Morphological Observation of Pre- and Postovulated Eggs from Artificially Maturated Wild and Feminized Eels (성성숙 유도된 자연산과 자성화 뱀장어의 채란 전·후 난의 형태학적 관찰)

  • This study about morphological observation of pre- or post ovulated eggs obtained from artificially maturated female eels. Female eels were divided with two groups as wild eels from nature and feminized eels from farm. Artificial maturation had been conducted with the established methods in this laboratory, and then maturate eggs sampled at fixed 3 times and these were observed with stereomicroscope. Rate of increased body weight (RIW) were measured with 2 times. Egg diameters and development of oil droplets are determined for standardization of egg maturation degree, and the transparency of egg cytoplasm and the homogeneous degrees of egg size were referred to determine of egg quality. Rate of increased body weight (RIW) were good in range about 10 % at final salmon pituitary extracts (SPE) injection time and in range about 20 % at $17{\alpha}$, $20{\beta}$-dihydroxy-4-pregnen-3-one (DHP) injection time. Good matured egg for fertilization had $900-1000{\mu}m$ diameter, and they had about 50 oil droplets in size about $100{\mu}m$ diameter. There were not predominant differences at egg size and oil droplet development between wild female and feminized eels.

High Ductile Fiber Reinforced Concrete with Micro Fibers (마이크로 섬유를 혼입한 고인성 섬유 보강 콘크리트)

  • In general, high strength and high performance fiber reinforced cement composites exclude coarse aggregates basically in order to have homogeneous distributions of material properties. However, these fiber-reinforced cement mortar without coarse aggregate have a tenancy that the modulus of elasticity is low and the unit weight of cement is high, resulting in low economic efficiency. Therefore, in this study, the development of high ductile fiber - reinforced concrete was conducted, which has the adequate level of coarse aggregate but still retains the high flexural toughness and strength and also has the crack - distributing performance. Experimental study was carried out by using the amount of coarse aggregate as an experimental parameter. The results showed that the best flexural toughness and crack dispersion characteristics was obtained when the coarse aggregate was added at 25% by weight of the fine aggregate to the typical mixtures of high ductile cement mortar. PVA fiber was effective in crack distribution and ductility enhancement, and steel fiber was effective in strengthening flexural strength rather than crack distribution.

Dosimetric comparison for Prostate VMAT of weight and photon energy change (전립선 암 입체적세기변조방사선치료 시 체형 및 에너지 변화에 따른 선량 평가)

  • Purpose : To compare the radiation doses of prostate cancer patients according to changes in abdominal body shape and energy during Volumetric modulated arc therapy(VMAT). Materials and Methods : Seven patients with prostate cancer were enrolled in this study. VMAT treatment plan was established at 6, 10, and 15 MV while changing from -2.0 cm to 2 cm by 0.5 cm. Conformal index(CI), homogeneous index(HI), $D_{max}$, $D_{95%}$, $D_{50%}$ and $D_{2%}$ of PTV were examined in order to evaluate the change of dose in the target organ according to body shape change. Normal organ of the femoral head, rectum and bladder was analyzed to evaluate dose changes. Results : The dose of $D_{max}$ 6 MV in PTV increased to 107.2 % in 1.0 cm body shape reduction, and 10 MV and 15 MV dose increased to 107.1 % and 107.0 % in 1.5 cm body reduction, respectively. The dose of $D_{50%}$ 6 MV in PTV decreased to 99.64 % in 1.0 cm body shape increase, and in 10 MV and 15 MV dose decreased to 99.79 % and 99.97 % in 1.5 cm body increase, respectively. In 2.0 cm body type increase, the dose was decreased to 99.30 % and 99.52 %, respectively. Doses for rectum and bladder gradually increased with decreasing weight, and dose decreased with decreasing weight. 6 MV, and $V_{70Gy}$ at 10 MV increased from 11.50 % to 12.76 % when the external shape decreased by 2.0 cm. The bladder $V_{70Gy}$ also increased from 14.0 % to 15.2 %. It was also shown that the dose increased as the body weight decreased in the femoral head. Conclusion : In the treatment of VMAT, dose distribution can be changed according to the change of abdominal shape. SSD and CBCT were used to decrease the body shape by more than 1cm or more than 1.0 cm at 6 MV and the body shape by more than 1.5 cm or more than 1.5 cm at 10 MV or 15 MV. It is considered that a new treatment plan should be established through re-simulation.

Purification and Characterization of Thermostable $\beta$-Mannanase from a Bacillus sp. YA-14

  • Thermostable $\beta$-mannanase from Bacillus sp. YA-14 was purified by acetone precipitation, CM-cellulose, Sephadex G-100 and hydroxyapatite column chromatography from culture supernatant. The final enzyme preparation appeared to be homogeneous on sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). $\beta$-Mannanase appeared to be a monomeric protein with a molecular weight of 67, 000 daltons. The optimal pH and temperature of the enzyme reaction were pH 6.0 and $75^{\circ}C$ , respectively. The enzyme was stable at a pH range of 6.0 to 9.0 and at temperatures between 45 and $85^{\circ}C$. The kinetic constants of $\beta$-mannanase as determined with a galactomannan (locust bean) as substrate were a Vmax of 25 unit/ml and a Km of 1.1 mg/ml. The enzyme had only limited activity on galactomannan substrate. It was suggested that mg $\beta$-mannanase activity is limited by the number of branched $\alpha$-galactose residues.

