흰쥐에서 대두단백질의 Hypolipidemic 효과에 관한 연구 (A Study on the Hypolipidemic Effects of Soyprotein in Rats)

  • The present study was designed to observe the effects of soyprotein and casein with or without cholesterol on serum and liver lipids in male rats. The 6 experimental groups were as fellows ; SF ; soyprotein, cholesterol-free diet. SC ; soyprotein, 0.5% cholesterol added diet. CF ; casein, cholesterol-free diet CC ; casein,0.5% cholesterol added diet. SCF ; protein mixture of soyprotein and casein(1 : 1), cholesterol-free diet SCC ; mixed protein, 0.5% cholesterol added diet. The hypocholesterolemic and hypotriglyceridemic effects of soyprotein were observed at 3 weeks, but these effects disappeared at 6 weeks. The hypocholesterolemic effect of soyprotein was more obvious when the 0.5% cholesterol was supplemented in the diets. The serum free cholesterol level was not affected by the dietary protein source or the dietary cholesterol, therefore, the difference in serum total cholesterol among groups seems due to the difference in cholesterol esters. There was a tendency of a higher percentage of HDL in soyprotein groups compared to casein groups at 1 week, however, this tendency disappeared with time. The liver cholesterol and triglyceride contents were not differ among cholesterol-free diet groups, however, with addition of cholesterol, those of soyprotein groups were significantly lower than casein groups. The higher serum arginine/lysine ratio of soyprotein groups may offer the part of explanation of its hypocholesterolemic effect.

식이지방 수준이 다른 식이를 섭취한 흰쥐의 체내 지질대사에 대한 난소화성 덱스트린의 섭취 효과 (Effects of Indigestible Dextrin on Lipid Metabolism in Rats Fed Normal or High Fat Diet)

  • The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. These effects are mediated by the increase of facal cholesterol and bile acid excretion. (Korean J Nutrition31(6) : 981-990, 1998)

흰쥐에서 김치식이가 조직과 분변의 지질조성과 Apo단백 및 Thyroxine 농도에 미치는 영향 (Effects of Kimchi on Tissue and Fecal Lipid Composition and Apolipoprotein and Thyroxine Levels in Rats)

  • This study was carried out to examine whether kimchi has hypolipidemic effect and to know how it exert lipid-lowering effect in rats. Male Sprague-Dawley rats were fed with kimchi-fee diet, or 3%, or 5%, or 10% kimchi diets for 6 weeks. Plasma cholesterol level was lowered in rats fed all concentrations of kimchi diets, and plasma triglyceride(TG) level was lowered in 10% kimchi diet group compared with that of control significantly(p<0.05). Th intake of kimchi lowered VLDL-cholesterol and VLDL-TG levels, whereas increased HDL-cholesterol level significantly(p<0.05). LDL-cholesterol level was lowered only in 5% kimchi diet group and LDL-TG level was lowered in all kimchi diet groups compared with those of control significantly (p<0.05). the intake of 5% and 10% kimchi diets also lowered the levels of hepatic cholesterol, TG, total lipid, and apolipoprotein B, whereas increased the levels of fecal total fat, cholesterol, TG, and apolipoprotein A-1 significantly(p<0.05). Triiodothyronine(T$_3$) level was elevated in rats fed kimchi diet, whereas thyroxine(T$_4$) level was not affected by kimchi treatment. These observations support that the intake of kimchin in rats loweres plasma and hepatic lipid levels by increasing the excretion of TG and cholesterol through feces, by the elevation of T$_3$ level, and by the altered lipoprotein metabolism.

Hypocholesterolemic Effect of Amaranth Squalene (Amaranth esculantus) in Rats Fed a High Cholesterol Diet

  • In experiment 1, rats (n=6) fed diet containing 10 g/kg cholesterol for 4 wk (control) with either no amaranth (control), amaranth grain (300 g/kg, AG) or amaranth oil (90 g/kg, AO). Both the AG and AO groups had lower concentration of serum and hepatic cholesterol and triglyceride than the controls (p < 0.05). Fecal excretions of cholesterol and bile acid in AO group increased about 4 fold and 2 fold, respectively, while AG affected only bile acid excretion (p < 0.05). In experiment 2, rats (n=6) were fed the cholesterol diet for 4 wk and injected intraperitoneally with saline (control) or amaranth squalene (AS) for 7d. The hypolipidemic effect of AS was evident in both serum and liver. Fecal excretions of cholesterol and bile acid were greater (p < 0.05) in AS than control. HMG-CoA (3-hydroxy-3-methylglutaryl coenzyme A) reductase activity was reduced in AS group (11.6%, p=0.13). This study suggests that the cholesterol-lowering effect of AS is mediated by greater fecal elimination of steroids through interference with cholesterol absorption.

