A total of 60 samples were collected from 35 swimming pools in Beijing, China, and the presence of Cryptosporidium and Giardia were investigated. The results showed that 16.7% and 15.0% of samples were positive for Cryptosporidium oocyst and Giardia cysts, respectively, with a mean concentration of 0.30 oocysts/10 L and 0.27 cysts/10 L. The oocysts and cysts were found to have higher rates of occurrence in August than in May. Genotyping confirmed the presence of Cryptosporidium hominis, C. parvum, and Giardia assemblages A and B, all of which were associated with human infections. The predominant species/assemblages were C. hominis and Giardia assemblage A. Analyses of the relationships between parasite oocysts/cysts, indicator bacteria, and physical-chemical parameters revealed that there was no correlation between 2 parasites and fecal bacterial indicators, whilst there was a significant correlation between protozoa and urea concentration, which indicates that urea concentration rather than fecal bacterial indicators might be an appropriate index for chlorine-resistant protozoa in swimming pools. This study provides useful information to improve the safety of swimming pool water and deduce the risk of protozoan infections.

The genus Sarcocystis is not usually considered as an important enteric pathogen in immune compromised patients. It might be expected that species for which humans are the final host (Sarcocystis hominis and Sarcocystis suihominis as well as possibly others) would be encountered increasingly often in immunodeficient persons. This study aimed to address how to detect and differentiate Sarcocystis oocysts and/or sporocysts from enteric protozoans in the diarrheal samples of immunodeficient patients in Shiraz, Iran. Diarrheal samples of 741 immunodeficient patients with recurrent persistent or chronic diarrhea were examined by microscopy and molecular biological analysis. Oocysts-positive samples were 68 Cryptosporidium spp., 9 Cystoisospora belli (syn. Isospora belli), 2 Cyclospora cayetanensis, and 15 microsporidia (Enterocytozoon bieneusi). Sarcocystis-like sporocysts found from a woman were identified as Sarcocystis cruzi through 18S rDNA amplification and phylogenetic analysis. To the best of our knowledge, this is the first report of S. cruzi from a human.

Background: Intestinal parasites in livestock cause huge economic setbacks. Moreover, these parasites can threaten human health when also present in companion animals. Objectives: The study examined the prevalence and burden of intestinal parasites among roaming/migrating animals (goats, sheep, cattle, and dogs) to provide insights into the risk of potential human parasitic infections. Methods: A total of 1,741 fecal samples from goats (n = 920), sheep (n = 335), cattle (n = 230) and dogs (n = 256) were obtained randomly across 18 local government areas in Edo State, Nigeria. The parasite samples were recovered and identified under a microscope. Molecular tools were used to identify Toxocara spp. Results: Eighteen different parasites were isolated. Among the different groups of parasites observed, nematodes occurred the most, followed by protozoans. Among nematodes, Haemonchus was most prevalent in goats (28.04%) and sheep (29.85%), while Strongyloides (10.86%) and Bunostomum (8.69%) were relatively high in cattle. Strongyloides (3.9%), hookworm (3.9%), and Toxocara (3.12%) were the predominant parasites in dogs. For protozoan parasites, Eimeria was most common in all 4 animal hosts. Several goats (2.39%) and sheep (2.38%) tested positive for Fasciola spp. Molecular analysis confirmed Toxocara canis in dogs for the first time in Nigeria. Conclusions: The major parasites recovered from these roaming/migrating animals have zoonotic potentials that can threaten human health.
This study was conducted in order to establish an in vitro method simulating the physiological function of fibers along the large intestine of humans. Commercial fibers including guar gum, apple pectin, citrus pectin, CM-cellulose, alginic acid and $\alpha$-cellulose, and dietary fiber residues obtained from rice bran, barley, soybean, Korea cabbage, apple, tangerine and sea mustard were employed to determine the water-holding capacity, weight of hydrated residue and fiber content after anaerobic fermentation using human fecal inoculum for 24 hours, followed by dialysis under osmotic suction pressure. The weight of hydrated residue in commercial fibers was in the decreasing order of CM-cellulose > alginic acid, $\alpha$-cellulose > apple pectin, citrus pectin > guar gum and that in food fiber residues was in the decreasing order of rice bran, sea mustard > soybean > tangerine, Korean cabbage > barley > apple. It was demonstrated that the larger the weight of hydrated residue was, the more the weight of human stool increased. Consequently this in vitro method can be used as a preceeding test before undertaking animal or human experiment to predict the physiological effects of fiber residues from diverse food samples as well as commercially refined fibers.

