Protoplast-source meterial and enzyme strength had a significant influence on protoplast yield from hybrid poplar, Populus alba ${\times}$ P. grandidentata. The yield of protoplasts from in vitro culture of 1 month-old plantlets was more than that from greenhouse grown 4 month-old stock plant. In vitro cultured plantlets regulary produced more viable protoplasts with E-I enzyme solution (0.5% cellulase and 0.1% macerase) than those with E-II enzyme solution (1.0% cellulase and 0.2% macerase) after overnight incubation. The mean yield of protoplasts from in vitro cultured plantlets was $4{\times}10^6$ with E-I enzyme solution. Cell division was observed in these protoplast cultures after 7-10 days. Protoplast-derived hybrid poplar cells survived over 3 weeks in culture and some continuous cell divisions were evident. Other aspects associated with protoplasts from in vitro cultured plantlet are also discussed.
The effects of the ectomycorrhizal fungus Pisolithus tinctorius and cadmium (Cd) on physiological properties and Cd uptake by Populus alba ${\times}$ glandulosa was investigated under greenhouse conditions. Cd treatment decreased the photosynthetic rate ($P_N$) of both non-mycorrhizal (NM) plants (16.3%) and ectomycorrhizal (ECM) plants (11.5%). In addition, the reduction in total dry weight by Cd treatment was greater in ECM plants (24.3%) than that in NM plants (17.6%). Mycorrhizal infection increased the $P_N$ and transpiration rate in both control and Cd-treated plants. Cd treatment increased superoxide dismutase (SOD) activity and decreased glutathione reductase activity, and the increase of SOD activity by Cd treatment was greater in NM plants (40.3%) than that in ECM plants (3.7%). Thiol content increased in both NM and ECM plants treated with Cd solution, and the increase in thiol content in NM plants (43.9%) was greater than that of ECM plants (15.6%). Cd uptake in the leaves, stems, and roots of ECM plants was 69.9%, 167.2% and 72.8%, respectively, higher than in the NM plants. However, the increase in Cd uptake ability of ECM plants resulted in a reduction in dry weight.

The vegetation period of trees might be prolonged by the delay of the leaf senescence in autumn. Thus, we focused on the generation of senescence-delayed transgenic trees to enhance biomass production. The PagMYC2, a gene containing the basic helix-loop-helix domain, was selected as a candidate for a senescence-delayed transgenic tree. The PagMYC2 gene was specifically induced after treatment with phytohormone jasmonic acid, and upregulated by abiotic stresses such as salinity, osmotic pressure and a low temperature. The constitutive overexpression of the PagMYC2 delayed the leaf senescence and inhibited chlorophyll degradation in the transgenic poplars. Leaf senescence analysis was performed in the leaf tissues of the PagMYC2-over-expression transgenic poplars. The transgenic poplars exhibited higher photochemical efficiency than did a wild type plant under a short-day condition (6 hours light/18 hours darkness) or a low temperature condition ($15^{\circ}C$) that was similar to the weather conditions of autumn. These results suggest that the PagMYC2 is a useful genetic resource to improve biomass production, which is able to sustain growth with senescence-delayed leaves for a long time in autumn.
Diploid plants were obtained by anther culture of tetraploid poplar(Populus alba L. X P.glandutosa Uyeki). The effect 2,4D on callus formation from anther culture was greater than any other auxins tested. The highest average number of multiple shoots per callus was obtained when zeatin was used at levels of 6-8 ${\mu}$M. Regenerated shoots were excised and transferred to MS basal medium. Rooted plantlets were subsequently transferred to pots containing artificial soil mix. Finally 100 plane were transplanted in nursery located in forest Genetics Research Institute. for the 300 anther clones growing in greenhouse for 6 months after transplanting, 33% were slow-growing, 47% were rapid-growing and 20% had huge leaf size with rapid-growing characteristics. Chromosome study showed a narrow range of variation from diploid to tetraploid. DNA polymorphism studies using various RAPD markers revealed some extend of differences among the anther-clones in their band pattern.

Suspended wood waste was being inflow into the dam and the problem of waste disposal has been occurred. In this study, ethanol production using woody floater wastes was performed to estimate value in use for raw material of renewable energy. To achieve the goal, experiments of acid hydrolysis and ethanol fermentation using dam woody floater as raw materials for bioethanol was carried out. In the results of field survey in the chungju dam, kind of woody floater was mainly Japanese larch (Larix leptolepis) and hybrid poplar (Populus tomentiglandulosa). The results of sugar extraction showed that sugar content was higher in Larix leptolepis than Populus tomentiglandulosa. Extracted sugar from wood waste was effective consumed by yeast(P. Stipitis and S cerevisiae). In the experiment consumption of sugar including glucose, galactose and xylose, the consumption rate of S. cerevisiae is faster than that of P. stipitis. and efficiency for ethanol production is higer in S. cerevisiae than P. stipitis. Also it can be confirmed that resource as ethanol production using wood waste was available.
Physiologically modified stem nodes derived from ex vitro and in vivo explants of hybrid aspen (Populus alba L.X P.grandidentata Michx. 'Crandon') were tested for their multiple shoot regeneration capacity using a broad spectrum dosage of cytokinins. Ex vitro derived stem nodes with excised axillary buds at the time of culture produced 11 to 13 multiple shoots on 20 to 30 $\mu$M zeatin containing Woody Plant Medium (WPM) after 6 weeks. Excision of axillary bud sprouts after 2 weeks of culture and culture of the remaining stem nodes on WPM with 1.0 to 2.0 $\mu$M BA or 10 to 30 $\mu$M zeatin produced 13 to 15 and 7 to 8 shoots per explant, respectively, Multiple tiny shoots were produced when in vivo derived stem nodes (on which all leaves were removed) were cultured on WPM with 30 to 50 $\mu$M 2iP or 20 to 50 $\mu$M zeatin. The greatest number of multiple tiny shoot proliferation (32 to 50 shoots per explant) were obtained when the explants were cultured on media containing 20 $\mu$M zeatin. Successful transplanting of these multiple shoots into the greenhouse and/or nursery was achieved.

