In vitro 상에서 유자(Citrus junos SIEB ex TANAKA) 용매 추출물의 암세포 억제효과 및 항산화성 (In Vitro Effect of Yuza (Citrus junos SIEB ex TANAKA) Extracts on Proliferation of Human Prostate Cancer Cells and Antioxidant Activity)

  • 유자는 성숙할수록 과피와 과육의 총페놀 함량은 많아지고 자유기 소거능이 높아지는 경향을 보였다. 유자로부터 추출한 18가지 용매 추출물의 항산화 활성과 전립선 암세포 DU 145, LN-CaP에 대한 세포독성 실험을 실시한 결과 과피와 과육 모두 미성숙한 유자 보다 성숙한 후 수확한 유자가 그 활성이 높게 나타냈다. 용매별로 살펴보면 과성숙 유자 과피, 과육의 클로로포름, 메탄올 추출물은 전립선 암세포 DU 145에 대하여 60% 이상의 세포사멸 효과를 나타났으며 메탄올 추출물의 효과도 농도에 따라 상당히 높은 것으로 나타났다. 또한 세포 조직을 현미경으로 살펴본 결과 추출물의 처리 농도가 증가할수록 세포 사멸이 쉽게 이루어지는 것을 확인 할 수 있었다. 이런 세포 사멸의 경향은 추출물의 자유기 소거 환성이 높을수록 높게 나타났다.

Increase in apoptotic effect of Panax ginseng by microwave processing in human prostate cancer cells: in vitro and in vivo studies

  • Background: Ginseng, which is widely used in functional foods and as an herbal medicine, has been reported to reduce the proliferation of prostate cancer cells by mechanisms that are not yet fully understood. Methods: This study was designed to investigate the changes in ginsenoside content in ginseng after treatment with a microwave-irradiation thermal process and to verify the anticancer effects of the extracts. To confirm the anticancer effect of microwave-irradiated processed ginseng (MG), it was tested in three human prostate cancer cell lines (DU145, LNCaP, and PC-3 cells). Involvements of apoptosis and autophagy were assessed using Western blotting. Results: After microwave treatment, the content of ginsenosides Rg1, Re, Rb1, Rc, Rb2, and Rd in the extracts decreased, whereas the content of ginsenosides 20(S)-Rg3, 20(R)-Rg3, Rk1, and Rg5 increased. Antiproliferation results for the human cancer cell lines treated with ginseng extracts indicate that PC-3 cells treated with MG showed the highest activity with an half maximal inhibitory concentration of $48{\mu}g/mL$. We also showed that MG suppresses the growth of human prostate cancer cell xenografts in athymic nude mice as an in vivo model. This growth suppression by MG is associated with the inductions of cell death and autophagy. Conclusion: Therefore, heat processing by microwave irradiation is a useful method to enhance the anticancer effect of ginseng by increasing the content of ginsenosides Rg3, Rg5, and Rk1.

Dracocephalum palmatum Stephan 잎 헥산 분획 추출물의 인간 유래 전립선 암세포 사멸에 대한 작용 (Effects of hexane fraction of Dracocephalum palmatum Stephan leaf on human-derived prostate cancer cell death)

  • Objectives : Dracocephalum palmatum Stephan (Lamiaceae) is a medicinal plant used by the East-Russian nomads but there were few studies on this plant. This study was to evaluate anti-cancer effects of D. palmatum Stephan leaf hexane fraction on human derived prostate cancer cell death. Methods : The dried leaves of D. palmatum were dissolved in methanol, and hexane fraction (DpLH) was again obtained from lyophilized methanol extract (DpLM). DpLH was investigated by measuring by MTT assay and annexin V/PI staining to evaluate its effects on the cell viability and apoptosis of PC-3 cells. The ROS generations were detected by DCF-DA dye. The protein expressions were confirmed by p-AKT, Bcl-2, Bax, procaspase-3 activities. Results : After treatment of DpLH to PC-3 cells, the cell proliferation was significantly inhibited, and in addition, DpLH treatment also accelerated apoptosis of PC-3 cells. When DpLH was treated to the PC-3 cells, its ROS production significantly decreased. The proportion of all proteins (p-AKT/actin, Bcl-2/Bax and procaspase-3/actin ratios) showed decreasing tendency of expression compared with the control group. Conclusions : As shown in the above results, the extract from D. palmatum inhibits ROS production and promotes cell death, which is considered to be a relatively safe induction of cell death when administered to a living body. In conclusion, these results suggested that DpLH may have anti-cancer effect in human prostate cancer cell.