We monitored the occurrence of human enteric viruses in urban rivers by cell culture-PCR and RT-nested PCR. Water samples were collected monthly or semimonthly between May 2002 and March 2003 in four urban tributaries. Enteric viruses were detected by RT-nested PCR and cell culture-PCR based on a combination of Buffalo Green monkey kidney (BGMK) and A549 cell lines, followed by phylogenetic analysis of amplicons. By RT-nested PCR analysis, 45 (77.6%), 32 (55.2%), 32 (55.2%), 26 (44.8%), 12 (20.7%), 2 (3.4%), 4 (6.9%), and 4 (6.9%) of 58 samples showed positive results with adenoviruses, enteroviruses, noroviruses (NV) genogroup I (GI) and II (GII), reoviruses, hepatitis A viruses, rotaviruses and sapoviruses, respectively. Adenoviruses were most often detected and only eight (13.8%) samples were negative for adenoviruses and positive for other enteric viruses in the studied sites. Thirty-one (77.5%) of the 40 samples were positive for infectious adenoviruses and/or enteroviruses based on cell culture-PCR, and the frequency of positive samples grown on A549 and BGMK (65.0%) was higher than that grown on BGMK alone (47.5%). The occurrence of each enteric virus, except reoviruses and hepatitis A viruses was not statistically correlated with the water temperature and levels of fecal coliforms according to Binary logistic regression model. By sequence analysis, most strains of adenoviruses and enteroviruses detected in this study are similar to the causative agent of viral diseases in Korea and most NV GI- and GII-grouped strains were closely related to the reference strains from China and Japan, and GII/4-related strains had similar sequences to strains recognized as a worldwide epidemic outbreak. Our results suggested that monitoring human enteric viruses is necessary to improve microbial quality and cell culture-PCR using the combination of A549 and BGMK cells and the adenovirus detection by PCR could be useful for monitoring viral contamination in the aquatic environment.
The objective of this study was to compare fecal microbes survival in soil between compost surface application and soil incorporation. The survival experiment was conducted in six styrofoam beds ($510{\times}325{\times}305(mm)$ in size) filled with sandy loam soil. A half of six boxes were received by compost surface application, while the other half were treated with compost-soil mixture. Duplicated surface and surbsurface soil (20 cm depth) samples were collected at various interval up to 50 days and analyzed for the determination of fecal coliforms and E. coli numbers. As expected, surface applied beds demonstrated two to three magnitudes order greater in both the study microorganisms as compared to soil incorporated beds. Microbial inactivation rate of soil surface was twice as great as subsurface soil condition probably due to exposure to sun light and environmental conditions including moisture loss. When rainfall occurred, microbes on the surface were transported into soil along with water movement. It was concluded that surface compost application may be easier to apply but pose higher risk of human exposure to microbes. Winter compost application may be favorable in alleviating health risk by giving some time for inactivation compared to spring application.
Non-human primates (NHPs) are confirmed as reservoirs of Cryptosporidium spp., Giardia intestinalis, and Enterocytozoon bieneusi. In this study, 197 fresh fecal samples from 8 NHP species in Qinling Mountains, northwestern China, were collected and examined using multilocus sequence typing (MLST) method. The results showed that 35 (17.8%) samples were positive for tested parasites, including Cryptosporidium spp. (3.0%), G. intestinalis (2.0%), and E. bieneusi (12.7%). Cryptosporidium spp. were detected in 6 fecal samples of Macaca mulatta, and were identified as C. parvum (n=1) and C. andersoni (n=5). Subtyping analysis showed Cryptosporidium spp. belonged to the C. andersoni MLST subtype (A4, A4, A4, and A1) and C. parvum 60 kDa glycoprotein (gp60) subtype IId A15G2R1. G. intestinalis assemblage E was detected in 3 M. mulatta and 1 Saimiri sciureus. Intra-variations were observed at the triose phosphate isomerase (tpi), beta giardin (bg), and glutamate dehydrogenase (gdh) loci, with 3, 1, and 2 new subtypes found in respective locus. E. bieneusi was found in Cercopithecus neglectus (25.0%), Papio hamadrayas (16.7%), M. mulatta (16.3%), S. sciureus (10%), and Rhinopithecus roxellana (9.5%), with 5 ribosomal internal transcribed spacer (ITS) genotypes: 2 known genotypes (D and BEB6) and 3 novel genotypes (MH, XH, and BSH). These findings indicated the presence of zoonotic potential of Cryptosporidium spp. and E. bieneusi in NHPs in Qinling Mountains. This is the first report of C. andersoni in NHPs. The present study provided basic information for control of cryptosporidiosis, giardiasis, and microsporidiosis in human and animals in this area.
