Effects of Gastrodia rhizoma on Lipid Components of Serum in Hypercholesterolemic Rats (천마분말이 고콜레스테롤혈증 흰쥐 혈청의 지질성분에 미치는 영향)

  • This study was designed to investigate the optimum recommendation level on Gatrodia rhizoma and the effects on the improvement of the lipids in the dietary hypercholesterterolemic rats. Experimental diets mixed with 5% Dioscorea batatas(Group 2), 10% Dioscorea batatas(Group 3), 15% Dioscorea batatas (Group 4) 5% Gastrodia rhizoma (Group 5), 10% Gastrodia rhizoma (Group 6), and 15% Gastrodia rhizoma (Group 7), were administered to the male rats of the Sprague Dawley for 3 weeks. Concentration of total cholesterol in serum was lower in the Gastrodia rhizoma groups than in the other groups, especially total cholesterol concentration of 10% Gastrodia rhizoma(Group 6) was the lowest in the Gastrodia rhizoma groups. Concentration of HDL-cholesterol in serum was higher in the 10% Gastrodia rhizoma and 15% Gastrodia rhizoma than in the other groups. Concentrations of cholesteryl ester, LDL, LDL-cholesterol in serum were the lowest in the 10% Gastrodia rhizoma. Concentration of glucose and activity of GPT in serum were the lowest in the 10% Gastrodia rhizoma group. The activity of GOT in serum was lower in the 10% Gastrodia rhizoma group and 15% Gastrodia rhizoma group than in the other groups. Therefore, we consider that there are effects on the improvement of the lipids in the Gastrodia rhizoma and the optimum recommendation quantity of Gastrodia rhizoma is 10% to quantity of food composition.

Effects of Dietary Calcium Levels on Lipid Metabolism in Rats Fed High Fat Diet with or without Supplemental Cholesterol (식이칼슘 수준이 콜레스테롤 첨가 또는 무첨가 고지방식 섭취 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of dietary calcium on blood and tissue lipids of adult rats fed a high fat diet with or without supplemental cholesterol for 4 weeks. Male Sprague-Dawley rats were fed the experimental diets containing 18%(w/w) beef tallow and three levels of calcium 0.1%, 0.5% and 1.5%. The contents of total lipid, cholesterol, triglyceride and phospholipid in blood, liver, small intestine, aorta, small intestinal contents and feces were determined. Only in rats fed the diets containing 1% cholesterol the concentration of cholesterol in aortic serum and various tissues significantly increased, and then decreased with increasing dietary calcium intake. Another observation was that high Ca intake significantly facilitated the fecal lipid and cholesterol excretion and bowel movement. These results suggest that possible hypocholesterolemic effects of dietary calcium could be related to the hypercholesterolemia and to the increase in excretion of fecal lipid and cholesterol.

Hypocholesterolemic Effects of Green Tea in Cholesterol-Fed Rats (고 콜레스테롤 식이 투여 흰쥐에 있어서 녹차의 콜레스테롤 저하 효과)

  • Green tea, which is high in polyphenols, is thought to have hypocholesterolemic effects. The present study was performed to further elucidate the hypocholesterolemic actions of green tea, specially the catechin and (-)-epigallocatechin gallate (EGCG) for their effects on the diet-induced hypercholesterolemia in rats. Male Sprague-Dawley rats were fed with green tea-free diet (control), diets containing 4% green tea powder (GTP), 1.0% green tea catechin (catechin) or 0.5% epigallocatechin gallate (EGCG) for 7 wks. All diets that were provided green tea contained approximately 0.5% EGCG Hypercholesterolemia was induced by adding 1% cholesterol and 0.5% cholic acid to all diets. There were no differences in food intake among groups. The green tea treatments showed significant improvement in the serum levels of total cholesterol, LDL-cholesterol, triacylglycerides and atherogenic index in the following order; EGCG>Catechin>GTP (p<0.05). The serum HDL-cholesterol level was highest in the EGCG-treated group. The catechin or EGCG diet up-regulated by 5 times the enzyme activity of hepatic cholesterol 7$\alpha$ -hydroxylase (CYP7Al) compared to control diet (p<0.05). Hepatic CYP7Al mRNA level paralleled tile increases in the CYP7Al activity. These results suggest that the EGCG in the green tea may account for the hypocholesterolemic effect by the induction of CYP7Al gene expression.

