까마중내 (Solanum nigrum L.) 항산화방어계의 항산화력 및 물질의 동정 (Evaluation of the Antioxidant Potential and ldentification of Active Principles of Solanum nigrum L. on Antioxidant Defense Systems)

  • 까마중의 부위별 DPPH free radical 소거활성은 뿌리에서 가장 높았고, 줄기, 전부위, 열매 잎 그리고 꽃 순으로 높았다. 추출방법간의 DPPH free radical 소거활성은 80%methanol에 침지하여 추출한것보다는 $80^{\circ}C$에서 80% meth-anol에 흰류추출한것이 높았다. Sephades LH-20 column chromatography에 의하여 분획된 L6의 DPPH free radical 소거활성은 ethy acetate 분획 추출물보다 6.7배 높았다. 분획물내의주요물질은 2, 6-methano-3-benzazocin-11-ol, 2[1h]-pyridinethione 및 2-hydroxy-5 methylbenzaldehyde로 phenolic 화 물이었다. 줄기와 뿌리의 POD및 SOD 활성은 타 부위에 비하여 높은 활성을 나타내었고, 생육기간에는 파종 후 60일후에서 활성이 가장 높았다. PODdmlehddnl gth 패턴은 뿌리에서 10개의 band로 가장 많았고 그중 8개의 band가 식물체의 부위별로 차이가 있어Tdmaul, SOD의 동위효소 패턴은 잎에 5개의 band로 가장 많았고 그중 2개의 band가 식물체의 부위별로 차이가 있었다.이가 있었다.

쇠비름(Portulaca oleracea) 추출물의 DPPH radical 소거능과 in vitro 지질과산화 억제 효과와 활성성분 (DPPH Radical Scavenging Effect and in vitro Lipid Peroxidation Inhibition Activity of Portulaca oleracea and Its Active Principles)

  • An antioxidative activity of Portulaca oleracea was tested by in vitro experimental models. The antioxidative activities were determined by evaluation the DPPH radical scavenging activity and by measuring lipid peroxide using 2-thiobarbituric acid (TBA). The crude extract was sequentially partitioned with n-hexane, 15% aq. MeOH, EtOAc, n-BuOH, $H_2O$. Among them, a remarkable antioxidative effect was observed in the fractions of EtOAc and n-BuOH. The DPPH radical scavenging effect $(IC_{50}=17.90{\mu}g/ml)$ of the n-BuOH soluble fraction was comparable with that of natural antioxidant, ${\alpha}-tocopherol(IC_{50}=\;6.99{\mu}g/ml)$ and the inhibitory effect of lipid peroxidation in mouse liver homogenate was similar to that of natural antioxidant, L-ascorbic acid at a concentration of 0.1mg/ml to 5mg/ml. From the BuOH soluble fraction yielded two biophenolic glycosides, 3-hydroxy-1-(2-hydroxyethyl)phenyl-4-O-${\beta}$-D-glucopyranoside(1) and 2-(3,4-dihydroxyphenyl)ethyl-O-${\beta}$-D-glucopyranoside(2) using column chromatography and revered-phase HPLC. In particular, the DPPH radical scavenging activity of 2 was comparable to that tocopherol$(IC_{50}=6.59{\mu}g/ml)$.

녹차 분말과 카테킨의 농도에 따른 항산화력 비교 분석 (Analysis on the Antioxidant Activity of Catechin Concentrations and Green Tea Extract Powder)

  • Antioxidative activities of catechin from green tea extracts were examined by the methods of 1,1-Diphenyl-2-picrylhydrazyl (DPPH) electron donating ability, hydroxy scavenging activity and the inhibitory effect on xanthine oxidase activity. The resulted demonstrated the fact that Catechin one (containing Green tea extracts plus catechin 51%) of green tea extracts product showed at 65.5% in electron donating activity on the DPPH. The electron donating ability on the DPPH of Catechin one was increased to 10% than purified catechin. Catechin one showed the activity at 64.5% in scavenging activity using hydroxy radical method. To the Catechin one provided to increase the hydroxy scavenging activity up to 3 fold. Inhibitory effects of the catechin one measured with xanthine oxidase method was 6.5%. Although the antioxidative activity of catechin (98% purified) was lower than that of Catechin one (containing Green tea extracts plus catechin 51%) in same catechin concentrations ($5{\mu}g$, respectively). Therefore, we may suggest that Catechin one can be used as a functional food additive possessing the potent antioxidative activity.