고지방식을 섭취한 흰쥐의 체내지질함량에 대한 단백질 가수분해물의 섭취 효과 (Effects of Protein Hydrolysates on Blood and Liver Lipids in Rats fed Fat-enriched Diet)

  • The experiments were performed to investigate the effects of protein and protein hydrolysates on lipid metabolism in the hyperlipidemic/hypercholesterolemic rat model induced by feeding fat-enriched diet. In Except 1 male rats were fed four semi-purified high fat and cholesterol diets that contained different nitrogen source, casein(C), casein hydrolysate(CH), corn gluten(G) and corn gluten hydrolysate(GH), for 6 weeks. In Expt. 2 rats were fed high fat and cholesterol diet for 4 weeks to induce hyperlipidemia and hypercholesterolemia. Then the rats were divided into 4 groups and were fed the four kinds of above experimental diets for 4 weeks consecutively. The contents of total lipid , cholesterol and triglyceride in blood, liver and feces were determined. Serum lipid concentrations of CH, G and GH were significantly lower than that of C. Serum cholesterol concentrations of hydrolysate groups(CH and GH) were significantly lower than those of intact protein groups(C and G). Serum HDL -cholesterol concentration tended to increase by hydrolysate intake. The total lipid, cholesterol contents in liver showed similarity results as above. Fecal lipid excretions of CH, G, and GH groups were significantly higher than that of C group. These results indicate that hypolipidemic and /or hypocholesterolemic effect of corn gluten or protein hydrolysates were detected in the process of inducing hyperlipidemia by high-fat and cholesterol diet or after inducing hyperlipidemia.

식이지방 수준에 따라 n6 와 n3 계 불포화지방산이 혈장 지질수준에 미치는 영향에 관한 비교연구 (Differential Effect of n6 and n3 Polyunsaturated Fatty Acids on Plasma Lipids in Rats Fed Low and High Fat Diets)

  • 식이지방의 수준에 따라 n6 와 n3 불포화지방산이 혈장 지질조성에 미치는 영향과 또 그 기전이 다른지를 연구하고자 Sprague Dawley 종 수컷쥐에게 저지방(LF, 10% Cal)식이와 고지방식이(HF, 40% Cal)를 각각 6주동안 투여하였으며, 사용된 기름은 포화지방산 급원으로는 쇠기름, n6 linoleic acid(LA) 급원으로 corn oil, n3 ${\alpha}-linolenic$ acid(LL) 급원으로 perilla oil, n3 eicosapentaenoic acid(EPA)와 docosahexaenoic acid(DHA) 급원으로 fish oil이었다. Ultracentrifugation 방법으로 VLDL fraction을 분리하여 thin layer chromatography에 의해서 화학적조성을 구하였다. Plasma cholestere 수준은 LF 와 HF 모두 n6 LA에 의해서는 오히려 증가되었고 n3 LL과 n3 EPA에 의해서는 감소되었으며, HF 에서 n3 EPA가 가장 cholesterol 저하효과가 있었다. HDL-Chol 농도는 n6 LA에 의해서 증가되었으나, HF 경우 n3 EPA에 의해서 유의성있게 감소되었다. Plasma TG 농도는 n3 EPA에 의해서 가장 감소되었고 간의 lipogenic enzyme 활성을 억제하였으며 VLDL fraction의 TG상대적양(%)이 유의성 있게 낮아졌다. 이때 LF 군에서는VLDL fraction의 apo-B의 상대적양(%)이 감소되지 않았으나 HF군에서 n3 EPA에 의해서 유의성 있게 낮았다. 그러므로 n3 EPA의 hvpotriglyceridemic effect는 간에서 lipogenesis를 억제하여 plasma VLDL로 TG 분비가 억제되었을 것이며, HF일 경우는 간에서 apo-B 생성도 억제되었다. 관상동맥성심장질환의 예방적 차원에서 평상시에도 쇠기름과 corn oil 보다는 n3 PUFA가 풍부한 들기름이나 생선을 더욱 이용하는 것이 바람직하다.

사람에서 식이지방의 불포화지방산과 불포화도가 혈장 지질조성에 미치는 영향 (Influence of Different Dietary Fats and Fat Unsaturation on Plasma Lipid Composition in Healthy Young Women)

  • Twenty college women were led experimental diet which composed ot basal diet plus different kinds of dietary rats at 27% Cal. Equal amount of 13.5g of corn oil, perilla oil or fish oil was supplied for 2 weeks as a source of n6 linoleic acid(LA). n3 $\alpha$-linolenic acid (LL). or n3 EPA + DHA. respectively. Plasma total Chol level was reduced by perilla and fish oils, significantly only by fish oil. Plasma Chol level was rather increased by corn oil(P<0.05), but was decreased by double amount of corn oil supplement. Therefore, hypocholesterolemic effect of fatty acids was in the order of n3 EPA+DHA>n3 LL>n6 LA and influenced by the degree of fat unsaturation. Plasma TG level was also significantly decreased by n3 EPA+ DHA and increased by n6 LA. Hypotriglyceridemic effect of fatty acids was also in the order of n3 EPA + DHA> n3 LL>n6 LA and influenced by the unsaturation. However, the reduction of plasma TG was more influenced by the fatty acid structure rather than the fat unsaturation. There were no significant effects on lipoprotein pattern 3nd chemical compositions of lipoprotein by different dietary PUFAs. but fish oil diet significantly increased the relative proportion of HDL-Chol. In conclusion. cholesterol- lowering effect of dietary PUFAS seemed to be a function of total fat unsaturation but hypotriglyceridemic effect seemed to be more linked to the ratty acid structure rather than the degree of unsaturation. The hypolipidemic effect of n3 PUFAs was significant so that fish oil or perilla oil may have important nutritional applications in the prevention and treatment of atherosclerotic disease.

