  • 본 연구에서는 우란바톨(Ulanbator)에서 북경(Beijing)사이의 지역에서 샘플링한 응회암 풍화토의 동상 특성을 규명하기 위해 상재하중 변화에 따른 동결점 저하를 중심으로 양단면 온도 일정$(상단 5^{\circ}C,\; 하단 5^{\circ}C)$의 동상실험 실시하였다. 본 실험을 통해, 빙편 성장에 따른 체적 팽창과 동결면으로의 수분이동을 확인하였다. 그리고 상재하중과 온도와의 관계식을 제안하였다. 상재하중이 증가함에 따라 동상속도는 비례적으로 감소하였고 빙편발생온도의 절대치는 증가하였다. 또한 동결부분의 함수비가 미동결 부분보다 크게, 특히 동결부분에서도 빙정에 가까울수록 함수비는 증가하였다.

Translation Initiation Factor IF1-Dependent Stimulation of 30 S Preinitiation Complex Formation: Rapid Isolation and fMEt-tRNA Binging Activity of IF1

  • Translation Initiation in prokaryotes involves the formation of a 30 S preinitiation complex, in which translation initiation factors play a role in the stimulation of fMet-tRNA (fMet) binding. However, the specific function and precise mechanism of initiation factor IF1 are still unclear. One a functionally active factor with a high purity. In the present study a large quantity of active IF was rapidly purified, obtained by the overexpression of the infA gene, and then used for a functional study. The induction of infA did not appreciably affect the growth rate of the protease-deficient strain E. coli AR68 harboring the IF1 overproducing plasmid. The level of IF1 obtained was approximately $1-2\%$ of the total cell protein, which enabled the yield of highly purified IF1 (>$98\%$ pure) to be increased to 0.15 mg of IF1/g of cells. The IF1 was isolated within one day by the centrifugatioin of the ribosomal washed fraction, by ammonium sulfate fractionation, chromatography on batch of phosphocellulose, and FPLC Mono S. The overexpressed IF1 was found to be comparable to the factor isolated from normal cells, as determined by migration in NEPHGE/SDS 2-D gels. For binding of fMet-tRNA(fMet) to the 30 S ribosomal subunitis, relatively high levels of binding were obtained when IF2 was present. The addition of IF1 up to 110 pmol proportionally stimulated the binding to a variable extent. This IF1-dependent stimulation of the 30 S preinitiation complex formation demonstrated that IF1 would appear to be exclusively essential for promoting the initiation phase of protein synthesis.

Use of Imaging Agent to Determine Postoperative Indwelling Epidural Catheter Position

  • Background: Epidural anesthesia is widely used to provide pain relief, whether for surgical anesthesia, postoperative analgesia, treatment of chronic pain, or to facilitate painless childbirth. In many cases, however, the epidural catheter is inserted blindly and the indwelling catheter position is almost always uncertain. Methods: In this study, the loss-of-resistance technique was used and an imaging agent was injected through the indwelling epidural anesthesia catheter to confirm the position of its tip and examine the migration rate. Study subjects were patients scheduled to undergo surgery using general anesthesia combined with epidural anesthesia. Placement of the epidural catheter was confirmed postoperatively by injection of an imaging agent and X-ray imaging. Results: The indwelling epidural catheter was placed between upper thoracic vertebrae (n = 83; incorrect placement, n = 5), lower thoracic vertebrae (n = 123; incorrect placement, n = 5), and lower thoracic vertebra-lumbar vertebra (n = 46; incorrect placement, n = 7). In this study, a relatively high frequency of incorrectly placed epidural catheters using the loss-of-resistance technique was observed, and it was found that incorrect catheter placement resulted in inadequate analgesia during surgery. Conclusions: Although the loss-of-resistance technique is easy and convenient as a method for epidural catheter placement, it frequently results in inadequate placement of epidural catheters. Care should be taken when performing this procedure.

