The goal of this research was (1) to describe the conditions and parameters required for the cell cycle synchronization and the accumulation of large number of metaphase cells in maize and other cereal root tips, (2) to isolate intact metaphase chromosomes from root tips suitable for characterization by flow cytometry, and (3) to construct chromosome-specific libraries from maize. Plant metaphase chromosomes have been successfully synchronized and isolated from many cereal root-tips. DNA synthesis inhibitor (hydroxyurea) was used to synchronize cell cycle, follwed by treatement with trifluralin to accumulate metaphase chromosomes. Maize flow karyotypes show substantial variation among inbred lines. thish variation should be sueful in isolating individual chromosome types. In addition, flow cytometry is a useful method to measure DNA content of individual chromosomes in a genotyps, and to detect chromosomal variations. Individual chromosome peaks have been sorted from the maize hybrid B73/Mol7. Libraries were generated form the DOP-PCR amplification product from each peak. To date, we have analyzed clones from a library constructed from the maize chromosome 1 peak. Hybridization of labeled genomic DNA to clone inserts indicated that $24\%,\;18\%,\;and\;58\%$ of the clones were highly repetitive, medium repetitive, and low copy, respectively. Fifty percent of putative low cpoy clones showed single bands on inbred screening, blots, and the remaining $50\%$ were low copy repeats. Single copy clones showing polymorphism will be mapped using recombinant inbred mapping populations. Repetitive clones are being characterized by Southern blot analysis, and will be screened by in situ hybridization for their potential utility as chromosome specific markers.
Current data on reproductive characters of endemic and native species are essential to provide a strategy for the conservation of these species. Red pine (Pinus densiflora Sieb. & Zucc.) is one of the dominant, native tree species in Korea, but its reproductive ecology is not well-known. In 1997, the pattern of variation in cone and seed yields contributing to the conservation of declining populations of red pines was examined. Plant height and dbh were measured, and several new cones were collected from each tagged tree after counting the number of cones on each tree. For a subset of cones sampled, the number of fertile scales, the number of seeds at three development stages (early/late aborted, and filled seed), seed wing size, wing color, and individual filled seed mass were measured. The three sites which differed significantly in mean plant size also differed in mean cone and seed production per plant. However further analyses showed that most variation in characters examined occurred among plants within sites, but not among sites. An average of 90% of the potential seeds on the cones aborted at an early developmental stage, demonstrating that early abortion is a major factor affecting the number of filled seeds per cone. Individual seed mass was the only character which exhibited significant variations among sites as well as among trees within sites. Individual seed mass was overall negatively correlated with both the percentage of late abortion and the number of old cones per plant, suggesting that both the past and current years' reproductive activities have caused variations in seed mass. The potential dispersal distance of red pine seeds is quite large. However, wing loading was correlated with seed mass and number in a complex pattern across the sites. Distribution of seeds with varied colored wings differed among sites and among trees within sites. These results suggest that red pines at different sites might possess different strategies to cope with selection pressures acting during the final phase of reproduction, from seed dispersal to establishment. Then the ‘fitted’ red pine trees at each site should be identified and managed to conserve or restore populations.

Natural habitats of the Korean long-tailed goral (Naemorhedus caudatus) have been fragmented by anthropogenic activities in South Korea in the last decades. Here, the individual identity, genetic variation, and population differentiation of the endangered species were examined via the multiple-tube approach using a non-invasive genotyping method. The average number of alleles was 3.16 alleles/locus for the total population. The Yanggu population (1.66) showed relatively lower average number of alleles than the Inje population (3.67). Of the total 19 alleles, only seven (36.8%) alleles were shared by the two populations. Using five polymorphic out of six loci, four and six different goral individuals from the captive Yanggu (n=24) and the wild Inje (n=28) population were identified, respectively. The allele distribution was not identical between the two populations (Fisher's exact test: P<0.01). A considerably low migration rate was detected between the two populations (no. of migrants after correction for size=0.294). Additionally, the F statistics results indicated significant population differentiation between them, however, quite low (FST=0.327, P<0.01). The posterior probabilities indicated that the two populations originated from a single panmictic population (P=0.959) and the assignment test results designated all individuals to both populations with nearly equal likelihood. These could be resulted from moderate population differentiation between the populations. No significant evidence supported recent population bottleneck in the total Korean goral population. This study could provide us with useful population genetic information for conservation and management of the endangered species.
