Aggressiveness in Plasmopara halstedii (sunflower downy mildew)

  • Aggressiveness was studied in seven Plasmopara halstedii (sunflower downy mildew) pathotypes: 100, 300, 304, 314, 704, 710 and 714. Aggressiveness criteria including percentage infection, latent period, sporulation density and reduction of hypocotyl length (dwarfing) were analysed in one sunflower inbred line showing a high level of quantitative resistance. Genetic relationships were detected between the seven pathotypes using 12 EST-derived markers. Pathotypes 100, 300, 304 and 314 were characterized with shorter latent period and higher sporulation density than pathotypes 710, 704 and 714. All pathotypes showed high percentage infection values and caused a large reduction in seedling size except for pathotype 314 involved in dwarfing. Pathotypes 714, 704 and 314 had an intermediary genetic position between the pathotypes 100 and 710. No correlation was detected between aggressiveness traits and EST genotypes.

  • To understand the relationship among different quantitative traits, correlation studies were performed by utilizing eighty-eight inbred strains of silkworm, Bombyx mori L. on twelve economic characters. Analysis resulted in correlation of 5th instar period with total larval period (r = 0.7882), cocoon weight with shell weight (r = 0.8326), shell weight with shell ratio (r = 0.5067), shell ratio with raw silk % (r = 0.7570), raw silk % with filament length (r = 0.3490), filament size (denier) with reelability (r = 0.3193) and boil-off loss % (r = 0.2792). Negative correlation was observed among filament length with filament size (r = 0.7582) and reelability with boil-off loss % (r = -0.3236). Correlation of different quantitative characters for quality silk production is discussed.

  • To understand the signal transduction pathway that leads to the activation of the wound-inducible proteinase inhibitor II (pin2) promoter. $F_2$ progenies of wound (-) mutant crossed with wild-type Arabidopsis plants were biochemically and genetically characterized. Wound (-) mutant was derived from transgenic Arabidopsis plants containing bacterial cytosine deaminase gene under the control of pin2 promoter. The cytosine deaminase assays indicated that wound (-) mutant is a dominant inhibitor of wound-inducibility as only 3 of the $20F_2$ progenies showed cytosine deaminase (CDase) activity, To construct a structural map of the wound (-) mutant chromosomal regions, cleaved, amplified polymorphic sequences (CAPS) markers that cover all Chromosomes were used. Chromosomal regions covered by three different CAPS markers could be candidates for further fine mapping of the location of the wound (-) mutation. g4026, RGA1 and ASA1 located at 84.9 on recombinant inbred (RI) map of chromosome I, at 1.75 on RI map of chromosome II, and 18.35 on RI map of chromosome V, respectively.

  • A new powdery mildew resistant squash (Cucurbita moschata Duch.) 'Miso' was bred from a cross between powdery mildew resistant true variety 'Sangol' and powdery mildew susceptible inbred line 'Seoulmadi' at National Institute of Horticultural & Herbal Science (NIHHS). The 'Miso' variety was vigorous and highly resistant to powdery mildew. It showed white green fruit color. The variety yielded 21.3MT/ha which is 52% more than control variety.
