Moderate Elevation of Extracellular $K^+$ Concentration Induces Vasorelaxation in Isolated Rat, Rabbit and Human Cerebral Arteries: Role of Na Pump and Ba-Sensitive Process

  • Cerebral blood vessels relax when extracellular $K^+$ concentrations $([K^+])_e$ are elevated moderately $(2{\sim}15$ mM, $K^+-induced$ vasorelaxation). We have therefore studied the underlying mechanism for this $K^+-induced$ vasorelaxation in the isolated middle cerebral arteries (MCAs). The effects of ouabain and $Ba^{2+}\;on\;K^+-induced$ vasorelaxation were examined to determine the role of sodium pump and/or Ba-sensitive process (possibly, inward rectifier K current) in the mechanism. Mulvany myograph was used to study 24 rats, 18 rabbits, and 10 humans MCAs $(216{\pm}3\;{\mu}m,\;347{\pm}7\;{\mu}m,\;and\;597{\pm}39\;{\mu}m$ in diameter when stretched to a tension equivalent to 55 mmHg). High $K^+$ (125 mM) and $PGF_{2{\alpha}}\;(1{\sim}10\;{\mu}M)$ induced concentration-dependent contractions in all 3 species, while histamine $(10{\sim}50\;{\mu}M)$ evoked contraction only in the rabbits and induced relaxation in the rats and humans. Addition of $K^+\;(2{\sim}10\;{\mu}M)$ to the control solution induced vasorelaxations. These effects were inhibited by the pretreatment with both ouabain $(10\;{\mu}M)$ and $Ba^{2+}\;(0.1{\sim}0.3\;mM)$ in the rat, but only with ouabain $(10\;{\mu}M)$ in the rabbit and human. These results suggest that $K^+-induced$ vasorelaxation occurs via the stimulation of electrogenic Na pump in the rabbit and human MCAs, while in the rat MCAs via the activation of both Na pump and Ba-sensitive process.

Production of Bovine Nuclear Transfer Embryos Using Fibroblasts Transfected with Single-Chain Human Follicle-Stimulating Hormone Gene

  • Human follicle-stimulating hormone (hFSH) is a pituitary glycoprotein that regulates follicular development and ovulation. Clinically, hFSH has been used to induce follicular growth in infertile women. The hormone is composed of heterodimers, including a common ${\alpha}$ subunit among the gonadotropin family and a hormone-specific ${\beta}$ subunit. Since assembly of the heterodimer is a rate-limiting step in the production of functional hFSH, transgenic clone cows carrying a single-chain hFSH transgene may efficiently produce functional hormone. Genes encoding the ${\alpha}$ and ${\beta}$ subunits of hFSH were linked using the C-terminal peptide sequence from the ${\beta}$ subunit of human chorionic gonadotropin. Bovine fetal fibroblasts were transfected with the gene construct, including the goat ${\beta}$-casein promoter and a single-chain hFSH coding sequence. Transfected fibroblasts were transferred into enucleated oocytes, and individual nuclear transfer (NT) embryos developed to the blastocyst stage were analyzed for the transgene by polymerase chain reaction. Seventy eight blastocysts (30.8%) were developed from 259 reconstructed embryos. Among these blastocysts, the hFSH gene was detected in 70.8% (34/48) of the embryos. Subsequent transfer of hFSH-transgenic clone embryos to 31 recipients results in 11 (35.5%) early pregnancies. However, all fetuses were lost before reaching day 180 of gestation. The results from this study demonstrated that bovine NT embryos carrying single-chain hFSH could be produced, and further extensive studies in which NT embryos are transferred to more recipients may give rise to single chain hFSH-transgenic cows for biomedical applications.

SQUID MEG Responses of the Human Auditory Cortex Generated by Acupuncture on GB43(Xiaxi) (협계혈(GB43) 자침시 침시술 방법에 따른 SQUID MEG 신호의 변화 관찰)

  • Using the 2-channel DROS SQUID (Korea Research Institute of Standards of Science, 1999), the present study was carried out to record changes elicited in the auditory cortex by acupuncture stimulus (right GB43, Xiaxi). Needle-retention and manual needle-twitching stimulation of GB43 and SP1 were done for acquiring the brain activities changed by acupuncture. Acupoint GB43 is known to be effective for the treatment of ear-related disease, such as deafness and tinnitus, and to be suspected to be related to the auditory cortex. Auditory evoked magnetic fields were recorded from the left hemisphere of five or four subjects, in response to contralateral ear stimulation by irregularly spaced 170msec long 1kHz tone busts (Korea Research Institute of Standards of Science). The result as follows The latency and amplitude of SQUID MEG responses at the human auditory cortex changed by needle-retention condition on GB43 were 7.2msec and 1.617, respectively, which were slower and larger than those of no-acupuncture condition. The amplitude of SQUID MEG responses at the human auditory cortex changed by needle-twitching condition on GB43 was 13.517, which was larger than that of no-acupuncture condition. The change in SP1 following GB43 needle-twitching condition were not observed in latency. The amplitude changed by needle-twitching condition on SP1 was 12.2fT, which was not significant. These results suggested that auditory cortex can be affected by acupuncture stimulus, though not specific or significant because of small number of subjects.

