Solubility and Storage Stability of Astaxanthin (Astaxanthin의 용해특성 및 저장 안정성)

  • Basic characteristics of astaxanthin including solubility and stability were investigated. Astaxanthin showed a very poor solubility in water, but it was highly soluble in organic solvents such as acetone and acetic acid. The solubility of astaxanthin in acidic condition was 10-20 times higher than those in neutral and basic conditions. Astaxanthin was very unstable in acidic condition under UV irradiation and in the presence of oxygen. Also, heating even for a very short time accelerated the degradation of astaxanthin. In conclusion, it is required to enhance the water-solubility and stability of astaxanthin for industrial application in food and cosmetic area.

Study on Fluorescence Excitation and Emission Bands for Detection of Fecal Residue on Poultry Carcasses (도계육 표면 내장 잔유물 오염 검사를 위한 형광 조사 및 방출파장 선정에 관한 연구)

  • Fecal contaminations on poultry carcasses, not easily discemable by human eyes, are potential harbor sites of pathogenic Escherichia Coli (E. coli O157:H7). Development of sensitive detection methods for fecal contamination is essential to ensure safe production of poultry products. Fluorescence has been shown to be very sensitive in detecting fecal and other biological substances that can harbor pathogens. In this study, fluorescence excitation-emission spectra of poultry fecal matter were compared with spectra for poultry skin and meat. Results indicated that the combinations of fluorescence intensities at the wavelength of 520 nm, 579 nm, 625 nm, and 635 nm with 411 nm excitation showed above 97% accuracy for differentiation of the contaminants from poultry tissues. Excitation and emission bands determined could be used for constructing a real-time fluorescence imaging device for detection of harmful residues on poultry carcasses.

  • The pulsatile nature of blood flow makes artefacts in 2D Fourier transform image. Spatial presaturation is known to be effective in eliminating flow artefacts when the spin echo acquisition is employed. However. this method requires additional RF pulse and spoiling gradient for presaturation. In this paper a new flow saturation technique which does not require additional saturation-RF and gradient is proposed. The proposed technique is equivalent to the existing saturation technique but the elimination of the flow component is achieved by a pair of tailored $90^{\circ}-180^{\circ}$ RF pulses in tile spin echo sequence. By use of two tailored RF pulses with opposite phase polarity, a linear phase gradient is generated for those moving materials and consequently all the spins of moving materials become dephased thereby no signal is observable. Computer simulations and experimental results obtained using both a phantom and a human volunteer with a 2.0 T whole body system are also presented.

  • The quantitative comparison of brain architecture across different subjects requires a common coordinate system, with respect to which the spatial variability of features form different individuals can be expressed. We have developed and implemented an image warping technique which is based on an elastic body deformation. The resulting 2D deformation map can be used to quantify anatomic differences between subjects. The technique's accuracy is tested, by warping 2D magnetic resonance images of age seventies into Talairach atlas.

Comparison of Analytical Method for Measuring Particulate Matter in Indoor Air (실내공간에서의 공기중 먼지 측정방법에 관한 비교분석)

  • Since most people spend a large majority of their time indoors (at least in the industrialized countries), indoor air may affect human health more than outdoor air. This study was carried out to characterize the reference and equivalent methods against the low volume method which was promulgated by the Ministry of Health and Social Affairs. The Laser and Piezo air sampler offer the advantage of real time data and low labor costs. The arithmetic mean concentrations were found to be 102.9% (Laser-2 min method) and 65.9% (Piezo method) against low volume method (100%). The statistical analysis procedure for this comparision is linear regression. The linear regression line of low volume method had slopes of 0.5487 and 0.9697 and Y intercepts of 0.0266 and 0.0110 $\mu$g/m$^3$ about Laser (2 min) and (24 h) method respectively. And the correlation coefficients were 0.7271 and 0.9433.

A Novel Anticoagulant Protein from Scapharca broughtonii

  • An anticoagulant protein was purified from the edible portion of a blood ark shell, Scapharca broughtonii, by ammonium sulfate precipitation and column chromatography on DEAE-Sephadex A-50, Sephadex G-75, DEAE-Sephacel, and Biogel P-l00. In vitro assays with human plasma, the anticoagulant from 'S. broughtonii, prolonged the activated partial thromboplastin time (APTT) and inhibited the factor LX in the intrinsic pathway of the blood coagulation cascade. But, the fibrin plate assay did not show that the anticoagulant is a fibrinolytic protease. The molecular mass of the purified S. broughtonii anticoagulant was measured to be about 26.0kDa by gel filtration on a Sephadex G-75 column and SDS-PAGE under denaturing conditions. The optimum activity in the APTT assay was exhibited at pH 7.0-7.5 and $40-45^{\circ}C$ in the presence of $Ca^{2+}$.

Toddler's Gender and Temperament by Maternal Stress as Predictors of Mothers' Parenting Behavior (걸음마기 아동의 성, 기질 및 어머니의 스트레스가 양육행동에 미치는 영향)

  • The objective of this study was to identify how child's gender, temperament, mother's stress and their interactions predict parenting behavior. The subjects were 97 2-year-olds(47boys; 50girls) and their mothers. Data were gathered by questionnaires. Results of a series of hierarchical regression analyses showed combinations of variables that predicted parenting behaviors : (1) mothers reported higher over-protectiveness toward girls and when they were under higher stress. (2) An interaction effect showed that mothers with sons who had higher 'difficult' temperaments reported greater over-protective parenting under greater stress. (3) There were no significant differences in over-protective parenting under low stress, regardless of the level of difficult temperament. (4) Mother's stress was a strong predictor of supportive parenting behaviors.

  • Bioassay-guided fractionation of the MeOH extract from the sponge Discodermia calyx collected off the coast of Jeju Island, South Korea, led to the isolation of a polyketide, icadamide C (1), along with previously reported theopederin K (3). Structure elucidation was performed by a combination of high resolution mass and 2D-NMR (principally COSY, HMBC, HSQC, and NOESY) spectroscopy. Stereochemistry of compound 1 was determined as 2R*, 3R*, 6R*, 10S*, 11S*, 12R*, 13S*, 15R* and 2'S by NMR data and Marfey analysis. Isolated metabolites displayed potent cytotoxic activity against a small panel of five human solid tumor cell lines with ED50 values of less than 0.1 μg/mL.

  • A column switching HPLC assay was developed to allow the separation and quantitation of cetirizine in human plasma by ultraviolet (UV) detection. Plasma samples were prepared by liquid-liquid extraction. After drying, the residue was reconstituted in 20 mM phosphate buffer (pH 2.8) containing 15% acetonitrile. The samples were initially injected onto a clean-up Capcell Pak MF C18 column. (50 mm $\times$ 4.6 mm I.D.), and the chromatographic region containing the peaks of interest was followed in an analytical C18 microcolumn (250 mm$\times$1.5 mm I. D.) via column switching device.