Anti-atherosclerotic Effect of the Methanol Extract of Sorbus commixta Cortex in the High Cholesterol-Diet Rats

  • Hypercholesterolemia is a pivotal pathogenic factor for the development and maintenance of atherosclerosis. The present study was designed to evaluate whether the methanol extract of Sorbus commixta cortex (MSC) restores vascular dysfunction in association with the aortic expressions of proinflarnmatory and adhesion molecules in high cholesterol (HC) diet-rats. Chronic treatment with low (100 mg/kg/day) or high doses (200 mg/kg/day) of MSC lowered the increase in plasma levels of triglyceride (TG) and low-density lipoprotein (LDL) cholesterol induced by a cholesterol-enriched diet without affecting on the plasma level of high density lipoprotein (HDL)-cholesterol. Vascular tone attenuated in the HC-diet rats was restored by administration with MSC. Treatment with MSC also suppressed the HC-induced increase in the monocyte chemoattractant protein-1 (MCP-1) and nuclear factor-$_K$B (NF-$_K$B) p65 expressions as well as expressions levels of adhesion molecules including intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (ICAM-1), and E-selectin in aorta. The present study also showed that MSC inhibited the HC-mediated induction of ET-1 and ACE expression. In histopathological examination, aortic segments in the HC-diet rat revealed thickening intima and media, which were blocked by administration with MSC. Taken together, MSC could suppress the development of atherosclerosis in the HC-diet rat model through the inhibition of the aortic expression levels of pro-inflammatory and adhesion molecules.

Effect of Dietary Fat Sources and L-arginine Supplementation on Endothelial function and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats (식이지방의 종류와 L-arginine 보충이 당뇨쥐의 ENdothelium 기능 및 체내 지질대사에 미치는 영향)

  • Alteration in the syntesis or enhanced inactivation of nitric oxide(NO) can induce impairment of endothelial cell function. Insulin dependent diabetes mellitus(IDDM) is characterized by impaired endothelial function and vascular disease. NO is produced through L-arginine pathway To elucidate the hypothesis that the decreased production on NO in IDDM reflects vascular damage and the NO production can be manipulated by either dietary fat(7% of kg diet) or the oral supplementation with L-arginine(2g/kg bw), plasma markers for vascular endothelial damage and plasma lipid profiles were measured in streptozotocin(STZ)-induced diabetic rats. Diabetic or normal Sprague-Dawley rats were fed 6 different experimental diets for 4 weeks(SO : soybean oil, SOA: soybean oil + L-arginine supplementation, BT : beef tallow, BTA_ beef tallow + L-arginine supplementation, OV olive oil, OVA : olive oil + L-arginine supplementation). Plasma glucose, total cholesterel, HDL-cholesterol, LDL-cholesterol and triglyceride were measured. Endothelial markers, plasma von Willebrand factor(vWf), thromboxane B$_2$, and 6-keto PGF1$\alpha$ of aorta were measured by ELISA. Plasma NO production was evaluated through the measurement of nitrite by EIA. Feeding saturated fatty acid(SFA, BT) increased relative liver size(RLS) in diabetic rats compared to either polyunsatunted fatty acid(PUFA, SO) or monounsaturated fatty acid(MUFA, OV) The supplementation of L-arginine inhibited the liver and kidney enlargement in olive oil find diabetic rats. Plasma glucose was lower in diabetic animal find the olive oil compared to fed beef tallow and the supplementation L-arginine decreased it in diabetic rats find beef tallow significantly(p < 0.05). Plasma TXB$_2$ levels were increased due to diabetes and the value of beef tallow group showed highest value. Plasma vWf concentration of beef tallow group was higher value in normal rats and was elevated more in diabetes. In diabetic groups, the vWf concentration of olive oil group was lower than beef tallow or soybean oil group. The supplementation of L-arginine in diabetic rats decreased plasma TXB$_2$ and vWf levels significantly(p < 0.05). NO production was higher in normal olive oil fed rats and was tend to be decreased in diabetic rats and the supplementation of L-arginine recovered to normal value(p < 0.05), Olive oil supplemented with L-arginine tended to lower plasma total cholesterol and LDL-cholesterol after 4 week treatment. These results suggest that generalized vascular endothelial changes based on plasma TXB$_2$and vWf occurs in diabetic rats. and olive oil with L-arginine supplementation contributes to a better control of the hyperglycemia, endothelial changes and hypercholesterolemia accompanying diabetes as compared with beef tallow or soy bean oil in this rat model.

