We investigated if Chlorella vulgaris (CV) has protective effects on cadmium (Cd) induced liver damage in male Sprague-Dawley (SD) rats. Forty rats, aged 5 weeks old and weighed 90-110g, were divided into a control (with Cd free water), 50 ppm of $CdCl_2$ in drinking water treated groups (Chlorella 0% diet group (Cd/CV0%), Chlorella 5% diet group (Cd/CV5%) or Chlorella 10% diet group (Cd/CV10%). All the rats had freely access to water and diet for 8 weeks. The results show that body weight gain and relative liver weight had significantly lower in Cd/CV0%-treated group than in Cd/CV-treated groups. Hepatic Cd contents showed significantly less by feeding CV (P<0.05). Cd/CV0%-treated rats had significantly (P<0.05) higher hepatic T-MTs, and Cd-MTs concentrations, compared to Cd/CV5% or Cd/CV10% treated rats. The MT I/II mRNA was expressed in the liver of all experimental rats. Its expression was more increased in Cd/CV5%- or Cd/CV10%-treated rats, compared to control and Cd-treated rats. Thus, this study suggested that CV would have a protective effect on Cd-treated liver injury by the reduction of Cd concentrations and stimulation of Cd-MT binds in the liver. However, more studies are needed to identify the proper mechanism of CV and liver toxicity.
Malachite Green (MG), a toxic chemical used as a dye, topical antiseptic and antifungal agent for fish, is highly soluble in water, cytotoxic to various mammalian cells and also acts as a liver tumor promoter. In view of its industrial importance and possible exposure to human beings, MG possesses a potential environmental health hazard. So, we performed with HepG2, a human hepatocellular carcinoma cell line, to identify the differentially expressed genes (DEGs) related to toxicity of MG. And we compared gene expression between control and MG treatment to identify genes that are specifically or predominantly expressed by employing annealing control primer (ACP)-based $GeneFishing^{TM}$ method. The cytotoxicity $(IC_{20})$ of MG was determined above the $0.867{\mu}M$ in HepG2 cell for 48 h treatment. And the DEGs of MG were identified that 5 out of 6 DEGs were upregulated and 1 out of 6 DEGs was down-regulated by MG. Also, MG induced late apoptosis and necrosis in a dose dependent in flow cytometric analysis. Through further investigation, we will identify more meaningful and useful DEGs on MG, and then can get the information on mechanism and pathway associated with toxicity of MG.
Conventional chemotherapy against hepatocellular carcinoma typically causes various side effects. Our previous study showed that cecropin of Musca domestica can induce apoptosis in human hepatocellular carcinoma BEL-7402 cells in vitro. However, whether cecropin inhibits BEL-7402 cell in vivo and the question of possible side effects remained undentified. The present study confirmed tumor-inhibitory effects of cecropin in vivo, and furthermore strongly suggested that cecropin cytotoxicity in BEL-7402 cells in vivo may be mainly derived from its pro-apoptotic action. Specifically, we found that cecropin exerted no obvious side effects in tumor-bearing mice as it had no significant hematoxicity as well as visceral toxicity. Therefore, cecropin may be a potential candidate for further investigation as an antitumor agent against hepatocellular carcinoma.

To investigate the toxic of herbs used in , the author analyzed herbs with toxic recorded in , , after searched toxic herbs literarily. Herbs with toxicity recorded in both and were total 17 species, and recorded in were total 13 species, and recorded in were total 12 species. The major cause that herbs generates a side effect in a human body is implicit use and abuse, inadequate processing the herbs, use of quack medicines of herbs and clinical use which is improper in symptoms and internal use, differences of constitution. The major toxic component in herbs is an alkaloid, essential oil and hydrargyrum (Hg), and the main invasion route of toxic herbs in human body is the liver, kidney, lungs, and gastrointestinal tract.
From the flowers of Chromolaena odorata (Eupatorium odoratum) four flavanones, isosakuranetin (5,7-dihydroxy-4'-methoxyflavanone) (1), persicogenin (5,3'-dihydroxy-7,4'-dimethoxyflavanone) (2), 5,6,7,4'-tetramethoxyflavanone (3) and 4'-hydroxy-5,6,7-trimethoxyfla-vanone (4), two chalcones, 2'-hydroxy-4,4',5',6'-tetramethoxychalcone (5) and 4,2'-dihydroxy-4',5',6'-trimethoxychalcone (6), and two flavones, acacetin (5,7-dihydroxy-4'-methoxyflavone) (7) and luteolin (5,7,3',4'-tetrahydroxyflavone) (8) were isolated and identified. Compound 1 exhibited moderate antimycobacterial activity against Mycobacterium tuberculosis with the MIC value of 174.8 $\mu$M, whereas compounds 4,7, and 8 exhibited weak activity with the MIC values of 606.0, 704.2 and 699.3 $\mu$M respectively. Compound 7 showed moderate cytotoxicity against human small cell lung cancer (NCI-H187) cells with the MIC value of 24.6 $\mu$M, whereas compound 8 exhibited moderate toxicity against NCI-H187 cells and week toxicity against human breast cancer (BC) cells with the MIC values of 19.2 and 38.4 $\mu$M respectively.