3,3',4,4'-tetrachlorobiphenyl (PCB77) enhances human Kv1.3 channel currents and alters cytokine production

  • Polychlorinated biphenyls (PCBs) were once used throughout various industries; however, because of their persistence in the environment, exposure remains a global threat to the environment and human health. The Kv1.3 and Kv1.5 channels have been implicated in the immunotoxicity and cardiotoxicity of PCBs, respectively. We determined whether 3,3',4,4'-tetrachlorobiphenyl (PCB77), a dioxin-like PCB, alters human Kv1.3 and Kv1.5 currents using the Xenopus oocyte expression system. Exposure to 10 nM PCB77 for 15 min enhanced the Kv1.3 current by approximately 30.6%, whereas PCB77 did not affect the Kv1.5 current at concentrations up to 10 nM. This increase in the Kv1.3 current was associated with slower activation and inactivation kinetics as well as right-shifting of the steady-state activation curve. Pretreatment with PCB77 significantly suppressed tumor necrosis factor-α and interleukin-10 production in lipopolysaccharide-stimulated Raw264.7 macrophages. Overall, these data suggest that acute exposure to trace concentrations of PCB77 impairs immune function, possibly by enhancing Kv1.3 currents.

Korean Red Ginseng extract ameliorates melanogenesis in humans and induces antiphotoaging effects in ultraviolet B-irradiated hairless mice

  • Background: Panax ginseng is a marvelous herbal remedy for all ailments of body. That may be why it is called Panax, which means "cure for all". Melanin is a pigment that gives color to our skin; however, increased melanin production can lead to tumor formation. Human exposure to ultraviolet B radiation has increased extensively owing to the increased sunlight due to global warming. Consequently, a phenomenon called photoaging has been observed for all skin colors and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases, which serve as degradation enzymes for extracellular matrix proteins, mainly collagen, is increased, causing depletion of collagen and resulting in early wrinkle formation. Methods: Therefore, in our study, we used the murine melanoma cell line B16/F10 to study the inhibition of melanogenesis by Korean Red Ginseng (KRG) extract in vitro and HRM-2 hairless mice exposed to artificial ultraviolet B to examine the efficacy of KRG in vivo. We prepared a 3% red ginseng extract cream and evaluated its effects on human skin. Results: Our results demonstrated that KRG induced potent suppression of tyrosinase activity and melanin production in B16/F10 cells; moreover, it reduced the transcription and translation of components involved in the melanin production pathway. In the in vivo experiments, KRG potently suppressed the expression of matrix metalloproteinases, reduced wrinkle formation, and inhibited collagen degradation. On human skin, ginseng cream increased skin resilience and skin moisture and enhanced skin tone. Conclusion: Therefore, we conclude that KRG is an excellent skin whitening and antiaging product.

  • Shenming(神明) exists everywhere in the universe; it affects human beings to feel and contact ideal and peaceful state of Shen(神). Shenming theory(神明思想) in ${\ulcorner}$Hwang Ti Nei Ching${\lrcorner}$ (黃帝內經) was established by ancient Northeastern Asians who were trying to apply paradigm called TianRenHeYi theory(天人合-思想), unifying man and the Great Spirit(絶對精神) of the universe. Based on present, Shenming(神明) is an idea that can estimate will of Shen(神) which has a key to the future; such concept is founded by both human's mind and action that are designed to seek for the uncertain future. Very pure and devoted mind can sympathize with Shen(神) to accept Shenming(神明). Sehnming theory(神明思想) in ${\ulcorner}$Hwang Ti Nei Ching${\lrcorner}$ (黃帝內經) has a basic concept of living balanced life as a human by accepting Shenming(神明), the Spirit of Universe. Thus, it can be summarized into a phrase, 'Shenming(神明) is a process that human conjures his ultimate destiny called the Great Spirit(絶對精神) in himself.' If a man would be guided by Shenming(神明), the Spirit of Universe, he can live peacefully. Therefore, it is essential to study Shenming theory(神明思想) in ${\ulcorner}$Hwang Ti Nei Ching${\lrcorner}$ (黃帝內經) to aid present people's mental health.

