Comparison Study on the Cell Increase and Immunity Activation Degree in The extracted liquid of Cervus Elaphus Xanthopgus and It's Distilled Liquid (녹용추출액과 노제의 세포증식 및 면역활성도 비교 연구)

  • In order to searched for the ways of use for distilled liquid of Cervus Elaphus Xanthopgus, the study examined the difference of immunological efficiency generally raised in the existing extracted liquid and distilled liquid of Cervus Elaphus Xanthopgus. As seen from the above results, both the extracted liquid of Cervus Elaphus Xanthopgus and distilled liquid have remarkable efficiency in the increase of cells, the efficient condition of immunity activation degree, the effect of anti-cancer possibly formed by these two and activation of cells and organism, in which distilled liquid in the beginning and the extracted liquid later tend to show its efficiency.

  • In order to search for the ways of use in distilled liquid of Shogunjung-tang, an oriental medical prescription, the study examined the differences in the activated degree of cells and immunological effects, which are supplied by restoratives such as the existing extracted liquid and distilled liquid of Shogunjung-tang. As seen from the above results, both the extracted liquid of Shogunjung-tang and distilled liquid have remarkable efficiency in the increase of cells, the efficient condition of immunity activation degree possibly formed by these two and activation of cells and organism, in which distilled liquid in the beginning and the extracted liquid later tend to show its efficiency.

  • In order to study the effect of dietary krill meal on the performance and the utilization of protein and energy, broiler chicks fed on the experimental diets containing 0.0%, 0.5% and 1.0% krill meal, respectively, and injected Salmonella typhimurium lipopolysacharide(LPS) (Immunological stress) in 2nd week of age during 3 weeks of experimental period. Dietary krill meal did not affect daily gain and feed efficiency but the immunological stress lowered daily gain and feed intake and nitrogen balance and increased liver and spleen weight and the energy utilization of bird. The uric acid excretion was accelerated by immunological stress.

Effect of Dietary Xylitol on Growth Performance and Nitrogen Retention in Male Broiler Chicks during Immunological Stimulation

  • The effect of dietary xylitol on growth performance and nitrogen retention was studied in male broiler chicks during immunological stimulation. In experiment 1, chicks (10 day of age) were fed a corn-soybean diet containing 10% glucose or 10% xylitol with identical metabolizable energy and crude protein for 14 days. In experiment 2, ten-day-old chicks were fed 10% glucose or 6% xylitol diet for 8 days. During the final 6 days of the experimental periods, a half of birds fed each diet were injected intraperitoneally with 0.5 mg/kg body weight of Escherichia coli lipopolysaccharide (LPS, 0127:B8) on days 1, 3 and 5, and with 250 mg/kg body weight of Sephadex-G50 superfine on days 2 and 4 to stimulate immune system in both experiments. Feeding of the xylitol diets partially prevented the reduction in body weight gain or feed efficiency due to LPS and Sephadex injections, but the glucose diet did not in both the experiments. LPS and Sephadex injections decreased nitrogen retention, whereas the diet containing xylitol partly in experiment 1 and almost completely in experiment 2, prevented the reduction due to immunological stimulation. These results indicate that dietary xylitol probably prevents the reduction in nitrogen retention with growth retardation due to LPS and Sephadex injection. The beneficial effect on nitrogen retention is obtained when chicks are given xylitol 2 days before stimulating the immune system.

  • Specific polyclonal and/or monoclonal antibodies (MAbs) to immunoglobulins (Igs) and their subunits have proved to be valuable tools in immunological research and in immunological assays. In this study, we developed and characterized MAbs against flounder serum Igs. To obtain the pure flounder serum Igs, mouse IgG (mIgG) was immunized to flounder. Flounder Igs were purified by using mIgG-agarose affinity column chromatography. The structure of purified flounder Ig was observed, on denatured SDS-PAGE, to be composed of two heavy chains (77 and 72 kd) and two light chains (28 and 26 kd). MAbs were produced by fusion of myeloma cells (SP2/0) with Balb/c mouse spleen cells previously primed with the flounder Igs. Finally, three hybridoma clones, FIM 511, FIM 519 and FIM 562 were established to recognize both 2 heavy chains, 26 kd of light chain and 28 kd of light chain, respectively. On the other hand, the flounder immune sera collected on the weekly basis were tested on ELISA and immunoblot analysis whether boosting effect is present in flounder humoral immune system. As a result, the secondary immune response in flounder was ascertained on ELISA, but not on immunoblot analysis. Further, we observed an alteration of serum protein levels following immunization. Our MAbs and basic information on flounder humoral immune system obtained in this study will be helpful to control and monitor the efficiency of fish vaccines and therapeutic process of flounder diseases.