  • Hydroxybenzothiophenes, dihydroxy-benzothiophenes, and benzothiophenedione were identified as inter-mediates of benzothiophene (BT) exposed to ultrasonic irradiation. It is proposed that benzothiophene is oxidized by OH radical to sequentially for m hydroxybenzothiophenes, dihydroxybenzothiophenes, and benzothiophenedione. Benzothiophene is decomposed rapidly following pseudo-first-order kinetics in a first-order manner by ultrasonic irradiation in aqueous solution. The toxicity of sonochemically treated solutions was checked by E. coli and a less inhibition in bacterial respiration was observed from the 120-min treated benzothiophene sample than from the untreated benzothiophene sample. Also evolution of carbon dioxide and sulfite was observed during ultrasonic reaction. A pathway for ultrasonic decomposition of benzothiophene in aqueous solution is proposed.

Antioxidant compounds from the stem bark of Sorbus commixta

  • The MeOH extract of Sorbus commixta (Rosaceae) exhibited strong DPPH radical scavenging activity, and through activity-guided fractionation two antioxidant compounds were isolated and identified as $catechin-7-O-{\beta}-D-xylopyranoside$ (1) and $catechin-7-O-\;{\beta}-D-apiofuranoside$ (2) by physicochemical and spectrometric methods. To evaluate the antioxidant effect of these compounds, some in vitro tests, such as the DPPH radical scavenging activity test, the superoxide radical scavenging activity test and the lipid peroxidation inhibitory activity test, were performed. Compounds 1 and 2 showed stronger activities than both a-tocopherol and butylated hydroxy anisole (BHA) in each assay.

복분자에 함유된 항산화물질의 동정 및 활성 (Identification and Activity of Antioxidative Compounds from Rubus coreanum Fruit)

  • 복분자를 착즙하여 착즙액과 잔사로 나누고, 각 추출물을 용매분획하여 얻어진 각 획분을 대상으로 DPPH radical-scavenging 활성을 측정하였더니, 각 추출물의 EtOAc 가용산성획분에서 비교적 강한 활성이 나타났다. 이들 획분에 함유된 활성물질을 규명하고자 silica gel column chromatography로 정제한 다음 활성획분을 GC-MS 분석을 실시한 결과, 복분자 착즙액의 EtOAc 가용 산성획분에 함유된 물질로 4-hydroxybenzoic acid, 4-hydroxy-3-methoxybenzoic acid, 3,4-dihydroxybenzoic acid, 3,4,5-trihydroxybenzoic acid, and 3,4-dihydroxycinnamic acid 등이 동정되었고, 잔사 MeOH 추출물의 EtOAc 가용 산성획분에 함유된 물질로 succinic acid, 3,4-dihydroxybenzoic acid, citric acid, 3,4,5-trihydroxybenzoic acid, 3,4-dihydroxycinnamic acid가 동정되었다. DPPH radical-scavenging 활성과 hydroxyl radical에 의한 malonaldehyde 생성억제 효과활성 등의 방법으로 동정된 물질의 항산화 활성이 평가되었다.

Synthesis, Antioxidant Activity and Fluorescence Properties of Novel Europium Complexes with (E)-2- or 4-hydroxy-N'-[(2-hydroxynaphthalen-1-yl)methylene]benzohydrazide Schiff Base

  • Two novel Eu(III) complexes with notable properties have been successfully prepared with hydrazone Schiff base ligands, (E)-2-hydroxy-N'-[(2-hydroxynaphthalen-1-yl)methylene]benzohydrazide (3a) and (E)-4-hydroxy-N'-[(2-hydroxynaphthalen-1-yl)methylene]benzohydrazide (3b). DFT, FMO energy and Mulliken charge distribution studies of the ligands allowed us to hypothesize that their HC=N, > C=O and -OH (naphthyl) groups were involved in coordinating with the $Eu^{3+}$ ion. The eight coordination sites of the $Eu^{3+}$ ion were occupied by the three functional groups of the two ligands (3a or 3b) mentioned above and two water molecules. Similar UV, IR and fluorescence spectra indicated the presence of comparable coordination environments for the $Eu^{3+}$ ion in both complexes. Both the ligands and their complexes exhibited moderate DPPH radical scavenging activity. Moreover, it was found that the Eu(III) complexes exhibited fluorescence properties.