Effects of Pharmacological Modulators of $Ca^{2+}-activated\;K^+$ Channels on Proliferation of Human Dermal Fibroblast

  • Employing electrophysiological and cell proliferation assay techniques, we studied the effects of $Ca^{2+}$ -activated $K^+$ channel modulators on the proliferation of human dermal fibroblasts, which is important in wound healing. Macroscopic voltage-dependent outward $K^+$ currents were found at about -40 mV stepped from a holding potential of -70 mV. The amplitude of $K^+$ current was increased by NS1619, a specific large-conductance $Ca^{2+}$-activated $K^+$ (BK) channel activator, but decreased by iberiotoxin (IBTX), a specific BK channel inhibitor. To investigate the presence of an intermediate-conductance $Ca^{2+}$-activated $K^+$ (IK) channels, we pretreated the fibroblasts with low dose of TEA to block BK currents, and added 1-EBIO (an IK activator). 1-EBIO recovered the currents inhibited by TEA. When various $Ca^{2+}$-activated $K^+$ channel modulators were added into culture media for 1∼3 days, NS1619 or 1-EBIO inhibited the cell proliferation. On the other hand, IBTX, clotrimazole or apamin, a small conductance $Ca^{2+}$-activated $K^+$ channel (SK) inhibitor, increased it. These results suggest that BK, IK, and SK channels might be involved in the proliferation of human dermal fibroblasts, which is inversely related to the channel activation.

TRPV1 in Salivary Gland Epithelial Cells Is Not Involved in Salivary Secretion via Transcellular Pathway

  • Transient receptor potential vanilloid subtype 1 (TRPV1) was originally found in sensory neurons. Recently, it has been reported that TRPV1 is expressed in salivary gland epithelial cells (SGEC). However, the physiological role of TRPV1 in salivary secretion remains to be elucidated. We found that TRPV1 is expressed in mouse and human submandibular glands (SMG) and HSG cells, originated from human submandibular gland ducts at both mRNA and protein levels. However, capsaicin (CAP), TRPV1 agonist, had little effect on intracellular free calcium concentration ($[Ca^{2+}]_i$) in these cells, although carbachol consistently increased $[Ca^{2+}]_i$. Exposure of cells to high temperature (> $43^{\circ}C$) or acidic bath solution (pH5.4) did not increase $[Ca^{2+}]_i$, either. We further examined the role of TRPV1 in salivary secretion using TRPV1 knock-out mice. There was no significant difference in the pilocarpine (PILO)-induced salivary flow rate between wild-type and TRPV1 knock-out mice. Saliva flow rate also showed insignificant change in the mice treated with PILO plus CAP compared with that in mice treated with PILO alone. Taken together, our results suggest that although TRPV1 is expressed in SGEC, it appears not to play any direct roles in saliva secretion via transcellular pathway.

Mechanisms of Siegesbeckia Glabrescens-induced Smooth Muscle Cell Apoptosis: Role of iNOS and PKC${\alpha}$ (희첨의 iNOS 발현과 PKC${\alpha}$ 억제를 통한 혈관평활근세포의 apoptosis 유도)

  • We have recently demonstrated that Siegesbeckia glabrescens(SG), a herbal medicine, induces apoptosis via nitric oxide(NO) production in human aortic smooth muscle cells(HASMCS). However, the molecular pathways involved in SG-mediated apoptosis are not fully understand. In the present study, we investigated the cellular mechanisms of SG-induced apoptosis in HASMCS. SG induced NO production through inducible nitric oxide synthase(iNOS) induction. The apoptotic effect of SG was attenuated by L-NNA, a NOS inhibitor. In the presence of L-NNA, the degradation of procaspase-3 by SG was inhibited. SG treatment induced a decrease in Bcl-2 expression but did not affect the expression of Bax. In addition, SG treatment evoked both down-regulation of PKC ${\alpha}$ and inhibition of PKC ${\alpha}$ phosphorylation. These downregulations were reversed by addition of L-NNA. It seems likely to De a downregulation of PKC${\alpha}$ due to long term treatment with PMA. Taken together, these results suggest that apoptotic effects of SG may be due to NO production via iNOS mRNA expression. Furthermore, Bcl-2 and PKC${\alpha}$ downregulation, and caspase-3 activation may be involved in the mechanisms for apoptotic effects by SG.
