Manganese in Seawaters of the Amundsen Sea, Antarctic (남극 아문젠해에서 해수 중 Mn의 분포 특성)

  • In order to investigate the behavior and seasonal variability of Mn as one of the bio-essential metals in the Amundsen sea, which is known as the most biologically productive coastal area around the Antartica, seawaters were collected using a clean sampling system for 10 stations (96 ea) in 2014 (ANA04B) and for 12 stations (139 ea) in 2016 (ANA06B) surveys of RV ARAON. Dissolved and particulate Mn concentration varied in the range of 0.15-4.43 nmol/kg and <0.01 to 2.42 nM in 2014 and in the range of 0.25-4.15 nmol/kg and 0.01-2.64 nM in 2016, respectively. From the sectional distribution of dissolved and particulate Mn, it might be suggested that dissolved/particulate Mn was provided from iceberg melting and diffusion/resuspension from sediments, respectively. Although this sea is highly productive, there was little evidence regarding the biological origin of dissolved Mn, but particulate Mn only in sea ice and offshore areas could be explained as originating from organic matters, e.g. phytoplanktons. And it could be suggested that the subsurface maximum of dissolved Mn was formed by isopycnal transport of melting materials from ice wall to offshore. Compared to early (2014) summer, temperature, salinity, biomass, dissolved and particulate Mn in late (2016) summer indicated that temporal variations might be resulted from the reduction of ice melting and mCDW flow, which induced a reduction in resuspension. In addition, in the late summer, particles including biomass were reduced, which brought about a reduction in the removal rate of dissolved Mn.

Clinical nurses' experiences of workplace verbal violence: a phenomenological study (병원 내 언어폭력에 노출된 임상 간호사의 경험: 현상학적 연구)

  • Purpose: This study aimed to describe clinical nurses' lived experiences of workplace verbal violence through qualitative research using descriptive phenomenology. Methods: Six female Korean nurses who had less than 5 years of clinical experience and had experienced verbal violence in the workplace within the past year participated in the study. Data were collected through one-on-one in-depth interviews with the participants and analyzed using Colaizzi's phenomenological method. Results: A total of 27 codes, eight themes, and four theme clusters were derived from the participants' statements. The four theme clusters of the clinical nurses' experiences of verbal violence in the workplace were as follows: "tip of the iceberg," "beyond me and my control," "fear and resignation," and "personal burden." The participants recognized that nurses experienced verbal violence daily, and that the causes of and responses to verbal violence were determined by external situational factors rather than nurses' individual problems. This suggests that nurses felt that they had no choice but to personally cope with verbal violence and bear the consequences due to systematic indifference and silence about verbal violence experienced by clinical nurses. Conclusion: The findings show that verbal violence was pervasive and unmerited, yet often endured at the cost of a personal burden to nurses. A clear definition of verbal violence and education for employees are needed, and a reporting system should be established to report all forms of violence regardless of the severity of the incident.

  • Ruminants possess a specialized four-compartment forestomach, consisting of the reticulum, rumen, omasum, and abomasum. The rumen, the primary fermentative chamber, harbours a dynamic ecosystem comprising bacteria, protozoa, fungi, archaea, and bacteriophages. These microorganisms engage in diverse ecological interactions within the rumen microbiome, primarily benefiting the host animal by deriving energy from plant material breakdown. These interactions encompass symbiosis, such as mutualism and commensalism, as well as parasitism, predation, and competition. These ecological interactions are dependent on many factors, including the production of diverse molecules, such as those involved in quorum sensing (QS). QS is a density-dependent signalling mechanism involving the release of autoinducer (AIs) compounds, when cell density increases AIs bind to receptors causing the altered expression of certain genes. These AIs are classified as mainly being N-acyl-homoserine lactones (AHL; commonly used by Gram-negative bacteria) or autoinducer-2 based systems (AI-2; used by Gram-positive and Gram-negative bacteria); although other less common AI systems exist. Most of our understanding of QS at a gene-level comes from pure culture in vitro studies using bacterial pathogens, with much being unknown on a commensal bacterial and ecosystem level, especially in the context of the rumen microbiome. A small number of studies have explored QS in the rumen using 'omic' technologies, revealing a prevalence of AI-2 QS systems among rumen bacteria. Nevertheless, the implications of these signalling systems on gene regulation, rumen ecology, and ruminant characteristics are largely uncharted territory. Metatranscriptome data tracking the colonization of perennial ryegrass by rumen microbes suggest that these chemicals may influence transitions in bacterial diversity during colonization. The likelihood of undiscovered chemicals within the rumen microbial arsenal is high, with the identified chemicals representing only the tip of the iceberg. A comprehensive grasp of rumen microbial chemical signalling is crucial for addressing the challenges of food security and climate targets.