The widely cultivated hybrid poplar Populus alba ${\times}$ P. glandulosa in Korea was transformed with Agrobacterium rhizogenes agropine type strain A4. Genetic transformation was confirmed by the presence of agropine. Alteration in growth rate of hairy roots was seen following changes in the dilution rate of medium and concentration of sucrose, suggesting that improved growth might be achieved by more precise manipulation of the nutrient medium. Plant regeneration occurred from transformed hairy roots on MS medium supplemented with 0.5 mg/l BAP. Transformed plantlets grown in vitro exhibited a more developed root system characterized by fast growth behavior in comparison to normal plantlets. This work demonstrates that the root transformation would be useful in improving plantlet establishment and growth through the effective root system.

This study was conducted to analyze growth responses of fast growing tree species(8 clones of hybrid poplars, Salix alba, Metasequoia glyptostroboides, Liriodendron tulipifera, Acer okamotoanum, and Quercus palustris), the chemical characteristics of soil and $NO_3-N$ concentration of groundwater in a plantation applied with liquid pig manure. Concentrations of nitrogen and phosphorous in the soil treated with liquid pig manure were higher than that of the soil treated without liquid pig manure. With the exception of S. alba, DBH(Diameter at Breast Height) growth of all the fast growing tree species treated with liquid pig manure was higher than that of the species treated without liquid pig manure. In liquid pig manure treatment group, P. euramericana 'Eco28' clone showed the best performance in height and DBH growth. Concentration of nitrogen in the leaf with liquid pig manure was higher than that of the leaf treated without liquid pig manure. Based on the $NO_3-N$ concentration of groundwater analyzed during the experimental period, there was no evidence that groundwater was polluted by the liquid pig manure applied at the plantation.
The influences of fertilizer treatment and clones of five willows and one hybrid poplar on above ground and soil carbon (C) accumulations in a willow bioenergy plantation were studied. The aboveground and soil samples were collected in the winter of 1992 and 1993 from the previously established willow plantation at Tully, New York, U.S.A. in 1987. Half of the plots were fertilized annually with 336kg/ha N, 112kg/ha P, and 224kg/ha K. All trees were harvested annually. The most productive clone, willow clone SV1 with fertilization, accumulated 5.4 and 6.8 t/ha/yr aboveground C contents during the sixth(1992) and seventh(1993) growing seasons, respectively. The average percentage of C in bolewood, bolebark, and branches for the five willow clones and one hybrid poplar clone ranged from 51.1 to 57.5, from 54.0 to 55.4, and from 55.6 to 56.5, respectively, among all treatment combinations. Only tyro of the six clones(SA22 and SA2) responded significantly to the addition of fertilizer by increasing the amount of aboveground C accumulated for the 1992 sampling period(clone-by-fertilizer interaction). No fertilization effect, on aboveground C content, was noted for the 1993 sampling period. No significant fertilization effect on soil C accumulation for all soil sampling depths(0-10, 10-20, and 20-40cm) was found in 1992 and 1993 sampling years. Little clone effect on soil C content was found in 1992 and 1993 sampling years, except at 0-10cm soil depth in 1992. The significant clonal effect on soil C content at 0-10cm soil depth could be because of stone content variation rather than clonal effect. The significant clone-by-fertilizer treatment interaction observed requires that evaluation of response to fertilization by willows be made for each clone individually.
Protoplasts isolated from leaf mesophyll tissues of Populus koreana ${\times}$ P. nigra var. italica were fused with those of P. euramericana cv. Guardi. Well expended healthy leaves of 5 to 7 week-old-plantlet grown in vitro were used as source materials. Leaves from P. koreana ${\times}$ P. nigra var. italica and P. euramericana cv. Guardi were digested in enzyme solution I (2.0% Cellulase, 1.2% Hemicellulase, 0.4% Macrozyme, 2.0% Driselase, 0.05% Pectolyase ; w/v) and enzyme solution II (1.0% Cellulase, 1.2% Hemicellulase, 0.4% Macrozyme, 2.0% Driselase, 0.05% Pectolyase ; w/v), respectively, The highest frequency of fusion among the protoplasts originated from the two source materials was approximately 21% using 40% PEG or 15% dextran. In addition, fusion frequency was enhanced by incorporating 30mM of $Ca^{2+}$ in eluting solution at pH 10.5. Dividing cells and/or mint-calli were obtained by culturing the fusion products in a liquid 8p-KM medium supplemented with 0.6M sucrose, $0.45{\mu}M$ 2, 4-D, and $0.5{\mu}M$ BA. Shoots were regenerated from the fusion product-derived calli after culture on MS medium containing $5.0{\mu}M$ zeatin. To verify the putative hybrid or cybrid, SDS-PAGE was carried out. From the 24 regenerants, just two plants showed intermediate protein band patterns compared with those of the original source plants.
