Leukotriene B4 receptors contribute to house dust mite-induced eosinophilic airway inflammation via TH2 cytokine production

  • Leukotriene B4 (LTB4) is a lipid mediator of inflammation that is generated from arachidonic acid via the 5-lipoxygenase pathway. Previous studies have reported that the receptors of LTB4, BLT1, and BLT2 play mediatory roles in the allergic airway inflammation induced by ovalbumin (OVA). However, considering that house dust mites (HDMs) are the most prevalent allergen and well-known risk factor for asthmatic allergies, we are interested in elucidating the contributory roles of BLT1/2 in HDM-induced allergic airway inflammation. Our aim in this study was to investigate whether BLT1/2 play any roles in HDM-induced allergic airway inflammation. In this study, we observed that the levels of ligands for BLT1/2 [LTB4 and 12(S)-HETE (12(S)-hydroxyeicosatetraenoic acid)] were significantly increased in bronchoalveolar lavage fluid (BALF) after HDM challenge. Blockade of BLT1 or BLT2 as well as of 5-lipoxygenase (5-LO) or 12-lipoxygenase (12-LO) markedly suppressed the production of TH2 cytokines (IL-4, IL-5, and IL-13) and alleviated lung inflammation and mucus secretion in an HDM-induced eosinophilic airway-inflammation mouse model. Together, these results indicate that the 5-/12-LO-BLT1/2 cascade plays a role in HDM-induced airway inflammation by mediating the production of TH2 cytokines. Our findings suggest that BLT1/2 may be a potential therapeutic target for patients with HDM-induced allergic asthma.

Efficacy of Modified Rush Allergen-Specific Immunotherapy on Canine Atopic Dermatitis

  • Modified rush ASIT protocol has been performed to identify the ideal schedule that allows the dose considered effective to be reached in the shortest possible time with the fewest adverse effects. Ten atopic dogs of this study includes fulfillment of Favrot's criteria. Offending allergens were identified by the use of IDST. During the induction period, the dogs were received a total of 15 injections. Ten injections were administrated every 30 minutes in a day with gradually increasing amounts and concentrations of allergens, and the last 5 injections were administered every 3 days. Disease severity was quantified by using the canine atopic dermatitis extent and severity index (CADESI). During induction period, reduction rate from baseline scores varied between 1% and 67% and the improvement of ${\geq}50%$ was recorded after induction period of therapy for CADESI-03 score in 6 of the 10 dogs. This study of ten dogs with atopic dermatitis provide evidence for the efficacy and safety of modified rush ASIT for clinical improvement.

Effect of Zedoariae rhizoma on Bronchial Inflammation and Allergic Asthma in Mice

  • There are detailed descriptions of the clinical experiences and prescriptions of asthma in traditional Korean medicine. Zedoariae rhizoma is one of the Korean herbal medicines used to treat bronchial asthma and allergic rhinitis for centuries. However, the therapeutic mechanisms of this medication are still far from clear, In this study, a house-dust-mite (Dermatophagoides pteronyssinus [Der p])-sensitized murine model of asthma was used to evaluate the immunomodulatory effect of Zedoariae rhizoma on the allergen-induced airway inflammation in asthma. Three different protocols were designed to evaluate the treatment and/or long-term prophylacitic effect of Zedoariae rhizoma in Der p-sensitized mice. Cellular infiltration and T-cell subsets in the bronchoalveolar lavage fluid (BALF)of allergen-challenged mice were analyzed. Intrapulmonary lymphocytes were also isolated to evaluate their response to allergen stimulation. When Zedoariae rhizoma was administered to the sensitized mice before AC (groups A and C), it suppressed airway inflammation by decreasing the number of total cells and eosinophil infiltration in the BALF, and downregulated the allergen- or mitogen-induced intrapulmonary lymphocyte response of sensitized mice as compared to those of controls. This immunomodulatory effect of Zedoariae rhizoma may be exerted through the regulation of T-cell subsets by elevation or activation of the CD8+ and double-negative T-cell population in the lung. However, the administration of Zedoariae rhizoma to sensitized mice 24 h after AC (group B) did not have the same inhibitory effect on the airway inflammation as Zedoariae rhizoma given before AC. Thus, the administration of Zedoariae rhizoma before AC has the immunomodulatory effect of reducing bronchial inflammation in the allergen-sensitized mice. On the other hand, to determine the potentiality of prophylactic and/or therapeutic approaches using a traditional herbal medicine, Zedoariae rhizoma, for the control of allergic disease, we examined the effects of oral administration of Zedoariae rhizoma on a murine model of asthma allergic responses. When oral administration of Zedoariae rhizoma was begun at the induction phase immediately after OVA sensitization, eosinophilia and Th2-type cytokine production in the airway were reduced in OVA-sensitized mice following OVA inhalation. These results suggest that the oral administration of Zedoariae rhizoma dichotomously modulates allergic inflammation in murine model for asthma, thus offering a different approach for the treatment of allergic disorders.

