Plant breeders have focused on improving plant architecture as an effective means to increase crop yield. Here, we identify the main-effect quantitative trait loci (QTLs) for plant shape-related traits in rice (Oryza sativa) and find candidate genes by applying whole genome re-sequencing of two parental cultivars using next-generation sequencing. To identify QTLs influencing plant shape, we analyzed six traits: plant height, tiller number, panicle diameter, panicle length, flag leaf length, and flag leaf width. We performed QTL analysis with 178 $F_7$ recombinant inbred lines (RILs) from a cross of japonica rice line 'SNU-SG1' and indica rice line 'Milyang23'. Using 131 molecular markers, including 28 insertion/deletion markers, we identified 11 main- and 16 minor-effect QTLs for the six traits with a threshold LOD value > 2.8. Our sequence analysis identified fifty-four candidate genes for the main-effect QTLs. By further comparison of coding sequences and meta-expression profiles between japonica and indica rice varieties, we finally chose 15 strong candidate genes for the 11 main-effect QTLs. Our study shows that the whole-genome sequence data substantially enhanced the efficiency of polymorphic marker development for QTL fine-mapping and the identification of possible candidate genes. This yields useful genetic resources for breeding high-yielding rice cultivars with improved plant architecture.
In an attempt to determine the susceptibility of inbred golden hamsters(Mesocricetus auratus) to the experimental infection with Clonorchis sinensis, twenty-five hamsters were divided into 5 groups and administered orally 5, 10, 20, 30 or 50 metacercariae each. The hamsters were killed on the 45th day after infection. The adult flukes were recovered from all hamsters. The overall recovery rate for the fluke was 57.9% with the range of 48.4% to 92.0%. As the number of the metacercariae given increased, a porportionate decrease in the recovery rate was found, the prepatent period of the fluke in the hamster varied from 15 to 17 days, with the average of 16 days. No significant differences in the development of the flukes were observed. Egg production expressed by EPG, increased step by step as the time elapsed. However, a tendency of decreasing egg laying capacity was observed with the increase of worm burden. It is suggested that the hamster is a suitable final host of C. sinensis.
This study was carried out to develope new zoysiagrass cultivar 'Millock'(Patent registration No. : 10-2005-0110051). Artificial selfing of collected line of MJ8 was conducted to develope F1 plant (MJ8S). Among the inbred progenies, MJ8S-9 (Millock) showed superior performance in color, density, and rust resistance. 'Millock' showed genetically dark green color, with medium-textured leaf ($4.2{\pm}0.44$ mm), short internode length ($3.5{\pm}0.28$ cm), and wide leaf angle ($52.5{\pm}10.8$ degree). Height to the lowest leaf blade of this cultivar was $1.9{\pm}0.91$ cm, which may allow low mowing height. 'Millock' has a yellowish green stolen. Also, specific bands with primer number OPB 8 by RAPD analysis can be used for a cultivar identification.
Brachypodium distachyon has been developed as a monocot model plant for temperate grasses and bioenergy crops. Although B. distachyon research is moving forward rapidly, the study of photoresponses has not been explored. To extend our knowledge of responses to light in monocots, we performed photoresponse analysis of B. distachyon using two inbred lines, Bd21 and Bd21-3. In this study, we first compared growing phenotypes between the two lines and investigated coleoptile and primary leaf growths under dark, far-red, red, and white light conditions. The results showed that the growth of the two lines were similar until tillering stage, but other developmental stages from heading to senescence were much delayed in Bd21-3, which resulted in increased height and tiller numbers. Under different light conditions, primary leaf lengths were kept increasing during the growth period, whereas the coleoptile extension was inhibited 4 to 7 days after growth depending on the light conditions applied. These results suggest that the responses to light in B. distachyon can be examined by measuring coleoptile lengths approximately 7 days after seedling growth. Moreover, we selected light-responsive genes known in Arabidopsis thaliana, such as chlorophyll A/B binding protein (CAB), light-harvesting chlorophyll binding protein (Lhcb) and chalcone synthase (CHS), and confirmed their light-induced gene expression in B. distachyon. Therefore, the present study suggests that the inhibition of coleoptile growth can be used as the parameter to analyze photoresponses in the monocot model plant, and also provide the reference genes whose expression is induced by far-red and red light treatment.