  • In China, primary purpose of policy concerning land tenure system was to maintain social homogeneity among people and growth of productivity. Even before reformed by Deng Shao Ping's government, left wing who had placed more importance on the social unity. When they gained political power, pressing against market oriented agricultural policy for the collective farming system. However right wing prefered to adopt the productivity oriented policy, which might lead to individualized farming system at the cost of social unity. Since Chinese government following principles of social economy put more weight on social unities rather than productivity growth, farming system and rural community became developed into homogeneous structure across the nation before Deng Shao Ping's reform. Process for People's Commune, followed by first and then second level coperations starting from group farming so called Hozozo, was historical reflection of developing such land tenure system. However, even under People's Commune, farmers' efforts could be found to increase their own productivity along with emerged private farming, in which Posandoho with week private farming system was gradually developed into Pogandoho. As Deng's government encouraged farmers to increase productivity through the market oriented measure, there had been wide spread of Pogandoho among the farmers even before legal desolution of Peoples' Commune was realized.

A Level Set Method to Image Segmentation Based on Local Direction Gradient

  • For image segmentation with intensity inhomogeneity, many region-based level set methods have been proposed. Some of them however can't get the relatively ideal segmentation results under the severe intensity inhomogeneity and weak edges, and without use of the image gradient information. To improve that, we propose a new level set method combined with local direction gradient in this paper. Firstly, based on two assumptions on intensity inhomogeneity to images, the relationships between segmentation objects and image gradients to local minimum and maximum around a pixel are presented, from which a new pixel classification method based on weight of Euclidian distance is introduced. Secondly, to implement the model, variational level set method combined with image spatial neighborhood information is used, which enhances the anti-noise capacity of the proposed gradient information based model. Thirdly, a new diffusion process with an edge indicator function is incorporated into the level set function to classify the pixels in homogeneous regions of the same segmentation object, and also to make the proposed method more insensitive to initial contours and stable numerical implementation. To verify our proposed method, different testing images including synthetic images, magnetic resonance imaging (MRI) and real-world images are introduced. The image segmentation results demonstrate that our method can deal with the relatively severe intensity inhomogeneity and obtain the comparatively ideal segmentation results efficiently.

Purification and Characterization of Aspartase from Hafnia alvei

  • Aspartase (EC 4.3.1.1) from Hafnia alvei was purified to homogeneity by a combination of DEAE-cellulose, Red A-agarose, and Sepharose 6B chromatography. The purified enzyme appeared homogeneous on denatured SDS-polyacrylamide gel electrophoresis. The purified enzyme was a tetrameric protein composed of identical subunits with a molecular weight of 55,000 daltons. The optimum pH for the enzymatic reaction was 8.5 and the optimum temperature for maximum activity was $45^{\circ}C$. The enzyme has an absolute requirement of divalent metal ions ($Mg^{2+}$, $Mn^{2+}$) at the alkaline pH. The enzyme, however, was inactivated in the presence of other divalent cations such as $Zn^{2+}$, $Ca^{2+}$. The helical content of the purified enzyme was estimated by CD spectropolarimetry to be 61%.

Optimal Design of Laminate Composites with Gradient Structure (경사형 구조 적층복합재료의 최적설계에 관한 연구)

  • In an effort to construct a structure under the design principle of minimal use of materials for maximum performances, a discrete gradient structure has been introduced in laminate composite systems. Using a sequential linear programming method, the gradient structure of composites to maximize the buckling load was optimized in terms of fiber volume fraction and thickness of each layer. The buckling load showed maximum value with the outmost [$0^{\circ}$] layer concentrated by almost all the fibers when the ratio of length to width(aspect ratio) was less than 1.0. But when the aspect ratio was 2.0, the optimum was determined in a structure where the thickness and fiber volume fraction were well-balanced in each layer. From the optimization of gradient structure, the optimal fiber volume fraction and thickness of each layer were proposed. Gradient structures have also shown an advantage in the weight reduction of composites compared with the conventional homogeneous structures.

  • We isolated a strain of Pseudomonas sp. from soil to utilize furfuryl alcohol as a carton source by enrichment culture. Alcohol dehydrogenase from this bacteria was purified 700-fold by Sephadex G-200 and affinity column chromatography to be homogeneous by electrophoresis and analytical centrifugation. This enzyme had a molecular weight of 120,000 and was composed of four subunits consisting of 266 amino acid residues. The optimal pH of the enzyme was pH 8.5 to 9, and the optimal temperature was, 45$^{\circ}C$. This enzyme was stable at 55$^{\circ}C$, but lost 80% of its activity in 10min at 6$0^{\circ}C$.

  • The cyclodextrin glucanotransferase (CGTase) was purified from the culture broth of Bacillus stearothermophilus by ammonium sulfate precipitation and affinity chromatography. The specific activity of the CGTase increased by about 31-fold from 111.5 U/mg protein to 3445.0 Ulmg protein. The SDSPAGE indicated that the purified CGTase was homogeneous and the molecular weight of the purified CGTase was about 78,000. The optimum pH and temperature was 6.0 and $60^{\circ}C$, respectively. This enzyme was stable from pH 5.5-10.0. The enzyme retained its full activity at the incubation temperature up to $60^{\circ}C$ and calcium ion increased the thermal stability. The isoelectric point was about 4.8.
