Effects of in vitro culture types on regeneration and acclimatization of yellow poplar (Liriodendron tulipifera L.) from somatic embryos

  • We compared germination efficiency for somatic embryos (SE) of Liriodendron tulipifera using semi-solid (SS), temporary immersion bioreactors (TIB), and continuous immersion bioreactors (CIB) to produce vigorous plants. The bioreactors were designed to be immersed in liquid media with plantlets with an adjustable immersion time. TIB and CIB improved germination rates up to 80.86% and 95.21%, respectively, however, CIB produced more hyperhydric plantlets than TIB. The height of plantlets in TIB was significantly higher than for those in CIB. Fresh weights of plantlets grown in CIB of were significantly lower than for those grown in TIB. The lowest chlorophyll concentration was found in in vitro plantlets from CIB. We examined abnormally developed leaves, stems, and apical zones of in vitro plantlets that were produced in CIB. Among the three types, SS showed the highest stomatal density and the shortest stomatal length in in vitro plantlets. After acclimatization, plants from CIB exhibited the lowest values in biomass, such as height, root collar diameter, leaf fresh weight, leaf length, leaf width, petiole length, petiole diameter, and leaf area. Photosynthesis and transpiration rates of ex vitro plants were not significantly different among the three culture types, but stomatal conductance was higher in TIB than in the SS and CIB. Therefore, the results suggest that TIB is the preferable bioreactor to improve in vitro plantlet regeneration of L. tulipifera. TIB-originated plants showed higher growth rate than SS and CIB after transferring to soil.

Microstructure and Mechanical Property Changes of Unidirectional and Plain Woven CF/Mg Composite Laminates after Corrosion (일방향 및 평직 CF/Mg 복합재 적층판의 부식에 따른 미세조직 및 기계적 특성 변화)

  • In this study, unidirectional and plain woven carbon fiber reinforced magnesium matrix composite laminates were fabricated by the liquid pressing infiltration process, and evolutions of the microstructure and compressive strength of the composite laminates under corrosion were investigated by static immersion tests. In the case of the unidirectional composite laminate, the main microstructural damage during immersion appeared as a form of corrosion induced cracks, which were formed at both CF/Mg interfaces and the interfaces between layers. On the otherhand, wrap/fill interface cracks were mainly formed in the plain woven composite laminate, without any cracks at the CF/Mg interface. The formation of these cracks was considered to be associated with internal thermal residual stress, which was generated during cooling after the fabrication process of these materials. As a consequence of the corrosion induced cracks, the thickness of both laminates increased in directions vertical to the fibers with increasing immersion time. With increasing immersion time, the compressive strengths of both composite laminates also decreased continuously. It was found that the plain woven composite laminates have superior corrosion resistance and stability under a corrosive condition than unidirectional laminates.

Effects of Immersion Liquid and High Hydrostatic Pressure on the Physicochemical Quality Characteristics of Scomber japonicus (침지액 및 초정수압의 병행처리에 의한 고등어(Scomber japonicus)의 물리화학적 품질 특성 변화)

  • This study investigated the effects of combined treatments of immersion liquid and high hydrostatic pressure (HHP, 100−400 MPa) on the quality of mackerel. In this study, we measured viable cell counts, pH level, color value, texture properties, and sensory evaluation of mackerel. Viable cell counts of mackerel treated with combined HHP were decreased by 2 log cycles at 300 MPa and 400 MPa compared to the control. The pH values of mackerel treated with HHP at 300 MPa and 400 MPa were higher than that of other samples. Hardness, gumminess, and chewiness decreased when treated with combined HHP and increased with increase in HHP. Lightness and whiteness of mackerel increased, but redness decreased, with increase in HHP. In case of sensory evaluation, texture of mackerel treated with HHP at 300 MPa and 400 MPa showed higher scores than that of other samples. These results suggest that immersion liquid and HHP treatments can increase microbiological safety and improve textural properties of mackerel.

