두류의 첨가가 당뇨병 쥐의 내당능과 지질대사 및 지질과산화에 미치는 영향 (Effects of Legume Supplementation on the Glucose and Lipid Metabolism and Lipid Peroxidation in Streptozotocin-Induced Diabetic Rats)

  • The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats.   • The aim of this study was to investigate the expression of PPAR-α, -β/δ, -γ, PGC-1α, GLUT-4 and p-AMPK-α2 protein in the skeletal muscle of diabetic induced-rats by endurance training combined with rosiglitazone. The expression of PPAR-α, -β/δ, -γ, PGC-1α, GLUT-4 and p-AMPK-α2 protein in red and white gastrocnemius by western blotting. The body weight was higher in diabetic induced-rats compared to the normal rats and after the treatment of exercise combined with rosiglitazone was significantly reduced in the all group. The levels of blood glucose was higher in diabetic induced-rats compared to the normal rats and after the treatment of exercise combined with rosiglitazone was significantly reduced in the all group. The expression of PPAR-α, -γ, PGC-1α in skeletal muscle of diabetic induced-rats were increased all groups and increased significantly in the group with exercise combined with rosiglitazone. The expression of GLUT-4 and p-AMPK-α2 protein in the skeletal muscle of diabetic induced-rats were increased all groups and increased significantly in the group with exercise combined with rosiglitazoneI. These results suggest that exercise training and rosiglitazone may act as complementary therapies for the treatment of insulin rasistance.

Effect of dietary Platycodon grandiflorum on plasma glucose and lipid metabolism in $KK-A^y$ mice and streptozotocin-induced diabetic rats

  • This study was designed to investigate the effect of dietary Platycodon grandiflorum on plasma glucose and lipid metabolism in $KK-A^y$ mice and STZ-induced diabetic rats. Both plasma triglyceride and plasma cholesterol levels in STZ-induced diabetic rats were significantly decreased by dietary Platycodon grandiflorum feeding for 4 weeks compared to those of control rats, but there were no marked differences in $KK-A^y$ mice. However, for plasma glucose values, Platycodon grandiflorum feeding resulted in a significant decrease in both STZ-induced diabetic rats and $KK-A^y$ mice. Also, dietary Platycodon grandiflorum slightly decreased the postprandial glucose level at 30 and 60 mins during oral glucose tolerance test in $KK-A^y$ mice. Although there was no statistical significance, the fasting plasma insulin levels of Platycodon grandiflorum dieted $KK-A^y$ mice tended to decrease when compared to that of control mice. Therefore, the present results suggested that dietary Platycodon grandiflorum may have a beneficial effect on preventing hypercholesterolemia and hyperlipidemia.

Effect of Berberis tinctoria leaf (Berberidaceae) extract on antidiabetic, antihyperlipidemic and antioxidant status in streptozotocin induced diabetes in rats

  • The present study was carried out to investigate the antidiabetic and antioxidant effect of methanolic extract of Berberis tinctoria leaves (MEBT), in streptozotocin induced diabetic rats. Oral administration of MEBT extract (150 mg/kg and 300 mg/kg) for a period of 14 days. Blood glucose levels, body weight food and liquid intake were measured on every $5^{th}$ day over a period of 14 days. In diabetic rats, MEBT at the dose of 150 mg/kg and 300 mg/kg body weight resulted in significant reduction in blood glucose levels. The study was further investigated to determine antioxidant and antihyperlipidemic potential of MEBT in streptozotocin (STZ) induced diabetic rats. These results suggest that the MEBT possess antidiabetic activity and is able to ameliorate biochemical damages in STZ induced diabetic rats and the results were found to be in a dose dependent manner.

Hepatoprotective and Anti-diabetic Effects of Pelvetia siliquosa, a Marine Algae, in Rats

  • The effects of various fractions from the whole plant of Pelvetia siliquosa Tseng et Chang (Fucaceae) on the $CCl_4$-induced hepatotoxicity as well as on streptozotocin (STZ)-induced diabetes in rats were investigated. The ether fraction exhibited a potent rat lens aldose reductase (RLAR) inhibition in vitro and showed a significant inhibition of not only serum glucose concentrations but also sorbitol accumulations in the lens, red blood cells and sciatic nerves in the STZ-induced diabetic rats. When administered orally in Sprague-Dawley rats, $H_{2}O$ fraction was found to cause a significant inhibition of the rise in the serum transaminase activities in $CCl_4$-intoxicated rats. These results suggested that this plant might possess constituents with hepatoprotective, anti-diabetic effects and those effects on diabetic complications.

  • To evaluate the hypoglycemic effect of nano powder propolis, streptozotocin (STZ) induced diabetic rats were divided into 2 groups : diabetic control group and nano powder propolis (0.9ml) group. Then the rats were fed with nano-powder-propolis for 4 weeks. After 4 weeks, oral glucose tolerance test (oral GTT) was performed and blood sugar levels after 16 hours fasting, body weights, and blood lipid levels were measured. Finally, pancreas were collected and examined by histopathology and immunohistochemistry. In conclusion, the nano-powder-propolis was effective in the treatment of diabetes due to the reduction of blood sugar level and the regeneration of damaged ${\beta}$-cells observed in streptozotocin-induced diabetic rats.