A single recessive gene, rxp, controls the bacterial leaf pustule (BLP) resistance in soybean and in our previous article, it has been mapped on linkage group (LG) D2 of molecular genetic map of soybean. A total of 130 recombinant inbred lines (RILs) from a cross between BLP-resistant SS2-2 and BLP-susceptible Jangyeobkong were used to identify molecular markers linked to rxp. Fifteen simple sequence repeat (SSR) markers on LG D2 were screened to construct a genetic map of rxp locus. Only four SSR markers, Satt135, Satt372, Satt448, and Satt486, showed parental polymorphisms. Using these markers, genetic scaffold map was constructed covering 26.2cM. Based on the single analysis of variance, Satt372 among these four SSR markers was the most significantly associated with the resistance to BLP. To develop new amplified fragment length polymorphism (AFLP) marker linked to the resistance gene, bulked segregant analysis (BSA) was employed. Resistance and susceptible bulks were made by pooling equal amount of genomic DNAs from ten of each in the segregating population. A total of 192 primer combinations were used to identify specific bands to the resistance, selecting three putative AFLP markers. These AFLP markers produced the fragment present in SS2-2 and the resistant bulk, and not in Jangyeobkong and the susceptible bulk. Linkage analysis revealed that McctEact97 $(P=0.0004,\;R^2=14.67\%)$ was more significant than Satt372, previously reported as the most closely linked marker.
In order to develop molecular markers related to stay green phenotype, AFLP analysis was conducted using near-isogenic lines for either stay green or non stay green trait. Both lines have characteristics of multi-ear and tillers (MET). Two out of 64 primer combinations of selective amplification identified three reproducible polymorphic fragments in MET corn with stay green. Both of E+AGC/M+CAC and E+AAG/M+CAA primer combinations produced two and one specific polymorphic fragments linked to stay green trait, respectively. For the conversion of AFLPs to sequence tag sites (STSs), primers were designed form both end sequences of each two polymorphic fragments. One fragment, which was amplified with E+AAG/M+CAA primer combinations, possessed 298 bp long and showed a $91\%$ homology with maize retrotransposon Cinful-l. One out of two polymorphic fragments produced with E+AGC/M+CAC primer combination had 236 bp long and matched a $96\%$ homology with an intron region of 22kDa alpha zein gene cluster in Zea mays. One out of two PCR fragments amplified with MET2 primer set in the stay green MET was not produced in the non-stay green MET. The developed AFLP and STS marker could be used as an efficient tool for selection of the stay green trait in the MET inbred.
This experiment was conducted to determine the isozymic differences between normal maize and maize inbreds of multiple ears and tillers (MET). Two maize inbreds Euisung, Iri and their hybrid having tillers and multiple ears were compared with normal maize. With usual electrophoresis using 6% polyacrylamide gel, peroxidase and esterase enzymes were studied. Matured leaf, culm, leaf sheath, root and young ear tissues showed different isozymic patterns between METs and normal maize in peroxidase. The Euisung inbred grown for 7 days under dark condition showed typical peroxidase. bands compared with checks in the tissues of coleoptile and stele. Better observation of isozymic bands was made during early part of maize growth. Parental inbreds showed more active and apparent band differences than their hybrids in esterase. Bands for esterase were also apparently different in the stele, coleoptile and young ear tissues of the METs and the checks. The maize lines infected with black streaked dwarf virus showed obvious differences in peroxidase and esterase isozymes.