목 적: Human astrovirus (HAstV)는 전세계적으로 영아 연령에서 중요한 위장관염의 중요 병원체로 알려지고 있지만 국내에서 HAtV 감염의 유병률에 대한 연구는 드문 편이다. 이에 저자들은 분자 유전학적 방법을 이용하여 위장관염으로 입원한 5세 이하의 소아에서 HAstV의 유병률과 유전형을 알아보기 위하여 본 연구를 시행하였다. 방 법: 2004년 2월부터 2005년 1월까지 12개월간 인제의대 상계백병원 소아과에 급성 위장관염으로 입원한 5세 이하의 소아에게서 수집된 대변 검체 812건을 대상으로 하였다. 대변 검체에 대해서 enzymeimmunoassay(EIA) 또는 RT-PCR을 이용하여 rotavirus, enteric adenovirus와 norovirus 감염여부를 진단하였다. HAstV 특이적인 시발체를 이용하여 RTPCR을 시행한 다음 염기서열 분석을 시행하였다. 결 과: 총 812건의 검체에서 rotavirus 양성률은 16.9% (138/812), norovirus 양성률은 11.5% (94/812), adenovirus 양성률은 4.0% (33/812)였다. HAstV의 유병률은 RT-PCR 방법에 의해 4.0% (33/812)였다. HAstV 양성인 환아의 연령은 생후 12개월 미만 82.0%, 1~2세 미만 6.0%, 2~3세 미만 6.0%, 3~5세 이하가 6.0%였다. HAstV의 월별 분포는 각각 4월 3건, 5월 5건, 6월 4건, 8월 12건, 9월 4건, 10월 2건, 11월 2건, 12월 1건으로 8월에 가장 높은 발생률을 보였다. 염기서열 분석이 가능하였던 HAstV 양성 검체 22건 중에서 유전형-1이 20건, 유전형-8이 2건이었다. 결 론: 2004년 2월부터 2005년 1월까지 12개월간 급성 위장관염으로 입원한 소아에서 HAstV 감염 유병률은 4.0%였으며, 영아기 연령에서 주로 발생하였다. 국내 HAstV 감염의 정확한 역학적 특성을 파악하기 위해서는 장기간에 걸친 추가 연구가 필요할 것으로 생각된다.
Saccharomyces cerevisiae (S. cerevisiae) and glucagon-like peptide-2 (GLP-2) have been employed to improve the intestinal development of weaned animals. The goal of this study was to determine whether either exogenous S. cerevisiae or GLP-2 elicits major effects on fecal microbiotas and cytokine responses in weaned piglets. Ninety-six piglets weaned at 26 days were assigned to one of four groups: 1) Basal diet (Control), 2) empty vector-harboring S. cerevisiae (EV-SC), 3) GLP-2-expressing S. cerevisiae (GLP2-SC), and 4) recombinant human GLP-2 (rh-GLP2). At the start of the post-weaning period (day 0), and at day 28, fecal samples were collected to assess the bacterial communities via sequencing the V1-V2 region of the 16S-rRNA gene, and piglets' blood was also sampled to measure cytokine responses (i.e., IL-$1{\beta}$, TNF-${\alpha}$, and IFN-${\gamma}$). This study revealed that, on the one hand, although S. cerevisiae supplementation did not significantly alter the growth of weaned piglets, it induced increases in the relative abundances of two core genera (Ruminococcaceae_norank and Erysipelotrichaceae_norank) and decreases in the relative abundances of two other core genera (Lachnospiraceae_norank and Clostridiale_norank) and cytokine levels (IL-$1{\beta}$ and TNF-${\alpha}$) (p < 0.05, Control vs EV-SC; p < 0.05, rh-GLP2 vs GLP2-SC). On the other hand, GLP-2 supplementation had no significant influence on fecal bacterial communities and cytokine levels, but it produced better body weight and average daily gain (p < 0.05, Control vs EV-SC; p < 0.05, rh-GLP2 vs GLP2-SC). Therefore, altered fecal microbiotas and cytokine response effects in weaned piglets were due to S. cerevisiae rather than GLP-2.
Intestinal parasitic infections remain a serious public health problem globally and have usually been associated with human malnutrition. This study was performed to observe the present status of intestinal helminthes infections among the primary school children in Iksan, Korea, during the period from June to August, 2006. A total of 974 fecal samples (male 479, female 495) were collected and examined by formalin-ether sedimentation technique for intestinal helminthes eggs. Of the 974 samples examined, 2 (0.2%) were egg positive for intestinal helminthes, and only eggs of Clonorchis sinensis were observed in the 2 cases. These C. sinensis egg were found in grade 5 (male, 1 case) and grade 6 (female, 1 case), respectively. The egg positive rate of male and female were 0.2%, respectively. Through this survey, I found that the C. sinensis infection is prevalent among primary school children in Iksan, Jeonbuk, and a continuous health education for school children is recommended to prevent the potential infection of C. sinensis.
