Relationship between Transferable Embryos and Major Metabolite Concentrations in Holstein Donor Cows

  • This research was investigated the relationship, in high-producing Holstein donor cows, between the number of the transferable embryos and the blood serum concentrations of Blood Urea Nitrogen (BUN), glucose and cholesterol, which affect the nutritional state of cows. CIDRs were inserted into the vaginas of twenty two heads of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. The total numbers of ova collected from 3 experimental groups whose blood BUN concentrations were <10 mg/dl, 11~18 mg/dl and ${\geq}19$ mg/dl were 8.9, 12.5 and 19.0, respectively; whereas the numbers of transferable embryos were 5.8 + 1.9, 7.9 + 2.8 and 5.2 + 1.4, respectively. When glucose concentration was <60 mg/dl, the total number of collected ova was 9.9, which was smaller than when the concentration was 60~70 mg/dl or ${\geq}70$ mg/dl. When glucose concentration was 60~70 mg/dl, the number of transferable embryos was 7.1 + 2.4, which was slightly larger than the numbers 6.4 + 2.1 and 6.1 + 1.7 that were obtained when the concentrations were <60 mg/dl and ${\geq}70$ mg/dl, respectively ; however, the differences were not significant (p>0.05). When cholesterol concentrations were <150 mg/dl, 150~200 mg/dl and ${\geq}200$ mg/dl, the total numbers of collected ova were 11.2, 11.3 and 8.6, respectively. Whereas the numbers of transferable embryos were 7.1 + 2.1, 7.3 + 1.9 and 5.6 + 1.3, respectively ; however, the differences were again not significant (p>0.05). The result of this research showed no significant difference in ovum recovery rate and the number of transferable embryos according to major metabolite concentrations in high-producing Holstein donor cows. However, it is considered that the failure of maintaining proper nutritional status would cause the fall in in vivo embryo productivity.

Relationship between Estrous Expression Rate, BCS and Transferable Embryos in Holstein Donor Cows

  • This research was investigated the relationship between the number of the transferable embryos and estrus expression rate, BCS (Body Condition Score), which affect the nutritional state of the cow, in Holstein donor cows. CIDRs were inserted into the vaginas of twenty two head of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. When BCS was $$\leq_-$$2.5, the total number of collected ova was 7.3 + 1.9, which is significantly lower (p<0.05) than the numbers 15.4 + 2.8 and 15.4 + 2.1 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively. Whereas the numbers of transferable embryos were 5.2 + 1.4 when BCS was $$\leq_-$$2.5, which was smaller than the numbers 6.0 + 2.1 and 8.5 + 1.8 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively; however, the differences were not significant. As for estrus induction rate, the cow groups whose BCSs were 2.75 and $$\geq_-$$3.0 showed 100.0% and 95.0%, respectively. Whereas the cow group whose BCS was $$\leq_-$$2.5 showed 57.1%, and the differences were significant (p< 0.05). As for estrous expression rate, the cow groups whose BCSs were $$\leq_-$$2.5, 2.75 and $$\geq_-$$3.0 showed 100.0%, 100.0% and 85.7%, respectively; however, the differences were not significant. According to the result of this research, it is considered that the total number of collected ova and the number of transferable embryos will be affected by the nutritional state before and after in vivo embryo production and superovulation treatment, and that although the mechanism is not clear, poor stockbreeding management and nutritional level would cause the decrease of ovum recovery rate and the number of transferable embryos in high-producing cows. On the other hand, diverse researches on the superovulation treatment method that is suitable for high-producing Holstein donor cows would contribute to preventing ovarian cyclicity disorder, as well as to the early multiplication of cows with superior genes by increasing the utilization value of donor cows.

