Heavy metals and metalloids removal can be considered as one of the most important world challenges because of their toxicity and direct impact on human health. Many processes have been introduced but biological processes of remediation seem to offer the most suitable solution in terms of efficiency and low cost. Actinobacteria constitute one of the major microbial populations in soil, and this can be attributed to their adaptive morphological structure as well as their exceptional metabolic power. Among microbes, actinobacteria are morphologic intermediate between fungi and bacteria. Studies on microbial diversities in metal contaminated lands have shown that actinobacteria may constitute a dominantly active microbiota in addition to ${\alpha}$ Proteobacteria. Furthermore, isolation studies have shown metal removal mechanisms which are reminiscent of notable multiresistant strains, such as Cupriavidus metallidurans. Apart from members of genus Streptomyces, which produce more than 90% of commercialized antibiotics, and the nitrogen fixing Frankia, little attention has been given to other members of this phylum. This is because of difficult culture condition requirements and maintenance. In this review, we focused on specific isolation of actinobacteria and their potential applications in metal bioremediation and plant growth promotion.
Most reactive oxygen species (ROS) are free radicals and implicated in the development of a number of disease processes including artherosclerosis, neurodegenerative disorders, aging and cancer. ROS are byproducts of a number of in vivo metabolic processes and are formed deliberately as part of nor-mal inflammatory response. On the other hand, ROS are generated either as by products of oxygen reduction during xenobiotic metabolism or are liberated as the result of the futile redox cycling of the chemical agents including several chemical carcinogens. A better understanding of the mechanisms of free radical toxicity may yield valuable clue to risks associated with chemical exposures that leading to the development of chronic diseases including cancer. The molecular biology of ROS-mediated alterations in gene expression, signal transduction and carcinognesis is one of the important subjects in free radical toxicology. Epidemiological studies suggest that high intake of vegetables and fruits are associated with the low incidence of human cancer. Many phytopolyphenols such as tea polyphenols, curcumin, resveratrol, apigenin, genistein and other flavonoids have been shown to be cancer chemopreventive agents. Most of these compounds are strong antioxidant and ROS scavengers in vitro and effective inducers of antioxidant enzymes such as superoxide dismutatse, catalase and glutathione peroxidase in vivo. Several cellular transducers namely receptor tyrosine kinase, protein kinase C, MAPK, PI3K, c-jun, c-fos, c-myc, NFkB, IkB kinase, iNOS, COX-2, Bcl-2, Bax, etc have been shown to be actively modulated by phyto-polyphenols. Recent development in free radical toxicology have provided strong basis for understanding the action mechanisms of cancer chemoprevention.
Genetically modified foods and food additives are derived from organisms that have been inserted foreign genetic materials by recombinant DNA techniques to improve the quality or any other pur-poses. The problems such as toxicity, allergenicity and antibiotics resistance in the safety of genetically modified foods are usually concerned. In Korea, the safety of foods is ensured by the Food Sanitation Act. Although there is no specific provision regarding the genetically modified foods in it, any foods that might cause negative effect(s) on public health or human life are prohibited to sell in the market. In order to systematically evaluate safety of genetically modified foods, the Korea Food and Drug Administration (KFDA) promulgated "Guidelines regarding review of safety assessment data for genetically modified foods and food additives (KFDA Notification 1999-46)". The objectives of these guidelines are to ensure safety of genetically modified foods and food additives. In order to evaluate the safety of genetically modified foods. KFDA operates a special expert committee composed by experts from government, universities, research institutes. and consumer's unions. Recently. manufacturers and consumers are interested in the issues on safety and labeling of genetically modified foods, because of increment of imported genetically modified crops and processed foods. Since government and consumers unions have different viewpoints, their positions regarding the issue are different each other. Therefore, the regulation of labeling on genetically modified foods is prepared and should be enforced at July 2000 in Korea. in Korea.
Cadmium is considered one of the most toxic, non biodegradable heavy metal for the human and animals. The purpose of the present study was to investigate the changes in biochemical parameters of blood and accumulation of cadmium in various tissue caused by various levels of dietary cadmium chloride ($CdCl_2$) in broiler chicks. $CdCl_2$ was administered through drinking water to broiler chicks. In spectral analysis, $CdCl_2$ treatment caused a significant increase in Glutamate pyruvate transaminase (GPT), creatinine and uric acid levels in all treated groups. Intriguingly, the GPT, creatinine, and uric acid levels were significantly higher at 75 mg/kg as compared to the groups treated with high doses (100, 125 and 150 mg/kg) of $CdCl_2$. Atomic Absorption Spectrophotometer (AAS) was used for the determination of Cd accumulation in kidney, liver and Breast muscles. AAS analysis revealed that Cd accumulation is increased in breast muscles as compared to liver and kidney at higher doses of Cd than 75 mg/kg.
Mining activity in the Gubong gold mine started in 1908 and lasted up to recent days. Heavy metals derived from the activity may be porentially toxic to human life and envirinment of this area. Because metal toxicity depends on chemical associations into five operationally defined groups: exchangeable, carbonate, reducible, oxidizable, and residual fractions, and the Most of heavy metals have significant little significance (alomost<1%). And Cu is mainly associated with the oxidizable from. Total concentration of heavy metals, pH, and mineralogy affect the chemical forms of the metals. Heavy metal concentrations. Significant amounts of metal elements (5∼65.1% in Pb, 6.2∼39.7% in Zn, 8.7∼54.7% in Cd, and 3.6∼24.7% in Cu) were present in carbonate form from mine wastes, contaminated soils and sediments. High pH value and cerussite (Pb bearing carbonate mineral) in mine wastes, contaminated soils and sediments. High pH value and cerussite (Pb beraring varbonate mineral) in mine waste support this result. Areas with high corbonate bound from would have higher potentoal conamination, however, because elements of carbonate bound forms are easily mobilized under lower pH conditions in the surface envionments due to acid to rain soil acidification.
