Nitric Oxide Prevents the Bovine Cerebral Endothelial Cell Death Induced by Serum-Deprivation

  • Endothelial cells play a central role in the inflammatory processes, and activation of nuclear factor kappa B ($NF-_{\kappa}B$) is a key component in that inflammatory processes. Previously, we reported that tumor necrosis factor alpha($TNF{\alpha}$) had protective effect of cell death induced by serum deprivation and this protection was related to $NF-_{\kappa}B$ activation. Inducible nitric oxide synthase (iNOS) is a member of the molecules which transcription is regulated mainly by $NF-_{\kappa}B$. And the role of nitric oxide (NO) generated by iNOS on cell viability is still controversial. To elucidate the mechanism of $TNF{\alpha}$ and $NF-_{\kappa}B$ activation on cell death protection, we investigate the effect of NO on the cell death induced by serum- deprivation in bovine cerebral endothelial cells in this study. Addition of $TNF{\alpha}$, which are inducer of iNOS, prevented serum-deprivation induced cell death. Increased expression of iNOS was confirmed indirectly by nitrite measurement. When selective iNOS inhibitors were treated, the protective effect of $TNF{\alpha}$ on cell death was partially blocked, suggesting that iNOS expression was involved in controlling cell death. Exogenously added NO substrate (L-arginine) and NO donors (sodium nitroprusside and S-nitroso-N-acetylpenicillamine) also inhibited the cell death induced by serum deprivation. These results suggest that NO has protective effect on bovine cerebral endothelial cell death induced by serum-deprivation and that iNOS is one of the possible target molecules by which $NF-_{\kappa}B$ exerts its cytoprotective effect.

Anti-Inflammatory Effects of Ethyl Acetate Fraction from Cnidium officinale Makino on LPS-Stimulated RAW 264.7 and THP-1 Cells

  • This work aimed to elucidate the anti-inflammatory effects of ethyl acetate fraction from Cnidium officinale Makino with a cellular system of LPS-stimulated RAW 264.7 and THP-1 cells. Some key pro-inflammatory cytokines and mediators including NO, iNOS, $PGE_2$, COX-2, TNF-${\alpha}$, NF-${\kappa}B$ p50 and NF-${\kappa}B$ p65 were studied by sandwich ELISA and western blot analysis. Ethyl acetate fraction could significantly inhibit the production of NO, $PGE_2$, TNF-${\alpha}$, iNOS and COX-2 in LPS-stimulated cell than that of single LPS-stimulated. And ethyl acetate fraction suppresses the activation of NF-${\kappa}B$ p50 and NF-${\kappa}B$ p65. All the results showed that ethyl acetate fraction had a good anti-inflammatory effect on LPS-stimulated RAW264.7 and THP-1 cells. Taken together, the anti-inflammatory actions of ethyl acetate fraction from Cnidium officinale Makino might be due to the down-regulation of NO, $PGE_2$, TNF-${\alpha}$, iNOS and COX-2 via the suppression of NF-${\kappa}B$ activation.

토복령(土茯笭)의 Raw 264.7 세포에 대한 항염효과 (Anti-inflammatory Effects of Smilacis Glabrae Rhizoma in Raw 264.7 Cells)

  • Objective : Inflammatory cytokines have a close relationship to insulin dependent diabetes mellitus (IDDM). The inhibitory effect of Smilacis Glabrae Rhizoma (SGR) were examined on production of nitric oxide (NO), prostaglandin $E_2$ $(PGE_2)$, synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and NF-${\kappa}$B activation in Raw 264.7 cells. Methods: Raw 264.7 cells were pretreated with SGR(20, 50, 100 ${\mu}g$/ml), and then cultured with lipopolysaccharides (LPS). Cell viability was measured by MTT assay; inhibition of NO, $PGE_2$, and TNF-${\alpha}$ production were measured by Griess reagent and enzyme-linked immunosorbent assay(ELISA). Induction of COX-2 and iNOS were determined by western blotting analysis. Inhibition of NF-${\kappa}$B was measured by immunofluorescence assay (IFA). Results: SGR inactivated NF-${\kappa}$B, and inhibited the production of NO, iNOS, and $PGE_2$. Inhibition of COX-2 and TNF-${\alpha}$ could not be confirmed. Conclusions: From the above result. SGR was found to have an anti-inflammatory effect of inhibition of NO, iNOS, and $PGE_2$ production via inhibition of NF-${\kappa}$B.

