In vitro anti oxidant activity of methanol extract of Clerodendrum infortunatum Linn

  • The antioxidant potency of methanolic extract Clerodendrum infortunatum Linn. (MECI), which are widely used in the Indian indigenous system of medicine for different purposes, was studied. The antioxidant potential was evaluated using different established in vitro antioxidant tests viz. determination of total amount of polyphenolics compounds, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, nitric oxide scavenging, superoxide anion radical scavenging, hydroxyl radical scavenging and reductive power assay. It was found that MECI contain a high amount of polyphenolics and possesses significant free radical scavenging activity in all the assay. The higher activity was may be due to presence of richest amount of polyphenolics and flavonoids in it.

동규자(冬葵子)와 경마자의 항산화(抗酸化) 효능(效能) 연구(硏究) (Effects of Malvae Semen and Abutili Semen on Anti-oxidation Activities)

  • Objectives : Anti-oxidation activities of Malvae Semen and Abutili Semen were compared and evaluated. Methods : We tested the anti-oxidant effect through in vitro experiment and in vivo experiment that induced oxidative stress using ethanol. Results : 1. In the anti-oxidative activities of Malvae Semen and Abutili Semen they had high effects in case of the scavenging activitie of DPPH and superoxide anion radical, and inhibitory effect of linoleic acid oxidation. 2. Malvae Semen had large quantities of phenolic compound than Abutili Semen, but the anti-oxidative activity of Malvae Semen was weaker than that of Abutili Semen. 3. The anti-oxidative effect of Abutili Semen was more strong than that of Malvae Semen in the rat stressed by ethanol. Conclusion : Anti-oxidation activities of Abutili Semen were stronger than those of Malvae Semen. Therefore, the complementary usage of Malvae Semen and Abutili Semen can be considered.

  • Objectives : To evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species, anti-platelet aggregative effects and anti-thrombosis effects in response to treatment with SA using various screening methods including biological and non-biological oxidants. Methods : The antioxidant activity concerning extract from SA was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on thrombin-induced platelet aggregation and thrombosis. Results : SA extracts were found to have a potent scavenging activity regarding oxidative stress as well as an inhibitory effect towards LDL oxidation, platelet aggregation, and thrombosis. Conclusions : The SA extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

A PROMISING NEW ANTI-WRINKLE INGREDIENT: Pericarpium castaneae extracts

  • Pericarpium castaneae extracts have variously potent activities, such as anti-oxidative activity and free radical scavenging activity. in vivo and in vitro studies both indicate that pericarpium castaneae extracts acts as a free radical scavenger($IC_{50}:7.6{\mu}g/ml$) stronger than gallic acid($IC_{50}:12.5{\mu}g/ml$) and ellagic acid($IC_{50}:15{\mu}g/ml$) which could prevent cutaneous UV damages and skin aging. The extracts showed a good effect as a anti-oxidant($IC_{50}:50{\mu}g/ml$). It was shown that the appearance of wrinkle in human skin was reduced by topical application of pericarpium castaneae extracts. And the treatment of human skin with the extracts increased the elasticity and moisture of the skin. We investigated the effect of the pericarpium castaneae extracts on production of extracellular matrix using cultured A431 fibroblast cells. The results indicated that the extracts had no detectable effect on collagen synthesis. But synthesis of cell adhesion protein was increased by the extracts. The results suggest that increase of cell adhesion protein synthesis by pericarpium castaneae extracts has closely related to reduction of wrinkle in skin.

접골산(接骨散)이 골절치유에 미치는 영향 (Effects of Jeopgolsan (JGS) Extract on Fracture Healing)

  • Objectives The purpose of this study was to evaluate the effect of Jeopgolsan (JGS) extract on anti-oxidant, anti-inflammatory activities in RAW 264.7 cells and on factors related with fracture healing in skull fractured rat. Methods Experimental animals were divided into four groups: normal group without any treatment (Normal), contral group were treated orally with distilled water (Control), Experimental group were treated orally with JGS at a concentration of 200 mg/kg/day (JGS 200) and Experimental group were treated orally with JGS at a concentration of 200 mg/kg/day (JGS 400). Rats in each group except the normal group were induced fractures in the skull. The 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity were measured to evaluate antioxidant activity. The production of nitric oxide (NO), $interleukin-1{\beta}$ ($IL-1{\beta}$), interleukin-6 (IL-6) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) in the RAW 264.7 cells were measured to evaluate anti-inflammatory activity. The production of osteocalcin calcitonin, carboxy-terminal telepeptides of type II collagen (CTX II), transforming growth $factor-{\beta}$ ($TGF-{\beta}$), bone morphogenetic protein-2 (BMP-2), Insulin and alkaline phosphatase (ALP) in serum of rats were measured to evaluate the effects of fracture healing at 0, 2, 4, and 6th week. X-rays were taken every 3 week from 0 to 6th week to evaluate fracture healing effect. Results 1. No cytotoxicity was observed. 2. DPPH and ABTS radical scavenging activity were increased in a concentration dependent manner, indicating anti-oxidant effect. 3. NO, $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ were not significantly changed, indicating no anti-inflammatory effect. 4. Osteocalcin, Calcitonin, $TGF-{\beta}$ and ALP were significantly increased in the experimental groups. 5. CTX II, insulin were significantly decreased in the expermental groups. 6. Radiologic examination showed that union of fracture was promoted. Conclusions From above results, JGS showed significant results in factors related with fracture healing and radiologic examination. Threfore, JGS is expected to be effective in the treatment of fracture.