사람 전립선암세포주인 LNCaP에서 셀레늄의 G1/S 세포주기억제에 관한 연구 (Selenium arrest G1/S phase of cell cycle in LNCaP human prostate cancer cells)

  • 우리 몸에 필수적인 미량원소인 셀레늄은 항암활성이 있는 것으로 알려져 있고, 역학적, 생태학적 그리고 임상시험을 종합해보면, 셀레늄은 종양, 특히 전립선암, 폐암, 대장암에 대한 위험성을 낮출 수 있을 것으로 생각되어진다. 기전연구에 따르면 셀레늄은 DNA single strand breaks와 활성산소종을 유도함으로써 종양을 억제하는 예방효능이 있는 것으로 밝혀지고 있다. 하지만, 아직까지 전립선암세포주에서 methylseloenol 대사체 중에 하나인 MSeA와 세포주기억제와의 관계에 대해서는 명확하게 밝혀져 있지 않은 실정이다. 따라서, 이번 연구에서는 전립선암세포주에서 셀레늄의 세포주기에 관한 효능을 확인해 보고자 하였다. 결과를 종합해보면, MSeA는 전립선암세포주에서 G1/S기 세포주기를 억제하고 DNA 합성을 방해하는 것으로 나타났고, 이러한 현상은 cyclin의존성 키나아제를 길항하는 단백질 중에 하나인 p27단백질의 발현을 증가시킴으로써 일어남을 확인하였다. 따라서, MSeA에 의해 유발되는 세포주기 억제가 전립선암 세포의 성장억제에 기여하는 것으로 사료된다.

인간 전립선암 PC-3 세포에서 Compound K에 의한 세포주기 조절 및 세포사멸 유전자 발현 변화 (Profile of Gene Expression Changes Treated with Compound K Induced Cell Cycle Arrest and Cell Death of Prostate Cancer PC-3 Cell Line)

  • Objectives : Previously, we reported that compound K isolated from fermented ginseng by Aspillus oryzae has a wide biochemical and pharmacological effect, including anti-cancer activity in prostate cancer PC-3 cells. Despite these findings, its signaling pathway and gene expression pattern are not clearly understood. Methods : To confirm the gene expression study of treated with compound K in PC-3 cells, a cDNA microarray chip composed of 44K human cDNA probes was used. MTT assay, western blot analysis, propidium iodide staining, and annexin V/propidium iodide staining were analyzed. Results : We confirmed the differences of gene expression profiles. Then, we analyzed with the cell cycle arrest, cell death and cell proliferation related genes using DAVID database. Conclusions : Our finding should be useful for understanding genome-wide expression patterns of compound K-mediated cell cycle arrest toward induction of cell death and be helpful for finding future cancer therapeutic targets for prostate cancer cells.

Bacterial Lipopolysaccharides Induce Steroid Sulfatase Expression and Cell Migration through IL-6 Pathway in Human Prostate Cancer Cells

  • Steroid sulfatase (STS) is responsible for the conversion of estrone sulfate to estrone that can stimulate growth in endocrine-dependent tumors such as prostate cancer. Although STS is considered as a therapeutic target for the estrogen-dependent diseases, cellular function of STS are still not clear. Previously, we found that tumor necrosis factor (TNF)-${\alpha}$ significantly enhances steroid sulfatase expression in PC-3 human prostate cancer cells through PI3K/Akt-dependent pathways. Here, we studied whether bacterial lipopolysaccharides (LPS) which are known to induce TNF-${\alpha}$ may increase STS expression. Treatment with LPS in PC-3 cells induced STS mRNA and protein in concentration- and time-dependent manners. Using luciferase reporter assay, we found that LPS enhanced STS promoter activity. Moreover, STS expression induced by LPS increased PC-3 tumor cell migration determined by wound healing assay. We investigated that LPS induced IL-6 expression and IL-6 increased STS expression. Taken together, these data strongly suggest that LPS induces STS expression through IL-6 pathway in human prostate cancer cells.