  • In the present study, we investigated the anti-inflammatory effects by kaempferol-3-O-${\beta}$-D-sophoroside (KS) isolated from Sophora japonica (Leguminosae) on the lipopolysaccharide (LPS)-induced nitric oxide (NO) and prostaglandin ($PGE_2$) production by RAW 264.7 cell line compared with kaempferol. KS significantly inhibited the LPS-induced NO and $PGE_2$ production. Consistent with these observations, KS reduced the LPS-induced expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the protein and mRNA levels in a concentration-dependent manner. In addition, the release and the mRNA expression levels of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and interleukin-6 (IL-6) were also reduced by KS. Moreover, KS attenuated the LPS-induced activation of nuclear factor-kappa B ($NF{-\kappa}B$), a transcription factor necessary for pro-inflammatory mediators, iNOS, COX-2, $TNF-{\alpha}$ and IL-6 expression. These results suggest that the down regulation of iNOS, COX-2, $TNF-{\alpha}$, and IL-6 expression by KS are achieved by the downregulation of $NF{-\kappa}B$ activity, and that is also responsible for its anti-inflammatory effects.

Shikonin Isolated from Lithospermum erythrorhizon Downregulates Proinflammatory Mediators in Lipopolysaccharide-Stimulated BV2 Microglial Cells by Suppressing Crosstalk between Reactive Oxygen Species and NF-κB

  • According to the expansion of lifespan, neuronal disorder based on inflammation has been social problem. Therefore, we isolated shikonin from Lithospermum erythrorhizon and evaluated anti-inflammatory effects of shikonin in lipopolysaccharide (LSP)-stimulated BV2 microglial cells. Shikonin dose-dependently inhibits the expression of the proinflammatory mediators, nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-${\kappa}B$ (TNF-${\alpha}$) as well as their main regulatory genes and products such as inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and TNF-${\alpha}$ in LPS-stimulated BV2 microglial cells. Additionally, shikonin suppressed the LPS-induced DNA-binding activity of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) to regulate the key regulatory genes of the proinflammatory mediators, such as iNOS, COX-2, and TNF-${\alpha}$, accompanied with downregulation of reactive oxygen species (ROS) generation. The results indicate that shikonin may downregulate the expression of proinflammatory genes involved in the synthesis of NO, $PGE_2$, and TNF-${\alpha}$ in LPS-treated BV2 microglial cells by suppressing ROS and NF-${\kappa}B$. Taken together, our results revealed that shikonin exerts downregulation of proinflammatory mediators by interference the ROS and NF-${\kappa}B$ signaling pathway.

괴화 추출물 루틴의 소염작용: 쥐의 복강대식세포로부터 NO와 TNF-alpha 생산에 있어서 괴화 루틴에 의한 억제효과 (Anti-inflammatory Function of the Sophora japonica Extract Rutin: The Inhibitory Effect of Rutin of Korean Sophora japonica on the Productions of NO and TNF-alpha from Mouse Peritoneal Macrophages)

  • Korean Sophora japonica has been found to posses an anti-inflammatory activity. In this study, Korean Sophora japonica extract, rutin, was used to know whether rutin inhibits to produce inflammatory mediators NO and TNF-$\alpha$ from the mouse peritoneal macrophages that were treated an inflammatory agent LPS. The rutin-1 hr pretreated macrophages were incubated with LPS for 0.5~5 hrs, and then collected the supernatant and the cell lysate for measurements of the level of iNOS, NO, TNF-$\alpha$ mRNA, TNF-$\alpha$, and p-NF-${\kappa}B$. Minimal and maximal effective doses of the rutin on them were 1 and $100{\mu}g/ml$, respectively. The maximal effective dose of rutin certainly inhibted the productions of iNOS, NO, TNF-$\alpha$ mRNA, TNF-$\alpha$and p-NF-${\kappa}B$ from the LPS-treated macrophages (p<0.0001). Its $ED_{50}$ for inhibition of TNF-$\alpha$ mRNA and p-NF-${\kappa}B$ was $5{\mu}g/m{\ell}$, and for iNOS, NO, and TNF-$\alpha$ was $10{\mu}g/m{\ell}$. The rutin did not have any cytotoxic effect. As the results, the Sophora japonica rutin could be a good candidate for an anti-inflammatory action.

