The Porcine FoxO1, FoxO3a and FoxO4 Genes: Cloning, Mapping, Expression and Association Analysis with Meat Production Traits

  • FoxO1, FoxO3a and FoxO4 belong to the FoxO gene family, which play important roles in the PI3K/PKB pathway. In this study, we cloned the porcine FoxO1, FoxO3a and FoxO4 sequences and assigned them to SSC11p11-15, SSC1p13 and SSC xq13 using somatic cell hybrid panel (SCHP) and radiation hybrid panel (IMpRH). RT-PCR results showed that these three genes are expressed in multiple tissues. Sequencing of PCR products from different breeds identified a synonymous T/C polymorphism in exon 2 of FoxO3a. This FoxO3a single nucleotide polymorphism (SNP) can be detected by AvaII restriction enzyme. The allele frequencies of this SNP were investigated in Dahuabai, Meishan, Tongcheng, Yushan, Large White, and Duroc pigs. Association of the genotypes with growth and carcass traits showed that different genotypes of FoxO3a were associated with carcass length and backfat thickness between 6th and 7th ribs (BTR) and drip loss (p<0.05).

  • 사진 촬영 당시의 외부표정요소 추정 방법에는 공선조건식 기반 후방교회법이 널리 사용되지만 초기값을 필요로 하고, 그 값에 민감하다는 단점이 있다. 본 연구에서는 초기값을 필요로 하지 않는 외부표정요소 알고리즘인 쿼터니언 기반 공간후방교회법과 PnP (Perspective-n-Point algorithm)을 소개하고 그 결과를 비교하였다. 두 결과를 비교하기 위하여 일반 스마트폰으로 취득한 영상을 사용하였고, 지상기준점 취득은 본 연구진이 보유하고 있는 하이브리드 MMS (Mobile Mapping System) 점군 자료를 이용하였다. 그 결과, 공선조건식 기반 SPR (Single Photo Resection)을 참값으로 할 때, 쿼터니언 기반 SPR이 PnP 알고리즘에 비해 자세각 추정 정확도가 높았다. 카메라 위치추정의 경우에는 두 알고리즘 모두 지상기준점과 비교했을 때 0.8m 내의 정확도를 보임을 확인하였다.

  • 플래시 메모리는 크기가 작고, 적은 전력을 사용하며 충격에 강하기 때문에 MP3 플레이어, 핸드폰, 디지털 카메라와 같은 휴대용 기기에서 저장장치로 널리 사용되고 있다. 플래시 메모리의 많은 장점 때문에 개인용 컴퓨터 및 노트북에서 사용되는 저장장치인 하드디스크를 플래시 메모리로 대체하고자 하는 연구도 진행되고 있다. 플래시 메모리는 덮어쓰기가 허용되지 않으며 읽기/쓰기의 기본 단위와 삭제의 기본 단위가 다르기 때문에 FTL(Flash Translation Layer)라는 플래시 변환 계층을 사용한다. 최근에는 기존의 플래시 메모리와 다른 물리구조와 특성을 갖는 대블록 플래시 메모리가 등장하여 기존의 FTL을 그대로 사용하게 되면 플래시 메모리를 효율적으로 사용할 수 없다. 본 논문에서는 기존의 FTL 중 가장 좋은 성능을 내는 FAST(Fully Associative Sector Translation)을 기반으로 데이타블록 내에서 페이지단위 사상을 적용하여 대블록 플래시 메모리의 특성에 맞는 FTL 기법을 제안한다.

Klebsiella pneumoniae NFB-320의 Pullulanase 유전자의 제한효소 분석과 효소학적 특성 (Restriction Mapping of Cloned Pullulanase Gene and Property of Pullulanase Produced in Escherichia coli (pYKL451) and Klebsiella pneumoniae NFB-320)