The Sensitization Rates for Inhaled Allergens by Skin Prick Test among Some Farmers in Gyeonggi Province, South Korea (피부 단자 검사로 평가한 경기도 일부 농업인의 흡입 알레르겐 감작률)

  • Objectives: Farmers are known to be exposed to a variety of allergens related to the work environment. This study was conducted to determine the sensitization rates as well as South Korea that they are sensitized to certain allergens farmers through the skin prick test. Methods: By targeting a total of 1143 people living in the rural town of Gyeonggi Province, it was conducted a questionnaire containing demographic and occupational risk factors and underwent skin prick tests with 15 types of allergens(including positive and negative controls). Multivariable logistic regression analysis was used to analyze the association between occupational risk factors and skin prick test positivity. Results: Except for the 30 people whose result is invalid, positive rate of the skin prick test was 18.6% in 1,113 people. The species of house dust mite, Dermatophagoides pteronyssinus and Dermatophagoides farinae was the highest at 8.7% and 8.6%. After adjusted by age, gender, smoking and education level, odds ratio of flower plant farmers is 4.467(95% CI: 2.094-9.527) and fruit farmer is 2.275(95% CI: 1.096-4.721). In addition, the rate of sensitization to grass pollen mixture of the flower plant farmers is significantly higher(15.9%, p<0.001) than other allergens. Conclusions: Even farmers, the rate of sensitization to allergens related to the general environment, such as house dust mite is relatively dominant. However, given the presence of potential cross-reactivity between the allergens or distribution showed that the unique aspects of allergen sensitization in the flower growers, occupational cause is not be completely ruled out.

The Effect of a Cooperative System of Oriental and Western Medicine in the Treatment of Allergic Rhinitis

  • Background and Purpose : Allergic rhinitis is a well-known, relatively controllable chronic disease. Although a number of methods for treating allergic rhinitis have been tried, many patients have not been satisfied with their treatment. Therefore, this study tried to evaluate the effect of a cooperative system of Oriental and Western medicine and to develop a new diagnosis protocol for treatment of allergic rhinitis. Methods : We measured improvement rate and acoustic rhinometry after the allergeninduction test and performed a filter paper test as a nonspecific hypersensitivity test with 60 patients who are allergic to house dust mite. Patients were divided into two groups, one treated with Western medicine only and one treated with both Western and Oriental herbal medicine. For the group with Western medicine only, antihistamine for one week and local steroid medicine for two weeks were prescribed. For the group with combined medicine, Oriental herbal medicine was prescribed according to the patient s constitution, along with Western medicine. After all treatments, the above tests were re-performed and the improvement rate was compared. Results and Conclusion : We observed better results in the group treated with both Western and Oriental herbal medicines, comparing improvement rate and the alteration of total nasal volume through acoustic rhinometry after the allergen induction test. In the filter paper test, there was no significant difference between the two groups. In conclusion, we showed the additive effect of Oriental herbal medicine without any severe side effects compared with treatment with Western medicine only. In this study, we set only two patient groups, but further study is required to create various experimental groups and compare among them. We suggest that it might enhance understanding of the improved effect of Oriental herbal medicine in the therapy of allergic rhinitis.