흰쥐의 혈청 및 간지질 저하에 미치는 쑥 에탄올 추출물의 영향 (Effects of Artemisia iwayomogi Kitamura Ethanol Extract on Lowering Serum and Liver Lipids in Rats)

  • Artemisia iwayomogi(A. iwayomogi) is a major edible vegetable in Korea. A iwayomogi containing a strong bitter taste, particularly, is known to manifest an effect on cough, abdominal pain, indigestion and bleeding. In this experiment, ethanol extract of A. iwayomogi(50mg/kg body weight) was fed to growing male Sprague-Dawley rats for four weeks in order to examine the lipid lowering effect of A. iwayomogi in the live and serum. High fat diet included 10% lard, 1% cholesterol and 0.25% sodium cholate. The administration of A. iwayomogi extract decreased serum cholesterol compared with the control. At the same time, serum triglyceride(TG) was decreased to 25%. Although atherogenic index and HTR tended to decrease, phospholipid level in the serum was shown to increase by A. iwayomogi administration. At the same token, total liver lipids were decreased to 40%. The group fed both high fat and ethanol extract showed 28% decrease of liver lipids compared with the group fed only high fat diet. On the other hand, HDL cholesteol level of high fat and extract fed group was increased to 30% compared with the non-extract-fed control. Liver TG also was decreased to about 21% in the extract fed groups. Liver phospholipids, particularly, exceeded more than double of the level of high fat control. The above results indicate than A. iwayomogi exert a strong hypocholesterolemic and hypolipidemic effect which may lead to the protection from hypertention, obesity, stroke and many other circulatory diseases.

인삼부산물 추출액의 ginsenosides 함량 및 고지방 식이에 있어 혈청 콜레스테롤 농도 개선에 미치는 효과 (Ginsenoside Contents and Hypocholesterolemic Effects of a By-Product in Ginseng Radix)

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from mixture of leaf and stem of ginseng radix (Panax Ginseng C.A. Meyer). We conducted analysis of the ginsenoside content by HPLC. Also we investigate the effects of the LSE on the reduction of serum lipid and improvement of blood parameters in rats fed high fat diet 5 weeks. We examined by analyzing the serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride and atherogenic index and hematological datas and serum metabolic variables. Sprague-Dawley rat weigh $150\;g\;{\pm}\;15\;g$, were ramdomly assigned to 4 groups, basal diet only(BDG), high fat diet weithout LSE(FDCG), high fat diet and 10% LSE(FD10G), high fat diet and 20% LSE(FD20G). The result of this study were as follow. Hematological datas of 4 groups were same level, which were not significant. The activities of ALP, GOT and LDH level were significantly different. Total cholesterol, LDL-cholesterol, triglyceride contentrations in serum and atherogenic index were remarkably reduced in LSE supplemented groups as compared high fat control groups. These result imply that LSE could be used as possible for decrease of serum lipid concentration.

김(Porphyra yezoensis)에서 분리한 porphyran이 고지혈증 및 고콜레스테롤혈증을 유발한 흰쥐의 지질대사에 미치는 영향 (Effect of Porphyran Isolated from Laver, Porphyra yezoensis, on Lipid Metabolism in Hyperlipidemic and Hypercholesterolemic Rats)

  • 김(Porphyra yezoensis)의 생리활성 물질인 porphyran을 분리 추출한 후, Sprague-Dawley계 흰쥐에 4주간 급여한 후의 혈액과 간의 지질농도와 간의 조직학적 변화를 관찰한 결과는 다음과 같다. 혈청의 중성지질, 총 콜레스테롤 및 LDL-콜레스테롤은 정상군에 비해 대조군에서 중성지질, 총콜레스테롤 및 LDL-콜레스테롤함량이 높게 나타났다. porphyran 1%, 5% 및 10% 급이에서 첨가량이 증가할수록 혈청중의 중성지질, 총콜레스테롤, LDL-콜레스테롤함량은 유의적으로 감소한 반면, HDL-콜레스테롤함량은 증가하였다. 간 조직의 현미경학적인 관찰 결과 정상군에 비해 대조군에서 수많은 지방입자가 세포내에 침착되어 있었으며, porphyran 1% 급이군에서도 유사한 현상을 나타내었다. 그러나 porphyran 5% 및 10% 급이군에서는 점차 지방 입자가 감소되었음을 확인할 수 있었으며, 특히 10% porphyran 급이군에서는 정상군과 유사한 경향을 나타내 porphyran의 급여수준이 높을수록 간의 지방침착이 감소됨을 알 수 있었다. 이러한 결과로 볼 때 고지혈증을 유발시킨 흰쥐의 식이에 김의 추출물인 porphyran의 첨가는 지방개선의 효과가 있음을 알 수 있었다.