Up-regulating of RASD1 and Apoptosis of DU-145 Human Prostate Cancer Cells Induced by Formononetin in Vitro

  • Prostate cancer is one of the most prevalent malignant cancers in men. The isoflavone formononetin is a main active component of red clover plants. In the present study, we assessed the effect of formononetin on human prostate cancer DU-145 cells in vitro, and elucidated posssible mechanisms. DU-145 cells were treated with different concentrations of formononetin and cell proliferation was assessed by MTT assay, cell apoptosis by Hoechst 33258 and flow cytometry, and protein levels of RASD1, Bcl-2 and Bax by Western blotting. The results showed that formononetin inhibited the proliferation of DU-145 cells in a dose-dependent manner. DU-145 cells treated with different concentrations of formononetin displayed obvious morphological changes of apoptosis under fluorescence microscopy. In addition, formononetin increased the proportion of early apoptotic DU-145 cells, down-regulated the protein levels of Bcl-2 and up-regulated those of RASD1 and Bax. The level of RASD1 reached its maximum at 48h post-treatment, and rapidly decreased thereafter. Together, we present evidence that formononetin triggered cell apoptosis through the mitochondrial apoptotic pathway by up-regulating RASD1.

  • Prostate cancer is one of the most common non-skin cancers in men. Armeniacae Amarum Semen has traditionally been used for the treatment of inflammation diseases, leprosy, leucoderma, and tumors. Apoptosis, which is also known as programmed cell death, is an important mechanism in cancer treatment.Objectives : We observed whether an aqueous extract of Armeniacae Amarum Semen induces apoptotic cell death in human DU145 prostate cancer cells.Methods : We treated DU145 cells with Armeniacae Amarum Semen extract and investigated characteristics of apoptosis. And investigated whether treated with Armeniacae Amarum Semen extract increased Bax mRNA expression, Bcl-2 mRNA expression, caspase-3 enzyme activity and their protein level.Results : We have shown that Armeniacae Amarum Semen extract can induce apoptotic cell death in human DU145 prostate cancer cells by caspase-3 activation through the down-regulation on Bcl-2 expression and the up-regulation on Bax expression.Conclusions : It can be expected that an aqueous extract of Armeniacae Amarum Semen may offer a valuable means for the treatment of prostate cancers.

Orphan Nuclear Receptor Nurr1 as a Potential Novel Marker for Progression in Human Prostate Cancer

  • A number of studies have indicated that Nurr1, which belongs to a novel class of orphan nuclear receptors (the NR4A family), is important for carcinogenesis. Here we investigated expression of Nurr1 protein in benign and malignant human prostate tissues and association with clinicopathologic features using immunohistochemical techniques. Moreover, we also investigated the ability of Nurr1 to influence proliferation, migration, invasion and apoptosis of human prostate cancer cells using small interfering RNA silencing. Immunohistochemical analysis revealed that the expression of Nurr1 protein was higher in prostate cancer tissues than in benign prostate tissue (P<0.001), levels being positively correlated with tumor T classification (P = 0.003), N classification (P = 0.017), M classification (P = 0.011) and the Gleason score (P = 0.020) of prostate cancer patients. In vitro, silencing of endogenous Nurr1 attenuated cell proliferation, migration and invasion, and induced apoptosis of prostate cancer cells. These results suggest that Nurr1 may be used as an indicator for prostate cancer progression and be useful for novel potential therapeutic strategies.

Anti-tumor activities of Panax quinquefolius saponins and potential biomarkers in prostate cancer

  • Background: Prostate carcinoma is the second most common cancer among men worldwide. Developing new therapeutic approaches and diagnostic biomarkers for prostate cancer (PC) is a significant need. The Chinese herbal medicine Panax quinquefolius saponins (PQS) have been reported to show anti-tumor effects. We hypothesized that PQS exhibits anti-cancer activity in human PC cells and we aimed to search for novel biomarkers allowing early diagnosis of PC. Methods: We used the human PC cell line DU145 and the prostate epithelial cell line PNT2 to perform cell viability assays, flow cytometric analysis of the cell cycle, and FACS-based apoptosis assays. Microarray-based gene expression analysis was used to display specific gene expression patterns and to search for novel biomarkers. Western blot and quantitative real-time PCR were performed to demonstrate the expression levels of multiple cancer-related genes. Results: Our data showed that PQS inhibited the viability of DU145 cells and induced cell cycle arrest at the G1 phase. A significant decrease in DU145 cell invasion and migration were observed after 24 h treatment by PQS. PQS up-regulated the expression levels of p21, p53, TMEM79, ACOXL, ETV5, and SPINT1 while it down-regulated the expression levels of bcl2, STAT3, FANCD2, DRD2, and TMPRSS2. Conclusion: PQS promoted cells apoptosis and inhibited the proliferation of DU145 cells, which suggests that PQS may be effective for treating PC. TMEM79 and ACOXL were expressed significantly higher in PNT2 than in DU145 cells and could be novel biomarker candidates for PC diagnosis.