The pod shattering or dehiscence is essential for the propagation of pod-bearing plant species in the wild, but it causes significant yield losses during harvest of domesticated crop plants. Identifying novel molecular makers, which are linked to seed-shattering genes, is needed to employ the molecular marker-assisted selection for efficiently developing shattering-resistant soybean varieties. In this study, a genetic linkage map was constructed using 115 recombinant inbred lines (RILs) developed from crosses between the pod shattering susceptible variety, Keunol, and resistant variety, Sinpaldal. A 180 K Axiom(R) SoyaSNPs data and pod shattering data from two environments in 2001 and 2015 were used to identify quantitative trait loci (QTL) for pod shattering. A major QTL was identified between two flanking single nucleotide polymorphism (SNP) markers, AX-90320801 and AX-90306327 on chromosome 16 with 1.3 cM interval, 857 kb of physical range. In sequence, genotype distribution analysis was conducted using extreme phenotype RILs. This could narrow down the QTL down to 153 kb on the physical map and was designated as qPDH1-KS with 6 annotated gene models. All exons within qPDH1-KS were sequenced and the 6 polymorphic SNPs affecting the amino acid sequence were identified. To develop universally available molecular markers, 38 Korean soybean cultivars were investigated by the association study using the 6 identified SNPs. Only two SNPswere strongly associated with the pod shattering. These two identified SNPs will help to identify the pod shattering responsible gene and to develop pod shattering-resistant soybean plants using marker-assisted selection.

Foxglove aphid, Aulacorthum solani (Kaltenbach), is a Hemipteran insect that infected a wide variety of plants worldwide and caused serious yield losses in crops. The objective of this study was to identify the putative genes to foxglove aphid resistance in wild soybean, PI 366121 (Glycine soja Sieb. and Zucc.). One hundred and forty-one F4:8 recombinant inbred lines developed from a cross between susceptible variety, Williams 82 and foxglove aphid resistance wild soybean, PI 366121 were used. The two type of resistance response, antibiosis and antixenosis resistance were evaluated through choice and no-choice test, graded by the degree of total plant damage and primary infestation leaf damage; a genome-wide molecular linkage map was constructed with 29,898 single-nucleotide polymorphism markers utilizing a Axiom(R) 180K soyaSNP array. Using inclusive composite interval mapping analysis for foxglove aphid resistance, one major candidate QTL on chromosome 7 was identified. The major QTL on chromosome 7 showed both antixenosis and antibiosis resistance responses. The newly identified major QTL was consistent with previously reported QTL, Raso2, which showed around 5 times narrow down interval range with 8 candidate genes. Furthermore, total 1,115 soybean varieties including Glycine soja and Glycine max were exposed to germplasm screening, and 31 varieties, which showed significant antibiosis type foxglove aphid resistance were identified. This result could be useful in breeding for new foxglove aphid resistant soybean cultivars and developing novel insecticides.

Pregnancy block by male pheromones in mice differs in incidence depending on the combination of strains. Female mice of BALB/cA strain mated with BALB/cA males show a 100% pregnancy block when exposed to males of inbred strain DDK shortly after copulation (Chung et al., Biol Reprod 1997). In the present study, BALB/cA females mated with the males of other strains (CBA/J, C3H/HeN, C57BL/6Cr, and IXBL) showed higher pregnancy rates (66.6-87.5%) even when they were exposed to DDK males. In the pharmacological induction of pregnancy block with dopamine agonist (Bromocriptine, 4mg/kg BW), BALB/cA females mated with BALB/cA males showed a 100% pregnancy block. In contrast, BALB/cA females mated with CBA/J, C3H/HeN, and C57BL/6Cr males showed higher pregnancy rates (40-70%). These results suggest that the better pregnancy rate of BALB/cA females mated with alien males may be due to the stronger viability of F 1 embryos. This interpretation was confirmed by an embryo transfer experiment in which a higher implantation rate was observed when BALB/cA embryos grown in BALB/cA females exposed to BALB/cA males were transferred into recipient BALB/cA females exposed to DDK males. These results suggest that the embryonic genotype or viability of the embryo is one factor contributing to the occurrence of pregnancy block by male pheromones in mice.