Purpose: Peri-implantitis, a clinical term describing the inflammatory process that affects the soft and hard tissues around an osseointegrated implant, may lead to peri-implant pocket formation and loss of supporting bone. However, this imprecise definition has resulted in a wide variation of the reported prevalence; ${\geq}10%$ of implants and 20% of patients over a 5- to 10-year period after implantation has been reported. The individual reporting of bone loss, bleeding on probing, pocket probing depth and inconsistent recording of results has led to this variation in the prevalence. Thus, a specific definition of peri-implantitis is needed. This paper describes the vast variation existing in the definition of peri-implantitis and suggests a logical way to record the degree and prevalence of the condition. The evaluation of bone loss must be made within the concept of natural physiological bony remodelling according to the initial peri-implant hard and soft tissue damage and actual definitive load of the implant. Therefore, the reason for bone loss must be determined as either a result of the individual osseous remodelling process or a response to infection. Methods: The most current Papers and Consensus of Opinion describing peri-implantitis are presented to illustrate the dilemma that periodontologists and implant surgeons are faced with when diagnosing the degree of the disease process and the necessary treatment regime that will be required. Results: The treatment of peri-implantitis should be determined by its severity. A case of advanced peri-implantitis is at risk of extreme implant exposure that results in a loss of soft tissue morphology and keratinized gingival tissue. Conclusions: Loss of bone at the implant surface may lead to loss of bone at any adjacent natural teeth or implants. Thus, if early detection of peri-implantitis has not occurred and the disease process progresses to advanced peri-implantitis, the compromised hard and soft tissues will require extensive, skill-sensitive regenerative procedures, including implantotomy, established periodontal regenerative techniques and alternative osteotomy sites.

This study characterized the fecal bacterial community structure and inter-individual variation in 30-week-old Duroc pigs, which are known for their excellent meat quality. Pyrosequencing of the V1-V3 hypervariable regions of the 16S rRNA genes generated 108,254 valid reads and 508 operational taxonomic units at a 95% identity cut-off (genus level). Bacterial diversity and species richness as measured by the Shannon diversity index were significantly greater than those reported previously using denaturation gradient gel electrophoresis; thus, this study provides substantial information related to both known bacteria and the untapped portion of unclassified bacteria in the population. The bacterial composition of Duroc pig fecal samples was investigated at the phylum, class, family, and genus levels. Firmicutes and Bacteroidetes predominated at the phylum level, while Clostridia and Bacteroidia were most abundant at the class level. This study also detected prominent inter-individual variation starting at the family level. Among the core microbiome, which was observed at the genus level, Prevotella was consistently dominant, as well as a bacterial phylotype related to Oscillibacter valericigenes, a valerate producer. This study found high bacterial diversity and compositional variation among individuals of the same breed line, as well as high abundance of unclassified bacterial phylotypes that may have important functions in the growth performance of Duroc pigs.
In this paper, we propose a gait recognition method based on gait silhouette sequences. Features of gait are affected by the variation of gait direction. Therefore, we synthesize silhouettes to canonical form by using planar homography in order to reduce the effect of the variation of gait direction. The planar homography is estimated with only the information which exist within the gait sequences without complicate operations such as camera calibration. Even though gait silhouettes are generated from an individual person, fragments beyond common characteristics exist because of errors caused by inaccuracy of background subtraction algorithm. In this paper, we use the Principal Component Analysis to analyze the deviated characteristics of each individual person. PCA used in this paper, however, is not same as the traditional strategy used in pattern classification. We use PCA as a criterion to analyze the amount of deviation from common characteristic. Experimental results show that the proposed method is robust to the variation of gait direction and improves separability of test-data groups.

Rain events are extremely important for phosphorus (P) dynamics in rivers since large portions of annual river P loads can be transported in particulate forms during only a few major events. Despite their importance, a precise estimation of P contribution in river sediments after rainy seasons has rarely been reported. This study estimated the longitudinal variation in the concentrations of different inorganic P fractions in bed sediments of the South Han River over a rainy season, through using the sequential extraction method. Non-apatite P was the dominant form, representing more than 60% of total inorganic P (TIP) content in sediments. Although no significant variation of TIP contents was observed, the proportion of bioavailable P in TIP pools decreased after the rainy season. The concentrations of individual inorganic P fractions ($NH_4Cl-P$, $NH_4F-P$, NaOH-P, and $H_2SO_4-P$) were significantly different across sites and after the rainy season (p < 0.05, two-way ANOVA). $NH_4F-P$ and NaOH-P concentrations in sediments increased in a downstream direction. After the rainy season, $NH_4Cl-P$ concentrations in sediments decreased whereas $NH_4F-P$ and $H_2SO_4-P$ concentrations increased. The redistribution of individual P fractions in sediments observed after rainy seasons were possibly due to the changing contribution of various sources of runoff and the variation in flow related particle size. Current estimation of P in bed sediments of the South Han River suggests a lower potential of internal P loading from sediments after the rainy season.