  • The objective of this study was to investigate the effects of abnormal ovarian cycles after superovulation treatment of Holstein Donor Cows. CIDRs were inserted into the vaginas of twenty two head of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_2{\alpha}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. The cows were considered to have resumed ovarian cyclicity on the day of ovulation if followed by regular ovarian cycles. Seventy two point seven percentage of the cows(16/22) had normal resumption of ovarian cyclicity(resumption within 40 days after superovulation), and 27.3%(6/22) had delayed resumption(resumption did not occur until>40 days after superovulation). Delayed resumption Type II(first ovulation did not occur until ${\geq}40$ days after superovulation, i.e. delayed first ovulation 13.6%) were the most common types of delayed resumptions. The mean numbers of total ova from < 10 and $10{\leq}$ of corpora lutea(CL) was $7.8{\pm}1.8$ and $12.7{\pm}2.7$, respectively. The number of transferable embryos differed between < 10 and $10{\leq}$ CL was $5.4{\pm}1.3$ and $8.1{\pm}3.4$, respectively. Four point five percentage of the cows(1/22) did not resumption their ovarian cyclicity until 60 days after superovulation treatment. Diverse researches on the superovulation treatment method that is suitable for high-producing Holstein donor cows would contribute to preventing ovarian cyclicity disorder, as well as to the early multiplication of cows with superior genes by increasing the utilization value of donor cows.

Further Modifications to the Mobile Nylon Bag Technique to Determine Nutrient Digestibility for Swine

  • Previous studies conducted with swine have reported that the mobile nylon bag technique (MNBT) does not always accurately predict in vivo nutrient digestibilities. Therefore, in this study, the MNBT was modified so that nutrient digestibilities would more closely resemble those from conventional (Con) digestibility studies obtained using the indicator method. A total of 19 feeds were tested including five cereal grains, five legumes, three high protein sources and six mixed diets. The principle changes to the MNBT included the use of a fecal collection harness which minimized the number of bags lost. In addition, previous protocols involved pooling of bags within pig while in the present experiment all bags were analyzed separately to increase the precision of the test. Finally, chemical analyses were done using the entire nylon bag plus residue rather than opening.the bags and scraping out the contents. With the exception of the barley sample (p=0.01), dry matter digestibility (DMD) coefficients obtained with the MNBT were not significantly different from those obtained with the indicator method. The linear regression equation relating the MNBT to the indicator method was Con DMD=-O.77+1.02 MNBT DMD ($r^2=0.93$: p<0.0001). There was no significant (p>0.05) difference in gross energy digestibility (GED) coefficients determined using the MNBT or the indicator method for any of the 19 feeds. The regression line equation relating the MNBT to the indicator method was Con GED=-5.68+1.06 MNBT GED ($r^2=0.94$: p<0.0001). The MNBT was less effective in predicting in vivo crude protein digestibility (CPD) than it was in predicting dry matter and energy digestibility. Differences greater than five percentage units were observed for two of the legumes, Kabuli chickpeas (p=0.02) and the extruded pea-canola seed mixture (p=0.01) as well as for three of the mixed diets including the unheated hulled barley-based diet (p=0.01), the unheated hulless-barley based diet (p=0.08) and the barley-soybean meal based diet (p=0.008). The regression equation relating the MNBT to the indicator method was Con CPD=5.75 + 0.90 MNBT CPO ($r^2=0.76$; p<0.0001). This study indicates that the modified MNBT can be used for the rapid determination of dry matter and energy digestibility in a wide variety of ingredients. For the measurement of crude protein digestibility, the technique produces results similar to conventional digestibility studies for cereal grains and high protein feeds but tends to overestimate protein digestibility for legumes and mixed diets.