Fruiting Body Extracts of Paecilomyces tenuipes Ameliorate Lipid and Antioxidant Metabolism in Rats Fed a High Fat-Cholesterol Diet

  • The present study examined the lipid lowering and antioxidant activity of Paecilomyces tenuipes. Eight week-old male Sprague-Dawley rats were fed one of the three diets, a reference diet without cholesterol addition (NC), a high fat (17 g/ 100 g)-high cholesterol (1 g/100 g) diet (HC) and a HC diet supplemented with 3% P. tenuipes (PT) for 30 days. Total lipid and total cholesterol were reduced significantly by 33 and 37%, respectively, in the PT diet group compared with controls. A similar reduction was found for low density lipoprotein (LDL) cholesterol levels, while plasma high density lipoprotein (HDL) cholesterol and triglyceride (TG) concentrations were not significantly different among groups. Hepatic total lipid and total cholesterol levels, but not hepatic TG levels, were significantly decreased in the PT group compared to the HC group. The administration of P. tenuipes increased the plasma total antioxidant potential and decreased the levels of lipid peroxidation. These results suggest that P. tenuipes exerts significant health benefits through the modulation of physiological functions including a variety of atherogenic lipid profiles and antioxidants in hypercholesterolemia.

Antihyperlipidemic Activity of Several Traditional Herbal Medicines on Experimental Hyperlipidemia in Rats (고지혈증 랫트를 이용한 수종 전통 한약제의 항고지혈 효과)

  • Three herbal medicine recipes were tried on the animal model of diet induced hypercholesterolemia to screen for the lipid-lowering effect. The recipes adopted were based on the prescription for atherosclerosis-related symptoms by folk-medicine practioners. Hyperlipidemia was induced in rats by giving the high lipid/cholesterol diet for 7 days. Then, the recipes started with the normal diet. Blood was sampled at 1,2,3 and 5 week-points of the treatment, and levels of total cholesterol, triglyceride, high density lipoprotein and lower density lipoprotein were measured. Dae-shiho-tang decreased total cholesterol level significantly. Hwangryun-haedok-tang and Samhwang-sasim-tang slightly decreased total cholesterol level, although it was not statistically significant.

Effect of St. John's Wort (Hypericum perforatum) on obesity, lipid metabolism and uterine epithelial proliferation in ovariectomized rats