Suppressive Effect of Der p 2 on Constitutive Neutrophil Apoptosis by Cytokine Secretion of Normal and Allergic Lymphocytes (정상인과 알레르기 환자의 림프구에서 Der p 2에 의한 사이토카인 분비가 호중구의 세포고사 억제에 미치는 효과)

  • Der p 2 is the major allergen of the house dust mite (HDM) associated with the development of allergic diseases. The pathogenic mechanism of the allergy is related to cytokine release of lymphocytes and constitutive apoptosis of neutrophils. In the present study, we examined whether Der p 2 induces cytokine release of lymphocytes, which is involved in regulation of neutrophil apoptosis. In normal and allergic subjects, Der p 2 enhanced the secretion of IL-6, IL-8, MCP-1, and GM-CSF in a time-dependent manner. Although Der p 2 was weakly effective against neutrophil apoptosis, conditioned media collected from normal and allergic lymphocytes after Der p 2 treatment inhibited the apoptosis of normal and allergic neutrophils. Der p 2 showed stronger inhibition of apoptosis of allergic neutrophils cocultured with allergic lymphocytes than normal neutrophils cocultured with normal lymphocytes. These findings improve our understanding of the role of Der p 2 in regulation of lymphocytes and neutrophils and will enable elucidation of allergy pathogenesis.

Changes of Causative Allergen in Patients with Allergic Diseases in Chonbuk Province (전북지역 알레르기 환자에서 원인 알레르겐의 변화)

  • Background: The prevalence of allergic disease has increased substantially over the past decades. The reasons for these trends are complex but include increased environmental allergen exposure. Environmental control measures are of particular importance in the prevention and management of allergic disease. Thus, we purposed to evaluate the changes of causative allergens in allergic patients in Chonbuk provincial area that began industralized recently. Method: To evaluate the allergens epidemiologically, we performed skin prick tests and RAST in 201 allergic patients from February, 1984 to February, 1985 and 339 patients from February, 1997 to February, 1998. Results: The Following results were obtained. 1) Total sensitization rates did not change significantly(74.2% vs 75.8%, p=0.664). 2) Positive reaction rates to pollens were similar(35.8% vs 30.1%, p=0.17). 3) Positive rates to HDM(house dust mite) were decreased(66.1% vs 56.3%, p=0.02). 4) Positive rates to fungi and animal danders were decreased significantly(p<0.001). Conclusion : Environmental allergens associated with hygine in Chonbuk provincial areas such as HDM and fungi are decreased significantly during 13 years.

  • Background : RANTES is associated with chemotaxis and activation of eosinophils. RANTES is up-regulated in allegic inflammation and play a critical role in the pathogenesis of allegic inflammation. Recently, circulating form of RANTES have been identified in the peripheral blood. Method : In the present study, we measured soluble RANTES levels in 17 patients with atopic brochial asthma (8 patients: early response to allegen challenge, 8 patients : early and late response to allergen challenge) on 30mins, 2hrs and 8 hrs after allergen challenge with house dust mite, prechallenge period. Result : RANTES levels in sera from patients with bronchial astma in prechallenge conditions were higher than in normal control subjects. But, RANTES levels in sera from patients with bronchial asthma in 30mins, 2hrs and 8hrs after challenge were no significantly higher than prechallenge conditions. Conclusion : These results suggest that RANTES plays a role in the pathogenesis of patients with atopic bronchial asthma and may be related to persistence of subclinical allergic inflammation.

S100A8 and S100A9 Secreted by Allergens in Monocytes Inhibit Spontaneous Apoptosis of Normal and Asthmatic Neutrophils via the Lyn/Akt/ERK Pathway (단구에서 분비되는 S100A8과 S100A9의 Lyn/Akt/ERK 경로를 통한 정상인과 천식질환 호중구의 세포고사 억제 효과)

  • Der p 1 and Der p 2 are essential allergens of house dust mite associated with the development of asthma. In the present study, we examined whether Der p 1 and Der p 2 induce a release of S100A8 and S100A9 in monocytes, which are involved in the regulatory mechanism of neutrophil apoptosis. We found that Der p 1 and Der p 2 significantly increased the secretion of S100A8 and S100A9 in normal monocytes. Moreover, S100A8 and S100A9 strongly suppressed the spontaneous apoptosis of normal and asthmatic neutrophils. The inhibitory effect of S100A9 was stronger than that of S100A8, and asthmatic neutrophils showed a higher inhibitory effect than normal neutrophils. S100A8 and S100A9 induced activation of Lyn, Akt, and ERK in a time-dependent manner. These findings elucidate the roles of Der p 1 and Der p 2 in the interaction between monocytes and neutrophils, as well as contributing to our knowledge of the pathogenesis of allergic diseases.