Effects of selenium on the survival and invasion of trophoblasts

  • Objective: Placental oxidative stress is known to be a factor that contributes to pregnancy failure. The aim of this study was to determine whether selenium could induce antioxidant gene expression and regulate invasive activity and mitochondrial activity in trophoblasts, which are a major cell type of the placenta. Methods: To understand the effects of selenium on trophoblast cells exposed to hypoxia, the viability and invasive activity of trophoblasts were analyzed. The expression of antioxidant enzymes was assessed by reverse-transcription polymerase chain reaction. In addition, the effects of selenium treatment on mitochondrial activity were evaluated in terms of adenosine triphosphate production, mitochondrial membrane potential, and reactive oxygen species levels. Results: Selenium showed positive effects on the viability and migration activity of trophoblast cells when exposed to hypoxia. Interestingly, the increased heme oxygenase 1 expression under hypoxic conditions was decreased by selenium treatment, whereas superoxide dismutase expression was increased in trophoblast cells by selenium treatment for 72 hours, regardless of hypoxia. Selenium-treated trophoblast cells showed increased mitochondrial membrane potential and decreased reactive oxygen species levels under hypoxic conditions for 72 hours. Conclusion: These results will be used as basic data for understanding the mechanism of how trophoblast cells respond to oxidative stress and how selenium promotes the upregulation of related genes and improves the survival rate and invasive ability of trophoblasts through regulating mitochondrial activity. These results suggest that selenium may be used in reproductive medicine for purposes including infertility treatment.

Comparison of the Differences in Survival Rates between the 7th and 8th Editions of the AJCC TNM Staging System for Gastric Adenocarcinoma: a Single-Institution Study of 5,507 Patients in Korea

  • Purpose: The aims of this study were to compare the 7th and 8th editions of the American Joint Committee on Cancer (AJCC) staging manuals on tumor, node, and metastasis (TNM) staging systems and to evaluate whether the 8th edition represents a better refinement of the 7th staging system, when applied for the classification of gastric cancers. Materials and Methods: A total of 5,507 gastric cancer patients, who underwent treatment from January 1989 to December 2013 at a single institute, were included. We compared patient survival rates across the disease groups classified according to the 7th and 8th editions of the AJCC TNM staging systems. Results: Stage migration was observed in 6.4% (n=355) of the patients. Of these, 3.5% (n=192) and 2.9% (n=158) of patients showed a higher stage and lower stage, respectively. According to the 8th edition of the AJCC TNM staging criteria, the 5-year overall survival rates of the patients with stage IIIB and IIIC showed a significant difference (40.8% vs. 20.2%, P<0.001) whereas no significant differences in the 5-year overall survival rates were observed according to the 7th edition criteria (37.6% vs. 33.2%, P=0.381). Conclusions: Restaging stage III cancers according to the 8th edition of the AJCC TNM classification criteria improved survival rate discrimination, particularly, in institutes where the stage III patients were not distinctly categorized.

  • Strontium (Sr) commonly exists in rock, groundwater, soil, plants, and animals. The Sr isotope ratio offers important information as a tracer on nature because the Sr isotopic composition is not fractionated by any biological process in these ecosystems. Hence, Sr isotope ratio has been used in several studies on tracing the Sr source for contaminated sites and human migration. In this study, we developed a separation method for Sr content, and then improved Sr isotope analysis using multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS). A powdered rock standard (NIST 2710a) was used to determine the removal of interference elements (Rb and Ca) and the recovery rate of Sr content. The results ranged from 98% to 106%. Additionally, three standard samples (NBS 987, IAPSO and NIST 1486) were analyzed to evaluate the precision and accuracy of the results. The measured $^{87}Sr/^{86}Sr$ ratio for all the samples were consistent with the reported values, within an error. These results indicate that our established Sr separation and Sr isotope measurement methods are reliable and can hence be useful in the fields of environmental and forensic sciences.

Mitochondrial DNA Sequence Variation of the Mason Bee, Osmia cornifrons (Hymenoptera: Apidae)

  • In order to understand geographic genetic variation and relationship among populations of the mason bee (Osmia cornifrons Radoszkowsky), which is used as pollinator for apple tree, we sequenced a portion of mitochondrial (mt) COI gene, which corresponds to "DNA Barcode" region (658 bp) from 81 O. cornifrons individuals collected over eight localities in Korea. The sequence data revealed overall moderate to low genetic diversity within species, with a maximum sequence divergence of 0.76%. Geographically, two haplotypes (BAROC01 and BAROC02) were widespread with a frequency of 82.7%, whereas several haplotypes were found in a locality as a single individual, suggesting that haplotype distribution can be summarized as coexistence of a few widespread haplotypes and several regionally restricted haplotypes. Overall, high rate of per generation female migration (Nm=$1.1{\sim}$infinite) and low level of geographic subdivision ($F_{ST}=0{\sim}0.315$) among localities were characteristic. Although two populations (p < 0.026) were genetically subdivided from the remaining localities, no clear polarity was observed. Taken together, the nature of genetic divergence of the mason bee populations is characterized as one that possessing moderate to low genetic diversity, high gene flow, and wide spread haplotypes with ahigh frequency, concordant with the capability of dispersal in connection with the lack of historical biogeographic barriers.