Changes of the surface roughness depending on immersion time and powder/liquid ratio of various tissue conditioners (수종의 조직 양화재의 침수시간과 분액비에 따른 표면 거칠기의 변화)

  • Statement of problem: Volume stability, microstructure reproducibility and fluidity along with compatibility with dental stone must be in consideration in order to use tissue conditioner as a material for functional impression. There are few studies concerning the influence of time factor in oral condition on surface roughness of the stone and optimal retention period in the oral cavity considering such changes in surface roughness. Purpose: The purpose of this study was to find out the influence of various kinds of tissue conditioner, its powder/liquid ratio and immersion time on surface roughness of the stone. Material and methods: Materials used in this study were the three kinds of tissue conditioners(Coe-Comfort, Visco-Gel, Soft-Liner) and were grouped into three: group R-mixed with standard powder/liquid ratio that was recommended by the manufacturers, group M-mixed with 20% more powder, group L-mixed with 20% less powder. Specimens were made with the size of 20 mm diameter and 2 mm width. Each tissue conditioner specimens were subdivided into 5 groups according to the immersion time(0 hour, 1 day, 3 days, 5 days, 7 days), completely immersed into artificial saliva and were stored under $37^{\circ}C$. Specimens of which the given immersion time elapsed were taken out and were poured with improved stone, making the stone specimens. Surface roughness of the stone specimens was measured by a profilometer. Results: Within the limitation of this study, the following results were drawn. 1. Major influencing factor on surface roughness of the stone model made from tissue conditioner was the retention period(contribution ratio($\rho$)=62.86%, P<.05) of the tissue conditioner in oral cavity to make functional impression. 2. In case of Coe-Comfort, higher mean surface roughness value of the stone model with statistical significance was observed compared to that of Soft-Liner and Visco-Gel as immersion time changes(P<.05). 3. In case of group L(less), higher mean surface roughness value of the stone model with statistical significance was observed compared to that of R(recommended) and M(more) group as immersion time changes(P<.05). Conclusion: We may conclude that as the retention period of time in oral cavity influences surface roughness of the stone model the most and as the kind of tissue conditioner and its P/L ratio may influence also, clinician should well understand the optimal retention period in oral cavity and choose the right tissue conditioner for the functional impression, thus making the functional impression with tissue conditioner usefully.

An experimental study on the atomizing characteristics of liquid column type coaxial sprays (액주형 동축노즐 분무의 무화특성에 관한 실험적 연구)

  • The main purpose of this study is to investigate the atomizing characteristics of a two phase spray by using a liquid column type coaxial nozzle. The experiments have been carried out to analyze the atomization behavior, the droplet size distributions, and the statistical properties of droplet size distributions. Immersion sampling method and the image processing technique were adapted for the measurements of particles, and the distributions of the droplet sizes were statistically analyzed. In the experiments, the mass ratio defined as Mr= $M_{\sigma}$/ $M_{1}$ has been changed from 1.0 to 3.4 and the measurements have been performed along the axis of the spray. As a result of this experimental study, the distributions of droplet size were satisfied with the Log-Normal distributions and arithmetic mean diameter and deviation of mass ratio. Droplet volume-surface mean diameter was denoted by a exponential function of mass-ratio and the exponent was denoted by linear relation according to the central axis from the nozzle. Dispersions, skewness factors and flatness factors had comparatively constant values regardless of mass ratio and location.

  • 1. INTRODUCTION : The phase inversion method is widely used to prepare a variety of polymeric membranes ranging from micro-filtration to gas separation. The final morphology obtained by immersion precipitation strongly reflects the thermodynamics and kinetics of the system involved. The equilibrium thermodynamics of the ternary system of polymer/solvent/ nonsolvent is still very important to understand and predict membrane structure. Polysulfone (PSf) and polyethersulfone (PES) are important polymers as membrane materials due to the chemical resistance, mechanical strength, thermal stability and transport properies. There are several reports on the experimental phase diagrams in ternary mixtures of PSf/solvent/nonsolvent, and PES/solvent/nonsolvent. It would be interesting to investigate the solution thermodynamics containing these two polymers since PES is slightly less hyclrophobic than PSf.

The effects of different polishing techniques on the staining resistance of CAD/CAM resin-ceramics

  • PURPOSE. The purposes of this study were to evaluate the staining resistance of CAD/CAM resin-ceramics polished with different techniques and to determine the effectiveness of the polishing techniques on resin-ceramics, comparing it with that of a glazed glass-ceramic. MATERIALS AND METHODS. Four different CAD/CAM ceramics (feldspathic ceramic: C-CEREC Blocs, (SIRONA) and three resin-ceramics: L-Lava Ultimate, (3M ESPE), E-Enamic, (VITA) and CS-CeraSmart, (GC)) and one light cure composite resin: ME-Clearfil Majesty Esthetic (Kuraray) were used. Only C samples were glazed (gl). Other restorations were divided into four groups according to the polishing technique: nonpolished control group (c), a group polished with light cure liquid polish (Biscover LV BISCO) (bb), a group polished with ceramic polishing kit (Diapol, EVE) (cd), and a group polished with composite polishing kit (Clearfil Twist Dia, Kuraray) (kc). Glazed C samples and the polished samples were further divided into four subgroups and immersed into different solutions: distilled water, tea, coffee, and fermented black carrot juice. Eight samples ($8{\times}8{\times}1mm$) were prepared for each subgroup. According to CIELab system, four color measurements were made: before immersion, immersion after 1 day, after 1 week, and after 1 month. Data were analyzed with repeated measures of ANOVA (${\alpha}=.05$). RESULTS. The highest staining resistance was found in gl samples. There was no difference among gl, kc and cd (P>.05). Staining resistance of gl was significantly higher than that of bb (P<.05). Staining resistances of E and CS were significantly higher than those of L and ME (P<.05). CONCLUSION. Ceramic and composite polishing kits can be used for resin ceramics as a counterpart of glazing procedure used for full ceramic materials. Liquid polish has limited indications for resin ceramics.