The Pros and Cons of SEW System - Review -

  • Early-weaning at an age of less than 21 days and removal of pigs to a second isolated site, which is commonly referred to as segregated early weaning (SEW), has been shown to substantially reduce disease transfer from the dam. This strategy has been successful in reducing the number of pathogens, but has not been successful in eliminating all pathogens. Although SEW has failed in most instances to totally eliminate pathogens, performance as measured by gain and efficiency through the nursery phase has been shown to be enhanced. In addition, SEW pigs have been shown to perform well on less complex nursery diets. Pigs which are continued on a similar isolation regime to market weight have been shown to have a remarkable improvement in feed intake, gain and efficiency. However, pigs which are co-mingled with conventional pigs after the nursery phase have either no improvement in performance or reduced performance. Backfat and carcass lean yield have been shown to be enhanced by SEW in high lean gain pigs, but not in lower lean gain pigs. Exposure of pigs to antigens which activate the immune system and increase the level of immunological stress has been suggested as the mechanism involved in depressing growth and performance of pigs.

  • Weaning is a complex phase when the mammal suffers the action of different stressors that contribute to negatively affect the efficiency of the intestinal mucosa and of the whole local integrated system, that acts as barrier against any nocuous agent. The components of this barrier are mechanical, chemical, and bacteriological; immunological and not. The development of contact with a saprophyte microflora and the maintenance of feed intake after the interruption of motherly nutrition are essential for the maturation of an equilibrated local immune function and for a functional integrity of villi. Opportunities and limits of some dietary strategies that can contribute to reduce negative effects of weaning on health and performance are discussed. Knowledges on the possible mechanism of action of probiotics are upgraded, particularly for their supposed role in the balance between different immune functions (effectory/regulatory). Some tools to control pathogen microflora are reviewed (acids, herbs, immunoglobulin sources) and practical feeding systems are proposed.

  • Cell-penetrating peptides (CPPs) are short amino acids that have been widely used to deliver macromolecules such as proteins, peptides, DNA, or RNA, to control cellular behavior for therapeutic purposes. CPPs have been used to treat immunological diseases through the delivery of immune modulatory molecules in vivo. Their intracellular delivery efficiency is highly synergistic with the cellular characteristics of the dendritic cells (DCs), which actively uptake foreign antigens. DC-based vaccines are primarily generated by pulsing DCs ex vivo with various immunomodulatory antigens. CPP conjugation to antigens would increase DC uptake as well as antigen processing and presentation on both MHC class II and MHC class I molecules, leading to antigen specific CD4+ and CD8+ T cell responses. CPP-antigen based DC vaccination is considered a promising tool for cancer immunotherapy due to the enhanced CTL response. In this review, we discuss the various applications of CPPs in immune modulation and DC vaccination, and highlight the advantages and limitations of the current CPP-based DC vaccination.

Indoor feeding combined with restricted grazing time improves body health, slaughter performance, and meat quality in Huang-huai sheep

  • Objective: The aim of this study was to evaluate the effects of three feeding systems, i.e., indoor feeding (CON), indoor feeding with 4-h daily access to grazing artificial pasture (ITGP), and indoor feeding with 8-h daily access to grazing artificial pasture (IEGP), on the plasma antioxidant and immunological capacity, slaughter characteristics, meat quality and economic efficiency of Huang-huai lambs. Methods: Thirty-three healthy Huang-huai rams with similar body weight (approximately 5 mo of age, 28.96±1.01 kg) were assigned equally to three experimental groups. When finished fattening, six lambs from each group were collect blood samples for plasma analyses and then slaughtered to determine slaughter characteristics and obtain biceps brachii muscle for further analysis of meat quality and fatty acid profile. Results: Compared to CON group, animals submitted to ITGP and IEGP groups resulted in greater contents of serum glutathione peroxidase, immunoglobulins (IgA, IgG, and IgM), polyunsaturated fatty acids (PUFA), n-6 PUFA, and PUFA/saturated fatty acid (FA) ratio and lower palmitic /oleic acid ratio (p<0.05). Moreover, animals in ITGP group exhibited a higher (p<0.05) loin eye area, content of meat crude protein (CP), and eicosetrienoic acid compared to CON group, while slaughter performance was superior (p<0.05) to that of the IEGP group. The economic efficiency of ITGP group was 70.12% higher than that of CON group, while the IEGP group exhibited a decrease of 92.54% in economic efficiency compared to the CON group. Conclusion: Restricted grazing time combined with indoor feeding was more effective in conferring superior body health, carcass traits and economic efficiency in Huang-huai lambs, as well as higher CP content and healthier FA composition in the resulting meat.

Direct Detection of Escherichia coli, Staphylococcus aureus, and Salmonella spp. in Animal-derived Foods Using a Magnetic Bead-based Immunoassay

  • In this study, an immuno-magnetic bead (IMB)-based assay was developed to simultaneously detect Escherichia coli, Staphylococcus aureus, and Salmonella spp. and was tested in four animal-derived foods: beef, ham, egg, and ricotta cheese. The IMB-based assay exhibited good specificity by binding to five E. coli serotypes [capture efficiency (CE) average (avg.) 90.4%], five S. aureus strains (CE avg. 91.4%), and five Salmonella serotypes (CE avg. 95.4%) but not binding to non-target bacteria (CE<10%). Furthermore, the assay detected all three pathogens with a detection limit of 10 CFU/g without the need for enrichment or additional platforms. Since the results demonstrated that the IMB-based assay can effectively separate and enrich target bacteria from a variety of animal-derived food matrixes, the assay exhibits good specificity for potential use in providing rapid, immunological, presumptive identification of pathogenic bacteria.