Effects of QZ-16 on blood glucose and lipids in Streptozotocin induced diabetic rats

  • The present study was designed to investigate the hypoglycaemic and hypolipidaemic activities of Qurs-e-Ziabetus 16 (QZ-16) in Streptozotocin (STZ) induced diabetic rats. QZ-16, a polypharmaceutical herbomineral formulation developed on the principles of Unani medicine is used for non-insulin dependent diabetes mellitus (NIDDM). The elevated levels of fasting blood glucose, serum levels of cholesterol, triglycerides and urea observed in rats treated with STZ (55 mg/kg body wt.) were significantly reduced by the treatment of QZ-16 (240 mg/kg, p.o.) and gliclazide (30 mg/kg, p.o.). The reduced HDL cholesterol levels were also increased by the QZ-16 and gliclazide treatments in the STZ induced diabetic rats. These data show that QZ-16 has hypoglycaemic, hypolipidaemic properties in STZ induced diabetic rats.

자하차약침(紫河車藥鍼)이 당뇨유발(糖尿誘發) 흰쥐의 신장보호기능(腎臟保護機能)에 미치는 영향(影響) (Renoprotective Effect of Hominis Placenta Aqua-acupuncture in Streptozotocin Induced Diabetic Rats)

  • Objective : This study was undertaken to determine whether Hominis Placenta(HP) aqua-acupuncture exerts renoprotective effect on diabetic nephropathy induced by streptozotocin(STZ) in rats. Methods : In order to study the renoprotective effect of HP aqua-acupuncture, experimental animals were divided into 5 groups and treated for 2 weeks as follows: control group was injected subcutaneously by saline aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, 5% HP aqua-acupuncture group was injected subcutaneously by 5% HP aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, 10% HP aqua-acupuncture group was injected subcutaneously by 10% HP aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, normal group was injected subcutaneously by saline aqua-acupuncture into Sinsu(BL23) in normal rats, and Captopril group was administrated with captopril at a dose of 50mg/kg in STZ induced diabetic rats. Results : While HP aqua-acupuncture did not reduce any body weight, index of kidney hypertrophy, the plasma glucose concentration and BUN respectively, HP aqua-acupuncture showed lowering urinary albumin excretion rate and serum creatinine as compared with the control group. Gene and protein expressions of $TGF-{\beta}1$ and fibronectin in kidney, one of the extracellular matrix proteins were investigated. There were significant differences in expression levels in HP aqua-acupuncture group as compared with the control group, and $100{\mu}g/m{\ell}$ and $500{\mu}g/m{\ell}$ of HP depressed apoptosis, showing in a dose dependent manner. In the HE staining, HP aqua-acupuncture inhibited the injury of glomerulus and proximal convoluted tubule. Conclusions : HP aqua-acupuncture showed the renoprotective effect possibly through suppressions of $TGF-{\beta}1$ and fibronectin expressions in kidney.

두릅나무과 열수추출물이 당뇨흰쥐의 혈당과 혈액 성분에 미치는 영향 (Effects of Araliaceae Water Extracts on Blood Glucose Level and Biochemical Parameters in Diabetic Rats)

  • This study was conducted to investigate the effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts on blood hemoglobin, $HbA_{1c}$, levels and biomarkers in the streptozotocin (STZ)-induced diabetic rats. Male Wistar rats divided into normal and diabetic groups. The diabetic groups subdivided into the control group (DM) and Araliaceae water extracts supplemented groups: Aralia elata (AE), Acanthopanacis cortex (AC) and Ulmus davidiana (UD). The extracts were supplemented in diet base on 11.42 g of raw Araliaceae/kg diet for 7 weeks. The diabetes was induced by injecting 572 (55 mg/kg B.W., i.p.) once 2 weeks before sacrifying. Relative weights of liver were significantly lowered in the DM group compared to the normal group, whereas those of kidney and heart were significantly increased in the DM group. Supplementation of the Araliaceae water extracts improved reduced liver weights in STZ-induced diabetic rats. Blood glucose level was significantly higher in the DM group than in the normal group, whereas insulin contents were significantly lowered in the DM groups. However, these parameters were normalized in the An, AC and UD supplemented groups, respectively. Blood hemoglobin and $HbA_{1c}$ levels were significantly higher in the DM group than in the normal group. When all of Araliaceae water extracts were supplemented to the diabetic rats lowered hemoglobin and $HbA_{1c}$ levels. Red blood cell, white blood cell and Lymphocyte were significantly higher in the DM group than in the normal group. The supplementation of Araliaceae family water extracts significantly lowered these parameters compared to the DM group. MCV, MCH contents were declined in the DM group, while the supplementation of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts elevated of these contents in STZ-induced diabetic rats. Accordingly, these results indicate that Aralia elata, Aeanthopanacis corex and Ulmus davidiana water extracts would seem to improve the blood biomarkers in STZ-induced diabetic rats.

Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과 (Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.