Inhibition of TNF-α-mediated NF-κB Transcriptional Activity in HepG2 Cells by Dammarane-type Saponins from Panax ginseng Leaves

  • Panax ginseng (PG) is a globally utilized medicinal herb. The medicinal effects of PG are primarily attributable to ginsenosides located in the root and leaf. The leaves of PG are known to be rich in various bioactive ginsenosides, and the therapeutic effects of ginseng extract and ginsenosides have been associated with immunomodulatory and anti-inflammatory activities. We examined the effect of PG leaf extract and the isolated ginsenosides, on nuclear factor (NF)-${\kappa}B$transcriptional activity and target gene expression by applying a luciferase assay and reverse transcription polymerase chain reaction in tumor necrosis factor (TNF)-${\alpha}$-treated hepatocarcinoma HepG2 cells. Air-dried PG leaf extract inhibited TNF-${\alpha}$-induced NF-${\kappa}B$transcription activity and NF-${\kappa}B$-dependent cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) gene expression more efficiently than the steamed extract. Of the 10 ginsenosides isolated from PG leaves, Rd and Km most significantly inhibited activity in a dose-dependent manner, with $IC_{50}$ values of $12.05{\pm}0.82$ and $8.84{\pm}0.99\;{\mu}M$, respectively. Furthermore, the ginsenosides Rd and Km inhibited the TNF-${\alpha}$-induced expression levels of the COX-2 and iNOS gene in HepG2 cells. Air-dried leaf extracts and their chemical components, ginsenoside Rd and Km, are involved in the suppression of TNF-${\alpha}$-induced NF-${\kappa}B$ activation and NF-${\kappa}B$-dependent iNOS and COX-2 gene expression. Consequently, air-dried leaf extract from PG, and the purified ginsenosides, have therapeutic potential as anti-inflammatory.

노화유도 쥐의 대동맥에서 L-arginine 투여와 규칙적인 운동의 효과: 염증인자와 혈관이완조절 인자의 변화 (Effects of L-Arginine Supplementation and Regular Exercise in D-Galactose Induced Aging Rat Aorta: Study on Inflammatory Factors, Vasodilation Regulatory Factors)

  • 이 연구는 L-arginine과 규칙적인 운동이 D-galctose (D-all)투여로 유발된 노화흰쥐의 대동맥에서 발현되는 NF-${\kappa}B$, TNF-a, iNOS, Cav-1, eNOS, Ang II의 변화양상을 관찰하는데 그 목적을 두고 있다. 노화유도 모델 쥐는 D-gal (50 mg/kg)를 숫컷 Strague-Dawley (SD)계 흰쥐의 복강에 1일 1회 총 12주간 투여하여 생산하였으며, 이 실험의 집단은 젊은 대조군(Y-con, n=8), 노화 대조군(A-con, n=8), 노화 운동군(A-Ex, n=8), 노화운동+아르기닌군(A-Ex+A, n=8), 노화 아르기닌군(A-A, n=8)의 5군으로 분류하여 실시하였다. L-arginine은 1일 150 mg/kg씩 총 12주간 경구투여 하였다. 운동방법은 트레이드운동으로 1일 60분씩 20 m/min 속도에서 훈련하였다. 분석결과 1) 유도된 노화군에서 NF-${\kappa}B$, TNF-a, iNOS, Cav-1, 그리고 Ang II 단백질의 발현은 젊은 대조군에 비해 유의하게 증가하였다. 그러나 규칙적인 운동과 L-아르기닌군에서 NF-${\kappa}B$, TNF-a, iNOS, Cav-1, 그리고 Ang II 단백질의 발현은 노화 대조군에 비해 유의하게 감소하였다. 2) 유도된 노화군에서의 eNOS 단백질 발현은 젊은 대조군에 비해 유의하게 감소하였다. 그러나 규칙적인 운동과 L-아르기닌군은 eNOS 단백질의 발현을 더욱 증가시킨 것으로 나타났다. 이상의 결과를 종합하면 12주간 L-아르기닌 투여와 규칙적인 운동 그리고 복합처치는 염증인자와 관련된 단백질인 NF-${\kappa}B$, TNF-a, iNOS 단백질들의 발현을 억제시켜 항염증효과를 보여주었으며, 혈관내피의 기능향상과 관련된 eNOS의 발현을 증가시키는데 긍정적인 역할을 수행할 것으로 기대된다.