Analysis Study on the use of Frequency and the Cooking Method of Leaf and Stem Vegetables in High School Foodservice (고등학교 급식식단의 엽경채류 식재료 사용 빈도 및 조리방법 분석 연구)

  • This study was conducted to extract the factors affecting the microbial safety of leaf and stem vegetables in the high school foodservice and to provide information for supplying the safe foodservice menu. The lunch and dinner menu (1,945 data) of the total 6 high schools at the Central and the South Region in March, June, September, and December were collected. The frequency analysis and the multiple correspondence analysis (MCA) based on the 3 factors (potentially hazardous food (PHF), leafy and stem vegetables in the menu, the cooking methods) were conducted. The most frequent PHF was the menu of blanched vegetables, salads, seaweeds and fried chicken. The most frequent consumed leaf and stem vegetables were spinach, chive, lettuce, Western cabbage, perilla leaf, iceberg lettuce, chicory, leek and broccoli. MCA based on the leaf and stem vegetables, the region, and the cooking method (cooked/non-cooked) showed that garlic stem and spinach were more used in the Central Region, while water drop-wort were more used in the South Region. Iceberg lettuce, Bok choy and leek were included frequently in the PHF menu. Plant products frequently used in PHF menu requires the food safety system such as Good Agricultural Practice (GAP) to reduce the microbial risk. The menu database according to raw materials based on cooking methods (heating or mixing) as well as the development and verification of menu based on the microbial safety will be contributed to provide the safer foodservice menu.

The Community Structure of Meiofauna in Marian Cove, King George Island, Antarctica (남극 킹조지섬 마리안소만의 중형저서동물 군집구조)

  • The temporal dynamics of the meiofauna community in Marian Cove, King George Island, Antarctica were observed from March 7 to December 21 2007. Nine meiofauna taxa were found, with nematodes the most dominant group, making up 92.97% of the total meiofauna density, followed by harpacticoid copepods (3.18%). Meiofauna abundance ranged from 123 to 874 individuals per 10 $cm^2$ (mean 464 inds.10 $cm^{-2}$), which is lower than that found in some polar and temperate regions. The lowest meiofauna abundance was found in the 26th April sample (III) and the highest meiofauna abundance was found in the March 23rd sample (II). There was no correlation between meiofauna abundance and season. The seasonal changes were likely caused by meltwater runoff, and there were the physical disturbances on the bottom sediment by huge iceberg. Biomass of meiofauna varied between 20.36 and 101.02 ${\mu}gC{\cdot}10\;cm^{-2}$, and overall mean biomass was 54.17 ${\mu}gC{\cdot}10\;cm^{-2}$ during the study periods. More than 80% of meiofauna was concentrated in the upper 2 cm of the sediment, and density decreased with depth. The mean diversity index was 0.37, and the ratio between the abundance of nematodes: and harpacticoids (N/C) ratio ranged from 7.31 to 95.04 (mean 26.39). NMDS analysis divided the community into three groups: A (III, IV, V, VII, VIII), B (II, IX, XI, XII) and C (I, V, X). The results of ANOSIM and SIMPER analysis revealed significant differences in community structure among three groups and major contributed meiofauna taxon in grouping were nematodes and copepods. No significant correlations were observed between major meiofauna taxon and environmental factors. Thirteen species in 12 genera representing nine families of harpacicoids were recorded. Ancorabolidae was the most diverse family, and Heteropsyllidae was the most abundant. The correlation analysis between benthic harpacticoid copepods and environmental factors showed that some species were affected by water temperature, sediment temperature, salinity, chlorophyll a concentration, grain size of the sediments and heavy metal contents of the sediments. These data describe the usefulness of benthic harpacticoid copepods as biological indicator species in Antarctic regions.