청아지황탕(靑蛾地黃湯)의 RAW264.7 Cell에서 항산화 및 항염증 효능 연구 (Anti-Oxidative and Anti-Inflammatory Effects of Cheongajihwang-Tang Extract on RAW264.7 Cells)

  • Objectives This study was designed to investigate whether the Cheongajihwang-Tang (CT) has an inhibitory effect association with oxidation or inflammation in RAW264.7 cells. Methods Cytotoxic activity of CT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Nitric oxide production was measured using Griess reagent system. The total phenolic contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the anti-oxidative effects of CT. Dichlorofluorescin diacetate (DCFH-DA) has been used as a substrate for measuring intracellular oxidant production. Results Cheongajihwang-Tang does not impair the cell viability in tested concentration. CT showed anti-oxidative effects in vitro by decreasing electron donating ability, and also showed anti-inflammatory effects suppressing NO and ROS expression in LPS induced RAW264.7 activation. CT inhibited the generation of intracellular ROS production as dose dependant manner. Conclusions CT has anti-oxidative effects and anti-inflammatory activities. These results indicate that CT extract has an anti-inflammatory activities via anti-oxidative effects.

산유자 잎 에탄올 추출물의 미백, 주름억제, 항염증 및 항산화 효능 (Anti-Melanogenic, Anti-Wrinkle, Anti-Inflammatory and Anti-Oxidant Effects of Xylosma congesta leaf Ethanol Extract)

  • 본 연구는 제주도 해변의 벌판에서 드물게 자라는 산유자(Xylosma congesta) 잎 에탄올 추출물의 화장품 기능성 소재로서의 활용 가능성을 알아보기 위하여 cell culture model 및 in vitro assay system을 이용하여 미백, 주름개선, 항염증 및 항산화 활성을 분석하였다. 그 결과 산유자 잎 에탄올 추출물은 B16F10 세포에 세포독성 없이 효과적으로 ${\alpha}$-MSH에 의해 유도된 melanin 합성을 억제함으로써 높은 미백 활성을 가지는 것이 관찰되었다. CCD-986SK 세포를 이용하여 산유자 잎 에탄올 추출물의 주름개선 활성 능력을 procollagen 합성시험을 통해 분석한 결과, 양성 대조군으로 사용한 TGF-${\beta}$ 만큼은 아니지만 농도별로 각각 120, 122%의 procollagen 합성을 증가시키는 것을 통하여 산유자 잎 에탄올 추출물의 주름개선 활성을 확인하였다. 또한 RAW 264.7 murine macrophage를 이용하여 NO 생성 억제능을 분석함으로써 산유자 잎 에탄올 추출물의 항염증 활성 정도를 측정한 결과, LPS에 의해 유도된 NO 생성이 산유자 잎 에탄올 추출물의 농도 의존적으로 감소하는 결과를 얻을 수 있었다. DPPH 법, ABTS 법, ORAC 법을 이용하여 항산화 활성을 분석한 결과, $50{\mu}g/mL$의 농도에서 DPPH 및 ABTS 라디칼 소거능이 각각 75.2, 99.1% 증가하는 것이 확인 되었다. ORAC activity assay kit를 이용하여 항산화 활성을 측정한 결과 산유자 잎 에탄올 추출물의 농도에 따라 높은 항산화 활성이 관찰 되었고, DPPH와ABTS 라디칼 소거능 결과와 유사하게 $50{\mu}g$/mL의 농도에서 가장 높은 항산화 활성($573.74{\pm}0.79{\mu}M$ TE/g)을 나타내었다. 이러한 결과를 통하여 산유자 잎 에탄올 추출물이 높은 미백과 주름개선, 항염증 및 항산화 활성을 나타내는 것을 확인 할 수 있었으며, 기능성 화장품 소재로서의 활용가능성을 제시 할 수 있었다.

