MS2 Labeling of Endogenous Beta-Actin mRNA Does Not Result in Stabilization of Degradation Intermediates

  • The binding of MS2 bacteriophage coat protein (MCP) to MS2 binding site (MBS) RNA stem-loop sequences has been widely used to label mRNA for live-cell imaging at single-molecule resolution. However, concerns have been raised recently from studies with budding yeast showing aberrant mRNA metabolism following the MS2-GFP labeling. To investigate the degradation pattern of MS2-GFP-labeled mRNA in mammalian cells and tissues, we used Northern blot analysis of ${\beta}$-actin mRNA extracted from the Actb-MBS knock-in and $MBS{\times}MCP$ hybrid mouse models. In the immortalized mouse embryonic cell lines and various organ tissues derived from the mouse models, we found no noticeable accumulation of decay products of ${\beta}$-actin mRNA compared with the wild-type mice. Our results suggest that accumulation of MBS RNA decay fragments does not always happen depending on the mRNA species and the model organisms used.

  • Brachytherapy, along with external beam radiation therapy (EBRT), is an essential and effective radiation treatment process. In brachytherapy, in contrast to EBRT, the radiation source is radioisotopes. Because these isotopes can be positioned inside or near the tumor, it is possible to protect other organs around the tumor while delivering an extremely high-dose of treatment to the tumor. Brachytherapy has a long history of more than 100 years. In the early 1900s, the radioisotopes used for brachytherapy were only radium or radon isotopes extracted from nature. Over time, however, various radioisotopes have been artificially produced. As radioisotopes have high radioactivity and miniature size, the application of brachytherapy has expanded to high-dose-rate brachytherapy. Recently, advanced treatment techniques used in EBRT, such as image guidance and intensity modulation techniques, have been applied to brachytherapy. Three-dimensional images, such as ultrasound, computed tomography, magnetic resonance imaging, and positron emission tomography are used for accurate delineation of treatment targets and normal organs. Intensity-modulated brachytherapy is anticipated to be performed in the near future, and it is anticipated that the treatment outcomes of applicable cancers will be greatly improved by this treatment's excellent dose delivery characteristics.

Radiation dosimetry of 89Zr labeled antibody estimated using the MIRD method and MCNP code

  • One important issue in using radiopharmaceuticals as therapeutic and imaging agents is predicting different organ absorbed dose following their injection. The present study aims at extrapolating dosimetry estimates to a female phantom from the animal data of 89Zr radionuclide accumulation using the Sparks-Idogan relationship. The absorbed dose of 89Zr radionuclide in different organs of the human body was calculated based on its distribution data in mice using both MIRD method and the MCNP simulation code. In this study, breasts, liver, heart wall, stomach, kidneys, lungs and spleen were considered as source and target organs. The highest and the lowest absorbed doses were respectively delivered to the liver (4.00E-02 and 3.43E-02 mGy/MBq) and the stomach (1.83E-03 and 1.66E-03 mGy/MBq). Moreover, there was a good agreement between the results obtained from both MIRD and MCNP methods. Therefore, according to the dosimetry results, [89Zr] DFO-CR011-PET/CT seems to be a suitable for diagnostic imaging of the breast anomalies for CDX-011 targeting gpNMB in patients with TNBC in the future.

Spectroscopic Detection of Alfvénic Waves in Chromospheric Mottles of a Solar Quiet Region

  • We present high resolution spectroscopic observations of transverse magnetohydrodynamic (MHD) waves in mottles located near the solar disk center. Different from previous studies that used transversal displacements of the mottles in the imaging data, we investigated the line-of-sight (LOS) velocity oscillations of the mottles in the spectral data. The observations were carried out by using the Fast Imaging Solar Spectrograph of the 1.6 meter Goode Solar Telescope of Big Bear Solar Observatory. Utilizing the spectral data of the Hα and Ca II 8542 Å lines, we measure the LOS velocity of a quiet region including the mottles and rosettes that correspond to the footpoints of the mottles. Our major findings are as follows: (1) Alfvénic waves are pervasive in the mottles. (2) The dominant period of the waves is 2 to 4 minutes. (3) From the time-distance maps of the three-minute filtered LOS velocity constructed along the mottles, it is revealed that the transverse waves in the mottles are closely related to the longitudinal waves in the rosettes. Our findings support the notion that Alfvénic waves can be generated by mode conversion of the slow magnetoacoustic waves as was shown in sunspot regions by Chae et al. (2021).

