Immature thymocyte antigen, JL1, as a possible immunodiagnostic and immunotherapeutic target for leukemia

  • The identification of tumor-specific antigens has represented a critical milestone in cancer diagnosis and therapy. Clinical research in this area for leukemia has also been driven over the past few decades by the hope that surface antigens with restricted tissue expression would be identified. Disappointingly, only a small number of the leukemic antigens identified to date, meet sufficient criteria to be considered viable immunophenotypic markers. In this paper, we nominate anti-JL1 monoclonal antibody as an immunodiagnostic and immunotherapeutic candidate for leukemia. The JL1 molecule appears to be a novel cell surface antigen, which is strictly confined to a subpopulation of limited stages during the hematopoietic differentiation process. Despite the restricted distribution of the JL1 antigen in normal tissues and cells, anti-JL1 monoclonal antibody specifically recognizes various types of leukemia, irrespective of immunophenotypes. On the basis of these findings, we propose JL1 antigen as a tumor-specific marker, which shows promise as a candidate molecule for diagnosis and immunotherapy in leukemia, and one that spares normal bone marrow stem cells.

Reproduction of the Shotted halibut in the southern Korean waters (한국 남해에 서식하는 물가자미, Eopsetta grigorjewi (Herzenstein)의 재생산 연구)

  • Maturation and spawning of the Shotted halibut, Eopsetta grigorjewi was investigated based on the samples captured in South Korean waters from January 2008 to December 2009. Gonadosomatic index began to increase in December, and reached maximum between January to March. After spawning it began to decrease from May. Reproductive season was estimated to January-April, with peak in February. Fecundity was proportional to the size of the female, with the clutch size varying from 170,000 eggs in the smallest female (total length, 28.9cm) to 1,300,000 eggs in the largest (total length, 41.5cm). Size at 50% sexual maturity (TL50), determined from mature females, was 28.8cm. Annual reproductive cycles of this species could be divided into six successive stages; immature stage (May-October), nucleolus stage (November-January), yolk vesicle stage (January-February), vitellogenic and ripe stage (January-April) and spent stage (April-May).

  • Effect of fluctuating temperatures on the development of the immature beet armyworm, Spodoptera exigua (H bner), was analyzed. At constant rearing temperature regimes, the estimated developmental threshold temperatures were varied among stages and instars, but had an average 13$^{\circ}C$ from egg hatch to adult emergence. Based on the 13$^{\circ}C$ threshold temperature, we set up three different rearing temperature regimes having the same day-degrees. Two fluctuating temperature regimes changed significantly the developmenta1 period expected by the constant rearing temperature regime. Under the same thermophase temperature (25$^{\circ}C$), the thermocycling regime with the higher cryophase temperature (10$^{\circ}C$) decelerated the developmental rate probably by lowering temperature limit thor development, but that with the lower cryophase temperature (5$^{\circ}C$) gave a negative developmental effect.

  • In vitro fertilization(IVF) derived morula and blastocyst embryos were bisected by a simple method and cultured in vitro without zona pellucida And also bisected embryos were frozen-thawed and cultured in vitro) to evaluate the survival rate. The results obtained were as follows : The average number of grade I or II immature follicular oocytes recovered by slicing method per ovary was 11.9 from 142 ovaries.   • Effects of $\alpha$-difluoromethylornithine (DFMO) and $\alpha$-difluoromethylarginine (DFMA), inhibitors of polyamine syntheic enzymes (ornithine decarboxylase, arginine decarboxylase), on ovary were investigated during pupal-adult development of Helicoverpa assulta. Two inhibitors (DFMO, DFMA) showed definite inhibition effects on ovarian development. The inhibition effect on ovaries weight was more marked in DFMA-injection than that observed in DFMO-injection. Two inhibitors (DFMO, DFMA) gave rise to a peculiar decrease in ornithine decarboxylase (ODC) or arginine decarboxylase (ADC) activity in ovaries, 72 hrs (5-day old pupa) post injection. However, DFMO clearly exhibited supression of ODC activity after 96 hrs (6-day old pupa). In addition, two inhibitors (DFMO, DFMA) diminished putrescine content in the ovary. The spermidine level was slightly decreased by each injection of the inhibitors. However, two inhibitors (DFMO, DFMA) raised the spermine content at certain developmental stages in the ovary. Although the effect of DFMA was less severe, two inhibitors (DFMO, DFMA) caused not only an overall delay in ovarian development, but also abnormalities in cellular differentiation. Noted effects in the pupal ovary were the appearance of irregular nurse cells and partial destruction of follicle epidermal cells. Adult ovary showed rapid degradation of nurse cells, a reduction in the number of follicle epidermal cells and immature oocytes that had a low yolk content.