  • 앞서 보고한 바와 같이 토양으로부터 분리한 K. pneumoniae NFB-320의 pullulanase 유전자를 pBR 322을 이용하여 E. coli에 cloning한 결과 약 14.4kb 의 재조합 plasmid DNA pYKL451을 얻었다. 이러한 pullulanase 유전자에 대한 유전적 정보를 얻기 위해 여러 가지 제한효소로 단일 혹은 이중 절단을 행하여 삽입된 pullulanase 유전자의 제한효소 절단지 도를 작성하였으며, E. coli(pYKL451)과 K. pneumoniae NFB-320이 생산하는 pullulanase의 효소적 특성을 조사하였다. 생산되는 두 균주의 효소는 50-55$^{\circ}C$ 부근에서 최적온도를 나타냈으며 최적pH는 모두 6.0이었다. 효소 안정성에 미치는 pH의 영향은 4$0^{\circ}C$에서 90min 간 방치했을 때 pH 5.0-10.0에서 안정하였으며 열안정성은 (pH6.0) 각 온도에서 한시간 처리하였을 때 4$0^{\circ}C$까지는 안정하였으나 5$0^{\circ}C$ 이상에서는 효소의 활성이 급격히 감소하였다.

High Resolution 3D Magnetic Resonance Fingerprinting with Hybrid Radial-Interleaved EPI Acquisition for Knee Cartilage T1, T2 Mapping

  • Purpose: To develop a 3D magnetic resonance fingerprinting (MRF) method for application in high resolution knee cartilage PD, T1, T2 mapping. Materials and Methods: A novel 3D acquisition trajectory with golden-angle rotating radial in kxy direction and interleaved echo planar imaging (EPI) acquisition in the kz direction was implemented in the MRF framework. A centric order was applied to the interleaved EPI acquisition to reduce Nyquist ghosting artifact due to field inhomogeneity. For the reconstruction, singular value decomposition (SVD) compression method was used to accelerate reconstruction time and conjugate gradient sensitivity-encoding (CG-SENSE) was performed to overcome low SNR of the high resolution data. Phantom experiments were performed to verify the proposed method. In vivo experiments were performed on 6 healthy volunteers and 2 early osteoarthritis (OA) patients. Results: In the phantom experiments, the T1 and T2 values of the proposed method were in good agreement with the spin-echo references. The results from the in vivo scans showed high quality proton density (PD), T1, T2 map with EPI echo train length (NETL = 4), acceleration factor in through plane (Rz = 5), and number of radial spokes (Nspk = 4). In patients, high T2 values (50-60 ms) were seen in all transverse, sagittal, and coronal views and the damaged cartilage regions were in agreement with the hyper-intensity regions shown on conventional turbo spin-echo (TSE) images. Conclusion: The proposed 3D MRF method can acquire high resolution (0.5 mm3) quantitative maps in practical scan time (~ 7 min and 10 sec) with full coverage of the knee (FOV: 160 × 160 × 120 mm3).

Synergy study on charge transport dynamics in hybrid organic solar cell: Photocurrent mapping and performance analysis under local spectrum

  • Charge transport dynamics in ZnO based inverted organic solar cell (IOSC) has been characterized with transient photocurrent spectroscopy and localised photocurrent mapping-atomic force microscopy. The value of maximum exciton generation rate was found to vary from $2.6{\times}10^{27}m^{-3}s^{-1}$ ($J_{sat}=79.7A\;m^{-2}$) to $2.9{\times}10^{27}m^{-3}s^{-1}$ ($J_{sat}=90.8A\;m^{-2}$) for devices with power conversion efficiency ranging from 2.03 to 2.51%. These results suggest that nanorods served as an excellent electron transporting layer that provides efficient charge transport and enhances IOSC device performance. The photovoltaic performance of OSCs with various growth times of ZnO nanorods have been analysed for a comparison between AM1.5G spectrum and local solar spectrum. The simulated PCE of all devices operating under local spectrum exhibited extensive improvement with the gain of 13.3-3.7% in which the ZnO nanorods grown at 15 min possess the highest PCE under local solar with the value of 2.82%.

  • 본 논문은 멀티스케일 공정-구조 해석의 방법론을 제안하고 단섬유층과 직물층으로 이루어진 배터리 하우징 파트에 적용한다. 특별히 마이크로스케일 대표체적요소(RVE: Representative Volume Element)안 기지의 불확정성을 고려하였다. 마이크로스케일의 RVE내 기지 물성의 랜덤한 공간내 분포는 KLE(Karhunen-Loeve Expansion)을 통해 구현하였다. 공간상 랜덤분포된 기지 물성을 갖는 RVE의 유효 물성을 전산균질화를 통해 얻어 매크로스케일 유한요소 모델에 매핑하였다. 또한 하이브리드 공정해석을 통해 압축 성형 해석으로부터 얻은 잔류응력과 섬유배향을 매핑한 유한요소 모델과 드레이핑 공정 해석결과로부터 얻어진 섬유배향을 매핑한 모델을 결합하였다. 본 연구에 제안된 방법은 배터리 하우징 뿐만 아니라 다양한 재료 구성을 갖는 복합재료의 공정-구조해석을 통해 설계요구도를 엄밀하게 평가할 수 있을 것이라 기대된다.