Yubu Island, the Important Waterbird Habitat on the West Coast of Korea and Its Conservation

  • Yubu Island is located within the estuary of the Geum River, South Chungcheong Province $(35^{\circ}59'N,\;126^{\circ}36'E)$, Korea. The island is surrounded by a broad and sandy mudflat, which is typical in the west coast of Korea, and is located 4km off from Gunsan City. Less than 100 humans live on the island, occupying 30 houses. After we discovered that this island was a very important waterbird habitat especially for the East Asian subspecies of the Eurasian oystercatcher, Haematopus ostralegus osculans. Waterbirds were monitored once every month from August 1999 to July 2000. The highest number of oystercatchers counted was 3,200 in December 1999, and the birds seemed to remain in the area continuously from September to next February. About 200 breeding and non-breeding birds remained during the breeding season. In August, early migrants returned to the island, with the number reaching 1,060 individuals. This island is also very important for other waterbirds. Endangered or significant species occurring at this site, and their maximum counts were: chinese egret Egretta eulophotes (5), black-faced spoonbill Platalea minor (17), brant Branta bernicla (1), common shelduck Tadorna tadorna (8,000), hooded crane Grus monacha (2), spoon-billed sandpiper Ewynorhynchus pygmeus (7), dunlin Calidris alpina (6,500), great knot Calidris tenuirostris (24,000), far eastern curlew Numenius madagascariensis (2,500), spotted greenshank Tringa guttifer (4) and Saunders's gull Larus saundersi (1,200). During the 12 month survey period, we observed 52 waterbird species and the total of the maximum counts for the separate species was 73,308, implying that perhaps 150,000 birds could be using the immediate area, if a turnover rate of 2 was assumed.

Subsidence of Cylindrical Cage ($AMSLU^{TM}$ Cage) : Postoperative 1 Year Follow-up of the Cervical Anterior Interbody Fusion

  • Objective : There are numerous reports on the primary stabilizing effects of the different cervical cages for cervical radiculopathy. But, little is known about the subsidence which may be clinical problem postoperatively. The goal of this study is to evaluate subsidence of cage and investigate the correlation between radiologic subsidence and clinical outcome. Methods : To assess possible subsidence, the authors investigated clinical and radiological results of the one-hundred patients who underwent anterior cervical fusion by using $AMSLU^{TM}$ cage during the period between January 2003 and June 2005. Preoperative and postoperative lateral radiographs were measured for height of intervertebral disc space where cages were placed intervertebral disc space was measured by dividing the sum of anterior, posterior, and midpoint interbody distance by 3. Follow-up time was 6 to 12 months. Subsidence was defined as any change in at least one of our parameters of at least 3 mm. Results : Subsidence was found in 22 patients (22%). The mean value of subsidence was 2.21 mm, and mean subsidence rate was 22%. There were no cases of the clinical status deterioration during the follow-up period No posterior or anterior migration was observed. Conclusion : The phenomenon of subsidence is seen in substantial number of patients. Nevertheless, clinical and radiological results of the surgery were favorable. An excessive subsidence may result in hardware failure. Endplate preservation may enables us to control subsidence and reduce the number of complications.

Relationship between roar sound characteristics and body size of Steller sea lion

  • Hundreds of Steller sea lions, Eumetopias jubatus, migrate from Sakhalin and the northern Kuril Islands to Hokkaido every winter. During this migration, they may use their roaring sounds to navigate and to maintain their groups. We recorded the roars of wild Steller sea lions that had landed on reefs on the west coast of Hokkaido, and those of captive sea lions, while making video recordings. A total of 300 roars of wild sea lions and 870 roars of captive sea lions were sampled. The fundamental frequency ($F_0$), formant frequency ($F_1$), pulse repetition rate (PRR), and duration of syllables (T) were analyzed using a sonagraph. $F_0$, $F_1$, and PRR of the roars emitted by captive sea lions increased in the order male, female, and juvenile. By contrast, the $F_1$ of wild males was lower than that of females, while the $F_0$ and PRR of wild males and females did not differ statistically. Moreover, the $F_0$ and $F_1$ frequencies for captive sea lions were higher than those of wild sea lions, while PRR in captive sea lions was lower than in wild sea lions. Since there was a linear relationship between body length and the $F_0$ and $F_1$ frequencies in captive sea lions, the body length distribution of wild sea lions could be estimated from the $F_0$ and $F_1$ frequency distribution using a regression equation. These results roughly agree with the body length distribution derived from photographic geometry. As the volume of the oral cavity and the length of the vocal cords are generally proportional to body length, sampled roars can provide useful information about a population, such as the body length distribution and sex ratio.