  • This study was carried out to screen the fruiting body formation-specific genes from the medicinal mushroom Cordyceps militaris. A cDNA synthesized using total RNA from 4 stages of mushroom development, mycelium, primordium, immature fruiting body and mature fruiting body. Differential expression gene screening was performed by DD-PCR(Differential Display Arbitrary Primer PCR) with cDNA, we sequenced partial 6 genes using pGEM cloning vector. The DNA Sequence of the six DD-PCR products derived from differentially expressed genes was compared to that in the GenBank database by using the NCBI BLAST search to identify similarities to known sequences. Sequence analysis showed that six of DD-PCR products have unknown sequence.

Comparative Spawning Ecology of Daggertooth Pike Conger Muraenesox cinereus and Common Pike Conger M. bagio in the South Sea of Korea (갯장어(Muraenesox cinereus)와 갈창갯장어(M. bagio)의 산란생태 비교)

  • We examined the comparative spawning ecology of daggertooth pike conger Mureanesox cinereus and common pike conger M. bagio in the South Sea of Korea from May 2018 to December 2019. In the South Sea of Korea, the mean total length (TL) of M. cinereus was significantly smaller than that of M. bagio. The sex ratios of both species were skewed toward females. The spawning period of M. cinereus was from June to October, while the spawning period of M. bagio was from August to October. The lengths at 50%, 75%, and 97.5% sexual maturity of M. cinereus females were estimated as 54.6 cm, 60.4 cm, and 74.1 cm TL, respectively. It was not possible to estimate lengths at sexual maturity for M. bagio in the South Sea of Korea due to the absence of immature stages.

In Vitro Developmental Competence of Vitrified Human Oocytes (유리화 동결된 인간난자의 체외발생능)

  • These studies were undertaken to evaluate morphological normality and development competence in vitro of hyman oocytes following vitrificatioin using ethylene glycol and electron microscopic grid. Human immature oocytes retrieved from natural and stimulated cycles was vitrified at 0 or 48 h and 0, 8 to 15 or 24 to 28 h after maturation culture, respectively. In oocytes retrieved from unstimulated cycle, no signifciant differences were found in morphological normality (56 to 63%) and fertilization (31 to 37%) rates between the times of vitrification. In stimulated patients, however, more oocytes were morphologically normal when vitrified at 24 to 28 h than when vitrified at 0 or 8 to 15 h after maturation culture. Regardlesss of the hormonal stimulation, high cleavage rates(83 to 100%) were obtained in all treatment groups but did not differ significantly. Twenty to 43% of cleaved oocytes developed to the blastocyst stage at 6 days after IVF. These results suggest that vitrified oocytes from unstimulated and stimulated cycles could develop to the blastocyst stage, regardless of the stages of vitrification.

Morphology and Ecological Notes on the Larvae and Pupae of Simulium (Simulium) from Korea

  • Morphological features of larvae and pupae of twelve species of Simulium (Simulium) from Korea are investigated and redescribed. Detailed descriptions of immature stages of each species are provided with photographs. For the Korean members of subgenus Simulium, combinations of the following characteristics can be used to separate them from other Korean subgenera: larval abdomen without a pair of conical-shaped ventral tubercles; postgenal cleft variously shaped, viz., miter shaped, spearhead, bullet shaped or triangular; rectal papillae 3 simple lobe or compound lobes; pupal gill of 6-16 filaments; cocoon shoe, boot or slipper shaped; and head trichomes 3 pairs (1 facial+2 frontal). Habitat information and other ecological aspects of each species are provided. Updated keys to larvae and pupae of the Korean species of the subgenus Simulium are also provided to facilitate species identification. Taxonomic treatment based on external morphologies of larvae and pupae of Korean species of the subgenus Simulium are focused and included in this study. To understand their interspecific relationships, phylogenetic analyses using multiple data from molecule, morphology, and ecology are warranted in future research. Also continued exploration for new character systems is needed to establish more stable boundaries for subgenus and species delimitations.