The photometric light curves of the W-type W UMa eclipsing contact binary system BB Pegasi have been found to be extremely asymmetric over all the observed 63 years in all wavelengths UBVR. The light curves have been characterized by occultation primary minima. Hence, the morphology of these light curves has been studied in view of these different asymmetric degrees. The system shows a distinct O'Connell effect, as well as depth variation. A 22.96 years of stellar dark spots cycle has been determined for the system. Almost the same cycle (22.78 yr) has been found for the depth variation of MinI and MinII. We also present an analysis of mid-eclipse time measurements of BB Peg. The analysis indicates a period decrement of $5.62{\times}10^{-8}$ day/yr, which can be interpreted in terms of mass transfer at a rate of $-4.38{\times}10^{-8}M_{\odot}$/yr, from the more to the less massive component. The O - C diagram shows a damping sine wave covering two different cycles of 17.0 yr and 12.87 yr with amplitudes equal to 0.0071 and 0.0013 day, respectively. These unequal durations show a non-periodicity which may be explained as a result of magnetic activity cycling variations due to star spots. The obtained characteristics are consistent with similar chromospherically active stars, when applying the Applegate's (1992) mechanism.
Background: Potentially avoidable hospitalizations (PAH) contribute to an increased post-discharge mortality. Methods: To investigate the between-hospital variation and the relationship between all predictors and mortality after discharge among older adults with PAH, we studied 15,186 older patients with PAH in 2,200 hospitals included in the National Health Insurance Service-Senior claims database from 2002 to 2013. Multivariable multilevel logistic regression analyses were performed to analyze the variance at between-hospital for mortality after accounting for differences in patient characteristics. Results: The between-hospital variation in mortality that could be attributed to hospital practice variations were 37.6% at 1-week to 13.9% at 12-month post-discharge, after adjustment for individual patient characteristics and hospital-level factors. Hospital-level factors significantly explained mortality at 3 weeks after discharge. Clinics, compared with general hospitals, demonstrated a 2.75 times higher likelihood of deaths at 3-week post-discharge (p<0.001). Compared with private hospitals, public hospitals exhibited 1.61 times higher odds of 3-week mortality (p=0.01). Conclusion: This study demonstrates considerable between-hospital variations in PAH-related mortality that could be attributed to hospital practices. Monitoring of hospitals to identify practice variations would be warranted to improve the survival of older patients with PAH.
The variation of stomatal density and stomatal length of four species of oaks was studied for the purpose of examining the differences among populations and among individual trees within population. Nine populations of Quercus mongolica, four populations of Q. serrata and Q. variabilis respectively, and three populations of Q. acutissima were selected in the natural stands of oaks distributed through the whole country. Twelve leaves were sampled from each of 20 trees from each population. The length of 20 stomata and ten replications of stomatal density were measured from collodion replicas of each leaf under a microscope. Average stomatal densities and lengths ranged through $600-1000/mm^2$ and $19-26{\mu}m$ respectively in all of the species studied. The stomatal densities and lengths presented significant differences statistically at the level of 1 or 5% among populations and among individual trees within population in all the species. Quercus mongolica, especially, showed large variation among populations, while Q. variabilis did very narrow variation compared to the other species. The coefficients of variation of stomatal densities and lengths among individual trees within population exhibited small values of 3.7-12.0% and 1.4-5.3% respectively in all the populations of the species. The average stomatal densities of Q. mongolica showed statistically significant correlation of multiple correlation coefficient of $R_{df{\cdot}2.6}=0.868^*$ and multiple regression equation of $Y=0.041X_1(G.M.T.S.)+0.489X_2(G.M.H.S.)+22.37$ with the sum of growing season mean daily temperature and the sum of growing season mean daily humidity of the stand studied. However the average stomatal lengths showed no relation with the same meteological variables. The figures of frequency distribution of the measurements of leaves or the mean values of individual trees did not show normal distribution curves in some populations. The curves, as well as the results of ANOVA, exhibited the differences among populations.