Rumen fermentation and performance of Hanwoo steers fed total mixed ration with Korean rice wine residue

  • Background: This study was conducted to evaluate the effects of adding Korean rice wine residue (RWR) in total mixed ration (TMR) on in vitro ruminal fermentation and growth performance of growing Hanwoo steers. Methods: For in vitro fermentation, the experimental treatments were Control (Con: 0 % RWR + TMR), Treatment 1 (T1: 10 % RWR + TMR), and Treatment 2 (T2: 15 % RWR + TMR). The rumen fluid was collected from three Hanwoo steers and mixed with buffer solution, after which buffered rumen fluid was transferred into serum bottles containing 2 g dry matter (DM) of TMR added with or without RWR. The samples were then incubated for 0 h, 12 h, 24 h, or 48 h at $39^{\circ}C$ and 100 rpm. For the in vivo experiment, 27 Hanwoo steers (6 months old) with an average weight of $196{\pm}8.66kg$ were subjected to a 24-week feeding trial. The animals were randomly selected and equally distributed into three groups. After which the body weight, feed intake and blood characteristics of each group were investigated. Results: The pH of the treatments decreased significantly relative to the control during the 12 h of incubation. Total gas production and ammonia nitrogen ($NH_3-N$) was not affected by RWR addition. The total volatile fatty acid (VFA) was lower after 24 h of incubation but at other incubation times, the concentration was not affected by treatments. Feed cost was 8 % and 15 % lower in T1 and T2 compared to control. Blood alcohol was not detected and a significant increase in total weight gain and average daily gain were observed in Hanwoo steers fed with RWR. Conclusion: Overall, the results of this study suggest that TMR amended with 15 % RWR can be used as an alternative feed resource for ruminants to reduce feed cost.

스트렙토조토신-당뇨쥐에서 지질과산화 및 항산화계의 경시적 변화 (Changes in in vivo Lipid Peroxidation and Antioxidant Defense System in Streptozotocin-Induced Diabetic Rats: a Time Course Study)

  • This study was carried out to examine a part of the mechanism for the etiology of diabetic complications. Thirty normal and forty streptozotocin(STZ)-induced diabetic rats were used as the animal models. The animals were sacrificed at the time points of 3 days, 1,2,4 and 6 weeks after STZ-injection and a time course changes in the concentrations of thiobarbituric acid-reactive substances(TBARS) in blood, urine, and tissues, along with the levels of conjugated dienes in tissues were measured as indices of in vivo lipid peroxidation. The activities of antioxidant enzymes, catalase, superoxide dismutase, glutathione peroxidase and the levels of blood retinol and alpha-tocopherol were also measured. The diabetic rats maintained a slightly higher plasma TBARS level throughout the experiment. The urinary TBARS level was significantly higher in diabetic group and gradually increased with time. Concentrations of TBARS in liver, heart, and kidney tissues from diabetic animals were higher than those from the normal group. An increase of conjugated dienes was also observed in the all tissues examined. The kidney tissue of diabetic animals revealed more significant lipid peroxidation state than any other organ tissues. The activities of hepatic antioxidant enzymes such as catalase, superoxide dismutase, glutathione peroxidase were higher in diabetic animals compared to the control ones and increased with the duration of diabetes mellitus. The plasma levels of vitamin A and E were loser in diabetic animals than in normal controls throughout the experimental period. The level of vitamin E in diabetic animals was significantly decreased with the duration of the disease. The results of this study suggest that an effective regimen to suppress the adverse changes in lipid peroxidation and antioxidant defense system is required from the early stage of the disease to prevent the development of diabetic complications.   • This study was conducted to anticipate nitrate reduction state in tree through measurement of nitrate reductase activity (NRA) and investigate the effect of nitrogen concentrations (100, 200, 400, and 600 $mg\;L^{-1}$) on growth, the nitrogen content of various tissue, and NRA of pear (Pyrus pyrifolia cv. Niitaka) seedlings in sand culture. Nutrient solutions used in this experiment were adjusted to pH 6.5 and fixed the ratio of ammonium and nitrate to 1:3 and trickle-irrigated 3 times a day. Tree height and dry weight of various organs in seedlings were higher in low nitrogen concentration (100 and 200 $mg\;L^{-1}$) than in high nitrogen concentration (400 and 600 $mg\;L^{-1}$). The shoot growth in 600 $mg\;L^{-1}$ was extremely poor by nitrogen over supply. Increasing the nitrogen concentration, the concentration of nitrate-N in leaves and roots were insignificantly changed but that of stems increased. The accumulation of total and reduced nitrogen in all organs with increasing concentrations of nitrogen supply were increased at 30 days after treatment but those of all organs at 60 and 90 days after treatment were highest in 600 $mg\;L^{-1}$, whereas there were no significant changes among other nitrogen concentration. The in vivo (${+NO_3}^-$) NRA of all organs did not relate to nitrogen concentration but the in vivo (${-NO_3}^-$) NRA of leaves except roots increased with increasing the nitrogen concentration. Therefore, the proper nitrogen concentration to promote growth and nitrate reduction of pear tree was 200 $mg\;L^{-1}$.