Environmental hazards of a chemical could be assessed by two different approaches : toxicity test and assessment of exposure potentials to human and environmental organisms. For the prediction of environmental fate of chemicals three available computer programs were compared each other and were verified. The results obtained by using these computer programs, PCHEM, EXAMS, and E4CHEM were summarized as follows. The estimated octanol/water partition coefficients by PCHEM were similar to the experimental values in the literature. But the other factors, water solubility and vapor pressure were different from the data in the literature. The simulation results of selected compounds by EXAMS showed similar tendency to the literature results of model field environment. Therefore, this computer program could be utilized to predict the environmental fate of chemicals. E4CHEM program is very simple and this program could predict the ultimate environmental fate of stable chemicals by input of two or three parameters. However, the validity should further be verified in the future field study using more compounds. It is suggested that these approaches could be fully utilized by understanding their limitations to predict the environmental fate of new chemicals under development, to screen the potential environmental pollutants among chemicals already-in use, and to devise measures to minimize the hazards to the environment.
Hexavalent chromium-contaminated soils are encounted at many unregulated discharge and improper handling of wastes from electroplating, leather tanning, steelmaking, corrosion control, and wood preservation industries. Contamination of hexavalent chromium in the soil is a major concern because of its toxicity and threat to human life and environment. Current technologies for hexavalent chromium-contaminated soil remediation are usually costly and/or cannot permanently prohibit the toxic element from entering into the biosphere. Thus, as an alternative technique, immobilization is seen as a cost-effective and promising remediation technology that may reduce the leachable potential of hexavalent chromium. The purpose of this paper is to develope an immobilization technique for the formation of the geochemically stabilized hexavalent chromium-contaminated soil from the reactions of labile soil hexavalent chromium forms with the added soluble phosphate and chromium reducing agent. From the liquid phase experiment, reaction order of chromium reducing agent, soluble phosphate, alkali solution shows the best removal efficiency of 95%. In addition, actual soil phase experiment demonstrates up to 97.9% removal efficiency with 1:1 molar ratio of chromium reducing agent and soluble phosphate. These results provide evidence for the potential use of soluble phosphate and chromium reducing agent for the hexavalent chromium-contaminated soil remediation.
BACKGROUND/OBJECTIVES: In Asia, various medicinal plants have been used as the primary sources in the health care regimen for thousands of years. In recent decades, various studies have investigated the biological activity and potential medicinal value of the medicinal plants. In this study, 100 methanol extracts from 98 plant species were evaluated for their biological activities. MATERIALS/METHODS: The research properties, including 1,1-diphenyl-2-pic-rylhydrazyl (DPPH) radical scavenging activity, ${\alpha}$-glucosidase and ${\alpha}$-tyrosinase inhibitory effects, anti-inflammatory activity, and anticancer activity were evaluated for the selected extracts. RESULTS: Fifteen of the extracts scavenged more than 90% of the DPPH radical. Among the extracts, approximately 20 extracts showed a strong inhibitory effect on ${\alpha}$-glucosidase, while most had no effect on ${\alpha}$-tyrosinase. In addition, 52% of the extracts showed low toxicity to normal cells, and parts of the extracts exhibited high anti-inflammatory and anticancer activities on the murine macrophage cell (RAW 264.7) and human colon cancer cell (HT-29) lines, respectively. CONCLUSIONS: Our findings may contribute to further nutrition and pharmacological studies. Detailed investigations of the outstanding samples are currently underway.

Free radicals including reactive oxygen species (ROS) are important chemicals in the research area of biology, pharmaceutical, medical, and environmental science as well as human health risk assessment as they are highly involved in diverse metabolism and toxicity mechanisms through chemical reactions with various components of living bodies. Electron spin resonance (ESR) spectroscopy is a powerful tool for detecting and quantifying those radicals in biological environments. In this work we observed the ESR signal of 2,2,6,6-Tetra-methyl piperidine 1-oxyl (TEMPO) in aqueous solution at various concentrations to estimate the uncertainty factors arising from the experimental conditions and signal treatment methods. As the sample position highly influences the signal intensity, dual ESR tube geometry (consists of a detachable sample tube and a position fixed external tube) was adopted. This type of measurement geometry allowed to get the relative uncertainty of signal intensity lower than 1% when triple measurements are averaged. Linear dependence of signal intensity on the TEMPO concentration, which is required for the quantification of unknown sample, could be obtained over a concentration range of ~103 by optimizing the signal treatment method depending on the concentration range.
Nor-aporphine derivatives have been discovered which interfere with the flux of Calcium into and out of the cell interior. It has been shown that adrenergic antagonists that block the Calcium exchange lead to an inhibition of Protein kinase C activity, thus blocking tyrosinase activation. Di-acetyl-dimethoxy-methyl-nor-aporphine is a semi-synthetic molecule of natural origin with very high potency. On B16 melanocytes as well as in normal human melanocytes the decrease in melanin synthesis reaches -50% at a level of 40 ppm in the culture medium. On a molar concentration basis, this is 50 to 70 times stronger than Kojic acid inhibition. Yet, the cell viability is not affected. Reversibility studies show that after washing out of the active compound, melanogenesis returns to normal levels. Possible mechanisms of the activity are discussed. Tests carried out on SkinEthic(R) three-dimensional models of the epidermis and in vivo clinical studies on Asian population confirm the strong inhibition of melanogenesis. Safety evaluation of these molecules, on the other hand, demonstrates good skin tolerance and absence of toxicity.