An Experimental Study on the Behavior of Twin-Spray with Flow Interaction in a Condensable Environment (주위기체내에서의 두 액체분무간의 유동간섭현상에 대한 정상적 고찰)

  • The effects of flow interaction between adjacent sprays in twin-spray system on the spatial distribution of injected liquid (water) and drop size distribution in condensable (steam) environment were carefully observed through experiments. The spatial distribution of injected liquid in twin-spray system appears to be more uniform than the simple superposition of the spatial distributions of liquid obtained from each individual spray. Drop size distribution was obtained by using the immersion sampling technique. It was found that, in the twin-spray, the larger numbers of small drops are collected throughout the spraying region due to the increase of entrainment velocity of ambient steam compared with the case of simple superposition of each individual spray. Moreover, in the overlapped portion of the twin-spray, the drop size distribution was changed also due to the collision between large drops. As a result, the behavior of twin-spray system (and eventually multiple-spray system) can not be predicted precisely by simple superposition of the behaviors of each constituting spray. Hence, for the design of multiple spray system, the effect of flow interaction between sprays should be taken into account seriously.

Characteristics of Surface Flashover of spacer in Liquid Nitrogen (액체 질소중에서 고체 Spacer에 의한 연면 방전특성)

  • Composite insulation system of liquid nitrogen and solid spacer is widely applied in high temperature superconducting power machine. This study has three step procedure. As follow, first step is composition of parallel deposited electrode and vertically deposited electrode along the direction of immersion in liquid nitrogen($LN_2$). Second step is investigation into surface flashover voltage of solid spacer under partially immersed in $LN_2$, and last step is comparison the result of this research with that of fully immersed in $LN_2$ and at cryogenic temperature gaseous nitrogen($GN_2$). This result presented that surface flashover voltage along solid spacer half immersed in $LN_2$ was almost the same as that of fully immersed spacer when the thickness of spacer(t) was t<10 mm. In the case of t>10 mm, however, spacer flashover voltage was equal to that obtained in $GN_2$ at cryogenic temperature.

The Effect of Digestive Medicine on Enamel Erosion (액상소화제가 법랑질 부식에 미치는 영향)

  • Consumption of liquid digestive medicine has continually grown in recent years. This present study was designed to evaluate the capability of liquid digestive medicine to erode dental enamel, relating the pH and titratable acidity of liquid digestive medicine. Three commercially available liquid digestive medicines were chosen these were Gashwalmyeungsu, Saengrokchun and Wicheongsu. The liquid digestive medicines were evaluated in respect to pH, titratable acidity and concentrations of calcium and phosphate, respectively. This measure was carried out three times for each digestive medicine and was recorded the data as mean (standard deviation). Bovine enamel specimens measured microhardness at base line and then were randomly assigned to 4 groups of 10 each. The specimens were immersed into each liquid digestive medicine for 1, 3, 5, 10, 15, and 30 minutes and then evaluated each time by VHN. All digestive medicines had a pH of less than 3.7. Wicheongsu had the lowest pH 2.93 and Gashwalmyeungsu had highest pH 3.63. In pH 5.5, titratable acidity of Wicheongsu was 1.27 ml. Gashwalmyeungsu was 0.63 ml. Saengrokchun was 0.60 ml. All liquid digestive medicines showed low concentration of calcium and phosphate. The microhardness of specimens after immersion into liquid digestive medicines was continuously reduced in all digestive medicines. After 30-minute treatment in liquid digestive medicines, Wicheongsu containing low pH and high tiratable acidity was shown to be lowest microhardness value ($207.80{\pm}15.52$). The three liquid digestive medicines caused surface softening of enamel erosion. We conclude that drinks, commonly consumed by functional dyspepsia patient can cause erosion of enamel.