Effects of Phytoncide Treatment on the Physicochemical, Microbiological, and Sensory Characteristics of Fresh-cut Lettuce (Phytoncide 처리가 신선편이 양상추의 저장 중 이화학적, 미생물학적 및 관능적 특성에 미치는 영향)

  • The effect of phytoncide solution treatment on the browning and quality of fresh-cut iceberg lettuce during storage was studied. The treatments were applied as four solutions adjusted at $10^{\circ}C$: distilled water (DW) as the control; edible ethanol (EE); 1% (v/v) phytoncide essential oil from pine needle diluted with distilled water (DP); and 1% (v/v) phytoncide essential oil diluted with edible ethanol (EP). Fresh-cut lettuce was dipped in each solution for 60 sec, was rinsed with distilled water, was packaged with an OPP film bag, and was then stored at $4^{\circ}C$ for 12 days. The EP group had a significantly high level of total soluble solids, titratable acidity, and carbon dioxide, and low total bacteria counts, pH, and oxygen. The sensory score of color in the EP group recorded a high value, but the EE and EP groups recorded low scores in aroma and taste during the storage period. Alcohol and phytoncide were vaporized by opening the package for two hours, and the score of the aroma and taste of EP showed no differences from those of the other groups. Based on the above results, it was determined that the phytoncide essential oil diluted in edible alcohol with 1% solution inhibited the browning of and microbial growth in fresh-cut lettuce, and will be a useful natural compound in maintaining the quality of fresh-cut produce.

Establishment of Sample Preparation Method to Enhance Recovery of Food-borne Pathogens from Produce (농산물 중 식중독세균 검출을 위한 전처리법 확립)

  • To establish sample preparation method for detection of food-borne pathogens from lettuce, perilla leaves, cucumber, pepper, and cherry tomato, the influences of diluent composition, processing time, and proportion of diluent to sample were examined. Each produce was inoculated with 6.0 log $CFU/cm^2$ of Escherichia coli O157:H7, Salmonella Typhimurium, Listeria monocytogenes, Staphylococcus aureus, and Bacillus cereus. Each produce was treated with 0.1% peptone water, and D/E neutralizing broth. Processing time of produce was 30, 60, 90, and 120s, and the proportion of diluent to sample was 2 : 1, 4 : 1, 9 : 1, and 19 : 1. The number of bacteria after treatment of D/E neutralizing broth was higher than that of 0.1% peptone water (P<0.05). In cherry tomato, the population of S. typhimurium recovered from treated with D/E broth was higher than that recovered from treated with 0.1% peptone water by 1.05 log $CFU/cm^2$ (P<0.05). No difference in numbers of pathogens was observed in processing time. Optimum proportion of diluent to perilla leaf, iceberg lettuce, cucumber, green pepper, and tomato was 9 : 1, 4 : 1, 2 : 1, 2 : 1, and 2 : 1, respectively. These data suggest that D/E neutralizing broth should be recommend as diluent, and the diluent volume applied to produce should be determined in proportion to produce surface area per weight (g).

Microbial and Pathogenic Contamination of Ready-to-eat Fresh Vegetables in Korea (한국에 유통중인 신선편이 채소류의 미생물 품질 및 병원성 세균의 오염도 조사)

  • The purpose of this study was to evaluate microbiological contamination of fresh vegetables in Korea. Twenty types of vegetables were tested for total aerobic bacteria, coliforms, Escherichia coli, yeast and mold, and pathogenic bacteria such as Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus, Salmonella, E. coli O157:H7, Cronobacter sakazakii, Shigella, and Campylobacter. Levels of total aerobic bacteria and coliform on 20 vegetables were between 3.74 and 8.04 log CFU/g, and 0.16 and 5.02 log CFU/g, respectively. The highest contamination levels of total aerobic bacteria were observed on water dropwort, sprouts, mungbean sprout, and ballflower root. There was no significant difference (p>0.05) in microbial contamination levels of total aerobic count, coliform, E. coli, yeast and mold between organic and nonorganic vegetables. When isolation methods using selective agars were applied, L. monocytogenes, B. cereus, Salmonella and Campylobacter were isolated from some fresh vegetable samples. Results of API kit tests showed that L. monocytogenes was identified on Chinese cabbage, cucumber, soybean sprouts, and iceberg lettuce while Salmonella was identified on Korean leek. Furthermore, Campylobacter jejuni was also identified in more than 50 of the 100 samples. However, when positive samples from API kit were tested for real-time PCR or 16S rRNA sequencing method, only B. cereus from perilla leaf, carrot, water dropwort, and sprouts showed positive results. These results indicate that selective agar and API kit detection methods might result in false positive results for some pathogens. Therefore, studies need to improve isolation or confirmation methods for such pathogens.