한국산 겨우살이의 항산화, 항노화 및 미백 효과 (Anti-oxidant, Anti-aging, and Whitening Effects of Viscum album var. coloratum In Vitro)

  • 본 연구에서는 한국산 겨우살이(Viscum album var. coloratum)의 항산화, 항노화, 미백 효능을 알아보았다. 겨우살이 분획물은 hexane (HX), ethyl acetate (EA), butanol (BU), methylene chloride (MC) 총 4 가지를 사용하였다. 실험 결과 HX 분획물이 4 가지 분획물 중 가장 높은 페놀 함량을 보였다. 또한 EA 분획물이 가장 높은 1,1-diphenyl-2-picrylhydrazyl (DPPH)자유 라디칼 소거능 및 catalase-유사 활성을 나타내었으며, MC 분획물이 가장 높은 superoxide dismutase (SOD)-유사 활성을 나타냈다. MC 분획물은 또한 가장 우수한 hyaluronidase와 elastase 억제능을 보였다. 미백 효능을 알아보기 위하여 tyrosinase 억제능을 알아보았는데, HX 분획물이 가장 우수한 억제능을 보여주었다. 본 연구 결과를 통하여 한국산 겨우살이는 항산화, 항노화 및 미백 활성을 가지는 기능성 화장품 소재로 사용이 가능할 것으로 보인다.

복숭아 품종별 핵과 종자의 항염증 및 항산화 효과간의 상관관계 (In vitro correlation between anti-inflammatory and anti-oxidant effects of stone and seed of peaches cultivars)

  • 본 연구에서는 현재 재배되고 있는 복숭아 품종 중 미황, 가납암백도 및 천홍의 핵과 종자를 이용하여 항산화 및 항염증 활성에 관한 연구를 하였다. 총 phenol 및 flavonoid 함량은 3품종 모두 종자에 비해 핵에서 월등히 높았으며, 품종간에는 천홍의 핵에서 가장 높은 함량을 나타내었다. DPPH radical 소거능과 FRAP 및 CUPRAC에 의한 항산화 활성은 3품종 모두 종자에 비해 핵에서 활성이 높았고, DPPH radical 소거활성은 가납암백도와 천홍, FRAP 활성은 천홍, CUPRAC 활성은 가납암백도에서 유의성 있게 높았다. NO 생성 억제 효과는 RAW 264.7 세포의 경우 핵 추출물에서 가납암백도 74%, 미황 70%, 천홍 58%이며, BV2 세포의 경우 미황 90%, 가납암백도 78%, 천홍 65%로 각각 염증 반응을 감소시켰다. 복숭아 3품종 핵과 종자의 총 phenol 함량과 총 flavonoid 함량은 높은 상관 관계를 나타내었고, 항산화 활성간의 상관성은 모두 유의적인 관계가 확인되었으나, 항염 활성과 항산화 활성, 항염 활성과 총 phenol 및 flavonoid 함량간은 낮은 상관관계를 나타내었다.

Ethanol Extract of Cynanchum wilfordii Produces Endothelium-Dependent Relaxation in Rat Aorta and Anti-inflammatory Activity in Human Aortic Smooth Muscle Cells

  • Objective: The present study investigated the effect of ethanol extract of Cynanchum wilfordii (ECW) on vascular relaxation and vascular inflammation in rat artery isolated from rats and anti-inflammatory activity in human aortic smooth muscle cells (HASMC). Methods: Vascular tone and guanosine 3',5'-cyclic monophosphate (cGMP) production were examined in rat artery isolated from Sprague Dawley rats, in the presence of ECW. HASMC were incubated with tumor necrosis factor-alpha (TNF-${\alpha}$) or Angiotensin II for 24 h. Matrix metalloproteinase (MMP)-2 and anti-oxidant activity of ECW was investigated by pretreatment with ECW in HASMC. Results: Cumulative treatment of ECW relaxed aortic smooth muscles of rats in a dose-dependent manner. ECW-induced vasorelaxation was significantly decreased by pretreatment of L-arginine methyl ester (L-NAME) or oxadiazolo-quinoxalinone (ODQ). Furthermore, ECW treatment of thoracic aorta significantly increased cGMP production. Incubation of ECW with ODQ or L-NAME markedly decreased ECW-induced cGMP production. ECW treatment dose-dependently suppressed TNF-${\alpha}$- or Angiotensin II-induced increase in matrix metalloproteinase-2 expression in HASMC. Also, ECW exhibited 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity in vitro and reduced TNF-${\alpha}$-induced increase in reactive oxygen species production in a dose-dependent manner. Conclusions: Taken together, the results suggest that ECW exerts vascular relaxation via NO/cGMP signaling pathway and decreases MMP-2 expression via anti-oxidant activity.