뇌조직의 활성산소 및 그 제거효소에 미치는 뽕잎 추출물의 영향 (Effects of Mulberry (Morus alba L.) Leaf Extract on Oxygen Radicals and Their Scavenger Enzymes in Brain of SD Rats)

  • This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in brain membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) lecels resulted in significant decreases (13.4% and 21.1%, 12.0% and 13.4%, respectively) in brain mitochondria and microsome of MLE-100 and MLE-300 groups compared with control group. Superoxide radical ($O_2$) levels were significantly decreased about 12% in brain cytosol of MLE-300 group compared with control group. Lipid peroxide (LPO) levels were effectively inhibited (18.1% and 12.3%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (14.2%, and 10.9%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Mn-SOD activities in brain mitochondria were significantly increased (13.5% and 18.6%, respectively) in MLE-100 and MLE-300 groups, and Cu/Zn-SOD activities in brain cytosol were also effectively increased (about 17.7%) in MLE-300 groups compared with control group. GSHPx activities in brain cytosol were remarkably increased (17.2% and 23.9%, respectively) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes in brain.

Evaluation of Antioxidative Activity of Agrimonia pilosa-Ledeb Leaves on Non-lipid Oxidative Damage

  • Present study was conducted to evaluate the anti oxidative activity of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidative damage. The antioxidative activity of methanolic (MeOH) extract of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidation, including liposome oxidation, deoxyribose oxidation, protein oxidation, chelating activity against metal ions, scavenging activity against hydrogen peroxide, scavenging activity against hydroxyl radical and 2'-deoxyguanosine (2'-dG) oxidation were investigated. The MeOH extract of the Agrimonia pilosa-Ledeb leaves exhibited high anti oxidative activity in the liposome model system. Deoxyribose peroxidation was inhibited by the MeOH extract of the Agrimonia pilosa-Ledeb leaves and MeOH extract of the Agrimonia pilosa-Ledeb leaves provided remarkable protection against damage to deoxyribose. Protective effect of MeOH extracts of the Agrimonia pilosa-Ledeb leaves on protein damage was observed at $600{\mu}g$ level (82.05%). The MeOH extracts of the Agrimonia pilosa-Ledeb leaves at $300{\mu}g$ revealed metal binding ability (32.64%) for hydrogen peroxide. Furthermore, the oxidation of 2'-deoxyguanosine (2'-dG) to 8-hydroxy-2-deoxyguanosine (8-OH-2'dG) was inhibited by MeOH extracts of the Agrimonia pilosa-Ledeb leaves and scavenging activity for hydroxyl radical exhibited a remarkable effect. From the results in the present study on biological model systems, we concluded that MeOH extract of the Agrimonia pilosa-Ledeb leaves was effective in the protection of non-lipids against various oxidative model systems.

한방약물로부터 항고지혈증 치료약물개발(I) -수종 한약재의 항고지혈증 활성검색- (Studies on the Development of Antihyperlipidemic Drugs from Oriental Herbal Medicines(I) -Antihyperlipidemic Activities of Oriental Herbal Medicines-)

  • Twenty-four Oriental herbal medicines including Platycodi Radix and Scutellariae Radix, etc., which have been used for the cure of hyperlipidemia, coronary heart, disease were evaluated for the 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA) reductase inhibition and antioxidant effect on a free radical, 1,1-diphenyl-2-picrylhydrazyl(DPPH) in vitro, and antihyperlipidemic effect on hyperlipidemic rats induced by Triton WR-1339 in vivo. 80% MeOH extract of eight herbs including Scutellariae Radix, Coptidis Rhizoma, Paeoniae Radix Rubra, Moutan Cortex Radicis, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam, Cinnamomi Ramulus and Crataegi Fructus inhibited the HMG-CoA reductase activities and exhibited a radical-trapping action on a stable free radical, DPPH. On Triton WR-1339-induced hyperlipidemic rats, four herbs including Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus showed respectively the significant suppression of serum total cholesterol, triglyceride, phospholipid and LDL-cholesterol levels and serum transaminase(ALT and AST) activities. From these results, it is suggested that each 80% MeOH extract of Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus have effective antihyperlipidemic action against hyperlipidemia.