Purpose: The aim of this study is to evaluate the effect of Xanthium sibiricum Patr. on carcinogenesis. Method: Water extract from Xanthium sibiricum Patr. (XPW) was prepared and investigated for the potential antitumor activity and inhibition of benzo[a]pyrene-DNA adduct formation and free radical formation. Result: It was shown that the water possess considerable toxicity toward tumor cell lines. Concentration of XPW at 1.0 mg/mL and 2.5 mg/mL resulted in more than 30% inhibition of growth in HeLa cells. Toxicity of XPW to A549 revealed that 54% inhibition of growth at concentration of 2.5 mg/mL. At concentrations of 0.5 mg/mL, 1.0 mg/mL and 2.5 mg/mL of XPW, the binding of [$^3H$]B[a]P metabolites to DNA of human Chang cell was inhibited by 19%, 33%, and 41%, respectively. There 18% and 32% inhibition in the free radical formation with XPW at the concentration of 1.0 mg/mL and 2.5 mg/mL, respectively. Conclusion: Water extract from Xanthium sibiricum Patr. (XPW) has antitumor and cancer chemopreventive activities.
To evaluate the cytogenetic toxicity, of cadmium and the reducing effect of selenium or zinc on cadmium toxicity, the induction of SCEs in cultured human lymphocytes by the concentraion of 0.5 $\mu$M to 16.0 $\mu$M of cadmium chloride and those of cadmium chloride combined with sodium selenite or zinc chloride 1.2 $\mu$M, respectively was investigated. The induction of SCEs by cadmium chloride in the range of 0.5 $\mu$M to 16.0 $\mu$M increased in a dose-dependent manner. A notable increase in SCEs by sodium selenite as well as zinc chloride was also observed. However, the frequency of SCEs by cadmium chloride was inhibited by the simultaneous addition of sodium selenite and zinc chloride 1.2 $\mu$M, respectively. The mitotic index significantly decreased in higher concentration of cadmium chloride but not was significantly different in any concentration of cadmium chloride with the simultaneous addition of sodium selenite or zinc chloride. The results showed that the decreased additive SCE effect was observed when induced by the combined treatment which could suggest that sodium selenite and zinc chloride have a protective effect on cadmium chloride.
Alzheimer's disease (AD) is neurodegenerative disease, characterized by the progressive decline of memory, cognitive functions, and changes in personality. The major pathological features in postmortem brains are neurofibrillary tangles and amyloid beta ($A{\beta}$) deposits. The majority of AD cases are sporadic and age-related. Although AD pathogenesis has not been established, aging and declining mitochondrial function has been associated. Mitochondrial dysfunction has been observed in AD patients' brains and AD mice models, and the mice with a genetic defect in mitochondrial complex I showed enhanced $A{\beta}$ level in vivo. To elucidate the role of mitochondrial complex I in AD, we used SH-SY5Y cells transfected with DNA constructs expressing human amyloid precursor protein (APP) or human Swedish APP mutant (APP-swe). The expression of APP-swe increased the level of $A{\beta}$ protein in comparison with control. When complex I was inhibited by rotenone, the increase of ROS level was remarkably higher in the cells overexpressing APP-swe compared to control. The number of dead cell was significantly increased in APP-swe-expressing cells by complex I inhibition. We suggest that complex I dysfunction accelerate amyloid toxicity and mitochondrial complex I dysfunction in aging may contribute to the pathogenesis of sporadic AD.

An attempt was made to assess metal ionic toxicity levels of different fishes in the polluted rivers viz., Buriganga and Turag. Fish samples collected from two polluted rivers were analyzed for the levels of metals such as Cd, Cr, Cu, Mn, Pb, and Zn in order to elucidate the status of these contaminants in fish meant for human consumption. The detected concentrations of Cr, Cu, Mn, and Zn ions in fish species collected from the polluted rivers were below the toxic levels and did not appear to pose a threat. Among the analyzed metals, Cd and Pb ions were detected above the permissible levels in liver and muscle tissues of stinging catfish (Heteropneustesfossilis), spotted snakehead (Channapunctata) and wallago (Wallagoattu) collected from the polluted rivers causing toxicity for human consumption. Stinging catfish (Heteropneustesfossilis) was the species found to highly bioaccumulate these metals. Fish species bioconcentrated appreciable amounts of Cd and Pb as toxic metals in the liver as compared to the muscle. Levels of these toxic metals varied depending on different tissues in fish species.
Background: Arsenic is a metalloid of public health significance due to its unique material properties and toxicity and the widespread pollution in the environment. Arsenic exists as inorganic arsenic and organic arsenic with distinct chemical properties. Its toxicity varies depending on the properties. Objectives: Although the carcinogenicity of arsenic has been identified, the various diseases that occur after acute and chronic exposure to arsenic are not yet clearly known. Methods: Research on the effects of chronic exposure to arsenic on human health was searched and the results were summarized. Results: It has been found that cancer occurs due to exposure to high concentrations of arsenic in areas with elevated exposure to arsenic, but research results have recently been presented on health effects caused by chronic exposure to low concentrations of arsenic. Cancers have also been identified to be related to inorganic arsenic, including skin cancer, lung cancer, and bladder cancer. Significant relationships with neurological diseases, cardiovascular diseases, and diabetes mellitus have been suggested as well. Conclusions: Our results suggest that it is necessary to evaluate the health impact on residents around abandoned metal mines and industrial complexes in South Korea.