Investigation of photon, neutron and proton shielding features of H3BO3-ZnO-Na2O-BaO glass system

  • The current study aims to explore the shielding properties of multi-component borate-based glass series. Seven glass-samples with composition of (80-y)H3BO3-10ZnO-10Na2O-yBaO where (y = 0, 5, 10, 15, 20, 25 and 30 mol.%) were synthesized by melt-quench method. Various shielding features for photons, neutrons, and protons were determined for all prepared samples. XCOM, Phy-X program, and SRIM code were performed to determine and explain several shielding properties such as equivalent atomic number, exposure build-up factor, specific gamma-ray constants, effective removal cross-section (ΣR), neutron scattering and absorption, Mass Stopping Power (MSP) and projected range. The energy ranges for photons and protons were 0.015-15 MeV and 0.01-10 MeV, respectively. The mass attenuation coefficient (μ/ρ) was also determined experimentally by utilizing two radioactive sources (166Ho and 137Cs). Consistent results were obtained between experimental and XCOM values in determining μ/ρ of the new glasses. The addition of BaO to the glass matrix led to enhance the μ/ρ and specific gamma-ray constants of glasses. Whereas the remarkable reductions in ΣR, MSP, and projected range values were reported with increasing BaO concentrations. The acquired results nominate the use of these glasses in different radiation shielding purposes.

Dosimetric Study Using Patient-Specific Three-Dimensional-Printed Head Phantom with Polymer Gel in Radiation Therapy

  • Purpose: In this study, we aimed to manufacture a patient-specific gel phantom combining three-dimensional (3D) printing and polymer gel and evaluate the radiation dose and dose profile using gel dosimetry. Methods: The patient-specific head phantom was manufactured based on the patient's computed tomography (CT) scan data to create an anatomically replicated phantom; this was then produced using a ColorJet 3D printer. A 3D polymer gel dosimeter called RTgel-100 is contained inside the 3D printing head phantom, and irradiation was performed using a 6 MV LINAC (Varian Clinac) X-ray beam, a linear accelerator for treatment. The irradiated phantom was scanned using magnetic resonance imaging (Siemens) with a magnetic field of 3 Tesla (3T) of the Korea Institute of Nuclear Medicine, and then compared the irradiated head phantom with the dose calculated by the patient's treatment planning system (TPS). Results: The comparison between the Hounsfield unit (HU) values of the CT image of the patient and those of the phantom revealed that they were almost similar. The electron density value of the patient's bone and brain was 996±167 HU and 58±15 HU, respectively, and that of the head phantom bone and brain material was 986±25 HU and 45±17 HU, respectively. The comparison of the data of TPS and 3D gel revealed that the difference in gamma index was 2%/2 mm and the passing rate was within 95%. Conclusions: 3D printing allows us to manufacture variable density phantoms for patient-specific dosimetric quality assurance (DQA), develop a customized body phantom of the patient in the future, and perform a patient-specific dosimetry with film, ion chamber, gel, and so on.

  • 4.7T의 고자장 자기공명 영상(Magnetic Resonance imaging : MRI) 장치에서 Tailored RF 경사자계반향(Tailored RF Gradient-Echo : TRGE)기법을 이용하여 phantom 영상과 고양이 뇌의 정맥혈관 영상을 얻었다. 증류수에 MgCl$_2$를 섞어 T1을 짧게한 지름 6cm의 원통형 phantom으로 TRGE기법에 대한 검증을 먼저 하였다. 이후 halothane으로 전신 마취된 몸무게 3.0~4.0kg의 고양이 뇌로부터 얻은 시단면(sagittal slice) 영상에서 Dorsal sagittal sinus, Straight sinus, Vein of corpus callosum과 Internal cerebral vein등의 혈관이 강조되어 나타남을 고양이 뇌의 해부학적 그림과 비교해 보였다. Tailored RF 파형은 ASPECT 3000 computer(스위스 BRUKER사 제품)의 PASCAL 언어를 이용하여 제작하였다. 사각형의 절편모양(slice profile)과 원하는 절편의 두께(slice thickness)에 선택절편내에서 절편방향으로 위상분포가 최대 2$\pi$가 되는 bi-linear ramp 모양을 갖는 절편함수를 공간상에서 Fourier 변환시켜 Tailored RF를 만들었다. TR/TE=205/10 msec, 절편두께 TH=7mm, 화소배열수(matrix size)=256$\times$256, 평면해상도(in-plane resolution)=0.62$\times$0.31mm$^2$, 관심영역 크기(field of view : FOV)=8cm의 영상조건으로 자기공명 TRGE 영상을 얻었다.