  • BACKGROUND/OBJECTIVES: This study was conducted to assess the potential of St. John's Wort (Hypericum perforatum) to prevent obesity and abnormalities in lipid metabolism induced by ovariectomy in a rat model without stimulatory activity on uterus. MATERIALS/METHODS: Ovariectomized (OVX) rats were treated for 6 weeks with 70% ethanol extracts of Hypericum perforatum [HPEs: whole plant (WHPE) and flower and leaves (FLHPE)], ${\beta}$-estradiol-3-benzoate at a dose of $50{\mu}g/kg/day$ (E2) or vehicle (distilled water). RESULTS: As expected, OVX increased body weight gain and adiposity and showed higher food efficacy ratio. OVX also increased the serum cholesterol as well as insulin resistance, while reducing uterus weight and uterine epithelial proliferation rate. HPEs (WHPE and FLHPE) showed estrogen-like effect on body weight gain, adipose tissue weight and food efficacy ratio in OVX rats. HPEs prevented hypercholesterolemia induced by OVX more effectively than E2. E2 increased uterus weight and epithelial proliferation rate in OVX rats, while HPEs maintained them at the level of the sham-operated animals. CONCLUSIONS: Our finding demonstrates that HPEs can be considered as an effective agent to prevent OVX-induced obesity without stimulatory activity on uterus.

Red Wine Prevents Brain Oxidative Stress and Nephropathy in Streptozotocin-induced Diabetic Rats

  • We have studied the effects of red wine on brain oxidative stress and nephropathy in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Wistar rats with a single intraperitonally injection of STZ (50 mg/kg). Two weeks before and four weeks after injection, red wine was given orally in both normal and diabetic rats. Blood samples were taken from the neck vascular trunk in order to determine the glucose, triglycerides, total cholesterol, HDL-cholesterol (HDL-c), atherogenic index (AI), total protein, blood urea nitrogen (BUN), creatinine, insulin, lipid peroxidation products, reduced glutathione (GSH) and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. As well, we estimated the lipid peroxidtion, GSH and SOD, GSH-Px and catalase activities in brain and renal homogenates, and the excretion of albumin, proteins and glucose in urine over 24 h period. The administration of STZ caused significant increases in levels of glycosuria, proteinuria, albuminuria, glycemia, total cholesterol and AI, as well as in lipid peroxidation products in the brain, plasma and kidney, whereas it decreased the GSH content and SOD, GSH-Px and catalase activities. Treatment with red wine significantly prevented the changes induced by STZ. These data suggested that red wine has a protective effect against brain oxidative stress, diabetic nephropathy and diabetes induced by STZ, as well as it protects against hypercholesterolemia and atherogenic risk.

Effect of Dietary Inclusion of Lactobacillus acidophilus ATCC 43121 on Cholesterol Metabolism in Rats

  • This study examined the effects of Lactobacillus acidophilus ATCC 43121 (LAB) on cholesterol metabolism in hypercholesterolemia-induced rats. Four treatment groups of rats (n=9) were fed experimental diets: normal diet, normal $diet+LAB(2{\times}10^6\;CFU/day)$, hypercholesterol diet (0.5% cholesterol, w/w), and hypercholesterol diet+LAB. Body weight, feed intake, and feed efficiency did not differ among the four groups. Supplementation with LAB reduced total serum cholesterol (25%) and VLDL+IDL+LDL cholesterol (42%) in hypercholesterol diet groups, although hepatic tissue cholesterol and lipid contents were not changed. In the normal diet group, cholesterol synthesis (HMG-CoA reductase expression), absorption (LDL receptor expression), and excretion via bile acids (cholesterol $7{\alpha}-hydroxylase$ expression) were increased by supplementation with LAB, and increased cholesterol absorption and decreased excretion were found in the hypercholesterol diet group. Total fecal acid sterols excretion was increased by supplementation with LAB. With proportional changes in both normal and hypercholesterol diet groups, primary bile acids (cholic and chenodeoxycholic acids) were reduced, and secondary bile acids (deoxycholic and lithocholic acids) were increased. Fecal neutral sterol excretion was not changed by LAB. In this experiment, the increase in insoluble bile acid (lithocholic acid) reduced blood cholesterol level in rats fed hypercholesterol diets supplemented with LAB. Thus, in the rat, L. acidophilus ATCC 43121 is more likely to affect deconjugation and dehydroxylation during cholesterol metabolism than the assimilation of cholesterol into cell membranes.