Evaluation of Selective Media for Isolation of Staphylococcus aureus from Agricultural Products (농산물 중 Staphylococcus aureus의 분리를 위한 선택배지 평가)

  • Five kinds of selective media, such as mannitol salt agar (MSA), Baird-Parker agar (BPA), Baird-Parker supplemented with rabbit plasma fibrinogen (BPA+RPF), CHROMagar Staphylococcus aureus (CSA), and Petrifilm Staph Express count system (Petrifilm), were compared to recommend the optimum selective media for isolation of Staphylococcus aureus from agricultural products. Seventy four target and non target bacteria were inoculated on five selective media to analyze sensitivity and specificity. In the recovery test of injured S. aureus cells, S. aureus was exposed to acid (1% lactic acid for 10 min), heat ($60^{\circ}C$ for 90s), and cold ($-20^{\circ}C$ for 1h) conditions. And artificially contaminated agricultural products (iceberg lettuce, green pepper, and cherry tomato) was enumerated on five selective media. The sensitivity of BPA+RPF, CSA, Petrifilm, MSA, and BPA were 100%, 100%, 100%, 90.5%, 90.5%, respectively. In addition, the specificity of BPA+RPF, CSA, MSA, BPA and Petrifilm were 100%, 100%, 84.6%, 75.0%, 67.3%, respectively. However, no difference among five selective media was observed in recovery on injured S. aureus cell and enumeration from agricultural products. This results suggest that BPA+RPF and CSA are the optimum media for detection of S. aureus from agricultural products.

A Comparative Study on the Measurement Model of Anti-Corruption Index : Anti-Corruption Index and CCEJ's Corruption Index (부패지수 측정모형에 대한 비교연구 : 서울시 시정청렴성지수 측정모형과 경실련 부패지수 측정모형을 중심으로)

  • Considering the importance of corruption index measurement model and the diffusion of popular awareness about the importance of anti-corruption, this research compared and analyzed existing corruption index measurement models. This paper has selected "Anti-Corruption Index model(ACI model)" that was developed by Seoul Development Institute and "CCEJ's Corruption Index model" which is under use by the Citizens' Coaltion for Economic Justice. These two models were analyzed in terms of appropriateness of 'the indicator and index choice method', 'the research design' and 'the use of statistical data'. This analysis shows that two models have some problems. First, at the level of indicator and index choice method, the indicators of CCEJ's corruption index model is too atomized and redundant. As not putting weight of indicator, it did not consider importance of each indicator. Citizens who evaluated the level of corruption didn't participate in the weight evaluation team, which results in failure of the model to reflect the citizens' opinion. Also, because the question about Seoul Metropolis' corruption level is not included, it is not detected difference between real corruption level and perceptive corruption level. Second, in terms of appropriateness of research design, the sample of the CCEJ' corruption index model is too small to get credibility. The objectivity of research method seems to be low because survey was performed by exit interview. In addition, 38 items are overfull in the questionnire of CCEJ's corruption index model compare to 13 items in ACI model. Also, in making up questions, the terms are redundant and unobvious. Third, in regarding with feasibility of the statistical data, the CCEJ's corruption index model uses regulation data for disciplinary punishment of 25 local governments in Seoul from 1995 to 1999. But, it is common ideas of scholars that statistical data indicates the tip of an iceberg in corruption. So for using a statistical data, it needs a data of enough quantity and has a high level credibility. In addition, objective method of giving weight is not developed. In this point of view, the use of statistical data has some limits. To solve this problem, ① objective data should be included in the indicators, ② various indicators should be developed and ③ a method of giving weight should be improved.