Potent Anticancer Effects of Lentivirus Encoding a Drosophila Melanogaster Deoxyribonucleoside Kinase Mutant Combined with Brivudine

  • Objective: Deoxyribonucleoside kinase of Drosophila melanogaster (Dm-dNK) mutants have been reported to exert suicide gene effects in combined gene/chemotherapy of cancer. Here, we aimed to further evaluate the capacity of the mutanted enzyme and its potential for inhibiting cancer cell growth. Methods: We altered the sequence of the last 10 amino acids of Dm-dNK to perform site-directed mutagenesis and constructed active site mutanted Dm-dNK (Dm-dNKmut), RT-PCR and western bloting studies were used to reveal the expression of lentivirus mediated Dm-dNKmut in a breast cancer cell line (Bcap37), a gastric cancer cell line (SGC7901) and a colorectal cancer cell line (CCL187). [3H]-labeled substrates were used for enzyme activity assays, cell cytotoxicity was assessed by MTT assays, cell proliferation using a hemocytometer and apoptosis induction by thenannexin-V-FITC labeled FACS method. In vivo, an animal study was set out in which BALB/C nude mice bearing tumors were treated with lentivirus mediated expression of Dm-dNKmut with the pyrimidine nucleoside analog brivudine (BVDU, (E)-5-(2-bromovinyl)-(2-deoxyuridine). Results: The Dm-dNKmut could be stably expressed in the cancer cell lines and retained its enzymatic activity. Moreover, the cells expressing Dm-dNKmut exhibited increased sensitivity in combination with BVDU, with induction of apoptosis in vitro and in vivo. Conclusion: These findings underlined the importance of BVDU phosphorylated by Dm-dNKmut in transduced cancer cells and the potential role of Dm-dNKmut as a suicide gene, thus providing the basis for future intensive research for cancer therapy.

소화관 내 압력 측정을 위한 텔레메트리 캡슐 구현 (Telemetry capsule for pressure monitoring in the gastrointestinal tract)

  • As the cause and the treatment about gastrointestinal disease has been issued recently, the importance of measuring the pressure in the gastrointestinal tract has been increased. However, the conventional measurement methods of the pressure in the gastrointestinal tract cause the patients' pain and inconvenience as well as an inaccurate pressure measurement. In this paper, the pressure monitoring telemetry system has been designed and implemented for an accurate pressure measurement inside the gastrointestinal tract with minimizing pain and inconvenience. The pressure monitoring telemetry system is composed of a pressure measurement capsule and an external receiver. The capsule has been miniaturized into the same size of a vitamin tablet so that the capsule can be swallowed through the oral cavity. After the capsule acquires and encodes the pressure data in the gastrointestinal tract, the encoded pressure data are modulated by frequency shift keying (FSK) and transmitted with ultrahigh frequency (UHF) band signal to the outside of a body. The performance of the telemetry capsule for monitoring pressure in the gastrointestinal tract is demonstrated by the results of animal in-vivo experiments.