Spectroscopic Confirmation of Galaxy Clusters at z~0.92

  • Galaxy clusters have provided important information to understand the evolution of the universe, since the number density and mass of clusters are tightly related to the cosmological parameters. In addition, galaxy clusters are an excellent laboratory to investigate the galaxy evolution in dense environments. However, finding galaxy clusters at high redshift ($z{\geq}1$) still remains as a main subject in astronomy due to their rareness and difficulty in identifying such objects from optical imaging data alone. Here, we report a spectroscopic follow-up observation of distant galaxy cluster candidates identified by a deep optical-NIR dataset of Infrared Medium-deep Survey. Through the galaxy spectra taken with the IMACS instrument on the Magellan telescope, we confirm at least 3 massive clusters at z~0.92. Interestingly, the maximum spatial separation between these clusters is ~8Mpc, which implies that this system is a new supercluster in the distant universe. We also discuss properties of galaxies in these clusters based on multi-wavelength photometric data.

New candidates of 1 < z < 2 galaxy clusters in 13.6 $deg^2$ of ELAIS-N1/N2 fields with a new colour-colour selection technique

  • Galaxy clusters, the largest gravitationally bound systems, are an important means to place constraints on cosmological models. Moreover, they are excellent places to test galaxy evolution models in connection to the environments. To this day, massive clusters have been found unexpectedly at high redshfit (Kang & Im 2009, Durret et al. 2011, Tashikawa et al. 2012), and evolution of galaxies in cluster has not been fully understood. Finding galaxy cluster candidates at z > 1 in wide, deep imaging survey data will enable us to solve such issues of modern extragalactic astronomy. We report new candidates of galaxy clusters in the wide and deep survey fields, European Large Area ISO Survey North1(ELAIS-N1) and North2(ELAIS-N2) fields, covering sky area of $8.75deg^2$ and $4.85deg^2$ each. We also suggest a new useful colour-colour selection technique to separate 1 < z < 2 galaxies from low-z galaxies by combining multi-wavelength data from the UKIRT Infrared Deep Sky Survey Deep Extragalactic Survey (UKIDSS DXS, JK bands), Spitzer Wise-area InfraRed Extragalactic survey (SWIRE, Optical-Infrared bands), Canada France Hawaii Telescope (CFHT, z band) and Infrared Medium-deep Survey(IMS, J band).

Monitoring observation of PG0934+013 using The Southern African Large Telescope

  • We performed spectroscopic and photometric monitoring observations of a QSO, PG0934+013 for a reverberation-mapping analysis, using the 9-m Southern African Large Telescope (SALT) for spectroscopy and the 2-m Faulkes Telescope North and the South for photometry. The monitoring campaign was carried out for 5 month between December 2012 to April 2013, providing 20 spectroscopic epochs and ~40 photometric epochs. Based on the obtained spectra, which typically have a signal-to-noise ratio to 30-60, we performed multicomponent decomposition using various components, i.e., power-law continuum, FeII emission complex, and broad and narrow emission lines, to properly measure the Hbeta line flux. After a flux normalization using [O III] 5007 line luminosity, we obtained a rms spectrum from all epochs, which shows clear variability of Hbeta line. We find that Hbeta line flux decreases by ~20% during the monitoring period while the continuum flux obtained from the aperture photometry based on the imaging data, shows similar variability. The current Hbeta light curve shows monotonic decrease and a reliable cross correlation analysis between Hbeta and continuum light is difficult. Nevertheless, we obtained a preliminary lag measurements as ~24 light days.