미세조류 유래 astaxanthin의 항염증 및 항산화 효과 (Anti-Inflammatory and Antioxidant Effect of Astaxanthin Derived from Microalgae)

  • Astaxanthin (ATX)은 다양한 생명체에서 생성되는 카로티노이드 색소이다. 본 연구에서는 ATX가 RAW264.7 cell에서 LPS에 의한 inducible nitric oxide synthase (iNOS), nitric oxide (NO), 염증성 사이토카인, nuclear factor-kappa B (NF-${\kappa}B$)와 reactive oxygen species (ROS)의 생성을 억제 시키는 지 또한, superoxide radical 소거능이 있는 지를 조사하였다. iNOS와 NF-${\kappa}B$는 immunoblot analysis로, interleukin (IL)-6와 tumour necrosis factor-${\alpha}$ (TNF-${\alpha}$)는 ELISA 법으로 분석하였다. NO 양은 nitrite의 양을 측정하였고, ROS는 2',7'-dichlorodihydrofluorescin diacetate (DCFH-DA) 법으로 superoxide radical 소거능은 superoxide radical scavenging activity assay로 검증하였다. 100 ${\mu}M$의 ATX 농도에서 LPS로 유도된 NO, IL-6 및 TNF-${\alpha}$ 같은 염증성 사이토카인의 생성 뿐만 아니라 iNOS 및 NF-${\kappa}B$의 발현도 억제되었다. 특히, IL-6 및 TNF-${\alpha}$ 생성에 있어 ATX의 최대 억제율은 각각 65.2% 및 21.2% 이었으며 LPS로 유도된 NF-${\kappa}B$의 전사활성을 억제하였다. 이러한 현상은 세포질에서 핵으로 NF-${\kappa}B$의 전위를 억제하는 것과 관련이 있다. 또한, 25-100 ${\mu}M$의 ATX 농도에서 세포 내 ROS 생성을 억제하였으며, 5 mg/ml 농도의 ATX는 동일농도의 ${\alpha}$-tocopherol에 비해 superoxide radical 소거능이 1.33배 높았다. 이러한 결과들은 ATX가 대식세포에서 ROS 생성 및 NF-${\kappa}B$ 활성을 저해하므로 iNOS의 발현, NO 및 염증성 사이토카인의 생성을 억제하며, 또한 우수한 superoxide radical 소거능을 보유한다는 것을 나타내었다. 결론적으로, ATX가 항염증제 및 항산화제로서 유용하게 사용될 수 있을 것으로 사료된다.

Anti-inflammatory Effects of the Methanol Extract of Polytrichum Commune via NF-κB Inactivation in RAW 264.7 Macrophage Cells

  • As an attempt to search for bioactive natural products exerting anti-inflammatory activity, we evaluated the effects of the methanol extract of Polytrichum commune Hedw (PCM) (Polytrichaceae) on lipopolysaccharide (LPS)-induced nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$) and pro-inflammatory cytokines release in murine macrophage cell line RAW 264.7. PCM potently inhibits the production of NO, $PGE_2$, tumor necrosis factor (TNF)-$\alpha$ and interleukin (IL)-6. Consistent with these results, PCM also concentration-dependently inhibited LPS-induced inducible NO synthase (iNOS) and cyclooxygase (COX)-2 at the protein levels, and iNOS, COX-2, TNF-$\alpha$ and IL-6 at the mRNA levels without an appreciable cytotoxic effect on RAW 264.7 macrophag cells. Furthermore, PCM inhibited LPS-induced nuclear factor-kappa B (NF-$\kappa$B) activation as determined by NF-$\kappa$B reporter gene assay, and this inhibition was associated with a decrease in the nuclear translocation of p65 and p50 NF-$\kappa$B. Taken together, these results suggest that PCM may play an anti-inflammatory role in LPS-stimulated RAW 264.7 macrophages through the inhibitory regulation of iNOS, COX-2, TNF-$\alpha$ and IL-6 via NF-$\kappa$B inactivation.