Cloning and Characterization of Bovine Titin-cap (TCAP) Gene

  • Titin-cap (TCAP), one of the abundant transcripts in skeletal muscles, was nvestigated in this study in cattle because of its role in regulating the proliferation and differentiation of myoblasts by interacting with the myostatin gene. From the 5, and 3, RACE experiments, full-length TCAP coding sequence was identified, comprising 166 amino acids. The amino acid comparison showed high sequence similarities with previously identified human (95.8%) and mouse (95.2%) TCAP genes. The TCAP expression, addressed by northern blot, is limited in muscle tissues as indicated by Valle et al. (1997). The radiation hybrid analysis localized the gene on BTA19, where the comparative human and porcine counterparts are on HSA17 and SSC12. A few muscle-related genetic disorders were mapped on HSA17 and some growth-related QTLs were identified on SSC12. The bovine TCAP gene found in this study opens up new possibilities for the investigation of muscle-related genetic diseases as well as meat yield traits in cattle.

Molecular Characterization, Chromosomal Localizations, Expression Profile, and Association Analysis of the Porcine PECI Gene with Carcass Traits

  • The full-length cDNA of the porcine peroxisomal ${\Delta}^3$,${\Delta}^2$-enoyl-CoA isomerase (PECI) gene encodes a monofunctional peroxisomal ${\Delta}^3$,${\Delta}^2$-enoyl-CoA isomerase. Cloning and sequencing of the porcine PECI cDNA revealed the presence of an 1185-base pair open reading frame predicted to encode a 394-amino acid protein by the 5'rapid amplification of cDNA ends (5'RACE) and EST sequences. The porcine PECI gene was expressed in seven tissues (heart, liver, spleen, lung, kidney, skeletal muscle, fat) which was revealed by reverse transcriptase-polymerase chain reaction (RT-PCR). The porcine PECI was mapped to SSC71/2 p11-13 using the somatic cell hybrid panel (SCHP) and the radiation hybrid panel (RH) (LOD score 12.84). The data showed that PECI was closely linked to marker S0383. A C/T single nucleotide polymorphism in PECI exon 10 (3'UTR) was detected as a PvuII PCR-RFLP. Association analysis in our experimental pig population showed that different genotypes of PECI gene were significantly associated with the Average Backfat thickness (ABF) (p<0.05) and Buttock backfat thickness (p<0.01).

Sequence Characterization, Expression Profile, Chromosomal Localization and Polymorphism of the Porcine SMPX Gene

  • The full-length cDNA of the porcine SMPX gene was obtained by the rapid amplification of cDNA ends (RACE). The nucleotide sequences and the predicted protein sequences share high sequence identity with both human and mouse. The promoter of SMPX was sequenced and then analyzed to find the promoter binding sites. The reverse transcriptase-polymerase chain reaction (RT-PCR) revealed that SMPX has a high level of expression in heart and skeletal muscle, a very low expression in lung and spleen and no expression in liver, kidney, fat and brain. Moreover, SMPX has a differential expression level in skeletal muscle, the expression in 65-day embryos being higher than other stages. The porcine SMPX was mapped to SSCXp24 by using a somatic cell hybrid panel (SCHP) and was found closely linked to SW1903 using the radiation hybrid panel IMpRH. An A/G single nucleotide polymorphism (PCR-RFLP) in the 3'-untranslated region (3'-UTR) was detected in eight breeds. The analysis of allele frequency distribution showed that introduced pig breeds (Duroc and Large White) have a higher frequency of allele A while in the Chinese indigenous pig breeds (Qingping pig, Lantang pig, YushanBlack pig, Large Black-White pig, Small Meishan) have a higher frequencies of allele G. The association analysis using an experimental population (188 pigs), which included two cross-bred groups and three pure-blood groups, suggested that the SNP genotype was associated with intramuscular fat content.
