callus induction and the organ differentiation from the explants and to find out the vegetative propagation method of Korean ginseng. The results obtained are summarized as follows; 1. NAA was significantly effective in forming roots from the ginseng stem segment and the number of roots was increased by increasing NAA concentration in the medium. The roots were formed from both distal and proximal ends of the ginseng stem segments grown on the medium containing more than 2mg/L of NAA. 2. The amount of callus growth increased proportionatly with NAA concentration in the range of 4.0mg per liter in the medium. The callus was easly induced from stem segment than leaf segment and 2, 4-D was more effective in callus induction and growth than NAA. 3. The benzyladenine showed the significant inhibition effect in forming roots from ginseng explant. The callus was not induced with BA alone, but in BA and 2, 4-D or BA and NAA added medium, the callus was easily induced and its growth was also accelerated. The interaction effects between 2, 4-D and BA on the callus induction and growth were significantly higher than those between NAA and BA. 4. As the ginseng embryos were cultured on the M.S. medium supplemented with 2mg per liter NAA, number of shoots was significantly increased and the percentage of embryo which had shown more than 4 shoots later was 22.2%. On the medium containing 8mg per liter NAA, the ginseng embryo showed the normal growth of shoots and leaves, but increased roots and callus induction on the basal part of shoots. 5. When the shoots with 3 leaflets were cut in 1.5cm long and grown on the Blayde's medium containing NAA 1.0mg per liter, roots were formed at the proximal end of shoot, and a new ginseng seedling was successfully obtained.

Callus induction and plant regeneration of Iris dichotoma Pall. in endangered species

  • Iris dichotoma Pall. is an important endangered plant belonging to the family Iridaceae. A method was developed for the rapid micropropagation of I. dichotoma through plant regeneration from leaf, rhizome, and root explant-derived calli. Leaf, rhizome, and root segments were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (2,4-D; $0-3.0mg{\cdot}L^{-1}$) for callus induction. Callus production was highest at $1.0mg{\cdot}L^{-1}$ 2,4-D, where 73.8% and 45.5% of cultured rhizome and root cuttings, respectively, produced calli. The viable calli were maintained at an induced concentration of 2,4-D ($3.0mg{\cdot}L^{-1}$). They were then transferred to MS medium supplemented with various concentrations of 2,4-D ($0-3.0mg{\cdot}L^{-1}$) in combination with 6-benzyladenine (BA: 0, 1.0 and $3.0mg{\cdot}L^{-1}$) for adventitious shoot regeneration. The addition of a low concentration of 2,4-D into BA-containing medium significantly increased the frequency of shoot regeneration in leaf, rhizome, and root-derived calli. The highest number of adventitious shoots (26.4 per callus) formed at $0.5mg{\cdot}L^{-1}$ 2,4-D and 1.0 mg/l BA. For rooting of the shoots, half- strength MS medium supplemented with different concentrations of indole 3-butyric acid (IBA) $0-3.0mg{\cdot}L^{-1}$ was tested. The optimal results were observed using half-strength MS medium supplemented with $1.0mg{\cdot}L^{-1}$ IBA, on which 98% of the regenerated shoots developed roots with an average of 3.5 roots per shoot within 45 days. The plantlets raised in vitro were acclimatized and transferred to soil with 95% success. This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.

Effect of Orchardgrass Varieties on Callus Culture and Plant Regeneration (오차드그래스의 품종이 캘러스배양과 식물체 재분화에 미치는 영향)

  • In order to investigate the effects of genetic variations of orchardgrass in tissue culture response, calli were induced from mature seeds of eight varieties, 'Hapsung 2', '93E', 'Amba', 'Ambassdor', 'Frode', 'Frontier', 'Potomac' and 'Roughrider', and plant regeneration frequency was compared. Significant differences were observed among the varieties in both callus induction and plant regeneration. Callus induction rate of viable seeds varied from 24.3% to 71.7%. Plant regeneration frequency ranged from 76.6% to 29.7%. 'Roughrider' varieties showed higher regenerability with the frequency of 76.6%. These results can be used not only to provide additional improvements in the plant regeneration frequency from transgenic callus, but also useful for molecular breeding of orchardgrass through genetic transformation.

Changes of RNA and Protein During Callus Induction and Plant Regeneration from Perilla frutescens (들깨로부터 Callus의 유기와 재분화에 따른 단백질 및 RNA의 변화)

  • Cotyledon and hypocotyl explants of perilla were cultured on MS medium containing a combined concentration of BA(0.5, 1.0 and 1.5mg/$\ell$) and NAA(0.1, 0.5 and 2.0mg/$\ell$) in order to regenerate the explant and induce the callus. The best regeneration of the explant and induction of the callus were observed in a combined concenteration of 0.5mg/$\ell$ of BA and 0.5mg/$\ell$ NAA both in cotyledon and hypocotyl explants. In cotyledon explants, rooting was achieved upon transferring shoots to MS medium containing 0.5mg/$\ell$ of BA and 0.1mg/$\ell$ of NAA. We also investigated the change of protein and RNA content on developmental stage of callus and plant regeneration of perilla. Protein content was increased but RNA content was decreased as the culture period increases. The banding pattern of polypeptide revealed that both 30KD and 45KD polypeptides were obvious in cotyledon obtained from pre-culture explants, but only 30KD polypeptide was further getting obvious as the culture period increases.

Establishment of an efficient regeneration system for Hibiscus syriacus 'Nanpa' (무궁화 품종 '난파'의 효율적인 재분화 체계 확립)

  • Hibiscus syriacus L., the national flower of Korea, is a commonly grown ornamental species found in parks, gardens and along roadsides in Korea. This study sought to establish a plant regeneration system of H. syriacus 'Nanpa' using mature leaves as an explant. Sterilized mature leaf explants were cultured on modified MS medium with combinations of NAA and BAP at various concentrations for 6 weeks. Among the various media evaluated, modified MS media containing 0.46 mg/L NAA and 0.5 mg/L BAP (CI-1) or 0.92 mg/L NAA and 1 mg/L BAP (CI-4) were the most effective for callus formation. Whitish-yellow calluses were observed on CI-1 medium, while green calluses formed on CI-4 medium. The whitish-yellow callus derived from CI-1 medium showed a higher frequency of shoot induction than the green callus derived from CI-4 on modified MS medium containing 0.5 mg/L BAP. Among the various media evaluated in this study, modified MS medium containing 0.46 mg/L NAA and 0.5 mg/L BAP was the most optimal for shoot-forming callus production. Our findings show that mature leaves of H. syriacus 'Nanpa' can be used for in vitro plant regeneration, and the regeneration system described here may be a powerful tool for molecular breeding of H. syriacus 'Nanpa' in the future.

Saikosaponin Contents in Bupleurum falcatum Root Produced by Tissue Culture (조직배양(組織培養)한 시호근(柴胡根)의 Saikosaponin 함량(含量))

  • The study for the quantitative analysis of saikosaponin of Bupleurum falcatum root produced by tissue culture was attempted. The optimal concentration of 2,4-D was 0.1 mg/l for inducing the callus. The induction and growth of the adventitious root from the callus was obtained in the suspension culture containing MS basal medium supplemented with no plant growth regulators. The results of the quantitative analysis of saikosaponins by high performance liquid chromatography (HPLC) showed that the content of saikosaponin a and c was high in the one-year-old root from somatic embryos and that of saikosaponin d was remarkably high in three-months-old adventitious root from callus.
